Der Ringschluss der 3-Hydrazinoderivate der allgemeinen Formel (VIIa) mit einem Orthoameisensäurealkylester mit niedrigmolekularer Alkylgruppe ergibt die Verbindungen der allgemeinen Formel (VIa), in der R, für ein Wasserstoffatom steht.
Durch Ringschluss der 3-Hydrazinoderivate der allgemeinen Formel (VIIa) mit einem niedrigmolekularen Alkancarbonsäureanhydrid, einem niedrigmolekularen Alkancarbonsäurechlorid oder einem Orthoester einer niedrigmolekularen Alkancarbonsäure erhält man die Verbindungen der allgemeinen Formel (VIb), in der R, für eine niedrigmolekulare Alkylgruppe steht. Der Ringschluss kann gegebenenfalls in Gegenwart von Katalysatoren mit Hilfe von Basen, wie Pyridin oder Trialkylaminen mit niedrigmolekularen Alkylgruppen erreicht werden. Den Ringschluss mit den Orthoameisensäurealkylestern mit niedrigmolekularen Alkylgruppen und den Orthoestern niedrigmolekularer Alkancarbonsäuren erreicht man vorzugsweise ohne Katalysatoren und ohne Lösungsmittel, wenngleich man ein inertes Lösungsmittel verwenden kann.
Der Ringschluss wird im allgemeinen dadurch erreicht, dass man die Reaktionsteilnehmer auf eine Temperatur von 50 bis 175 C erhitzt, wobei man gegebenenfalls in Gegenwart eines Lösungsmittels arbeitet.
Die Salze der erfindungsgemäss erhältlichen Verbindungen können zweckmässigerweise in einem neutralen Lösungsmittel mit Säuren, wie Schwefelsäure, Phosphorsäure, Chlorwasserstoffsäure, Brom-

salze sind in nicht-polaren, organischen Lösungsmitteln, wie Diäthyläther, Benzol, Toluol u. dgl., relativ unlöslich, in Wasser jedoch beträchtlich löslich. Im Rahmen der Erfindung sind die freien
Basen ihren nichttoxischen Säureadditionssalzen äquivalent. Die erfindungsgemäss erhältlichen neuen
Verbindungen besitzen bei nichttoxischen Dosierungen eine antihypertensive Wirkung und können daher als hypotensive Mittel verwendet werden.
Die pharmakologische Untersuchung dieser Ver- bindungen hat gezeigt, dass sie Eigenschaften besitzen, bei denen ein ausreichend grosser Sicher- heitsabstand zwischen den Dosierungen besteht, bei denen eine Verminderung des Blutdrucks erreicht wird und bei denen toxische Symptome auftreten. Die hypotensiven Eigenschaften der erfindungsge- mäss erhältlichen Verbindungen wurden bei der oralen Verabreichung an Säugern, insbesondere Warm- blüter, wie folgt untersucht : mit Hilfe von Leinwand und Gliederbändern befestigt man nichtbetäub- te, männliche Albinoratten des Stammes Wistar mit normalem Blutdruck und einem Durchschnittsge- wicht von etwa 250 g in der Rückenlage auf Ratten-Befestigungsbrettern.
Dann betäubt man die
Oberschenkelbereiche (durch subcutan Infiltration von Lidocain) und legt die linken oder rechten gemeinsamen Darmbeinarterien frei und klemmt sie proximal mit Hilfe einer Arterienklemme und distal mit einem Faden ab. Dann bewirkt man Einschnitte in der Nähe der abgebundenen Stelle und führt kurze Nylonkatheter ein, die an Ort und Stelle befestigt werden. Die andern Enden der Katheter versieht man mit nadelansatzfreien Nadeln (24 gauge), die an dickwandigen Polyäthylenröhren befestigt sind. Man verabreicht die zu untersuchenden Verbindungen auf oralem Wege über eine Magensonde. Die Verbindungen werden in einer Dosis von 100 mg/kg Körpergewicht untersucht und in Form einer 2%igen wässerigen Stärkelösung oder-suspension eingesetzt, die in einer Menge von 0, 2 ml/100 g Körpergewicht die gewünschte Dosierung ergibt.
Dann werden 4 h und 24 h nach der Verabreichung der Verbindungen die durchschnittlichen Arterienblutdrücke gemessen. Ein Vergleich erfolgt gegenüber dem durchschnittlichen Kontrollblutdruck von 162 mbar, bei dem es sich um den Durchschnittswert einer Anzahl von Kontrollieren handelt, die während mehrerer Monate untersucht wurden. Die Blutdruckmessungen erfolgen mit acht Dehnungsmessgeräten, die mit einem Aufzeichnungsgerät (Beckman Dynograph Recorder) verbunden sind, das mit acht DehnungsmessgerätVorverstärkern ausgerüstet ist, die zur Bestimmung des mittleren Arterienblutdrucks Mittelwert-bildende Schaltkreise aufweisen. Die Ergebnisse, die mit repräsentativen erfindungsgemäss herstellbaren Verbindungen erhalten worden sind, sind in der folgenden Tabelle I aufgeführt.
Die erfindungsgemäss erhältlichen Verbindungen wurden auch hinsichtlich ihrer antihypertensiven Wirkung mit Hilfe der folgenden Verfahrensweise untersucht, bei der spontan hypertensive Ratten (SHR) eingesetzt wurden : man verabreicht einer männlichen, ausgewachsenen (mit einem Alter von 16 bis 20 Wochen) spontan hypertensiven Ratte mit einem Gewicht von etwa 300 g über eine Sonde 100 mg der zu untersuchenden Verbindung pro kg, gegebenenfalls in einer 0,9%gen Natriumchloridlösung in einer Belastung von 25 mg/kg, wobei diese Verabreichung zum Zeitpunkt 0 erfolgt. Eine zweite identische Dosis verabreicht man 24 h später, worauf man den durchschnittlichen Arterienblutdruck (MABP) der nichtbetäubten Ratte nach 28 h direkt über eine Punktur der Oberschenkelarterie misst.
Die Ergebnisse dieser Untersuchung repräsentativer erfindungsgemäss er- hältlicher Verbindungen sind ebenfalls in der folgenden Tabelle I zusammengestellt.

Es zeigt sich somit,
dass die erfindungsgemäss erhältlichen Verbindungen äusserst wirksam dafür sind, den erhöhten Blutdruck von Säugern zu senken, wenn sie in Mengen von etwa
1,0 bis etwa 25,0 mg/kg Körpergewicht/Tag verabreicht werden. Eine bevorzugte Dosierung zur
Erzielung optimaler Ergebnisse erstreckt sich von etwa 5,0 mg bis etwa 15,0 mg/kg Körperge- wicht/Tag, wobei diese Dosiseinheiten in der Weise verwendet werden, dass einem Patienten mit einem Körpergewicht von etwa 70 kg/24 h etwa 0, 35 bis etwa 1,0 g des Wirkstoffs gegeben werden.
Die Dosierung kann in der Weise eingestellt werden, dass sich ein optimaler therapeutischer Effekt einstellt. Beispielsweise kann man täglich mehrere Einzeldosierungen geben oder man kann die Dosis proportional vermindern, je nachdem, wie es die therapeutische Situation erfordert. Ein wesentlicher praktischer Vorzug der Erfindung ist darin zu sehen, dass die aktiven Verbindungen auf irgendeinem geeigneten Weg verabreicht werden, beispielsweise auf oralem, intravenösem, intramuskulärem oder subcutanem Wege.
Die erfindungsgemäss erhältlichen Verbindungen haben sich ferner als äusserst wirksam zur Linderung der Angst bei Säugern dann erwiesen, wenn sie in Dosierungen von etwa 0,03 bis etwa 10,0 mg/kg Körpergewicht/Tag gegeben werden. Eine bevorzugte Dosis zur Erzielung optimaler Ergebnisse beträgt etaw 0, 1 mg bis etwa 5, 0 mg/kg Körpergewicht/Tag, wobei diese Dosiseinheiten in der Weise verwendet werden, dass einem Patienten mit einem Körpergewicht von etwa 70 kg im Verlaufe von 24 h etwa 7,0 mg bis etwa 0,35 g des Wirkstoffs gegeben werden. Diese Dosierung kann jedoch in der Weise eingestellt werden, dass ein optimaler therapeutischer Effekt erreicht wird. Beispielsweise kann man mehrere Einzeldosierungen täglich geben oder man kann die Dosis proportional in Abhängigkeit von der therapeutischen Situation vermindern.
Die erfindungsgemäss erhältlichen Verbindungen besitzen somit bei nichttoxischen Dosierungen eine Wirkung auf das zentrale Nervensystem und können daher als angstlösende Mittel benützt werden. Dies bedeutet, dass sie bei gewissen Standarduntersuchungen an Laboratoriumstieren gewisse Reaktionen hervorrufen, von denen bekannt ist, dass sie in guter Korrelation zu einer Angstlösung beim Menschen stehen. Die Verbindungen wurden pharmakologisch untersucht, wobei sich gezeigt hat, dass sie dieses Verhalten besitzen bei einem erwünscht weiten Abstand zwischen den Dosierungen, die eine angstlösende Wirkung haben und den Dosierungen, die toxische Symptome nach sich ziehen.
Die gegen die Angst wirkenden Eigenschaften der erfindungsgemäss erhältlichen Verbindungen wurden bei einer Untersuchung ermittelt, bei der die angstlösende Wirkung über den Schutz vor Krämpfen gemessen wird, die durch das Verabreichen von Pentylentetrazol verursacht werden. Man verabreicht die erfindungsgemäss erhältlichen Verbindungen auf oralem Wege in bestimmten Dosierungen in einem 2%igen Stärketrägermaterial an Gruppen von mindestens fünf Ratten. Zu dem geschätzten Zeitpunkt der stärksten Wirkung behandelt man die Ratten auf intravenösem Wege mit Pentylentetrazol in einer Dosis von 21 bis 23 mg/kg Körpergewicht. Die gegen Angstzustände gerichtete Wirkung wurde mit Hilfe der Methode von J. R.
Vogel, B. Beer und D. E. Clody ("A Simple and Reliable Conflict Procedure for Testing Anti-Anxiety Agents", Psychopharmacologia, Vol. 21 [1971] 1 bis 7) untersucht, bei der ein passives Vermeidungsverhalten ohne Gewöhnung angewandt wird. Durch eine Änderung dieser Methode wird eine Konfliktsituation bei Ratten hervorgerufen.
An Gruppen von jeweils sechs naiven Ratten (Sprague-Dawley mit einem Gewicht von 200 bis 220 g). die man zuvor während 48 h ohne Wasser und während 24 h ohne Futter gehalten hat, verabreicht man auf oralem Wege bestimmte Dosierungen der zu untersuchenden Verbindungen in Form von Suspensionen in einem 2%igen Stärketrägermaterial, das jeweils zwei Tropfen Polyäthylenglykol und Polysorbat 80 enthält, oder lediglich das Trägermaterial (für die Kontrollen).
Zum Zeitpunkt der stärksten Wirkung überführt man jede Ratte in einen Plexiglaskäfig, der mit einem Tränk-Schaltkreis ausgerüstet ist, der den aus einem Netz aus rostfreiem Stahl bestehenden Fussboden mit einem Trinkröhrchen aus rostfreiem Stahl verbindet, das in einem Loch in

die Ratte während der Periode, während der ein Schock verabreicht wird, erhalten hat, und eine Verzögerungseinrichtung von 1/2 s zwischen aufeinanderfolgenden Schocks umfasst.
Nachdem die Ratte in den Käfig eingebracht worden ist, lässt man sie die Umgebung erforschen und eine 10%ige Dextroselösung trinken, die über das Tränkröhrchen zugeführt wird. Nachdem man die Ratte während 20 s ohne Strafe hat trinken lassen, werden die Zeitschalteinrichtung und die Tränk-Schalteinrichtung in Gang gesetzt, worauf alternierend jeweils Perioden von 5 s, bei denen kein Schock verabreicht wird bzw. bei denen ein Schock verabreicht wird, durchgeführt werden. Gleichzeitig wird die Anzahl der Schocks ermittelt, denen die Ratte während der Untersuchungsdauer von 5 min ausgesetzt ist. Der Prozentsatz der Ratten, die bei jeder Dosis in 4 bis 5 min neun oder mehr Schocks hinnehmen müssen, wird als positives Ansprechen beim Berechnen der mittleren wirksamen Dosis lED 5 0 ) verwendet.
Die Ergebnisse dieser Untersuchung repräsentativer erfindungsgemäss erhältlicher Verbindungen sind in der folgenden Tabelle III im Vergleich zu bekannten Substanzen des Standes der Technik wiedergegeben.

Arzneimittel mit den erfindungsgemäss erhältlichen Wirkstoffen, die die erwünschte Klarheit, Stabilität und Eignung für die parenterale Verabreichung besitzen, erhält man durch Auflösen von 0, 10 Gew.-% bis 10, 0 Gew.-% des Wirkstoffs in einem Trägermaterial, das einen mehrwertigen aliphatischen Alkohol oder mehrere solche Alkohole in Mischung umfasst. Besonders, geeignet sind Glycerin, Propylenglykol und Polyäthylenglykole.
Die Polyäthylenglykole umfassen eine Mischung aus nichtflüchtigen, normalerweise flüssigen Polyäthylenglykolen, die sowohl in Wasser als auch in organischen Flüssigkeiten löslich sind und Molekulargewichte von etwa 200 bis etwa 1500 besitzen. Obwohl die Menge, in der der Wirkstoff in dem genannten Trägermaterial gelöst ist, von 0, 10 bis 10, 0 Gew.-% variieren kann, ist es bevorzugt, dass die verwendete Wirkstoffmenge etwa 3, 0 bis etwa 9, 0 Gew.-% beträgt. Obwohl man verschiedene Mischungen der oben erwähnten nichtflüchtigen Polyäthylenglykole anwenden kann, verwendet man vorzugsweise eine Mischung von Poly- äthylenglykolen mit einem durchschnittlichen Molekulargewicht von etwa 200 bis etwa 400.
Neben dem Wirkstoff können die parenteral zu verabreichenden Lösungen verschiedene Konservierungsmittel enthalten, die eine Verunreinigung durch Bakterien oder Pilze verhindern. Die für diesen Zweck verwendeten Konservierungsmittel sind beispielsweise Myristyl- Y-picolinium-chlorid, Benzalkoniumchlorid, Phenäthylalkohol, Chlorpheny1- a -glycerin äther, Methyl-paraben, Propyl-paraben und Thimerosal. Aus praktischen Gründen ist es ferner geeignet, Antioxydantien zu verwenden.
Geeignete Antioxydantien sind beispielsweise Natriumbisulfit, Natriummetabisulfit und Natriumformaldehyd-sulfoxylat. Im allgemeinen verwendet man das Antioxydans in Konzentrationen von etwa 0, 05 bis etwa 0, 2%.
Zum Zwecke der intramuskulären Injektion verwendet man den Wirkstoff in einer Konzentration von vorzugsweise 0, 25 bis 0,50 mg/ml des fertigen Präparats. Die erfindungsgemäss erhältlichen Verbindungen können auch nach dem Verdünnen mit Wasser oder Verdünnungsmitteln, die üblicher-

weise bei der intravenösen Verabreichung von Wirkstoffen verwendet werden, wie isotonische Glucose- lösungen, in entsprechend geeigneten Mengen eingesetzt werden. Bei der intravenösen Verwendung sind Anfangskonzentrationen des Wirkstoffs von bis zu etwa 0, 05 bis 0, 25 mg/ml zufriedenstellend.
Die erfindungsgemäss erhältlichen aktiven Verbindungen können auch auf oralem Wege verabreicht werden, beispielsweise zusammen mit einem inerten Verdünnungsmittel oder mit einem assimilierbaren essbaren Trägermaterial oder man kann sie in harte oder weiche Gelatinekapseln einbringen, zu Tabletten verpressen oder direkt dem Nahrungsmittel oder dem Futter zusetzen. Für die orale therapeutische Verabreichung kann man die Wirkstoffe in entsprechende Bindemittel bzw.
Trägermaterialien einarbeiten und in Form von Tabletten, Pastillen, Kapseln, Elixieren, Suspensionen, Sirupen, Oblaten u. dgl. verwenden.
Diese Zubereitungen und Präparate sollten mindestens 0, 1% des Wirkstoffs enthalten. Die prozentualen Konzentrationen der Wirkstoffe in den Zubereitungen und Präparaten können natürlich va- riieren und können üblicherweise zwischen etwa 2 und etwa 60% des Gewichts der Dosisform liegen.
Die Menge des Wirkstoffs in diesen therapeutisch nützlichen Zubereitungen ist derart, dass eine geeignete Dosierung letztendlich erreicht wird. Vorzugsweise werden die Zubereitungen oder Präparate mit erfindungsgemäss erhältlichen Wirkstoffen derart hergestellt, dass eine oral zu verabreichende Dosiseinheit zwischen etwa 0, 1 und 5, 0 mg des Wirkstoffs enthält.
Die Tabletten, Pastillen, Pillen, Kapseln u. dgl. können weiterhin folgendes enthalten : ein Bindemittel, wie Tragantgummi, Akaziagummi, Maisstärke oder Gelatine ; Trägermaterialien, wie Dicalciumphosphat ; ein den Tablettenzerfall begünstigendes Mittel, wie Maisstärke, Kartoffelstärke, Alginsäure u. dgl. ; ein Schmiermittel, wie Magnesiumstearat ; und einen Süssstoff, wie Saccharose, Lactose oder Saccharin sowie einen Aromastoff, wie Pfefferminzöl, Wintergrünöl oder Kirsoharoma.
Wenn die Dosiseinheitsform eine Kapsel darstellt, kann sie neben den Materialien der obigen Art ein flüssiges Trägermaterial, wie ein fettes Öl, enthalten. Es können verschiedene andere Materialien als Beschichtungsmaterialien vorliegen oder in anderer Weise die physikalische Form der Dosiseinheit modifizieren. Beispielsweise können die Tabletten, Pillen oder Kapseln mit Schellack, Zucker oder beidem beschichtet sein. Die Sirupe oder Elixiere können den Wirkstoff, Saccharose als Süssstoff, Methyl-paraben und Propyl-paraben als Konservierungsmittel, einen Farbstoff und einen Aromastoff, wie Kirscharoma oder Orangenaroma, enthalten. Natürlich sollte das Material zur Herstellung irgendwelcher Dosiseinheitsformen pharmazeutisch rein und im wesentlichen in den verwendeten Mengen nicht toxisch sein.
