L'invention est relative à la préparation de 1-(ss-oxyéthyl)-4-phényl- pipéridines substituées nouvelles et de leurs sels avec des acides inorganiques et organiques et des composés d'ammonium quaternaireso
Les nouveaux composés obtenus sont des médicaments de valeur qui se distinguent surtout par des propriétés analgésiques excellentes.

R5 et R6, qui peuvent être iden- tiques ou différents, étant de l'hydrogène, des restes alcoyle inférieurs ou, le cas échéant, des restes aryle substitués ou forment, avec l'atome d'azote voisin, le reste d'un noyau à 5 ou 6 éléments, le reste pouvant, le cas échéant, contenir encore d'autres atomes hétéro, tels que l'oxygène ou l'azote ou pouvant encore ê- tre substitué davantage.
Les nouveaux composés peuvent être préparés par les procédés usuels qui conviennent pour la fabrication de substances de ce genre. Les procédés de préparation suivants conviennent tout particulièrement à cet effet.
1. - Transformation d'une 4-phénylpipéridine correspondante, substi- tuée en 4 (il) à l'aide d'un oxyde d'halopropylène (III) et transformation ulté- rieure de l'oxyde de 4-phénylpipéridyl-N-propylène, substitué en 4, ainsi obtenu (IV) avec un composé ayant pour formule HZ selon le schéma suivant :

Dans le cas où R3 désigne un reste acyle, les composés (V) obtenus sont acylés par des méthodes connues.
La première phase du procédé, décrit plus haut, à savoir la prépara- tion de l'oxyde de 4-phényl-pipéridyl-N-propylène (IV), a lieu à une température inférieure à 100 , de préférence à 40-45 . On opère avantageusement dans un sol- vant, par exemple de l'eau. Afin que la réaction soit complète on ajoute encore, après un certain temps, un agent de condensation tel que l'hydroxyde de sodium ou un amine organique tertiaire, par exemple la pyridine ou de la diméthylanili- ne.
La transformation des oxydes de 4-phényl-pipéridyl-N-propylène (IV), à l'aide des produits susdits qui participent à la réaction, peut se faire en pré- sence ou en l'absence de solvants, de préférence dans la chaleur. Suivant la réactibilité et le point d'ébullition des participants à la réaction utilisés et suivant le point d'ébullition du solvant utilisé, la réaction a lieu à la tempé- rature d'ébullition des participants à la réaction ou du solvant sous une pres- sion normale ou à des températures supérieures dans l'autoclave. La température préférée correspond à 50-150 . Des solvants appropriés sont, par exemple, des alcools, tels que le méthanol ou l'éthanol, ainsi que le benzène, le toluol, etc.
2. - Transformation d'une 4-phénylpipéridine correspondante (II), substituée en 4, avec un oxyde de propylène substitué correspondant (VI). Cette

Dans le cas où l'on veut préparer, par cette transformation, des composés dans lesquels R3 est un reste acyle on doit acyler les composés obtenus (V) par des méthodes connues.
La transformation susindiquée peut se faire en présence ou l'absence de solvants, de préférence dans un milieu chauffé. Suivant la réactibilité et le point d'ébullition des participants utilisés pour la réaction ou le point d'é- bullition du solvant utilisé, on exécute la transformation avantageusement à la température d'ébullition des constituants pour la réaction ou du solvant à la pression normale. Des solvants appropriés pour cette réaction sont, par exemple, des alcools, tels que le méthanol ou l'éthanol, ainsi que le benzène ou le toluol.
3. - Condensation d'une 4-phénylpipéridine correspondante (II), sub- stituée en 4, avec un 1-halo-propanolène-(2) substitué correspondant (VII). La réaction a lieu selon le schéma suivant :

Pour cette transformation on peut ajouter les produits, participant à la réaction, à l'état de solution ou de suspension et on peut utiliser, comme milieu de réaction, des solvants organiques, par exemple des alcools, des hydro- carbures, tels que le benzène, le toluol ou le xylol, ainsi que des éthers.
La réaction a lieu avantageusement en chauffant jusqu'au point d'é- bullition du solvant correspondant. En outre, on peut avantageusement effectuer la réaction en présence d'un agent de condensation basique,par exemple du carbo- nate de sodium ou de potassium, de l'hydroxyde de sodium, de la pyridine ou la diméthylaniline.

Les composés,, préparés selon l'invention, peuvent, si on le désire, être transformés avec des acides organiques et inorganiques en leurs sels ou en leurs composés d'ammonium mono- ou bisquaternaires, La préparation de ces sels ou composés quaternaires est réalisée de façon en soi connue. Les nouveaux composés, obtenus selon l'invention, possèdent des propriétés pharmacodynamiques de valeur.
Ils se distinguent plus spécialement par des propriétés analgésiques et spasmoly- tiques excellentes et sont, d'une manière surprenante, supérieurs, en ce qui con- cerne leur action, à d'autres.composés connus qui ont une structure analogue,
Les exemples ci-dessous servent à expliquer l'invention avec plus de détails mais ils n'ont aucun caractère restrictif ni limitatif.

EXEMPLE I. - Préparation du 4-phênyl-1-N-(3-o-crésyloxy-2-hydroxy-pro- pyl)-pipéridyl- (4)-éthyl-cétone. a) On chauffe 200 g. d'épichlorhydrine avec 30 ml. d'eau à'35 . On ajoute ensuite, goutte à goutte et en agitant, 450 g. de 4-phényl-pipéridyl-(4)- éthylcétone de manière telle que la température de 45 ne soit pas dépassée. Le mélange de réaction est maintenu, pendant deux heures, à une température de 40- 45 et est traité, ensuite, avec 1 litre d'une lessive de soude à peu près 6n a- près quoi il est agité encore pendant 2 heures à la même température. Ensuite on traite le mélange de réaction avec de l'eau et on l'extrait à l'éther.
Après avoir séparé le solvant par distillation, il reste un produit de réaction hui- leux à partir duquel le composé désiré est obtenu par distillation fractionnée.
Son P.E.O,3 = 170-173 . Le rendement = 85-95% de la valeur théorique.

On mélange 23 g. de 4-phényl-pipéridyl-(4)-éthyloétone avec 16 go d'oxyde de 1- phénoxy-2,3-propylène et on chauffe pendant environ 5 heures à 120 . Le produit de réaction désiré est purifié par distillation fractionnée.
Le P.E.O 2 = 231-234 . Le P. F. = 110-112 (éthanol). Le rendement = 90 % de la valeur théorique. Le chlorhydrate a un P.F. = 194-196 .

The invention relates to the preparation of novel substituted 1- (ss-oxyethyl) -4-phenyl-piperidines and their salts with inorganic and organic acids and quaternary ammonium compounds.
The new compounds obtained are valuable drugs which are distinguished above all by their excellent analgesic properties.
Their general formula is:

wherein R1 is hydrogen or a hydroxyl group; R2 an alkyl residue, saturated or not, with 1 to 5 carbon atoms, an alkoxy residue with 1 to 5 carbon atoms, an aryl residue, more especially a phenyl residue or an amino group; R3 is hydrogen or an acyl residue; Z is a residue XR4 or / R5 N, with X corresponding to oxygen or sulfur; R4 is an aryl residue, optionally substituted with lower alkyl, hydroxyl, alkoxy, acyloxy or nitro groups, or an alicyclic residue;
