Abstract

Methods for the synthesis of various novel substituted dipeptide derived nitrogen-containing heterocyclic compounds (I) and their pharmaceutically acceptable salt derivatives are provided. The compounds of the invention are claimed and are useful as medicaments for the treatment or prevention of disease in a mammal, for example a human. In particular the compounds are useful as immuno therapeutics and anti -microbial drugs or vaccines. These heterocyclic derivatives can be used as an active agent in a pharmaceutical, as well as a diagnostic utility.

Description

COMPOSITIONS AND METHODS FOR SYNTHESIZING NOVEL HETEROCYCLIC THERAPEUTICS Field of the Invention 5 The present invention relates to novel compositions and methods for synthesizing 7 and 8-membered ring dipeptide-derived nitrogen-containing heterocyclic compounds that are useful for the treatment of disease, and pharmaceutically acceptable salts thereof In addition, the invention relates to compositions useful for the treatment and prevention of disease in a mammal, for 10 example a human. Background of the Invention Any discussion of the prior art throughout the specification should in no way be considered as an admission that such prior art is widely known or forms part of 15 common general knowledge in the field. Approximately 40% of the world's population lives in areas where a significant risk of contracting malaria exists. Each year, 300-500 million people suffer from acute malaria, and 0.5-2.5 million die from the disease. Although malaria has been widely eradicated in many parts of the world, the global number of 20 cases continues to rise. The most important reason for this alarming situation is the rapid spread of malaria parasites that are resistant to antimalarial drugs, especially chloroquine, which is by far the most frequently used. The Plasmodium genus of protozoal parasites (mainly Pifalciparum, P. vivax, P.ovale, and P.malariae) have a life cycle which is split between a vertebrate host 25 and an insect vector. The Plasmodium species, with the exception of P.malariae (which may affect the higher primates) are exclusively parasites of man. The mosquito is always the vector, and is always an Anopheline mosquito, although, out of the 380 species of Anopheline mosquito, only 60 can transmit malaria. Only female mosquitos are involved as the males do not feed on blood. 30 During a blood meal by the female Anophelus mosquito the Plasmodium sporozoites from the mosquito salivary gland are injected into the human Once in the human bloodstream, the sporozoites arrive in the liver and penetrate hepatocytes, where they remain for 9-16 days, multiplying within the cells and form 2 merozoites. On release, they penetrate red blood cells in which they produce either merozoites or micro and macrogametocytes, which have no further activity within the human host. Another mosquito arriving to feed on the blood may suck up these 5 gametocytes into its gut, where exflagellation of microgametocytes occurs, and the macrogametocytes are fertilized. The resulting ookinete penetrates the wall of a cell in the midgut, where it develops into an oocyst. Sporogeny within the oocyst produces many sporozoites and, when the oocyst ruptures, the sporozoites migrate to the salivary gland, for injection into another host. 10 Once injected into the human host, all Plasmodium species will penetrate hepatocytes. However, Pfalciparum and P.malariae sporozoites trigger immediate schizogony whereas P.ovale and P.vivax sporozoites may either trigger immediate schizogony or have a delayed trigger, resulting in dormant hypnozoites. Some strains, such as the North Korean strain, seem to consist of sporozoites with 15 universally delayed triggers, so they all form long lasting hypnozoites. P.vivax may have an incubation period of up to 10 months. A biting mosquito transfers about 10% of its sporozoite load into the capillaries or perivascular tissue. At this stage the sporozoites must begin their evasion of the host defenses, possibly by binding serum proteins in order to avoid 20 the host's immune defenses. Some are destroyed by macrophages, or by antigen specific antibodies in immune individuals, but in non immune individuals, they reach the hepatocytes and initiate schizogeny or become hypnozoites depending on their delay trigger. Within 45 minutes nearly all sporozoites will be absent from the peripheral circulation. 25 Invasion of host hepatocytes by sporozoites represents an early step in the life cycle of the malaria parasite. Sporozoites develop in the liver and give rise to merozoites which subsequently invade erythrocytes after release into the bloodstream. The parasites reproduce asexually in the red blood cells. Eventually the red blood cells burst releasing merozoites into the blood. This blood phase 30 continues, destroying significant numbers of red blood cells, and is responsible for the paroxysms (i.e., "chills and fever" cycles) that are characteristic of malaria 2a infections. The research and development of new antimalarial drugs has been largely neglected since theI970s owing to the end of colonialism, changes in the areas of military engagement, and the restricted market potential. Only in recent years, in 5 part supported by public funding programs, has interest in the development of antimalarial drugs been renewed. This effort has been aided by new data available from the recently sequenced genome of the malaria parasite, Plasmodium falciparum, and the application of methods of modern drug design. The application of modern drug development strategies promises to bring 10 significant advances in the fight against this disease. Currently, few drugs against the malaria liver stage parasites are available. Because of technical difficulties, low yield and the complexity of the setting necessary to produce liver stage parasites, no new drugs are assessed systematically against these stages. Nevertheless, liver stages are a strong 15 potential for drug targeting because; a) they possess a more complex and distinct metabolism than their blood stage counterpart, and b) they precede the pathogenic blood stage and thus offer prophylactic possibilities against malaria. Thus, the evaluation of new drugs against the liver stage offers new therapeutic horizons. Moreover, the completion of the parasite genome, which 20 will lead to the identification of parasite metabolic pathways different from those of the host, will aid the research toward the identification of new drugs. This, combined with the recent development in combinatorial chemistry (synthesis and evaluation of a great number of compounds, creation of a wide molecular diversity) is leading to the identification of new lead molecules with antimalarial 25 activity. An additional observation is that some antimalarial compounds, for example, chloroquine also exhibit anti-HIV-1 activity. It is an object of the present invention to overcome or ameliorate at least 2b one of the disadvantages of the prior art, or to provide a useful alternative. It is an object of an especially preferred form of the present invention to provide for antimalarial drugs to also be useful for HIV and AIDS therapy. Unless the context clearly requires otherwise, throughout the description 5 and the claims, the words "comprise", "comprising", and the like are to be construed in an inclusive sense as opposed to an exclusive or exhaustive sense; that is to say, in the sense of "including, but not limited to". Although the invention will be described with reference to specific examples it will be appreciated by those skilled in the art that the invention may 10 be embodied in many other forms. Summary of the Invention According to a first aspect of the present invention there is provided a therapeutic composition comprising a compound of the formula B: 0 R5N N R1 R4 R2 15 R 3 0 (B) or a pharmaceutically acceptable salt thereof, wherein, R', R2, R', R4, and R 5 represent, each independent from the other, a member selected from the group consisting of: a hydrogen atom; an amino acid side chain; a (Cl-C1O) alkyl; (C1-C1O) alkenyl; (Cl-C1O) alkynyl; 20 (C5-C12) monocyclic or bicyclic aryl; (C5-C14) monocyclic or bicyclic aralkyl; monocyclic or bicyclic (C5-C14) heteroaralkyl; and (Cl-CO) monocyclic or bicyclic heteroaryl group having up to 5 heteroatoms selected from N, 0, S, and P said groups being able to be non-substituted or substituted by 1 to 6 substituents further selected from the group 25 consisting of: a halogen atom, an NO 2 , OH, amidine, benzamidine, imidazole, 1,2,3-triazole, alkoxy, (CI-C4) alkyl, amino, piperazine, piperidine, dialkylamino, guanidine group, bis alkylated or bis acylated guanido group, carboxylic acid, carboxamide, ester, hydroxamic acid, 2c phosphinic acid, phosphonate, phosphonamidate, sulfhydryl and any combination thereof, and at least one other ingredient selected from the group consisting of a carrier, an excipient, an additional biologically active agent or a 5 combination thereof in a pharmaceutically acceptable form. According to a second aspect of the present invention there is provided a therapeutic compound of the formula B: 0 RN NR R4 N 2 R 3 0 (B) 10 or a pharmaceutically acceptable salt thereof, wherein, 1 2 3 45 R', R , R , R4, and R represent, each independent from the other, a member selected from the group consisting of: a hydrogen atom; an amino acid side chain; a (Cl-C10) alkyl; (Cl-C0) alkenyl; (Cl-C0) alkynyl; (C5-C12) monocyclic or bicyclic aryl; (C5-Cl4) monocyclic or bicyclic 15 aralkyl; monocyclic or bicyclic (C5-C14) heteroaralkyl; and (Cl-CI) monocyclic or bicyclic heteroaryl group having up to 5 heteroatoms selected from N, 0, S, and P said groups being able to be non-substituted or substituted by 1 to 6 substituents further selected from the group consisting of: a halogen atom, an NO 2 , OH, amidine, benzamidine, 20 imidazole, 1,2,3-triazole, alkoxy, (Cl-C4) alkyl, amino, piperazine, piperidine, dialkylamino, guanidine group, bis alkylated or bis acylated guanido group, carboxylic acid, carboxamide, ester, hydroxamic acid, phosphinic acid, phosphonate, phosphonamidate, sulthydryl and any combination thereof, 25 with the proviso that when R1 and R 5 are H; and R 2 and R 4 are aralkyl, then R 3 is other than alkyl, for use as a medicament.
2d According to a third aspect of the present invention there is provided a method for the treatment of malaria, HIV infection and/or AIDs, said method comprising administering to a subject in need thereof an effective amount of a 5 compound of the formula B: 0 R5N NR' Nt

In one aspect, the invention relates to novel dipeptide derived heterocyclic compounds synthesized using the WO 2007/074171 PCT/EP2006/070258 3 methods of the invention. WO 2007/074171 PCT/EP2006/070258 4 Detailed Description of the Invention [00016] When describing the compounds, compositions and methods of the invention, the following terms have the following meanings, unless otherwise indicated. [00017] "Pharmaceutically acceptable salt" means those salts which retain the biological effectiveness and properties of the parent compounds and which are not biologically or otherwise harmful as the dosage administered. The compounds of this invention are capable of forming both acid and base salts by virtue of the presence of amino and carboxy groups respectively. Pharmaceutically acceptable base addition salts may be prepared from inorganic and organic bases. Diluents are carefully selected to match the specific formulation with attention paid to the compression properties. Suitable lubricants, including agents that act on the flowability of the powder to be compressed are, for example, colloidal silicon dioxide such as Aerosil 200; talc; stearic acid, magnesium stearate, and calcium stearate. These compounds were found to be active by screening a small "prospecting" library against the malaria liver stage (LS). [0001031 Interest in designing and evaluating the dipeptide-derived 1,3,5 triazepan-2,6-dione scaffold stems from the remarkable biological activities exhibited by molecules with diazepine and triazepine skeletons. In particular seven membered cyclic ureas have attracted much attention in recent years with application in the development of HIV-protease and reverse transcriptase inhibitors, Factor Xa inhibitors, beta-lactamases inhibitors, phospholipase C inhibitors, and chemokine receptor antagonists. [000104] With reference to Figure 2, 1, 3, 5-triazepan-2,6-diones 4 are constructed in only four steps by cyclization of simple activated dipeptide derivatives, and the approach benefits from the considerable diversity of commercial alpha-amino acids. The cyclization strategy parallels the previously reported approach to the preparation of enantiopure macrocyclic oligoureas. Succinimidyl carbamates 2 were prepared from Boc-dipeptides 1 as previously described for N-protected alpha- and beta-amino acids. Selective removal of the Boc group by treatment of crude 2 with trifluoroacetic acid (TFA) afforded 3 which cyclized to 4 in the presence of diisopropylethylamine (DIEA). The cyclization proceeded rapidly with concomitant release of N-hydroxysuccinimide and formation of the TFA salt of DIEA. These byproducts were removed by either recrystallization of crude 4, liquid-liquid extraction or polymer supported sequestration (PSS) with tris-(2-aminoethyl)amine polystyrene (PS-triamine). Alternatively polymer supported-DIEA (PS-DIEA) was found to be very effective in promoting cyclization and simultaneously removing byproducts. Interestingly, in the presence of KF/AI20 3 or NaH (2 equiv.) cyclo(Xaa-gSar-CO) 4a, 4c and cyclo(Phe gPro-CO) 4i were converted to 8 and 9 monoalkylated at the gem-diamino urea nitrogen with good to excellent selectivities (8:6 and 9:7 in the range 80:20 to 100:0). Crystal structures of dialkylated and monoalkylated derivatives 7 and 9(I), respectively, show a rearrangement of the ring geometry with R' side-chain now pointing in quasi axial orientation (Figure 5).
WO 2007/074171 PCT/EP2006/070258 26 [000110] Invasion of host hepatocytes by sporozoites represent an early step in the life cycle of the malaria parasite. Malaria liver stages hold great promise for drug targeting because i) they possess a more complex and distinct metabolism than their blood stage counterpart and ii) they precede the pathogenic blood stage, thus offering prophylaxis possibilities against malaria. However, very few drugs against LS parasites are available and most of them like primaquine display severe side effects or loose efficiency due to the development of parasite resistance. [000111] Seventeen different 1, 3, 5-triazepan-2,6-diones were randomly selected and were first tested at different doses for toxicity on primary mouse hepatocyte culture. From these, nine were shown to be toxic at all doses tested (6, 25-100 pg/ml) and two at high doses(over 50 pg/ml). The non toxic remaining six molecules were thus tested to evaluate their effets on sporozoite invasion and development in hepatocytes. Claims (16)

1. A therapeutic composition comprising a compound of the formula B: 0 R5~)L R 1 RN NR R 3 0 (B) 5 or a pharmaceutically acceptable salt thereof, wherein, 1 2 3 4 R , R , R , R4, and R 5 represent, each independent from the other, a member selected from the group consisting of: a hydrogen atom; an amino acid side chain; a (Cl-C10) alkyl; (Cl-C0) alkenyl; (Cl-C10) alkynyl; (C5-Cl2) monocyclic or bicyclic aryl; (C5-C14) monocyclic or bicyclic 10 aralkyl; monocyclic or bicyclic (C5-C14) heteroaralkyl; and (Cl-C10) monocyclic or bicyclic heteroaryl group having up to 5 heteroatoms selected from N, 0, S, and P said groups being able to be non-substituted or substituted by 1 to 6 substituents further selected from the group consisting of: a halogen atom, an NO 2 , OH, amidine, benzamidine, 15 imidazole, 1,2,3-triazole, alkoxy, (CI-C4) alkyl, amino, piperazine, piperidine, dialkylamino, guanidine group, bis alkylated or bis acylated guanido group, carboxylic acid, carboxamide, ester, hydroxamic acid, phosphinic acid, phosphonate, phosphonamidate, sulfhydryl and any combination thereof, 20 and at least one other ingredient selected from the group consisting of a carrier, an excipient, an additional biologically active agent or a combination thereof in a pharmaceutically acceptable form.
2. A therapeutic compound of the formula B: 0 R.N NR* R4 R 2 25 R 3 0 (B) 37 or a pharmaceutically acceptable salt thereof, wherein, 1 2 3 45 R , R , R
3 , R 4 , and Rs represent, each independent from the other, a member selected from the group consisting of a hydrogen atom; an amino acid side chain; a (Cl-C1O) alkyl; (Cl-C1O) alkenyl; (Cl-CIO) alkynyl; 5 (C5-C12) monocyclic or bicyclic aryl; (C5-C14) monocyclic or bicyclic aralkyl; monocyclic or bicyclic (C5-C14) heteroaralkyl; and (Cl-C1O) monocyclic or bicyclic heteroaryl group having up to 5 heteroatoms selected from N, 0, S, and P said groups being able to be non-substituted or substituted by I to 6 substituents further selected from the group 10 consisting of: a halogen atom, an NO 2 , OH, amidine, benzamidine, imidazole, 1,2,3-triazole, alkoxy, (CI-C4) alkyl, amino, piperazine, piperidine, dialkylamino, guanidine group, bis alkylated or bis acylated guanido group, carboxylic acid, carboxamide, ester, hydroxamic acid, phosphinic acid, phosphonate, phosphonamidate, sulfhydryl and any 15 combination thereof, with the proviso that when R1 and R5 are H; and R2 and R4 are aralkyl, then R 3 is other than alkyl, for use as a medicament. 20 3. A method for the treatment of malaria, HIV infection and/or AIDs, said method comprising administering to a subject in need thereof an effective 5 amount of a compound of the formula B: 0 N N R4* R2 N R 3 O (B) or a pharmaceutically acceptable salt thereof, wherein, 2 2 R', R , R 3 , R 4 , and R 5 represent, each independent from the other, a member selected from the group consisting of: a hydrogen atom; an amino 10 acid side chain; a (Cl-C0) alkyl; (Cl-C0) alkenyl; (Cl-C0) alkynyl; (C5-C12) monocyclic or bicyclic aryl; (C5-C14) monocyclic or bicyclic aralkyl; monocyclic or bicyclic (C5-C14) heteroaralkyl; and (Cl-C0) monocyclic or bicyclic heteroaryl group having up to 5 heteroatoms selected from N, 0, S, and P said groups being able to be non-substituted 15 or substituted by I to 6 substituents further selected from the group consisting of a halogen atom, an NO 2 , OH, amidine, benzamidine, imidazole, 1,2,3-triazole, alkoxy, (C1-C4) alkyl, amino, piperazine, piperidine, dialkylamino, guanidine group, bis alkylated or bis acylated guanido group, carboxylic acid, carboxamide, ester, hydroxamic acid, 43 phosphinic acid, phosphonate, phosphonamidate, sulfhydryl and any combination thereof

Use of a compound of the formula B: 0 R5 ) R' N N, 5 R 3 0 (B) or a pharmaceutically acceptable salt thereof, wherein, R', R 2 , R 3 , R 4 , and R 5 represent, each independent from the other, a member selected from the group consisting of: a hydrogen atom; an amino acid side chain; a (Cl-C10) alkyl; (Cl-C0) alkenyl; (Cl-C0) alkynyl; 10 (C5-C12) monocyclic or bicyclic aryl; (C5-C14) monocyclic or bicyclic aralkyl; monocyclic or bicyclic (C5-C14) heteroaralkyl; and (Cl-CO) monocyclic or bicyclic heteroaryl group having up to 5 heteroatoms selected from N, 0, S, and P said groups being able to be non-substituted or substituted by 1 to 6 substituents further selected from the group 15 consisting of a halogen atom, an NO 2 , OH, amidine, benzamidine, imidazole, 1,2,3-triazole, alkoxy, (Cl-C4) alkyl, amino, piperazine, piperidine, dialkylamino, guanidine group, bis alkylated or bis acylated guanido group, carboxylic acid, carboxamide, ester, hydroxamic acid, phosphinic acid, phosphonate, phosphonamidate, sulfhydryl and any 20 combination thereof; in the preparation of a medicament for treating malaria, HIV infection and/or AIDs.

A therapeutic composition according to claim 1; a therapeutic compound 25 according to claim 2; a method for the treatment of malaria, HIV infection and/or AIDs; or use according to claim 15, substantially as herein described with reference to any one of the embodiments of the invention illustrated in the accompanying drawings and/or examples.
