Die Erfindung betrifft neue Cefalosporinderivate der Formel

Bickel in Helvetica Chimica Acta vol.57, Nr. 219, Seiten 2044ff (1974) aus 7-Phenylacetamido-3-hydroxymethyl-3-cephem-4-carbonsaurebenzhydrylester die entsprechende 3-Formylverbindung durch Oxidation hergestellt und anschliessend die mit Phenylessigsäure acylierten Analoga der Formel I mit R, = H oder Methyl durch Spaltung mit Trifluoressigsäure (in Gegenwart von Orthoameisensäuremethylester) hergestellt. Das Verfahren hat neben der aufwendigen Schutzgruppentechnologie vor allem den Nachteil, dass die Oxidation des Alkohols zum Aldehyd durch unerwünschte A-2 Isomensierung und Lactonislerung nicht sauber abläuft.
Daneben sind Oxidationsmittel wie Chrom (VI) oxld und Spaltreagenzien wie Trifluoressigsäure aus Ökologiegründen praktisch nicht einsetzbar.
Ein Verfahren zur Herstellung von 7-Phenylacetylamino-3-formyl-3-cephem-4-carbonsäure-p-methoxybenzylester, ausgehend von der entsprechenden 3-Jodmethylverbindung, wird von H. Tanaka et al in Synlett Seite 660 Nov. 1990 beschrieben. Als Oxidationsmittel wird Os mit Rhodiumchlond und Aluminium als Katalysatoren verwendet. Das Oxidationsprodukt muss chromatographisch gereinigt werden und die Ausbeuten liegen bei maximal 66 %.

In DE 2 360 620 werden 7-Acylamino-3-hydroxymethyl-3-cephem-4-Carboxylate mit Chromsäure/Schwefelsäure zu den entsprechenden Lactolen oxidiert. In dieser Patentschrift wird auf die Stabilität- probleme mit den entsprechenden tautomeren Aldehyden im Vergleich zu der in Formel I wiedergegebenen Lactolform hingewiesen. Der Einsatz von Chromverbindungen ist aus Ökologiegründen praktisch ausgeschlossen.
In Chem. Pharm. Bull. Vol. 28, Seiten 1339 ff, 1980 wird ein Verfahren zur Herstellung von 7-Acylamino- 3-formyl-3-cephem-4-carbonsäure-hydroxy-lactolen, ausgehend vom entsprechenden Lacton, durch Bromierung gefolgt von Halogen-Hydroxyaustausch beschrieben. Dabei müssen sorgfältig die Bedingungen zur Vermeidung einer Bromierung im Thiazinkern vermieden werden.
Nach der gegenständlichen Erfindung werden nicht nur die oben genannten Probleme - Ökologie, Wirtschaftlichkeit und unerwünschte Nebenreaktionen wie A-2 Bildung-vermieden, sondern gleichzeitig auch die zentralen Zwischenprodukte der Formel i zur beliebigen Derivierung in Position 7 und 3 - zur Herstellung von Cephalosporinen mit der Oxidationstufe eines Aldehyds in Position 3 - erhalten, und zwar ohne extensive Technik in sehr einfacher Welse und hohen Ausbeuten.
Verbindungen der Cefalosporinreihe mit freier Amino-Gruppe in 7-Stellung, einer ungeschützten Aldehydgruppe in Position 3 und einer freien Carbonsäuregruppe in Position 4 sind unbekannt, ihre Existenzfähigkeit ist bei der bekannten Labilität der Cefalosporine mit Aldehyd-Funktion in 3-Stellung überraschend und neu.
Einige 7-Acylderivate der Verbindungen der Formel I sind bekannt, sie existieren grösstenteils in der isomeren Hydroxylactonform der Formel

Als inerte Lösungsmittel sind Alkohole, beispielsweise Methanol, Nitnile, z. B. Acetonitril, Ketone, z. B.
Aceton, Ester oder halogenierte Lösungsmittel, Wasser oder Gemische der oben genannten Lösungsmittel geeignet. Als Base eignen sich aliphatische oder aromatische Amine, deren konjugierte Säuren in den genannten Lösungsmitteln löslich sind. Beispiele für Amine sind Triethylamin oder Pyridin. Arbeitet man in Wasser oder in Gegenwart von Wasser, kann als Base auch eine anorganische Base, beispielsweise ein Carbonat oder Hydrogencarbonat, eingesetzt werden.
Die Verbindungen der Formel I, worin Ri für eine Silylschutzgruppe steht, können aus den Verbindungen der Formel I, worin R, für Wasserstoff steht, oder direkt aus den Verbindungen der Formel VII durch Silylierung mit einem Silylietungsmittel erhalten werden.

Die Menge an Silylierungsmittel muss ausreichen, sowohl die Carbonsäure als auch die Aminogruppe quantitativ zu silylieren, um eine Selbstkondensation der freien Aminogruppe mit der Aldehydfunktion zu vermeiden. Vorzugsweise werden mindestens zwei bis drei Mol Silylierungsmittel pro Mol zu silylierendem Ausgangsmaterial eingesetzt.
Die neuen Verbindungen der Formel I (R, = H) sind genügend stabil, um auch aus wässriger Lösung isoliert zu werden ; sie können gegebenenfalls in Bissilylderivate der Forme ! ! (Ri = Silylschutzgruppe) überführt werden. Diese sind In Lösung stabil und werden vorzugsweise als Lösung weiter umgesetzt, gegebenenfalls an der Aminogruppe acyliert oder an der Aldehydgruppe weiter deriviert.
Als Acylierungsmittel eignen sich aktivierte Carbonsäurederivate, beispielsweise Säurechloride, gemischte Anhydride oder Aktivester. Die Isolierung der acylierten Derivate geschieht in bekannter Weise, entweder durch Entsilyiierung mit einem protischen Lösungsmittel. z.
B. einem Alkohol oder Wasser, wobei die Produkte direkt ausfallen, oder durch Fällung mit einem Antilösungsmittel oder als Carbonsäuresatze. Eine weitere Möglichkeit besteht in der Isolierung der Produkte als Ester, beispielsweise als Benzhydrylester, durch Umsatz der entsllyllerten Acylierungslösung mit Diphenyidiazomethan
Die Aldehydfunktion der Verbindung der Forme ! i (Ri = Silylschutzgruppe) ist der Derivierung mit gängigen stickstoffhältigen Aldehydreagentien, beispielsweise Aminen, Hydroxylaminen, Hydrazinen oder Semicarbaziden, zugänglich. Enthalten die Reagentien silylierbare Funktionen, werden diese zweckmässig als deren Silylderivate eingesetzt.
Der Umsatz mit den entsprechenden Aldehydreagentien wird in den oben zur Silylierung erwähnten Lösungsmitteln durchgeführt. Gegebenenfalls kann die Löslichkeit des Derivierungsreagens durch Zusatz eines dipolar aprotischen Lösungsmittels, z. B. DMF oder Sulfolan, erhöht werden. Die Umsetzungstemperatur ist nicht kritisch, vorzugsweise wird jedoch bei Raumtemperatur oder unter Kühlung gearbeitet. Die Isolierung der Derivate erfolgt in üblicher Weise, entweder durch Entsilylierung mit einem protischen Lösungsmittel, z. B. einem Alkohol oder Wasser, wobei das Produkt ausfällt, oder auch durch Extraktion und anschliessender Isolierung aus Wasser oder einem organischen Lösungsmittel- (gemisch).
Es ist sehr wesentlich, die Schutzgruppen an beiden funktionellen Gruppen einzuführen (Amin und Carboxylgruppe). Bei Monosilylierung nur an der Carboxylgruppe erfolgt, erwartungsgemäss Polymerisation unter Zersetzung.
In den folgenden Beispielen, die die Erfindung näher erläutern, ohne jedoch ihren Umfang einzuschränken, erfolgen alle Temperaturangaben in Celsiusgraden.
Beispiel 1: (6R-trans)-7-Amino-3-formyl-8-oxo-5-thia-1-azabicyclo[4.2.0]oct-2-en -2-carbonsäure (=7Amino-3-formyl-3-cephem-4-carbonsäure)
2. 64 g Hydrochlorid des 6-Amino-5a, 6-dihydro-3-hydroxy-3H, 7H-aceto[2, 1-b]furo[3, 4-d][1, 3]thiazin-1, 7- (4H) -dions (Hydroxylacton des Hydrochlorids der 7-Amino-3-formyl-3-cephem-4-carbonsäure) werden in 50 ml Methanol gelöst. Zu dieser Lösung wird dann unter Rühren und Eiskühlen eine Losung von 0. 78 g Pyridin in 10 ml Methanol zugetropft. Das ausgefallene Produkt wird unter Stickstoff und Feuchtigkeitsausschluss abfiltriert, mit etwas Methanol gewaschen und dann über Sicapent im Vakuum bei Raumtemperatur getrocknet. Man erhält die Titelverbindung In Form eines hellbraunen Pulvers.
IR (KBr) : 1799 cm-1 (-Lactam), 1672 cm-1 (CHO). 1606 und 1542 cm-1 (Carboxylat) UV-Spektrum : \max in Hz 0=302 nm.

Beispiel 2 :7-Trimethylsilylamino-3-formyl-3-cephem-4-carbonsäure-trimethylsitylester
Eine Suspension von 100 mg 7-Amino-3-formyl-3-cephem-4-carbonsäure-hydroxy-lactonhydrochiond in 1 ml Deuterochloroform wird bei Raumtemperatur mit 0. 28 ml BSA versetzt. Anschliessend wird das Reaktionsgemisch 10 Minuten bei Raumtemperatur gerührt, wobei eine klare Lösung entsteht. Die Reak- tionslösung zeigt folgendes 1 H-NMR-Spektrum.

Nach 3 Stunden Rühren bel 0'ward die Reaktionslösung in 30 ml kaltes Ethanol eingerührt. wobei die Iminoverbindung ausfallt. Nach 30 Minuten Nachrühren Im Kühlbad wird der Niederschlag abfiltnert, mit Ethanol gewaschen und getrocknet.

Acetonitrilhellgelben Lösung tropft man 1. 32 ml Phenylessigsäurechlorid. Nach der Zugabe rührt man das Reaktionsgemisch noch 30 Minuten bel O* und hydrolysiert dann mit 0. 4 ml Wasser. Die braune, trübe Lösung wird klarfiltriert und das Dichlormethan im Vakuum abgezogen.
Der Rückstand wird mit 20 ml Acetonitril verdünnt und dann unter Rühren mit 1. 7 g Natnum-2-ethylhexanoat versetzt. Man rührt noch 10 Minuten bei Raumtemperatur und filtriert dann das ausgefallene Produkt. Nach Trocknen im Vakuum erhält man die Titelverbindung.

In Chem. Pharm. Bull. Vol. 28, pages 1339 ff, 1980, a process for the preparation of 7-acylamino-3-formyl-3-cephem-4-carboxylic acid hydroxy-lactols, starting from the corresponding lactone, is followed by bromination described by halogen-hydroxy exchange. The conditions for avoiding bromination in the thiazine nucleus must be carefully avoided.

The aldehyde function of the connection of the forms! i (Ri = silyl protective group) is amenable to derivation with common nitrogen-containing aldehyde reagents, for example amines, hydroxylamines, hydrazines or semicarbazides. Example 2: 7-Trimethylsilylamino-3-formyl-3-cephem-4-carboxylic acid trimethylsityl ester
A suspension of 100 mg of 7-amino-3-formyl-3-cephem-4-carboxylic acid-hydroxy-lactone hydrochione in 1 ml of deuterochloroform is mixed with 0.28 ml of BSA at room temperature.
