Abstract
OF THE DISCLOSURE
The present disclosure discloses an SDF-1 induced rabbit knee osteoarthritis model building
device. A rabbit is fixed in an inwards sunken groove of an operation table through fixing belts
of the building device, and then is operated with operation kits and injection syringes which are
collected in a collection drawer. The rabbit is effectively irrigated using a designed irrigating
water tank before and after the operation. Meanwhile, a designed drainage hole is used to collect
sewage, and a designed magnifying lens connected by a shaping rod is used to magnify and
display the injected part during injection. The present disclosure assists in building a reasonable
and effective knee osteoarthritis model to effectively study the pathogenesis mechanism of the
knee osteoarthritis, shorten the study period and provide reliable measures for effective diagnosis
and treatment.

SDF-1 INDUCED RABBIT KNEE OSTEOARTHRITIS MODEL BUILDING DEVICE
[0001] The present disclosure belongs to the technical field of animal disease modeling, and specifically relates to an SDF-1 induced rabbit knee osteoarthritis model building device.
[0002] Knee osteoarthritis is a disease that is widespread in the joints, and is the degeneration of the articular cartilage. Meanwhile, cartilage cells cannot repair the articular cartilage matrix, ending up with changes in composition and structure. This disease mainly clinically manifests as chronic pain, joint instability, stiffness, deformity and narrowing of the joint space. The pathological changes of the joint suffering from the knee osteoarthritis include degeneration of the articular cartilage, thickening of the subchondral bone, formation of osteophytes, changes in the degree of synovitis, degeneration of ligaments, degeneration of the meniscus of the knee joint, and hypertrophy of the joint capsule. Changes in the muscles, nerves, mucous sacs, and partial fat pads around the joints may also occur, leading to arthritis or arthritis symptoms. However, the pathogenesis of the knee osteoarthritis is not yet fully clear.
[0003] Animal disease models are applied to researches in experimental physiology, experimental pathology, experimental diagnostics, and therapeutics to explore the occurrence, development, diagnosis, treatment, etc. of diseases. They are an important part of modern medical research. The development process of the knee osteoarthritis disease is slow and complicated. Studies on the diseases relying on clinical cases alone or taking humans as experimental subjects develop slowly and are restrained in many aspects. By the use of experimental animal models to study the diseases, the occurrence mechanisms of the diseases can be observed more accurately, the development law of the diseases can be learned more effectively, and the advancement of medical technology can be promoted. Therefore, by establishing a reasonable and effective knee osteoarthritis animal model, the pathogenesis of the knee osteoarthritis can be accurately and effectively studied, the study period is shortened, and effective diagnosis and treatment methods can be sought.
[0004] The present disclosure aims to provide an SDF-1 induced rabbit knee osteoarthritis model building device which assists in building a reasonable and effective rabbit knee osteoarthritis model to effectively study the pathogenesis mechanism of knee osteoarthritis, shorten the study period and provide reliable measures for effective diagnosis and treatment.
[0005] In order to solve the above-mentioned problems, the technical solution adopted in the present disclosure is as follows.
[0006] The SDF-1 induced rabbit knee osteoarthritis model building device includes an animal fixing device which is an operation table provided with an inwards sunken groove in an upper end surface. A lower end surface of the operation table is provided with a collection drawer; several fixing belts used to fix limbs are provided on the inwards sunken groove of the operation table; an irrigation water tank is provided at one end of the right side of the operation table; the lower end of the irrigation water tank is connected with an irrigation pipe and head; one side of the irrigation water tank is connected with a magnifying lens through a shaping rod; one end of the operation table close to the irrigation water tank is provided with a run-through drainage hole; and the lower end of the drainage hole is connected with a guide pipe connected to a collection barrel at the bottom.
[0007] Further, the inwards sunken groove on the operation table is a slope, and the lowest point is provided with the drainage hole.
[0008] Further, the collection drawer collects injection syringes of different specifications, operation kits, and 2-0 Mousse stitches.

A lower end surface of the operation table 1 is provided with a collection drawer 3; several fixing belts 4 used to fix limbs are provided on the inwards sunken groove 2 of the operation table 1; an irrigation water tank 5 is provided at one end of the right side of the operation table 1; the lower end of the irrigation water tank 5 is connected with an irrigation pipe and head 8; one side of the irrigation water tank 5 is connected with a magnifying lens 7 through a shaping rod 6; one end of the operation table 1 close to the irrigation water tank 5 is provided with a run-through drainage hole 9; and the lower end of the drainage hole 9 is connected with a guide pipe connected to a collection barrel 10 at the bottom.

[0020] In the present disclosure, 45 adult 3 to 5-month-old healthy Japanese white rabbits purchased by the Department of Experimental Zoology of Kunming Medical University, female or male or both, each 2 to 2.5 kg, are selected to verify the reliability of the present disclosure by means of grouping.
[0021] 1. Preparation of main ingredients
[0022] 1) SDF-1 solution: 1000 uL of PBS solution is added into 1000 ug of SDF-1 lyophilized powder to prepare SDF-1 stoste at 1000 ug/mL, and 0.2 mL of solutions at corresponding concentrations are prepared according to 10 ug/kg, 20 ug/kg, and 40 ug/kg.
[0023] 2) 10% chloral hydrate: The chloral hydrate is prepared according to a volume ratio of chloral hydrate anhydrous to normal saline of 1: 9.
[0024] 3) 0.01 mol/L phosphate buffer solution (PBS): PBS powder is dissolved in 2000 ml of distilled water, pH=7.4.
[0025] 4) preparation of 0.01 mol/LPBST: 0.1% Tween20 is added into the 0.01 mol/LPBS, and the product is used after full and uniform mixing.
[0026] 5) preparation of 5% sheep serum solution: a pipette sucks up 30 ul of TritonX-100 (0.3%); 500 ul of normal sheep serum stoste (5%) is used for closure; finally, the volume is fixed at 10 ml with the PBS; the solutions are uniformly mixed and then placed in a 4°C refrigerator for later application.
[0027] 2 Experimental method
[0028] 2.1 Building of a rabbit knee osteoarthritis model and animal grouping
[0029] 1) 45 3 to 5-month-old healthy Japanese white rabbits are randomly selected as a blank group, a Hulth operation group, a low-dosage group, a medium-dosage group, and a high-dosage group; the knee joint of the right posterior limb is an operation part; normal saline is injected to the blank group; the anterior cruciate ligament, the posterior cruciate ligament, and the medial collateral ligament of the Hulth operation group are cut off, and the medial meniscus of the Hulth operation group is excised; and the SDF-1 solution atlOug/kg, 20ug/kg, and 40ug/kg is injected to the low-dosage group, the medium-dosage group, and the high-dosage group respectively according to weights.
[0030] 2) Personnel should wear hats and masks in strict aseptic operation, and the 10% chloral hydrate is injected to the ear vein as per 1 mL/kg for anesthesia.
[0031] 4) After the anesthesia is completed, the rabbit is fixed on the operation table; rabbit hairs on the surface of the posterior limb are shaved with a shaver to expose the operation part; and the operation part is disinfected for 3 times with a sterile cotton ball dipped with Type I iodine.
[0032] 5) In the blank group, the articular cavity of the right knee joint is located and then punctured point by point; if a part is punctured through, it is indicated that this part is the articular cavity; 0.2 mL of normal saline is injected into the articular cavity; the prepared SDF-1 solution is injected into the corresponding articular cavities of the low, medium, and high-dosage groups; and the above-mentioned operations are repeated for all the injection groups on the 3rd day and the 5th day.
[0033] 6) In the Hulth operation group, the rabbit is fixed; after the right knee joint is disinfected and is covered with sterile drapes, the knee joint of the right posterior limb is cut with a scalpel from the center, forming a skin incision that is about 3 cm long to expose tissues such as the patella, the patellar tendon, and the muscles, and is then cut longitudinally along the patella and the patellar tendon to open the articular cavity; the knee joint is flexed, and the patella is eversed, fully exposing the articular cavity; the operation field is irrigated with the normal saline (irrigation is realized by the irrigation water tank); and the anterior cruciate ligament, the posterior cruciate ligament meniscus and the medial collateral ligament are cut off. The articular cavity is irrigated; the knee joint is reset; the operative incision is sutured layer by layer (the magnifying lens may be used in the injection and suturing processes); the operative wound is irrigated with the Type I iodine and covered with aseptic dressing; penicillin is injected daily according to 20,000 units/kg 3 days after the operation to prevent postoperative infection; and the dressing is changed once every other day to observe the wound healing.
[0034] 7) After the treatment, the rabbits are routinely fed in SPF animal rooms, and the wounds, activities and feeding are observed.
Claims (3)
1. An SDF-1 induced rabbit knee osteoarthritis model building device, comprising an animal fixing device which is an operation table provided with an inwards sunken groove in an upper end surface, wherein lower end surface of the operation table is provided with a collection drawer; several fixing belts used to fix limbs are provided on the inwards sunken groove of the operation table; an irrigation water tank is provided at one end of the right side of the operation table; the lower end of the irrigation water tank is connected with an irrigation pipe and head; one side of the irrigation water tank is connected with a magnifying lens through a shaping rod; one end of the operation table close to the irrigation water tank is provided with a run-through drainage hole; and the lower end of the drainage hole is connected with a guide pipe connected to a collection barrel at the bottom.
2. The SDF-1 induced rabbit knee osteoarthritis model building device according to claim 1, wherein the inwards sunken groove on the operation table is a slope, and the lowest point is provided with the drainage hole.
3. The SDF-1 induced rabbit knee osteoarthritis model building device according to claim 1, wherein the collection drawer collects injection syringes of different specifications, operation kits, and 2-0 Mousse stitches.
