On obtient alors le 3-(bêta-pyridyl)-5- diéthylaminométhyl-1,2,4-oxadiazole avec un rendement pratiquement quantitatif (98%); ce produit se présente sous forme d'une huile de couleur brune assez claire.
En faisant passer bulle à bulle de l'acide @ chlorhydrique gazeux sec dans une solution alcoolique du produit ainsi préparé, il se forme un dichlorhydrate que l'on recueille à l'état cristallin et qui a une tem- pérature de fusion de 200 C.
Par réaction du 3-(bêta-pyridyl)-5-diéthyla- minométhyl-1,2,4-oxadiazole (lmole) avec 1 mole d'acide salicylique dans 3 litres d'acétone on obtient, après chauffage à reflux pendant 4 heures, refroidissement à -20 C, filtration et lavage un sel de l'acide salicylique et du composé oxadiazole ayant un point de fusion de 57 C (Kafler), Le produit obtenu est séché sous vide, à froid, en,présence de P2O5; il est conservé à l'abri de la lu- mière et en atmosphère sèche.
Son analyse pondérale a con- duit aux résultats suivants:

L'activité antitussive des composés selon l'invention a également été comparée à celle de la codéine et du Prilon dans le test suivant.
On a provoqué la toux selon une technique dérivée de celle de Winter et Flataker. Les cobayes sont places individuellement dans une enceinte en verre, cylindrique, munie d'un bouchon traverse par deux cana- lisations permettant l'arrivée et la sortie de vapeurs d'ammoniaque; sur la canalisation de sortie est branchée en dérivation une capsule de Marey dont la sensibilité permet d'enregistrer les variations de pression de l'en- ceinte lors des accès de toux de l'animal, cette sensibi- lité étant ajustée parle débit gazeux de sortie.
Les vapeurs irritantes sont obtenues par barbotage ù'un cuurant d'oxygène à travers une solution contenant 10 ml d'ammoniaque à 22 Bé pour 100mi d'eau.
L'exposition du l'animal aux vapeurs dure toujours 5 mi- nutes. Chaque animal est soumis il un essai témoin; une heure après il reçoit la substance à l'étude puis il est replacé dans l'enceinte 30 minutes après l'administration de la drogue.
Par comparaison à un groupe de cobayes n'a- yant reçu que du sérum sale isotonique, lots de 5 cobayes ont reçu le Prllen et les composes selon l'invention par voie intrapéritonéale. en solution aqueuse, à la dose de 40mg/kg; la codéine a été administrée par la même voie aux doses de 30 et @ mg/kg.

Les résultats figurent dans les tableaux II à IV ci-joints; la signification statistique de l'inhibition procentuelle a été calculée selon la méthode des couples.
Il apparaît à l'examen des tableaux II à IV que le composé de l'exemple 4 possède une action antitussive semblable à celle de la codéine pour dus doses "absolues" comparables.
Dans lc tableau IV on a envisagé l'action des produits sur le'nombre de toux d'expectoration; le composé de l'exemple 4 le diminue de façon sensible.
