Absorbent voor cultuurmedium en een daarvan voorziene houder
De onderhavige uitvinding heeft betrekking op een absorbent voor cultuurmedium en op een daarvan voorziene houder. Meer in het bijzonder heeft de uitvinding betrekking op een absorbent dat amorfe kooldeeltje bevat, welke kooldeeltjes echter niet gedispergeerd kunnen raken in het cultuurmedium.
Bij het kweken in vitro van cellen, bijvoorbeeld bij de in vitro fertilisatie van eicellen met zaadcellen, is gebleken, dat in het cultuurmedium stoffen terecht kunnen komen die een negatieve invloed hebben op het kweekresultaat. Bijvoorbeeld kunnen deze toxische stoffen in het kweekmedium geraken doordat bijvoorbeeld rubber van operatiehandschoenen in het cultuurmedium deze stoffen afscheiden.
De onderhavige uitvinding beoogt een absorbent te verschaffen, waarmee deze toxische stoffen kunnen worden afgescheiden uit het cultuurmedium en daardoor de kweekresultaten worden verbeterd.
Dit absorbent volgens de uitvinding bevat als werkzame stof amorfe kooldeeltjes die op zich geen negatief effect uitoefenen op de kweekresultaten, maar bij gebruik op zieh als gevolg van de vorming van een dispersie van de kooldeeltjes om praktische redenen niet bruikbaar zijn. Derhalve omvat het absorbent volgens de uitvinding tevens een kunststof die de dispersie van de amorfe kooldeeltjes in het cultuurmedium tegengaat.
Daarbij moet de kunststof permeabel zijn voor de uit het cultuurmedium te verwijderen toxische stoffen.
Het absorbent voor cultuurmedium volgens de uitvinding omvat derhalve een absorbent voor kweekmedium, omvattende amorfe kooldeeltjes die zijn omgeven door een kunststof die impermeabel is voor de kooldeeltjes en

permeabel voor in het kweekmedium aanwezige toxische stoffen. De amorfe kooldeeltjes kunnen afzonderlijk, als groep of als persstuk, zoals een pellet, bekleed worden met de semi-permeabele kunststof omhulling. Volgens een andere uitvoeringsvorm kunnen de kooldeeltjes of de persstukken, zijn opgenomen in een omhulling van de semipermeabele kunststof. Deze omhulling kan de vorm hebben van een zakje, of van een scheidingswand die de amorfe kooldeeltjes afscheidt van in een houder op te nemen cultuurmedium.
De te gebruiken kunststof dient permeabel te zijn voor de in het cultuurmedium aanwezige toxische stoffen.
Deze toxische stoffen kunnen door de kunststof heen diffunderen naar het oppervlak van de amorfe kooldeeltjes, dan wel naar de ruimte waarin de amorfe kooldeeltjes aanwezig zijn. Het zwellen van de kunststof onder invloed van cultuurmedium bevordert de diffusie.
Verder is het mogelijk, dat de kunststof poreus is en poriën bevat die kleiner zijn dan de kleinste afmeting van de koolstofdeeltjes. In het algemeen is de poriegrootte kleiner dan 40 pm, meer bij voorkeur kleiner dan 20 pm, en met de meeste voorkeur kleiner dan 10 pm.
Kunststoffen die kunnen worden toegepast bij het absorbent volgens de onderhavige uitvinding, omvatten kunststoffen die zijn gekozen uit de navolgende groepen : cellulose esters, fluorhoudende polymeren, acrylpolymeren, rubber, vinylpolymeren, polyalkyleen polymeren, polyamiden en polystyreen.
Volgens een voorkeursuitvoeringsvorm, is een met cultuurmedium in contact komende houder vooraf voorzien van het absorbent volgens de uitvinding. Dit absorbent kan bijvoorbeeld zijn gehecht aan een houderwand. Houders die kunnen zijn voorzien van het absorbent omvatten proefbuizen, perischalen en kweekflessen.
Tevens heeft de uitvinding betrekking op een houder die is voorzien van een absorbent volgens de uitvinding.
Genoemde en andere kenmerken van het absorbent voor cultuurmedium volgens de uitvinding, zullen hierna verder

verduidelijkt worden aan de hand van een aantal nietlimitatieve uitvoeringsvoorbeelden, waarbij wordt verwezen naar de bijgevoegde tekening.
In de tekening is figuur 1-4 elk een perspectievisch aanzicht van een houder voor cultuurmedium, waarin een absorbent volgens de uitvinding is opgenomen.
De amorfe kooldeeltjes die worden gebruikt bij de onderhavige uitvinding, hebben de vorm van poeder, korrels en eventueel persstukken, zoals een pellet. De amorfe kool is afkomstig uit koolstofhoudend materiaal, zoals hout, veen, ligniet, botkool (verkregen door het calcineren van bot), notendoppen, kolen en dergelijke. De kool wordt geactiveerd, waardoor een zeer poreuse structuur ontstaat. De activering kan bijvoorbeeld worden uitgevoerd door oxidatie met stoom, kooldioxide, brandbaar gas, lucht en dergelijke. Een andere mogelijkheid is de behandeling met chemicaliën, zoals metaalchloriden, metaalsulfiden, metaalsulfaten.
De geactiveerde, amorfe kool heeft een poriestructuur met poriën in het bereik van 1-100 nm, meer bij voorkeur poriën in het bereik van 20-100 nm. Het specifiek oppervlak bedraagt in het algemeen 200-2000 m2 per gram, meer bij voorkeur 500-1500 m2 per gram. De

deeltjesgrootte van de kooldeeltjes bedraagt in het algemeen 50-1000 um, meer bij voorkeur 100-500 um.
De amorfe kool is in de handel verkrijgbaar bijvoorbeeld onder de handelsmerken Carbomix, Carboraffin, Medicoal, Norit, Ultracarbon.
De semi-permeabele kunststof kan worden gekozen uit een aantal toepasbare kunststofgroepen. Voorbeelden hiervan zijn cellulose esters ; fluorhoudende polymeren, zoals polytetrafluoretheen, polychloortrifluoretheen, polyvinylideenfluoride ; acrylpolymeren, zoals polymethylmetacrylaal ; rubber, zoals isopreen rubber ; vinylpolymeren, zoals polyvinylacetaat en polyvinylchloride ; polyaklyleenpolymeren, zoals polyetheentherefthalaat ; polyamiden, zoals nylon-6 ; en

polystyreen. Essentieel voor de te gebruiken kunststof is, dat de in het cultuurmedium aanwezige toxische stoffen door de kunststof heen kunnen diffunderen dan wel passeren door in de kunststoflaag aanwezige poriën.
Het cultuurmedium kan een zeer diverse samenstelling bezitten. Veelal bevat het cultuurmedium serum, zoals humaanserum, runderserum, foetaalkalverserum of navelstrengserum. Voorbeelden van een cultuurmedium zijn Earles-medium, dat is gebufferd met HEPS en voorzien van 2% door warmte geinactiveerd humaan navelstrengserum.
Een ander cultuurmedium is HTF (Human Tubal Fluid) waaraan is toegevoegd 15% met warmte geinactiveerd humaan navelstrengserum en 0. 3 mM EDTA.
Figuur 1 toont een proefbuis 1 die is gevuld met cultuurmedium 2 voor het in vitro kweken van cellen. Voor het afscheiden van toxische stoffen, is een absorbent 3 volgens de uitvinding in de proefbuis opgenomen. Het absorbent omvat pellets 4 van amorfe kooldeeltjes, die worden omgeven door een omhulling 5 van een cellulose ester. Aan het einde van het kweken, kan het adsorbent 3 op eenvoudige wijze worden afgescheiden van het cultuurmedium.
Figuur 2 toont een proefbuis 6 volgens de uitvinding die is gevuld met cultuurmedium 2. De proefbuis is aan zijn binnenoppervlak voorzien van absorbent 7 volgens de uitvinding, in de vorm van amorfe kooldeeljes die aan het inwendig oppervlak van de proefbuis zijn gehecht en van het cultuurmedium zijn afgescheiden door een omhulling 8 in de vorm van een laag kunststof die impermeabel is voor de kooldeeltjes en permeabel voor in het cultuurmedium 2 aanwezige toxische stoffen.
Figuur 3 toont een petrischaal 9, waarvan de zijwand 10 is bekleed met een laag kooldeeltjes 11 die is afgedekt met een laag kunststof 12 volgens de uitvinding.

zijn bekleed met kooldeeltjes 16, die afzonderlijk zijn omhuld door een laag kunststof 17 volgens de uitvinding.
Bij de in vitro fertilisatie van eicellen met zaadcellen in een houder voor cultuurmedium die is voorzien van een absorbent volgens de uitvinding, zal de overleving van de zaadcellen, het percentage bevruchtingen en de kwaliteit van de kweek in vitro, en mogelijk ook het aantal latere zwangerschappen sterk zijn verbeterd, doordat overeenkomstig de uitvinding door het absorbent tijdens de in vitro kweek in het cultuurmedium aanwezige of daarin geraakte toxische stoffen zijn geabsorbeerd door het absorbent, zonder dat daarbij de amorfe kooldeeltjes tot in het cultuurmedium konden dispergeren.

The present invention relates to an absorbent for culture medium and to a container provided with it. More particularly, the invention relates to an absorbent containing an amorphous carbon particle, which, however, cannot be dispersed in the culture medium.
In vitro cultivation of cells, for example in the in vitro fertilization of egg cells with sperm cells, it has been found that substances which have a negative influence on the culture result can enter the culture medium. For example, these toxic substances can get into the culture medium because, for example, rubber from surgical gloves in the culture medium secrete these substances.
The present invention aims to provide an absorbent with which these toxic substances can be separated from the culture medium and thereby improve the culture results.
This absorbent according to the invention contains as active substance amorphous carbon particles which in themselves do not have a negative effect on the cultivation results, but are not usable for practical reasons when used because of the formation of a dispersion of the carbon particles. The absorbent according to the invention therefore also comprises a plastic which prevents the dispersion of the amorphous carbon particles in the culture medium.
In addition, the plastic must be permeable to the toxic substances to be removed from the culture medium.
The culture medium absorbent according to the invention therefore comprises a culture medium absorbent, comprising amorphous carbon particles surrounded by a plastic impermeable to the carbon particles and

permeable to toxic substances present in the culture medium. The amorphous carbon particles can be coated individually, as a group or as a pellet, such as a pellet, with the semi-permeable plastic casing. In another embodiment, the carbon particles, or the compression pieces, may be contained within an envelope of the semipermeable plastic. This envelope can be in the form of a bag, or of a dividing wall that separates the amorphous carbon particles from culture medium to be contained in a container.
The plastic to be used must be permeable to the toxic substances present in the culture medium.
These toxic substances can diffuse through the plastic to the surface of the amorphous carbon particles, or to the space in which the amorphous carbon particles are present. The swelling of the plastic under the influence of culture medium promotes diffusion.
Furthermore, it is possible that the plastic is porous and contains pores smaller than the smallest size of the carbon particles. Generally, the pore size is less than 40 µm, more preferably less than 20 µm, and most preferably less than 10 µm.
Plastics that can be used in the absorbent of the present invention include plastics selected from the following groups: cellulose esters, fluorine-containing polymers, acrylic polymers, rubber, vinyl polymers, polyalkylene polymers, polyamides, and polystyrene.
According to a preferred embodiment, a container that comes into contact with culture medium is pre-provided with the absorbent according to the invention. This absorbent can for instance be adhered to a container wall. Containers that can be provided with the absorbent include test tubes, perish trays, and culture bottles.
The invention also relates to a container provided with an absorbent according to the invention.

The amorphous carbon particles used in the present invention are in the form of powder, granules and optionally pellets, such as a pellet. The amorphous coal is derived from carbonaceous material such as wood, peat, lignite, bone cabbage (obtained by calcining bone), nut shells, coal and the like. The carbon is activated, creating a very porous structure. The activation can be carried out, for example, by oxidation with steam, carbon dioxide, combustible gas, air and the like. Another possibility is the treatment with chemicals, such as metal chlorides, metal sulphides, metal sulphates.

polystyrene. It is essential for the plastic to be used that the toxic substances present in the culture medium can diffuse through the plastic or pass through pores present in the plastic layer.
The culture medium can have a very diverse composition. The culture medium often contains serum, such as human serum, bovine serum, fetal calf serum or umbilical cord serum. Examples of a culture medium are Earles medium, which is buffered with HEPS and provided with 2% heat-inactivated human umbilical cord serum.
Another culture medium is HTF (Human Tubal Fluid) to which is added 15% heat-inactivated human umbilical cord serum and 0.3 mM EDTA.
Figure 1 shows a test tube 1 filled with culture medium 2 for culturing cells in vitro. For the separation of toxic substances, an absorbent 3 according to the invention is included in the test tube. The absorbent comprises pellets 4 of amorphous carbon particles, which are surrounded by an envelope 5 of a cellulose ester. At the end of the culture, the adsorbent 3 can be easily separated from the culture medium.
Figure 2 shows a test tube 6 according to the invention which is filled with culture medium 2. The test tube is provided on its inner surface with absorbent 7 according to the invention, in the form of amorphous carbon particles adhered to the inner surface of the test tube and with the culture medium are separated by an envelope 8 in the form of a layer of plastic which is impermeable to the carbon particles and permeable to toxic substances present in the culture medium 2.

In vitro fertilization of egg cells with sperm cells in a culture medium container provided with an absorbent according to the invention, the survival of the sperm cells, the percentage of fertilization and the quality of the culture in vitro, and possibly also the number of subsequent pregnancies have been greatly improved in that, according to the invention, the absorbent in the culture medium during the in vitro culture has contained or got into the culture medium, without the amorphous carbon particles being able to disperse into the culture medium.
