"Lamelle destinée à des tests pour diagnostics et procédés pour sa fabrication"
La présente invention est relative à une lamelle ou plaque, principalement destinée à des tests pour diagnostics, comprenant, sur au moins une de ses deux grandes faces, au moins un puits, dont la surface est mouillable et sur laquelle sera déposé l'échantillon à analyser, qui est délimité par une couche de substance hydrophobe faisant corps avec la grande face susdite.
Les lamelles, de type susdit, généralement utilisées actuellement ont leur couche de substance hydrophobe constituée soit par une encre ou peinture classique compatible avec la surface à recouvrir, soit par une peinture à base de résine epo- xy ou de silicone. L'angle de contact E) que fait une goutte d'eau pure avec ces substances et qui définit la caractéristique hydrophobe de ces dernières est compris entre 40 et 50 pour les encres ou peintures classiques et 85 et 95 pour les peintures epoxy ou siliconées.
Ces lamelles connues présentent divers inconvénients.
A savoir, parmi ceux-ci : angle de contact e trop faible pour éviter, pour des lamelles les plus utilisées à puits multiples, la contamination d'un puits à l'autre ; angle de contact e trop important de la surface du puits qui empêche l'échantillon de s'étaler parfaitement sur cette dernière, et résistance insuf- fisante de la substance hydrophobe, d'une part, aux attaques des produits chimiques, tels qu'acétone, méthanol, et, d'autre part, aux températures d'autoclavage auxquelles lesdites lamelles sont soumises soit lors de leur utilisation, soit pour leur traite- ment en vue d'utilisations successives.

et de procurer une lamelle du type susdit dont le ou les puits qu'elle présente ont une surface totalement et parfaitement mouillable et dont la substance hydrophobe qui délimite ce ou ces puits a un angle de contact 0 nettement supérieur à celui des lamelles connues, cette surface hydrophobe, à grand angle de contact, présentant en outre l'avantage d'offrir une grande résistance à l'abrasion, aux agents chimiques agressifs ainsi qu'aux hautes températures d'autoclavage.
A cet effet, suivant l'invention, la substance hydro- phobe associée à la grande face susdite de la lamelle est consti- tuée par un mélange homogène de peinture ou d'encre, choisie pour sa bonne adhérence à la matière de cette grande face, de polytétrafluoroéthylène (PTFE) en poudre et de diluant appro- prié, les proportions des constituants de ce mélange étant choisies pour que l'angle de contact 0, que fait une goutte d'eau pure avec ladite substance et qui définit la caractéris- tique hydrophobe de cette dernière, soit supérieur à 1000 et de préférence de l'ordre de 1500.
Suivant une forme de réalisation particulièrement avan- tageuse de l'invention, le mélange précité, d'une part, com- prend, en poids, 1 kg d'encre thermodurcissable et de 1,75 à
2,25 kg de PTFE en poudre et, d'autre part, s'effectue en présence de 1,8 à 2,2 litres de diluant.
L'invention a également pour objet des procédés pour la préparation de la substance hydrophobe qui est appliquée sur la face précitée de la lamelle, pour l'application de cette substance sur ladite face et pour la fabrication de lamelles sur support verre.
D'autres détails et particularités de l'invention ressortiront de la description des dessins annexés au présent mémoire et qui, à titre d'exemples non limitatifs, représentent des formes de réalisation particulières de la lamelle suivant l'invention et illustrent les procédés précités suivant l'invention.

La figure 2 est une section partielle suivant la ligne II-II de la figure 1 et à plus grande échelle que celle de cette dernière, montrant notamment les angles de contact 9 que font une goutte d'eau pure, d'une part, avec la substance hydrophobe couvrant la face susdite de la lamelle et, d'autre part, avec la surface d'un puits de cette lamelle.
La figure 3 est une vue en élévation d'une lamelle suivant l'invention destinée plus particulièrement à des test au latex pour agglutination.
La figure 4 est une vue en coupe, suivant la ligne IV-IV de la figure 3.
La figure 5 est une vue analogue à la figure 4 et montre une variante de la lamelle représentée aux figures 3 et 4.
Dans les différentes figures, les mêmes notations de référence désignent des éléments identiques ou analogues.
Les lamelles 1, suivant l'invention et illustrées aux dessins, sont destinées soit à être commercialisées en vrac, soit à faire partie de nécessaires de tests pour diagnostics.
Elles comprennent, sur une de leurs deux grandes faces 2, des puits 3 régulièrement répartis, dont la surface est mouillable et sur laquelle est déposée l'échantillon 4 à analyser. Ces puits 3 sont isolés les uns des autres par une couche de substance hydrophobe 5 qui adhère parfaitement à la face 2 des lamelles de manière à faire corps avec cette dernière. La substance hydrophobe 5, déposée sur la face 2 susdite, est constituée par un mélange homogène de peinture ou d'encre, telle que peinture ou encre thermodurcissable, choisie pour sa bonne adhérence à la face 2 précitée, de polytétrafluoroéthylène (PTFE) en poudre et de diluant compatible avec cette peinture et le PTFE.
Les proportions des constituants dudit mélange sont choisies pour que le degré hydrophobe de la substance 5 soit défini par un angle de contact E) supérieur à 100 et de préférence de l'ordre de 150 .

modurcissable et de 1,75 à 2,25 kg de PTFE en poudre s'effectue en présence de 1,8 à 2, 2 litres de diluant. Pour obtenir l'an- gle de contact @ de 1500, on utilise avantageusement, comme encre, une encre de la série 13500 de la S. A. R. L. DUBUIT et, comme PTFE en poudre, le"PTFE-FLUOR PLUS"blanc vendu par
LAWTER INTERNATIONAL, INC., avec, pour diluant, un diluant Z à base de cyclohexanol.
La lamelle 1 montrée aux figures 1 et 2 est une lamel- le réutilisable qui est notamment destinée à la réalisation de tests d'immunofluorescence (IFA) à l'aide d'un microscope à lumière ultraviolette. Cette lamelle est réalisée en verre, matériau qui n'est pas mouillable par nature. Pour que les échantillons 4 susdits puissent se répandre sur la surface des puits 3, la lamelle est traitée, après application de la substan- ce hydrophobe 5 sur la plus grande partie de la face 2 laissée libre par les puits, pour que cette surface soit mouillable, de préférence avec un angle de contact E) compris entre 30 et 80.
La lamelle illustrée aux figures 3 et 4 est également réutilisable et est principalement destinée à la réalisation de tests au latex pour agglutination qui s'effectuent à l'oeil nu.
Elle est réalisée en verre et ses puits 3 sont délimités par un cordon 6 en forme d'ellipse réalisé à l'aide de la substance hydrophobe 5 précitée, la lamelle étant traitée, après applica- tion de la substance 5, pour que sa surface non recouverte par cette dernière soit rendue mouillable avec un angle de contact qui est avantageusement compris entre 3 et 8 .
La lamelle 1 représentée à la figure 5 est une lamelle à usage unique qui peut avantageusement être utilisée pour effec- tuer des tests au latex pour agglutination. Elle est constituée par un support en carton 7 sur lequel est laminé un film 8 natu- rellement mouillable qui constituera la surface des puits 3.
Comme film, on utilise avantageusement soit un film en polypropylène traité par effet Corona, soit un film mat en acétate de vinyle ou de cellulose. Ce film est choisi pour que l'angle de contact e soit compris entre 30 et 80.

Suivant l'invention, la substance hydrophobe 5 est préparée en incorporant peu à peu à l'encre ou peinture thermo- durcissable le PTFE en poudre et le diluant. Le mélange ainsi obtenu est ensuite homogénéisé par passage dans un broyeur à cylindres en acier, pendant une durée d'environ 30 minutes.
Cette substance 5 parfaitement homogénéisée, est avantageuse- ment, suivant l'invention, appliquée par sérigraphie soit sur la plaque de verre constituant la lamelle, soit sur le film 8 préci- té laminé sur le support carton 7.
Dans le cas des lamelles en verre telles qu'illustrées aux figures 1 et 3, elles sont, suivant l'invention, après que la substance hydrophobe 5 y a été déposée, placées dans un four à fermeture hermétique qui élève leur température entre 215 et
2250C et ce, pendant un laps de temps compris entre 10 et 15 minutes. Cette élévation de température et son maintien dans la fourchette susdite, pendant le laps de temps précité, confère à la substance hydrophobe 5, qui comprend de la peinture ou de l'encre thermodurcissable, la grande résistance décrite ci- dessus et confère à la surface des puits 3, la mouillabilité susdite.
Il doit être entendu que l'invention n'est nullement limitée aux formes de réalisation décrites et que bien des modifications peuvent être apportées à ces dernières sans sortir du cadre du présent brevet.

and to provide a lamella of the aforementioned type, the well or wells of which it presents have a totally and perfectly wettable surface and the hydrophobic substance which delimits this or these wells has a contact angle 0 much greater than that of known lamellas, this hydrophobic surface with a large contact angle, which also has the advantage of offering high resistance to abrasion, aggressive chemical agents and high autoclaving temperatures.
To this end, according to the invention, the hydrophobic substance associated with the aforementioned large face of the strip is formed by a homogeneous mixture of paint or ink, chosen for its good adhesion to the material of this large face. , powdered polytetrafluoroethylene (PTFE) and suitable diluent, the proportions of the constituents of this mixture being chosen so that the contact angle 0, which a drop of pure water makes with said substance and which defines the characteris- hydrophobic tick of the latter, ie greater than 1000 and preferably of the order of 1500.
According to a particularly advantageous embodiment of the invention, the above-mentioned mixture, on the one hand, comprises, by weight, 1 kg of thermosetting ink and from 1.75 to

The strips 1, according to the invention and illustrated in the drawings, are intended either to be sold in bulk, or to be part of test kits for diagnostics.
They comprise, on one of their two large faces 2, wells 3 regularly distributed, the surface of which is wettable and on which the sample 4 to be analyzed is deposited. These wells 3 are isolated from each other by a layer of hydrophobic substance 5 which adheres perfectly to the face 2 of the strips so as to form a body with the latter. The hydrophobic substance 5, deposited on the aforesaid side 2, consists of a homogeneous mixture of paint or ink, such as paint or thermosetting ink, chosen for its good adhesion to the aforementioned side 2, of powdered polytetrafluoroethylene (PTFE) and thinner compatible with this paint and PTFE.

The strip 1 shown in FIGS. 1 and 2 is a reusable strip which is in particular intended for carrying out immunofluorescence tests (IFA) using a microscope with ultraviolet light. This strip is made of glass, a material which is not wettable by nature. So that the aforementioned samples 4 can spread over the surface of the wells 3, the coverslip is treated, after application of the hydrophobic substance 5 on most of the face 2 left free by the wells, so that this surface is wettable, preferably with a contact angle E) of between 30 and 80.
The coverslip illustrated in FIGS. 3 and 4 is also reusable and is mainly intended for carrying out latex tests for agglutination which are carried out with the naked eye.
It is made of glass and its wells 3 are delimited by a bead 6 in the form of an ellipse produced using the aforementioned hydrophobic substance 5, the strip being treated, after application of the substance 5, so that its surface not covered by the latter is made wettable with a contact angle which is advantageously between 3 and 8.
The slide 1 shown in FIG. 5 is a disposable slide which can advantageously be used to carry out latex tests for agglutination. It consists of a cardboard support 7 on which is laminated a naturally wettable film 8 which will constitute the surface of the wells 3.
As film, advantageously is used either a polypropylene film treated by Corona effect, or a matte film of vinyl acetate or cellulose. This film is chosen so that the contact angle e is between 30 and 80.

According to the invention, the hydrophobic substance 5 is prepared by gradually incorporating the powdered PTFE and the diluent into the ink or thermosetting paint. The mixture thus obtained is then homogenized by passage through a steel roller mill, for a period of approximately 30 minutes.
This perfectly homogenized substance 5 is advantageously, according to the invention, applied by screen printing either to the glass plate constituting the strip, or to the above-mentioned film 8 laminated on the cardboard support 7.
In the case of glass strips as illustrated in FIGS. 1 and 3, they are, according to the invention, after the hydrophobic substance 5 has been deposited there, placed in an oven with hermetic closure which raises their temperature between 215 and
2250C and this, for a period of time between 10 and 15 minutes. This rise in temperature and its maintenance in the above-mentioned range, during the abovementioned period of time, gives the hydrophobic substance 5, which comprises paint or thermosetting ink, the great resistance described above and gives the surface wells 3, the above wettability.
It should be understood that the invention is in no way limited to the embodiments described and that many modifications can be made to these without departing from the scope of this patent.
