Prostaglandinendoperoxyde (PGG2 und PGH2) werden aus Arachidonsäure durch ein membrangebundenes Cyclooxygenaseenzymsystem, von Hamberg und Samuelsson in Biochem. Biophys. Acta 326,448-461, 1974, beschrieben, erzeugt und danach in PGFa. PGE2, PGD2 oder Thromboxan A2 übergeführt. Thromboxan A2 besitzt gemeinsam mit den Prostaglandinendoperoxyden die wichtigen biologischen Eigenschaften, dass es Streifen der Kaninchenaorta kontrahiert und Blutplättchen aggregiert.
Es wurde gefunden, dass von einer Vielzahl von Säugetiergeweben stammende Mikrosomen die enzymatische Umwandlung der Prostaglandinendoperoxyde in ein Prostaglandinderivat katalysieren. Dieses Prostaglandinderivat wird im nachstehenden"Prostacyclin"bezeichnet ; es kontrahiert Streifen von Kaninchenaorta nicht, entspannt Streifen von Kaninchenbauchmesenterial- und Coronararterien, hat eine starke Antiaggregationswirkung auf Blutplättchen, ist bei Tieren ein starker Vasodilatator und besitzt andere Eigenschaften, die im nachstehenden beschrieben werden.
Von Kaninchen- oder Schweineblutgefässen, wie Venen und Arterien, und Rattenmägen stammende Mikrosomen bewirken eine etwa 80 bis 90%ige Umwandlung der Prostaglandinendoperoxyde.

die Auflösung bereits vorhandener Aggregate.
Prostacyclin kann biosynthetisch dadurch hergestellt werden, dass man PGG2 oder PGH2 mit Aortamikrosomen in einer geeigneten Pufferlösung, wie Tris-Puffer, etwa 2 min lang bei einer Temperatur von etwa 22 C inkubiert. Die Umwandlung der Prostaglandinendoperoxyde erfolgt etwa zu 85%.
Das Extrahieren von Prostacyclin erfolgt durch Zusetzen von kaltem (0 C) trockenem Diäthyl- äther zu der Inkubationsmischung. Der Zusatz des kalten Äthers stoppt die Enzymreaktion und Prostacyclin gelangt in die Ätherphase, die von der wässerigen Phase abgetrennt werden kann.
Das Abdampfen des Äthers nach Standardmethoden, wie durch Blasen von Stickstoff durch die Lösung, führt zum Prostacyclin, das als Rückstand verbleibt und anschliessend zur weiteren Prüfung in einer wässerigen Lösung wieder gelöst oder zur späteren Verwendung in wasserfreiem Aceton gelöst und bei einer Temperatur von etwa-20 C gelagert werden kann.
Prostacyclin kann auch unter Verwendung anderer Gewebe, die oben angeführt wurden, auf im wesentlichen die gleiche Weise hergestellt werden. In einer derartigen Inkubationsmischung gebildetes Prostacyclin scheint sich von den andern Produkten von PG-Endoperoxyden, so weit beschrieben wurde, zu unterscheiden. Seine biologischen Eigenschaften auf den isolierten Geweben, seine Instabilität und seine starke Antiaggregationswirksamkeit zeigen, dass Prostacyclin weder PGE2 noch PGF2 ist. Die Anwesenheit von Prostaglandin D2 -Isomerase in Homogenaten mehrerer Gewebe wurde beschrieben. Da Prostacyclin instabil und ein stärkeres Antiaggregationsmittel ist als PGD2, kann es nicht als PGD2 angesehen werden. Weiterhin ist die PGDz-Isomerase in der bei 100000 g überstehenden Flüssigkeit vorhanden, eine Fraktion, die Prostacyclin nicht aus PG-Endoperoxyden bildete.
Ausserdem benötigt PGD2-Isomerase Glutathion als Cofaktor und die Inkubationen wurden ohne Cofaktor durchgeführt. PGE2, PGFo und PGD2 waren keine Substrate für Aortamikrosomen und daher konnten 15-Keto-PGs und andere Produkte von Prostaglandinkatabolismus nicht als Prostacyclin angesehen werden. Prostacyclin ist wahrscheinlich auch kein bekanntes 15-Hydroperoxy-PG, erstens weil 15-Hydroperoxy-PGE2 auf Kaninchenaortastreifen eine kontrahierende Wirksamkeit ausübt und zweitens weil das (die) Produkt (e) der spontanen Zersetzung von Prostacyclin bei Bioprüfung sich nicht wie PGE2, PGF oder PGD2 verhält (verhalten).
Da Prostacyclin eine Antiaggregationswirksamkeit aufweist, kann es mit Thromboxan A2 oder B2 nicht identisch sein, da es sich bei diesen um aggregationsfördernde Substanzen handelt.

Die erfindungsgemäss erhaltenen Verbindungen der allgemeinen Formel (III) können als Zwischenprodukte zur Herstellung von Prostacyclin der allgemeinen Formel (I) verwendet werden.
Dabei wird von einer Verbindung der Formel (II) ausgegangen, worin Zl und/oder Z2 Wasserstoff oder eine Schutzgruppe, wie Acyl oder Trialkylsilyl (beispielsweise Trimethylsilyl) bedeuten. Der oxydative Angriff durch Jod oder Kaliumtrijodid in Anwesenheit eines Metallhydrogencarbonats an der 5, 6-Doppelbindung einer Verbindung der Formel (II) mit gleichzitiger oder nachfolgender Cyclisierung bewirkt, dass die 9-Hydroxygruppe eine Verbindung der Formel (III) bildet. Nach Behandlung mit einer geeigneten Base, wie einer organischen Base oder einem Metallalkoxyd kann eine Verbindung der Formel (III) dehydrohalogeniert werden, was zur Einführung einer 5, 6-Doppelbindung führt.

Sie sind weiterhin bei Menschen und Säugetieren zur Verminderung und Regulierung übermässiger Magensekretion verwendbar, wobei sie gastrointestinale Ulcusbildung reduzieren oder verhindern und die Heilung derartiger Geschwüre und krankhafter Veränderungen, die bereits im Magen-Darmtrakt vorhanden sind, beschleunigen.
Prostacyclin und seine Salze zeigen weiterhin auf Blutgefässe eine vasodilatatorische Wirkung und sind daher als Antihypertonika zur Behandlung von hohem Blutdruck bei Menschen und Säugetieren besonders geeignet. Plättchen können im vaskulären Endothel assimiliert oder sogar in Endothelzellen einverleibt werden. Das biochemische Zusammenwirken zwischen Plättchen und vaskulärem Endothel bei der Bildung von Prostacyclin trägt zur Wiederherstellung von vaskulärem Endothel bei, und Prostacyclin und dessen Salze besitzen eine weitere Verwendungsfähigkeit bei der Förderung der Heilung von Wunden bei Menschen und Säugetieren.
Prostacyclin und seine Salze können immer dann verwendet werden, wenn es erwünscht ist, die Blutplättchenaggregation zu hemmen, den adhäsiven Charakter von Plättchen zu vermindern und die Bildung von Thromben bei Menschen und Säugetieren zu behandeln oder zu verhüten.
Sie können auch als Additive für das Blut, Blutprodukte, Blutsubstitute und andere Flüssigkeiten verwendet werden, die bei der künstlichen ausserkörperlichen Zirkulation und Perfusion von isolierten Körperteilen, z. B. Gliedern und Organen, egal ob sie mit dem Originalkörper verbunden, losgelöst und zur Transplantation konserviert oder vorbereitet oder mit einem neuen Körper verbunden sind, verwendet werden. Während dieser Zirkulationen und Perfusionen besitzen aggregierte Plättchen die Tendenz, die Blutgefässe und Teile der Zirkulationsapparatur zu verstopfen. Eine derartige Verstopfung wird durch die Anwesenheit von Prostacyclin vermieden.
Das folgende Beispiel soll die Erfindung näher erläutern.
Beispiel : Eine gerührte Lösung von 50 mg PGFo -Methylester in 1 ml Äther wurde mit 115, 0 mg (10 Moläquivalente) Natriumhydrogencarbonat und 1 ml Wasser behandelt und dann tropfenweise während 2 h mit 0, 261 ml (0, 7 molarem) wässerigem Kaliumtrijodid. Nach Rühren über Nacht wurde die Reaktionsmischung mit Äther und wässerigem Natriumthiosulfat geschüttelt ; die ätherische Phase wurde abgetrennt, mit Wasser gewaschen, mit Magnesiumsulfat getrocknet und eingedampft, wobei 5-Jod-9-deoxy-6, 9a-epoxyprostaglandin-F -methylester als gelber Gummi erhalten wurde.
Anwendungsbeispiel 1 : Herstellung von Prostacyclin aus der erfindungsgemäss erhaltenen Ver- bindung (III)
Eine Lösung von 100 mg 5E-Jod-9-deoxy-6E :, 9a-epoxyprostaglandin-Fl -methylester in methanolischem Natriummethoxyd, hergestellt aus 46 mg Natrium und 0, 70 ml trockenem Methanol, wurde 5 h lang unter trockenem Stickstoff zur Seite gestellt, worauf das Lösungsmittel im Hochvakuum entfernt wurde. Der verbleibende amorphe Feststoff wurde mit Benzol gewaschen, in Luft über Nacht stehengelassen und dann mit 0,5 ml wässerigem n Natriumhydroxyd gerührt, wobei eine Suspension aus farblosen feinen Nadeln erhalten wurde.
Die Kristalle wurden gesammelt, mit einigen Tropfen wässerigem n Natriumhydroxyd gewaschen und an der Luft getrocknet, wobei das Natriumsalz von 9-Deoxy-6, 9a-epoxy-A'-prostaglandin-Fl. erhalten wurde. Die Hemmung der durch Arachidonsäure induzierten Aggregation von menschlichen Blutplättchen bei einer Konzentration von 0,2 ng/ml durch dieses Salz und dessen Instabilität in Wasser bei saurem PH-Wert stimmt auch mit der zugeschriebenen Konfiguration (5) -5, 6-Didehydro-9-deoxy-6, 9a-epoxyprostaglandin-F la -na- triumsalz überein.
Das hoch aufgelöste 13 C-NMR-Spektrum einer Lösung der Kristalle in Dimethylsulfoxyd-d6 zeigte die erwarteten 20 Resonanzen, deren chemische Verschiebungen mit der für Prostacyclin festgesetzten chemischen Struktur vollkommen übereinstimmten. Es wurden keine Verunreinigungs- - Peaks festgestellt.
Anwendungsbeispiel 2 : 500 mg 5-Jod-9-deoxy-6, 9a-epoxyprostaglandin-F -methylester wurden mit methanolischem NaOMe, hergestellt aus 0, 23 g (10 Äquivalente) Na und 3, 5 ml MeOH, über Nacht bei Raumtemperatur unter N2 gerührt. Der gelben Reaktionslösung wurden 2, 5 ml In wässerige NaOH zugesetzt, um die Hydrolyse der Estergruppe zu bewirken, und nach 2 h wurde das Methanol im Vakuum bei Raumtemperatur abgedampft.
