Description

DESCRIPTION APPLICATION OF A BILE ACID COMPOSITE BACTERIAL AGENT IN THE PREPARATION OF SWINE FEED ADDITIVES
TECHNICAL FIELD The present invention generally relates to the field of poultry and livestock breeding, and particularly to application of a bile acid composite bacterial agent in the preparation of swine feed additives. BACKGROUND Nowadays, swine breeding is generally in large scale. In order to achieve high profits, farmers will choose to over-breed and give excessive feeds. These problems will cause chaos in the pig breeding environment and affect the health of swine. At present, the main problems of swine breeding include body poisoning, malnutrition, immunosuppression, and inflammation, etc. Especially for sows before parturition, the breeding environment problems can cause in vivo accumulation of toxins in swine, and develop metabolic diseases and nosotoxicosis, leading to the reduction of body's self-purification ability and weakening the physical condition of the piglets, and causing diarrhea, etc. The diarrhea of piglets may induce pathological changes of liver and kidneys and reduce the survival rate of piglets. Presently, antibiotics are often used to solve the problems of endotoxin accumulation and diarrhea in sows and piglets, but antibiotics can make the body to produce resistance, which is not conducive to the swine breeding. SUMMARY In order to solve the drawbacks and deficiencies of the prior art, the present invention provides the application of a bile acid composite bacterial agent in the preparation of swine feed additives. The bile acid composite bacterial agent can significantly improve the immunity of sows and restore the healthy liver and intestines of piglets. In order to achieve the object, the present invention adopts the following technical solutions: The present invention provides application of a bile acid composite bacterial agent in the preparation of swine feed additives. Further, the bile acid composite bacterial agent comprises Parabacteroides
DESCRIPTION
distasonis bacterial suspension, Bacteroides thetaiotaomicron bacterial suspension, bile acid and Eucalyptus polybractea essential oil. Further, the volume-to-mass ratio of the Parabacteroides distasonis bacterial suspension, Bacteroides thetaiotaomicron bacterial suspension, bile acid and Eucalyptus polybractea essential oil is 2-3:2-4:2-3:1. Further, the total bacterial count of the bile acid composite bacterial agent is not less than 6x108 CFU/mL. Further, the method of using the bile acid composite bacterial agent is: adding the bile acid composite bacterial agent directly to a feed for prenatal sow breeding, 3 times a day until sows give birth. Further, the additive amount of the bile acid composite bacterial agent is 2-2.5 L/t feeds for breeding. Further, after the bile acid composite bacterial agent is diluted with water in a volume ratio of 1:1-2, the diluent of the bile acid composite bacterial agent is directly administered to sick piglets, one daily for 8 to 12 consecutive days. Further, the amount of the diluent of the bile acid composite bacterial agent is 6 to 10 mL/piglet. Compared with the prior art, the present invention has the following advantages and beneficial effects. The composite bacterial agent provided in the present invention has natural components, without toxic and side effect. It can significantly restore the intestinal health and non-specific immunity of the parturient sows, restore their body's self-purification ability, promote the elimination of toxins from the body and protect the health of sows, thereby reducing the problem of diarrhea caused by maternal toxin infection in piglets; in addition, it can effectively increase the sow's milk yield and ensure the nutritional intake of suckling piglets. The bile acid composite bacterial agent can be directly administered to sick piglets to enhance their physical conditions, increase their survival rate and milk intake, and restore their liver and intestine functions, thereby protecting the liver and gallbladder and repairing the damage to liver and gallbladder. In addition, it has the antibacterial, anti-inflammatory and deworming effects, to make the piglet skin red and bright, thereby increasing the income of swine breeding. Therefore, it has broad application prospects.

spots; the left figure indicates sow with tear spots, and the right figure indicates the sows after using a bile acid composite bacterial agent. DETAILED DESCRIPTION Example 1: 1. Preparation of Parabacteroides distasonis bacterial suspension (1) Inoculate Parabacteroides distasonis into a liquid nutrient medium (tryptone 15.0g, soytone 5.0g, sodium chloride 5.0g, sterile water 10OOmL, pH 7.4), then activate and cultivate for 48 h in an anaerobic environment at 37°C, to get Parabacteroides distasonis bacterial solution; (2) Add a sucrose solution with a mass concentration of 20% to the bacterial solution at a volume ratio of 1:1, mix well and centrifuge to obtain a bacterial strain precipitate; (3) Re-dissolve the above bacterial strain precipitate with a sucrose solution with a mass concentration of 10%, with a ratio of the volume of the sucrose solution to the mass of the bacterial strain precipitate of 2:1, mix well to obtain Parabacteroides distasonis bacterial suspension with bacterial count of not less than 2x10' CFU/mL. 2. Preparation of Bacteroides thetaiotaomicron bacterial suspension Bacteroides thetaiotaomicron was inoculated into GAM liquid medium and activated under anaerobic environment at 35°C for 48 h. 1 mL of the resulting activated bacterial solution was re-inoculated into 50 mL GAM liquid medium and cultured for 24 to 76 h, centrifuged and the bacterial strain precipitate was resuspended in sterile normal saline, and centrifuged again and resuspended in sterile normal saline for the second time, to obtain Bacteroides thetaiotaomicron bacterial suspension with bacterial count>1x109 CFU/mL. 3. Preparation of bile acid (1) Saponification: Add 1kg of porcine gall powder crushed through a 40-mesh sieve into a reactor, then add 1OL of sodium hydroxide solution with a mass concentration of 10%, heat and stir until boiling, and keep boiling and stir for 14h, then cool the saponification fluid until solid-liquid stratification, then remove the supernatant, to obtain the remaining solid of bile acid saponification product; (2) Decolorization: Add 6L of water to the bile acid saponification product, heat it to completely dissolve the bile acid saponification product, then pour it into a decolorization tank, add hydrogen peroxide and stir evenly, react at room temperature
DESCRIPTION
for 24 h, then filter the reaction solution to an acidification tank, to obtain the filtrate; (3) Acidification: Cool the filtrate to room temperature, slowly add 10% hydrochloric acid solution while stirring, and stop adding acid when the solution pH reaches 3, and centrifuge and filter to obtain a white solid; (4) Purification: Rinse the white solids with water continuously, and complete the water rinsing when the filtered water is detected neutral; place the white solids after centrifugal filtration into an oven to obtain the crude bile acid; (5) Recrystallization: Place the crude bile acid into an extraction tank, add ethyl acetate of 10 times the weight of the crude bile acid to stir until dissolved completely, then add 0.08g of anhydrous sodium sulfate for dehydration, filter and collect the filtrate, concentrate the filtrate under reduced pressure and recover ethyl acetate to obtain liquid bile acid; (6) Drying: Place the liquid bile acid into an oven to obtain a bile acid finished product. The bile acid finished product contains 78.6% of hyocholic acid and hyodeoxycholic acid and 20.0% of chenodeoxycholic acid. 4. Preparation of Eucalyptus polybractea essential oil The Australian Eucalyptus polybractea leaves were placed into a dryer for drying after washing in a washing tower, then pulverized in a pulverizer, and then entered a steam distillation kettle, a condenser and an oil-water separator to separate the oil phase and water phase. The water phase entered an ether extractor to separate the waste water, the liquid containing ether entered the ether recovery tower to recover the ether. The recovered ether and oil phase liquid were added to a fixed-bed adsorption dessicator, to further remove the moisture in the oil phase. Finally the oil phase liquid was added to a vacuum rectifying tower for rectification to remove the impurity oil and obtain the Eucalyptus polybractea essential oil. The prepared Eucalyptus polybractea essential oil was stored in a sealed, opaque container and protected from light. 5. Preparation of bile acid composite bacterial agent The above prepared bile acid, Eucalyptus polybractea essential oil, Bacteroides thetaiotaomicron bacterial suspension and Parabacteroides distasonis bacterial suspension were mixed evenly at a mass-to-volume ratio of 2:1:2:2, to obtain a composite bacterial agent with the total bacterial count of no less than 6x108 CFU/mL. Example 2 A total of 536 pigs were bred in a pig farm. During the breeding period, pregnant
DESCRIPTION
sows were found to have the tear spots (FIG. 1), and 30 of them were chosen to study the effect of the bile acid composite bacterial agent. Thirty sows were divided into different pigsties for breeding. The bile acid composite bacterial agent was mixed into the feed at a ratio of 2L/ton feed. The sows were fed 3 times a day, for 20 consecutive days, as shown in FIG.1. Twenty days later, the tear spots of sows alleviated apparently and even disappeared. After 20 day, the bile acid composite bacterial agent was evenly mixed into the feed at a ratio of 1 L/ton feed to consolidate the effect until the sows farrowed. Compared with the sows whose feeds were not added the bile acid composite bacterial agent, the experimental sows could produce more milk with obviously improved milk quality. The piglets produced by the experimental sows had obviously improved health conditions, with stronger vitality and brighter red skin than other newborn piglets, indicating that the bile acid composite bacterial agent of the present invention had good safety effects, could effectively treat the tear spots of sows and would not bring negative effects to sows and piglets. In addition, because the components are natural and endogenous substances, the bile acid composite bacterial agent of the present invention can be recycled in the body along with the enterohepatic circulation of bile acid, with stable effect; furthermore, it is not easy to be catabolized out of the body.
