Werkwijzen en produkten voor het bleken van tanden.
Technisch vakgebied Deze uitvinding betreft nieuwe methoden en nieuwe formuleringen voor het "bleken" of "witten" van tanden. Beide termen hebben dezelfde betekenis in het kader van dit dokument. Doel is om verkleuringen aan de buitenkant en binnenin de tand te verwijderen, om aldus helderwitte tanden te bekomen.
Stand van de techniek Patienten, tandartsen en consumenten beogen de witheid van tanden te behouden of te herstellen.
Er is heel wat bekend over de oorzaak van verkleuringen. Ze kunnen zich aan de buitenkant van de tand voordoen, in het glazuur of in het dentine dat zich dichtbij het tandoppervlakte bevindt en worden"extrinsieke verkleuringen"genoemd ; ze kunnen zich ook dieper in de tand manifesteren, de "intrinsieke verkleuringen".
Extrinsieke verkleuringen worden veroorzaakt door : kolonisatie van diverse soorten bacterien, neerslag van glycoproteinen en chromoforen van bacterien (plaque), afbraakprodukten van bloed, hemoglobuline en zwavel bevattende proteinen ; of door opname van zware metalen (zoals cadmium, lood, nikkel en koper) of van geneesmiddelen met chlorhexidine, mangaan of zilver. Voedingsmiddelen zoals koffie, tee, wijn, sommige fruitsoorten zoals krieken, bessen, druiven. behoren tot de belangrijke oorzaken van extrinsieke verkleuringen. Vooral het voedingsingredient "tannine" draagt bij tot de verkleuring. Het vormt water onoplosbare complexen met proteinen en zetmelen en slaat neer op de tanden.
Tabaksrook is een andere bron van vervuiling.
Minstens een gedeelte van de extrinsieke vervuiling kan verwijdert worden bij middel van een fysische, schurende, behandeling van het tandoppervlakte ; chemische en enzymatische bleekmiddelen kunnen echter wel helpen.
Na verloop van tijd kunnen extrinsieke verkleuringen dieper penetreren in de tand en intrinsieke verkleuringen opleveren. Deze verkleuringen kunnen ook een interne oorzaak hebben en het gevolg zijn van afbraakprodukten van bloed, consumptie van bepaalde soorten antibiotica (tetracyclines) of van overmaat van fluorides, of opname van voedingsingredienten die de tanden bereiken via de bloedbaan. Ziekten zoals cikkel-cel anemie, thalassemie en ziekten van lever en nieren, kunnen ook verkleuringen veroorzaken.

Intrinsieke verkleuringen kunnen alleen maar verwijderd worden met bleekmiddelen die in staat zijn de tand in de diepte te penetreren.
In het verleden zijn een aantal bleekmethoden ontwikkeld die hoofdzakelijk gebaseerd zijn op het gebruik van een peroxide derivaat, zoals waterstofperoxide, urea-peroxide of een peroxide/natriumperboraat mengsel. In sommige gevallen wordt de reaktiviteit van het peroxide verhoogd door behandeling met hitte of licht.
De peroxide concentratie in de bleekformuleringen varieert tussen 1 % en 35 % (op eindgewicht formulering). En de bleekkracht van de formulering hangt in hoge mate af van de concentratie van het bleekprodukt. In geval een substantieel bleek resultaat vereist wordt in een behandeling, is een waterstofperoxide concentratie van minstens 30 % vereist. Bij gebruik van een 10 % urea-peroxide formulering zijn substantiele bleekresultaten pas te bekomen na dagenlange, soms wekenlange, herhaling van de behandeling. De resultaten met waterstofperoxide concentratie beneden de 10 % zijn nog meer bescheiden.
Peroxiden oxideren niet op selektieve wijze. Ze reageren niet alleen met stoffen die tanden verkleuren, maar ook met andere bio-moleculen die aanwezig zijn in de tand, die niet bijdragen tot de verkleuring, en die bij voorkeur onaangetast zouden moeten blijven. In aanwezigheid van ijzer reageren peroxiden zelfs bij voorkeur met suiker-moleculen.
Peroxiden irriteren tandvlees, mond en keel en reageren met essentiele aminozuren en restauratieve materialen. Het tandvlees moet nauwlettend afgeschermd worden op het moment dat de tanden behandeld worden, een vereiste waaraan moeilijk volledig kan voldaan worden. Het meest zorgwekkend is het effekt van de bleekbehandeling op de sterkte eigenschappen van de tanden. Het is bekend dat peroxiden de tanden brozer maakt.
Het doel van deze uitvinding, is de ontwikkeling van een bleekmethode en van formuleringen die de nadelen van het gebruik van peroxiden verminderen en vermijden, terwijl een hoge bleekkracht behouden blijft.
Onlangs werd een niewe techniek ontwikkeld (patent US5989526), waarbij tanden gebleekt worden met oxiderende enzymen, in hoofdzaak laccase enzymen in combinatie met een produkt (versneller) dat de reaktie versterkt.
Het gaat over een éen-staps-bleekproces met een bleek-formulering die minstens een oxiderend enzym

bevat al dan niet in combinatie met een kleine hoeveelheid waterstofperoxide (max. 1%). Men stelt dat het gaat om een veiliger proces omdat de concentratie van waterstofperoxide beperkt wordt.
Echter de bleekkracht (zoals blijkt uit de delta-L resultaten die vermeld staan in het patent) is bescheiden. Enzymen zijn niet in staat tanden te witten tot op het niveau gewenst door patient, tandarts en consument. De nadelen zijn eventueel vermindert in vergelijking met peroxiden, maar de bleekkracht evenzeer. Het patent is vaag over het al dan niet gebruik van hogere concentraties waterstofperoxide in het éénstaps-bleek-proces. Het verwijst naar een eventueel gebruik tot 10 % peroxide, maar waarschuwt tegelijkertijd voor afbraak van het enzym onder deze omstandigheden. Ook het versnellende co-produkt zal niet stabiel zijn in een dergelijke formulering.
Enzymen kunnen verkleuringen slechts in beperkte mate verwijderen en het patent bevat, in tegenstelling tot de claims, geen aanwijzingen dat intrinsieke verkleuringen verwijdert worden (tanden die behandeld worden met chloorhexidine-digluconate bieden geen model voor interne verkleuringen).
Een methode met sterkere bleekkracht is gewenst om te kunnen voldoen aan de verwachtingen van patient, tandarts en consument.

Uitvinding Het onderwerp van deze uitvinding is een nieuwe multistap-bleek-procedure die een maximale bleekkracht combineert met een minimum gebruik aan chemisch bleekmiddel, waardoor de nadelige neveneffekten van een klassieke peroxide behandeling vermindert of vermeden worden.
Wij hebben, verassenderwijze, ontdekt dat blekende enzymen, meer bepaald oxidoreductases, en meer specifiek laccases en verwante enzymen in aanwezigheid van een versnellend co-product, niet alleen een deel van de kleurstoffen verwijderen van de tand (en dus bleken) maar ook een effekt hebben op de kleurstoffen die na behandeling op de tand blijven. De strukturen van deze kleurstoffen worden dermate aangetast door het enzym, dat ze kwetsbaar worden voor chemische bleekmiddelen (peroxiden en verwante bestaande chemische bleekmiddelen) in de daarop volgende bleekstap. Een voorbehandeling met enzymen oefent aldus een synergetisch effekt uit op de daaropvolgende behandeling met een chemisch bleekmiddel.
Aldus en onafgezien het gebruik van een lagere concentratie bleekmiddel, minder stringente reactieomstandigheden en kortere behandelingstijden wordt, desondanks, een groter bleekeffekt te bekomen.
Dankzij het synergetisch effekt, levert deze multistap bleek procedure (met minstens twee, mogelijk meer, stappen) de gewenste combinatie van hoge bleekperformantie en hogere veiligheid en minder neveneffekten, met mindere risico voor het ontstaan van broze tanden. Dit contrasteert met de bekende bleekprocedures die alleen chemische bleekmiddelen gebruiken (en onveiliger zijn) en de methoden op basis van enzymen (die minder bleekkracht hebben).
Meerdere bleekstappen kunnen uitgevoerd worden met chemische bleekmiddelen ; minstens één stap moet voorafgegaan worden door een enzymatische behandeling.

De blekende enzymen zijn oxidoreductasen (bij voorkeur laccasen of verwante enzymen zoals catechol oxidase, biluribin oxidase, mono-phenol-monooxygenase. The concentratie van de enzymen ligt tussen 0. 0001 % en 25 % op basis van het eindgewicht van de formulering. De enzymatische reaktie wordt uitgevoerd in het pH gebied 4. 5 tot 9, en bij voorkeur tussen pH 5. 5 tot 7. 5.

Het co-produkt dat de enzymatische reaktie versneld kan elke bekend versnellend produkt zijn zoals vermeld in de conclusie 3 ofwel derivaten van adenine, indole, imidazole, benzothiazole, benzothiadiazole, benzotriazole, purine, benzoxazole, benzimidazole, pteridine, indazole, pyrazolopyrimidine, isoindole, benzisoxazole, carbazole.
Het chemisch bleekprodukt kan elk bekend chemisch bleekprodukt zijn of zoals vermeld in de conclusie 3.
De gebruikte concentratie ligt tussen 0. 0001 % en 25 % op basis eindgewicht van de formulering.
In een aanvullend aspekt van de uitvinding werd, verassenderwijze, gevonden dat met het toevoegen van een wasstap aan de multi-stap bleek-procedure, en volgend op de behandeling van tanden met blekende enzymen (meer specifiek oxidoreductasen, en bij voorkeur laccasen), extra verkleurend materiaal verwijderd kon worden, hetgeen de witheid van de tanden verder verbetert. De wasstap dient uitgevoerd te worden met een alkalische formulering die een hydroxide produkt (eg. natriumhydroxide), een carbonaat produkt (eg. natriumcarbonaat), of "lime" bevat. De pH is tussen 8 en 14, bij voorkeur tussen 9 en 12.
In aan ander aanvullend aspekt van de uitvinding werd, verassenderwijze, ontdekt dat de bleekkracht van de bleekstap waarbij gebruik wordt gemaakt van chemische bleekmiddelen zoals waterstopperoxide, urea-peroxide of organische peroxiden zoals cumeneperoxide, verbetert wordt door het voorafgaandelijk wassen van tanden met een metaalion-complexerend produkt zoals EDTA (ethylenediamine tetraacetic acid) of DTPA (diethylenetriaminepenta acetic acid). The concentratie van het complexerend produkt is bij voorkeur tussen 0. 001 % en 1 %, maar kan ook meer dan 1% bedragen.
In een ander aanvullend aspekt van de uitvinding werd verassenderwijze gevonden dat het toevoegen van magnesium-ionen, bij voorkeur magnesiumsulfaat, aan een formulering met een chemisch bleekmiddel op basis van peroxiden, het bleekresultaat verbetert. De concentratie van magnesiumsulfaat is tussen 0. 001 % en 1 % op eindgewicht van de formulering, bij voorkeur tussen 0. 05 % en 0. 2%.
In een ander aanvullend aspekt van de uitvinding werd verassenderwijze gevonden dat het toevoegen van koper-ionen, bij voorkeur kopersulfaat, en van een koper-coordinerend-produkt, aan een formulering met

een chemisch bleekmidddel op basis van peroxiden, de bleekkracht versterkt. Het koper zout en het koper coordinerend produkt worden gebruikt in de concentratie tot 20 % op eindgewicht van de formulering, bij voorkeur tussen 0. 001 % en 5 %.
Het koper coordinerend produkt kan elk bekend coordinerend produkt zijn zoals verbindingen met stikstof (eg pyridine), met een hydroxylgroep (eg catechol), met een ether group (eg 18 crown 6) of een verbinding met zwavel (eg mercaptosuccinic acid).
Het chemisch bleekmiddel kan elk bekend peroxide zijn, bij voorkeur waterstofperoxide of een organisch peroxide zoals cumeneperoxide. The concentratie van het chemisch bleekmiddel is tussen 0. 001 % en 25 %, bij voorkeur tussen 0. 1 % en 10 %. Het effekt van het gebruik van koper ionen en een koper coordinerend produkt op de oxidatiekracht van een peroxide is bekend voor zover het de oxidatie van lignine in pulp betreft (patent W099/0924 ; tittel"chemical method for lignin depolymerization"). Kleurstoffen in en op tanden hebben echter een andere chemische struktuur en de bereikbaarheid ervan is veel problematischer zodat een gelijkaardig effekt bij tanden vooraf niet kon voorspeld worden.
Het gebruik van deze stoffen in combinatie met chemische peroxide bleekstoffen kan zowel nuttig ingezet worden bij een multi-stap-bleek-procedure als bij een mono-stap-bleek procedure.
In een ander aanvullend aspekt van de uitvinding, werd verassenderwijze ontdekt dat de bleekkracht van de bleekstap met bleek-enzymen, zoals laccasen en verwante enzymen, en met versnellende co-producten, verbeterd kan worden door verzadiging van de bleekformulering met moleculaire zuurstof.
Het verzadigingsproces kan zowel voor de behandeling van tanden als tijdens de behandeling uitgevoerd worden. De verbetering is het resultaat van een grotere beschikbaarheid van zuurstof in de bleekformulering.
Het blijkt tevens mogelijk het bleekeffekt te versterken door toevoeging van een perfluorcarbon product (bijvoorbeeld perfluoro-1, 3-dimethyl cyclohexane) en een emulgator (bijvoorbeeld lecithine van ei) aan de formulering met bleekenzymen zoals laccase en verwante enzymen. Deze formulering wordt verzadigd met moleculaire zuurstof.
In een ander aanvullend aspekt van de uitvinding werd ontdekt dat het tannase enzym, een enzym dat tannine (een voedingsingredient dat verkleuring van tanden veroorzaakt) afbreekt, in staat is om tanden te bleken. Het tannase kan afkomstig zijn van Asp.
Oryzae of van elk ander organisme dat tannase

produceert. Het enzyme wordt gebruikt bij een temperatuur tussen 0 en 70 graden celcius, bij voorkeur bij 20 tot 45 graden celcius ; de pH is tussen 3 en 8, bij voorkeur tussen 5. 5 en 6. 5. de enzyme concentratie is tussen 0. 001 % en 25 % op basis eindgewicht van de formulering (bij verdunde oplossing kan de concentratie boven 25 % uitstijgen).
Het is gebruikelijk dat de enzym aktiviteit van de enzym oplossing zieh bevindt tussen 50 U/gr en 50000 U/gr.
Tannase kan alleen ofwel in combinatie met een oxiderend enzym zoals laccase of verwant enzym, ingezet worden in een bleek formulering.
Tannase enzymen kunnen gebruikt worden in een multi stap-en in een één-staps bleek procedure.
In een ander aanvullend aspekt van de uitvinding werd verassenderwijze ontdekt dat bleek-formuleringen met Mn-peroxidase in staat zijn om tanden te bleken.
Mn-peroxidase kan ook in combinatie met andere oxidoreductasen of met tannase ingezet worden en dit zowel in multi stap als in één staps bleek formuleringen.
In een ander aanvullend aspekt van de uitvinding werd verassenderwijze ontdekt dat het mogelijk is om "interne" verkleuringen te bleken met een formulering op basis van enzymen zoals oxidoreductasen, bij voorkeur laccasen of verwante enzymen, en/of Mnperoxidase al dan niet in aanwezigheid van een kleine hoeveelheid waterstofperoxide. Tot op heden zijn geen experimentele gegevens in de openbaarheid bekend die een dergelijk effekt aantonen (Patent US5989526 hanteert een dergelijke conclusie op basis van experimenten met tanden die gekleurd werden met chlorhexidine-digluconate, maar dit produkt vormt geen interne verkleuringen).
In een ander aanvullend aspekt van de uitvinding werd ontdekt dat een formulering op basis van oxidoreductase enzymen, bij voorkeur laccasen of verwante enzymen, of Mn-peroxidase of tannase in staat is om verkleuringen te bleken die veroorzaakt werden door tabaksrook. Tot op heden zijn geen experimenten bekend die zulks aantonen.
De bovenstaande aspekten van de uitvinding worden toegepast bij het bleken van tanden ; de produkten kunnen zowel gebruikt worden door tandartsen als door schoonheidsinstituten en door consumenten thuis.

Methode en Materialen Aan de Dental School, Vrije Universiteit Bussel, V. Geneeskunde, werden tanden verzameld.
De tanden werd in twee gelijke delen gesneden volgens hun langste axis en het versneden oppervlak van elke tandhelft werd aan een glazen plaatje gelijmd met cyanoacrylaat lijm.
Elke tandhelft werd behandeld met een verschillende bleekprocedure ; ofwel met een experimentele bleek procedure ofwel, ter vergelijking, met een conventionele bleekprocedure.
De behandele tanden werden gefotografeerd met een Canon reflex camera uitgerust met een speciale medische lens op statief. Diapositieven werden gemaakt van de experimentele tanden, maar ook van commercieel beschikbare standaard tanden (shade guide, Vita) teneinde effekten veroorzaakt door variaties in het filmmateriaal op te heffen.
De diapositieven werden geanalyseerd met computer software volgens Bentley et al (J. Amer. Dent. Assoc ;, 1999 ; 130 : 809-816). Het aantal gemeten blauwe pixels werd als maatstaf genomen voor de witheid van de tand.

Methods and products for teeth whitening.
Technical Field This invention relates to new methods and new formulations for "whitening" or "whitening" teeth. Both terms have the same meaning in the context of this document. The aim is to remove discolouration on the outside and inside of the tooth, so that clear white teeth are obtained.
State of the art Patients, dentists and consumers aim to maintain or restore the whiteness of teeth.
A lot is known about the cause of discolouration. They can occur on the outside of the tooth, in the enamel or in the dentin that is close to the tooth surface and are called "extrinsic discolorations"; they can also manifest themselves deeper in the tooth, the "intrinsic discolorations".
Extrinsic discolorations are caused by: colonization of various types of bacteria, precipitation of glycoproteins and chromophores of bacteria (plaque), degradation products of blood, hemoglobulin and sulfur-containing proteins; or by incorporating heavy metals (such as cadmium, lead, nickel and copper) or from drugs with chlorhexidine, manganese or silver. Foods such as coffee, tee, wine, some fruits such as cherries, berries, grapes. are among the leading causes of extrinsic discolouration. The food ingredient "tannin" in particular contributes to the discoloration. It forms water-insoluble complexes with proteins and starches and deposits on the teeth.
Tobacco smoke is another source of pollution.
At least a portion of the extrinsic contamination can be removed by physical, abrasive, treatment of the tooth surface; however, chemical and enzymatic bleaching agents can help.
Over time, extrinsic discolorations can penetrate deeper into the tooth and produce intrinsic discolorations. These discolorations can also have an internal cause and result from breakdown products from blood, consumption of certain types of antibiotics (tetracyclines) or from excess fluorides, or uptake of food ingredients that reach the teeth through the bloodstream. Diseases such as cell-cell anemia, thalassemia and liver and kidney diseases can also cause discolorations.

Intrinsic discolorations can only be removed with bleaching agents capable of penetrating the tooth into the depth.
In the past, a number of bleaching methods have been developed which are mainly based on the use of a peroxide derivative, such as hydrogen peroxide, urea peroxide or a peroxide / sodium perborate mixture. In some cases, the reactivity of the peroxide is increased by treatment with heat or light.
The peroxide concentration in the bleaching formulations varies between 1% and 35% (on final weight formulation). And the bleaching power of the formulation is highly dependent on the concentration of the bleaching product. In case a substantially pale result is required in a treatment, a hydrogen peroxide concentration of at least 30% is required. When a 10% urea-peroxide formulation is used, substantial bleaching results can only be obtained after days, sometimes weeks of, repetition of the treatment. The results with hydrogen peroxide concentration below 10% are even more modest.
Peroxides do not oxidize selectively. They react not only with substances that discolor teeth, but also with other bio-molecules that are present in the tooth, that do not contribute to the discoloration, and that should preferably remain unaffected. Peroxides even preferentially react with sugar molecules in the presence of iron.
Peroxides irritate gums, mouth and throat and react with essential amino acids and restorative materials. The gums must be carefully screened off when the teeth are treated, a requirement that is difficult to fully meet. The most worrying is the effect of the bleaching treatment on the strength properties of the teeth. Peroxides are known to make the teeth brittle.
The object of this invention is to develop a bleaching method and formulations that reduce and avoid the disadvantages of using peroxides while maintaining a high bleaching power.
Recently a new technique was developed (patent US 5989526), in which teeth are bleached with oxidizing enzymes, mainly laccase enzymes in combination with a product (accelerator) that enhances the reaction.
It is about a one-step bleaching process with a bleaching formulation that has at least one oxidizing enzyme

contains or not in combination with a small amount of hydrogen peroxide (max. 1%). It is said that it is a safer process because the concentration of hydrogen peroxide is limited.
However, the bleaching power (as shown by the delta-L results mentioned in the patent) is modest. Enzymes are unable to whiten teeth to the level desired by patients, dentists and consumers. The disadvantages are possibly reduced in comparison with peroxides, but the bleaching power is just as much. The patent is vague about whether or not to use higher concentrations of hydrogen peroxide in the one-step bleaching process. It refers to a possible use up to 10% peroxide, but at the same time warns against degradation of the enzyme under these conditions. The accelerating co-product will also not be stable in such a formulation.
Enzymes can only remove discolorations to a limited extent and, contrary to the claims, the patent contains no evidence that intrinsic discolorations are removed (teeth treated with chlorhexidine digluconate do not provide a model for internal discoloration).
A method with stronger bleaching power is desirable to be able to meet the expectations of the patient, dentist and consumer.

Invention The subject of this invention is a new multi-step bleaching procedure that combines maximum bleaching power with a minimum use of chemical bleach, thereby reducing or avoiding the adverse side effects of a traditional peroxide treatment.
We have surprisingly discovered that bleaching enzymes, more specifically oxidoreductases, and more specifically laccases and related enzymes in the presence of an accelerating co-product, remove not only part of the dyes from the tooth (and thus bleaching) but also an effect on the dyes that remain on the tooth after treatment. The structures of these dyes are so affected by the enzyme that they become vulnerable to chemical bleaching agents (peroxides and related existing chemical bleaching agents) in the subsequent bleaching step. Pre-treatment with enzymes thus exerts a synergistic effect on the subsequent treatment with a chemical bleaching agent.
Thus and irrespective of the use of a lower concentration of bleaching agent, less stringent reaction conditions and shorter treatment times, a greater bleaching effect is nevertheless obtained.
Thanks to the synergistic effect, this multi-step bleaching procedure (with at least two, possibly more, steps) delivers the desired combination of high bleaching performance and higher safety and fewer side effects, with less risk of brittle teeth. This contrasts with the known bleaching procedures that only use chemical bleaching agents (and are more unsafe) and the enzymatic methods (which have less bleaching power).
Multiple bleaching steps can be performed with chemical bleaching agents; at least one step must be preceded by an enzymatic treatment.

The bleaching enzymes are oxidoreductases (preferably laccases or related enzymes such as catechol oxidase, biluribin oxidase, mono-phenol-monooxygenase. The concentration of the enzymes is between 0.0001% and 25% based on the final weight of the formulation. The enzymatic reaction is carried out in the pH range 4.5 to 9, and preferably between pH 5.5 to 7.5.

The co-product that accelerates the enzymatic reaction can be any known accelerating product as stated in claim 3 or derivatives of adenine, indole, imidazole, benzothiazole, benzothiadiazole, benzotriazole, purine, benzoxazole, benzimidazole, pteridine, indazole, pyrazolopyrimidine, isoindole, benzisoxazole, carbazole.
The chemical bleaching product can be any known chemical bleaching product or as stated in claim 3.
The concentration used is between 0.0001% and 25% based on the final weight of the formulation.
In an additional aspect of the invention, it was surprisingly found that by adding a washing step to the multi-step bleaching procedure, and following treatment of teeth with bleaching enzymes (more specifically oxidoreductases, and preferably laccases), additional discolouring material could be removed, which further improves the whiteness of the teeth. The washing step must be carried out with an alkaline formulation containing a hydroxide product (eg sodium hydroxide), a carbonate product (eg sodium carbonate), or "lime". The pH is between 8 and 14, preferably between 9 and 12.
In another additional aspect of the invention, it was surprisingly discovered that the bleaching power of the bleaching step using chemical bleaching agents such as water stopperoxide, urea peroxide or organic peroxides such as cumene peroxide is improved by previously washing teeth with a metal ion complexing product such as EDTA (ethylenediamine tetraacetic acid) or DTPA (diethylenetriaminepenta acetic acid). The concentration of the complexing product is preferably between 0. 001% and 1%, but can also be more than 1%.
In another additional aspect of the invention, it was surprisingly found that the addition of magnesium ions, preferably magnesium sulfate, to a formulation with a chemical bleaching agent based on peroxides improves the bleaching result. The concentration of magnesium sulfate is between 0. 001% and 1% on the final weight of the formulation, preferably between 0.05% and 0. 2%.

a chemical bleaching agent based on peroxides, the bleaching power enhanced. The copper salt and the copper coordinating product are used in the concentration up to 20% on final weight of the formulation, preferably between 0. 001% and 5%.
The copper coordinating product can be any known coordinating product such as compounds with nitrogen (eg pyridine), with a hydroxyl group (eg catechol), with an ether group (eg 18 crown 6) or a compound with sulfur (eg mercaptosuccinic acid).
The chemical bleaching agent can be any known peroxide, preferably hydrogen peroxide or an organic peroxide such as cumene peroxide. The concentration of the chemical bleaching agent is between 0. 001% and 25%, preferably between 0. 1% and 10%. The effect of the use of copper ions and a copper coordinating product on the oxidation power of a peroxide is known insofar as it concerns the oxidation of lignin in pulp (patent WO99 / 0924; title "chemical method for lignin depolymerization"). However, dyes in and on teeth have a different chemical structure and their accessibility is much more problematic so that a similar effect on teeth could not be predicted in advance.
The use of these substances in combination with chemical peroxide bleaching agents can be used effectively in a multi-step bleaching procedure as well as in a mono-step bleaching procedure.
In another additional aspect of the invention, it was surprisingly discovered that the bleaching power of the bleaching step with bleaching enzymes, such as laccases and related enzymes, and with accelerating co-products, can be improved by saturating the bleaching formulation with molecular oxygen.
The saturation process can be carried out both for the treatment of teeth and during treatment. The improvement is the result of a greater availability of oxygen in the bleach formulation.
It also appears possible to enhance the bleaching effect by adding a perfluorocarbon product (e.g. perfluoro-1, 3-dimethyl cyclohexane) and an emulsifier (e.g. egg lecithin) to the formulation with bleaching enzymes such as laccase and related enzymes. This formulation is saturated with molecular oxygen.
In another additional aspect of the invention, it was discovered that the tannase enzyme, an enzyme that degrades tannin (a food ingredient that causes discoloration of teeth), is capable of bleaching teeth. The tannase can be from Asp.
Oryzae or from any other organism that has tannase

produces. The enzyme is used at a temperature between 0 and 70 degrees Celsius, preferably at 20 to 45 degrees Celsius; the pH is between 3 and 8, preferably between 5.5 and 6. The enzyme concentration is between 0. 001% and 25% on the basis of the final weight of the formulation (in the case of dilute solution, the concentration may exceed 25%).
It is common for the enzyme activity of the enzyme solution to be between 50 U / g and 50,000 U / g.
Tannase can only be used in a bleaching formulation either in combination with an oxidizing enzyme such as laccase or related enzyme.
Tannase enzymes can be used in a multi-step and in a one-step bleaching procedure.

To date, no experimental data in the public domain are available to demonstrate such an effect (Patent US 5989526 uses such a conclusion based on experiments with teeth stained with chlorhexidine digluconate, but this product does not form internal discolorations).
In another additional aspect of the invention, it was discovered that a formulation based on oxidoreductase enzymes, preferably laccases or related enzymes, or Mn peroxidase or tannase is capable of bleaching discolorations caused by tobacco smoke. To date, no experiments are known to prove this.
The above aspects of the invention are used in teeth whitening; the products can be used by dentists as well as by beauty institutes and consumers at home.

Method and Materials At the Dental School, Free University Bussel, V. Medicine, teeth were collected.
The teeth were cut into two equal parts along their longest axis and the cut surface of each tooth half was glued to a glass plate with cyanoacrylate glue.
Each tooth half was treated with a different bleaching procedure; either with an experimental bleaching procedure or, for comparison, with a conventional bleaching procedure.
The treated teeth were photographed with a Canon reflex camera equipped with a special medical lens on a tripod. Slide positives were made from the experimental teeth, but also from commercially available standard teeth (shade guide, Vita) in order to eliminate effects caused by variations in the film material.
The slides were analyzed with computer software according to Bentley et al (J. Amer. Dent. Assoc;, 1999; 130: 809-816). The number of measured blue pixels was taken as a measure of the whiteness of the tooth.
