WERKWIJZE VOOR DE WINNING VAN CEFALEXINE
De uitvinding betreft een werkwijze voor de winning van cefalexine uit een mengsel dat cefalexine en 7-aminodesacetoxycefalosporinezuur (7-ADCA) bevat.
Bij de bereiding van cefalexine waarbij 7-ADCA wordt geacyleerd met een D-fenylglycinederivaat, is de winning van het cefalexine en de opwerking van het reaktiemengsel in het algemeen moeilijk. Zo staat bijvoorbeeld beschreven in WO-A-93/12250 dat de acyleringsreaktie nooit volledig verloopt en de uiteindelijke zuivering van het eindprodukt wordt bemoeilijkt doordat de zuur/base eigenschappen en oplosbaarheden van sommige onzuiverheden (in het bijzonder 7-ADCA en fenylglycine zoals beschreven in US-A-4003896) weinig verschillen van die van het eindprodukt. Bijgevolg treedt coprecipitatie op waardoor onzuiver cefalexine wordt verkregen. In WO-A-93/12250 en US-A-4003896 wordt het gebruik van een complexeringsmiddel zoals naftol voorgesteld. Een nadeel hiervan is echter dat een extra procesvreemde stof dient te worden toegevoegd.
Een andere methode om cefalexine zuiver te isoleren uit een mengsel dat cefalexine en geringe hoeveelheden 7-ADCA bevat, wordt beschreven in JP-A- 52111584. Volgens de in deze Japanse publikatie beschreven methode wordt het mengsel dat 7-ADCA en cefalexine bevat onderworpen aan een herkristallisatie in een organisch oplosmiddel, bijvoorbeeld een mengsel van dichloormethaan, dimethylsulfoxide en een lagere alcohol, in aanwezigheid van een basische component, bijvoorbeeld diisopropylamine.
Deze methode heeft eveneens het nadeel dat procesvreemde stoffen moeten worden toegevoegd. Bovendien treedt bij deze zuivering 18% verlies aan cefalexine op.

Het doel van de uitvinding is een eenvoudige werkwijze te verschaffen die het mogelijk maakt cefalexine zuiver te winnen zonder gebruik te maken van dergelijke procesvreemde organische verbindingen.
Dit wordt volgens de uitvinding bereikt doordat een mengsel dat cefalexine en 7-ADCA bevat met een pH groter dan 7, dat, afgezien van eventueel aanwezige vaste cefalexine, bij een pH tussen 7 en 8, 5 homogeen is, wordt onderworpen aan een pH-verlaging tot een pH tussen 0 en 3, 5 is bereikt gevolgd door een pH-verhoging tot een pH tussen 5 en 7, 8 is bereikt en de aanwezige vaste stof wordt gewonnen.
Aanvraagster heeft gevonden dat het mogelijk is door de pH van het mengsel te verlagen tot een waarde tussen 0 en 3, 5, bij voorkeur tussen 0, 5 en 2, 5 en vervolgens de pH van het mengsel te verhogen tot een waarde tussen 5 en 7, 8, in het bijzonder tussen 6, 0 en 7, 6, afhankelijk van de samenstelling van het reaktiemengsel, cefalexine met een zuiverheid groter dan 90 massa-%, in het bijzonder groter dan 98 massa-% te laten uitkristalliseren zelfs wanneer een grote hoeveelheid 7-ADCA in het mengsel aanwezig is, waarna het kan worden gewonnen. In het reaktiemengsel zijn naast cefalexine vaak nog waardevolle componenten aanwezig zoals bijvoorbeeld de 7-ADCA. Om te komen tot een commercieel aantrekkelijk proces is het dan ook van belang tevens de verliezen aan 7-ADCA en cefalexine te minimaliseren.
Gebleken is tevens dat wanneer vervolgens de pH van bovengenoemd reaktiemengsel wordt verlaagd tot een waarde lager dan 6, 5, in het bijzonder tussen 1, 5 en 5, 5, 7-ADCA en de resterende cefalexine nagenoeg volledig uitkristalliseren, waarna dit mengsel van 7-ADCA en cefalexine kan worden gewonnen, bijvoorbeeld door filtratie. Het verkregen mengsel van 7-ADCA en cefalexine kan eventueel worden hergebruikt zodat een werkwijze wordt verkregen waarin zuiver cefalexine wordt verkregen zonder

significante verliezen aan 7-ADCA en cefalexine.
De werkwijze volgens de uitvinding is in het bijzonder geschikt om te worden toegepast in de opwerking van het reaktiemengsel dat wordt verkregen na de enzymatische acyleringsreaktie waarin 7-ADCA wordt geacyleerd met D-fenylglycineamide (PGA) of esters van Dfenylglycine. Zo kan bijvoorbeeld de werkwijze volgens de uitvinding worden toegepast op een uitgangsmengsel dat wordt verkregen door het reaktiemengsel van een bij relatief hoge pH, bijvoorbeeld een pH tussen 8 en 10, uitgevoerde enzymatische acyleringsreaktie, achtereenvolgens te filtreren, waarbij (geimmobiliseerd) enzym wordt afgescheiden, de pH te verlagen tot een waarde tussen 7, 0 en 8, 5, afhankelijk van het mengsel zodanig dat PG is uitgekristalliseerd en cefalexine nog niet, de gevormde vaste stof, die meestal in hoofdzaak zal bestaan uit D-fenylglycine (PG) dat tijdens de enzymatische reaktie is gevormd,
af te scheiden, en de pH eventueel verder te verlagen tot een waarde tussen 7 en 8, 5.
Een andere uitvoeringsvorm is die waarbij als uitgangsmengsel het mengsel wordt toegepast dat wordt verkregen na een enzymatische acyleringsreaktie die eindigt bij een relatief lage pH, bijvoorbeeld een pH tussen 7 en 8, 5, en na afscheiding van de vaste stof welke in hoofdzaak het gelmmobiliseerd enzym en D-fenylglycine bevat.
In het uitgangsmengsel dat 7-ADCA en cefalexine bevat kan zich een significante hoeveelheid 7-ADCA bevinden. Meestal is de hoeveelheid 7-ADCA lager dan 75 mol% berekend ten opzichte van de totale hoeveelheid 7ADCA plus cefalexine, bij voorkeur 2-60%, in het bijzonder 5-50%.
Een pH-verlaging kan in het kader van de uitvinding bijvoorbeeld worden bewerkstelligd, door toevoegen van een zuur aan het mengsel of gelijktijdig doseren van het mengsel en het zuur. Geschikte zuren zijn

bijvoorbeeld minerale zuren, in het bijzonder zwavelzuur, zoutzuur of salpeterzuur. Een pH-verhoging kan bijvoorbeeld worden bewerkstelligd door toevoegen van een base aan het mengsel of door gelijktijdig doseren van het mengsel en de base. Geschikte basen zijn bijvoorbeeld anorganische basen, in het bijzonder ammonia, kaliloog of natronloog. De optimale pH waarbij cefalexine wordt gewonnen hangt af van de samenstelling van het mengsel en wordt zo gekozen dat een optimale scheiding tussen 7-ADCA en cefalexine wordt verkregen.
De optimale pH is in de praktijk een compromis tussen enerzijds hoge zuiverheid van het gewonnen cefalexine die wordt bereikt wanneer de pH waarbij cefalexine wordt gewonnen relatief hoog is zodat de cefalexine nog gedeeltelijk en de 7-ADCA nog volledig in oplossing is ; en anderzijds hoge opbrengst die wordt bereikt wanneer de pH waarbij cefalexine wordt gewonnen relatief laag is zodat de cefalexine nagenoeg volledig is neergeslagen maar waarbij tegelijkertijd ook een deel van de 7-ADCA is neergeslagen. De deskundige is eenvoudig in staat de voor zijn situatie optimale pH vast te stellen.
De temperatuur waarbij de opwerking wordt uitgevoerd ligt meestal lager dan 35 C, bij voorkeur tussen 0 en 30 C, in het bijzonder tussen 10 en 30oC.
Door toepassing van de werkwijze volgens de uitvinding voor de winning van zuiver cefalexine in combinatie met recirculatie van het na verdere pHverlaging verkregen mengsel van 7-ADCA en cefalexine, op het mengsel dat verkregen wordt na enzymatische acylering van 7-ADCA met PGA kan in totaal een hoge selectiviteit op 7-ADCA, in het bijzonder groter dan 80%, worden verkregen.
Het resterende filtraat, dat hoofdzakelijk nog geringe hoeveelheden PGA bevat, kan desgewenst verder worden opgewerkt, bijvoorbeeld door het op een pH groter dan 8, in het bijzonder tussen 8, 5 en 10, te brengen. Desgewenst kan nog worden geconcentreerd en gekoeld tot een

temperatuur lager dan 10 C, bijvoorbeeld tussen 0 en 8OC.
Op deze wijze wordt een werkwijze verkregen waarmee zowel 7-ADCA als PGA met hoog rendement kunnen worden toegepast.
In de enzymatische acyleringsreaktie kunnen als acyleringsmiddel bijvoorbeeld PGA of esters van PG worden toegepast.
Als enzym kan in principe elk enzym worden toegepast dat geschikt is als katalysator in de koppelingsreaktie. Dergelijke enzymen zijn bijvoorbeeld de enzymen die bekend zijn onder de algemene aanduiding penicilline amidase of penicilline acylase. Voorbeelden van geschikte enzymen zijn enzymen afgeleid van Acetobacter, Aeromonas, Alcaligenes, Aphanocladium, Bacillus so., Cephalosporium, Escherichia, Flavobacterium, Kluvvera, Mvcoplana, Protaminobacter, Pseudomonas en Xanthomonas, in het bijzonder Acetobacter pasteurianum, Alcaliaenes faecalis, Bacillus meaaterium, Escherichia coli en Xanthomonas citrii.

De temperatuur waarbij de enzymatische acyleringsreaktie wordt uitgevoerd ligt meestal lager dan 35 C, bij voorkeur tussen 0 en 28 C. De pH waarbij de enzymatische acyleringsreaktie wordt uitgevoerd ligt meestal tussen 6 en 10, bij voorkeur tussen 6, 5 en 9.
De enzymatische acyleringsreaktie, en de opwerking van het reaktiemengsel wordt in de praktijk meestal uitgevoerd in water. Desgewenst kan het

reaktiemengsel ook een organisch oplosmiddel of een mengsel van organische oplosmiddelen bevatten, bij voorkeur minder dan 30 vol. %. Voorbeelden van organische oplosmiddelen die kunnen worden toegepast zijn alcoholen met 1-7 C-atomen, bijvoorbeeld een monoalcohol, in het bijzonder methanol of ethanol ; een diol, in het bijzonder ethyleenglycol of een triol, in het bijzonder glycerol.
In het kader van deze uitvinding kunnen de verschillende componenten in de vrije vorm of als zouten in het reaktiemengsel aanwezig zijn. Met de genoemde pHwaarde wordt steeds de pH-waarde gemeten bij kamertemperatuur bedoeld.
De uitvinding zal verder worden toegelicht aan de hand van de voorbeelden, zonder evenwel daartoe te worden beperkt.

Een mengsel van 38, 3 g PGA, 54, 6 g 7-ADCA en 375 ml water werd gekoeld tot 5OC. De resulterende suspensie werd toegevoegd aan 70 g gelmmobiliseerd Pen-G-acylase suspensie van Pharma Biotechnology Hannover. Dit enzym werd voor gebruik gewassen met 3 x 100 ml water. Het gecombineerde filtraat werd onder roeren bij 250C aan 100 ml water toegedruppeld, waarbij de pH op 1, 6 werd gehouden d. m. v. gelijktijdig doseren van geconcentreerd H2SO4 m. b. v. een autotitrator.
Vervolgens werd de pH op 7, 0 gebracht door toevoegen van geconcentreerd ammonia onder roeren bij 25 C.
Na 30 min. werd gefiltreerd over een G-3 glasfilter en gewassen met 3 x 25 ml water en 3 x 25 ml aceton. Het residu werd gedroogd, waardoor 48, 5 9 CEX. H2O verkregen werd met een chemische zuiverheid (op watervrije basis) van > 99, 5%. Het filtraat en het waswater werden gecombineerd en onder roeren werd bij 250C aangezuurd m. b. v. geconcentreerd H2S04 tot pH = 4, 4. Na 1 uur werd gefiltreerd en gewassen met achtereenvolgens 1 x 25 ml water en 3 x 25 ml aceton. Het residu werd gedroogd, waardoor 23, 5 g vaste stof werd verkregen. Deze vaste stof bevatte een mengsel van CEX en 7-ADCA in een molaire verhouding 11 : 89.

In the preparation of cephalexin in which 7-ADCA is acylated with a D-phenylglycine derivative, recovery of the cephalexin and work-up of the reaction mixture is generally difficult.
