Abstract

Orgateins are isolated from mixtures of soluble proteins using >=1 of the following purification stages (1) an aq. soln of the protein mixture is treated with a protidolytic enzyme which selectively degrades one fraction of the proteins other than orgotein (2) at least the erythrocyte fraction of whole blood is heated at 60-80 degrees C and pH 5-8 to precipitate haemoglobin, the precipitate is separated and orgotein isolated from supernatant liquid (3). A mixture of orgotein and other proteins is dissolved in an aqs. buffer at pH 6 to give a soln of ionic strength 0-01 M and chromatographed on a weakly basic ion exchange resin (e.g. DEAE-cellulose TEAE-cellulose or cross-linked dextran having diethylaminoethyl gps). The resin is selectively eluted with an eluent having greater ionic strength (pref. increased progressively from 0.02 M to >0.03 M) at pH 6 with simultaneous determination of the absorption at 2650 and/or 2800 angstroms and the Cu and/or Zn content of the eluate and separation of the eluate which has a maximum non-ionised metal content and a minimum absorption at 265 or 280 nm. When the soluble protein mixture is obtained from erythrocytes the haemoglobin and carbonic-anhydrase are removed by washing the column prior to the elution of the orgotein.
