Die Erfindung betrifft ein Verfahren zur Herstellung neuer Alkylthiocyclopentanderivate der allgemeinen Formel

R2 = 0, (H, OH) oder (H, Acyloxy mit bis zu 4 C-Atomen),

R4 gegebenenfalls durch F, OH, Acyloxy mit bis zu 4 C-Atomen, Phenyl oder p-Tolyl substituiertes

A gegebenenfalls ein-oder mehrfach durch F substituiertes Alkylen mit bis zu 8 C-Atomen bedeuten, und die durch-gekennzeichnete Bindung a- oder ss-Konfiguration anzeigt.
Es wurde gefunden, dass die Verbindungen der Formel (I) bei guter Verträglichkeit neben einer blutdrucksenkenden Wirkung auch prostaglandinartige und antiprostaglandinartige Wirkungen besitzen. So besitzen sie vasodilatorische, antiphlogistische, diuretische, bronchienentkrampfende, die Magensaftsekretion, die Thrombozyten-Aggregation, den Lipidabbau und die Noradrenalin Freisetzung hemmende, sowie Nasenschleimhaut abschwellende Eigenschaften. Ausserdem beeinflussen sie die Funktion des corpus luteum, den Eitransport durch den Eileiter, die Nidation und die Fertilität des Mannes. Die Verbindungen der Formel (I) können daher als Arzneimittel und auch als Zwischenprodukte zur Herstellung anderer Arzneimittel verwendet werden.
Die Verbindungen der Formel (I) enthalten mindestens zwei asymmetrische C-Atome im Fünfring. Ist R2 ungleich 0 oder R3 ungleich H, so ist ein drittes C-Atom im Ring asymmetrisch ; ist R2 ungleich 0 und R3 ungleich H, so sind im Ring 4 Asymmetriezentren vorhanden. Handelt es sich bei A bzw. R4 um substituiertes Alkylen bzw. Alkyl, so können in den beiden Seitenketten weitere Asymmetriezentren auftreten.
Die Verbindungen der Formel (I) können daher in einer Vielzahl stereoisomerer Formen auftreten und liegen in der Regel als Gemische von Racematen vor.
Die Verfahrensprodukte umfassen auch optisch aktive Verbindungen der Formel (I) sowie deren Racemate. Besonders bevorzugt sind die optisch aktiven Stereoisomeren der Formeln (Ia) bis (Ih), ihre optischen Antipoden und ihre Racemate :

Solche Verbindungen können wie folgt hergestellt werden : Nitrile der Formel (II) (Ri' = CN) erhält man z. B. nach an sich bekannter Methode aus 2-Oxocyclopentylcarbonsäureäthylester durch Umsetzen mit M-Jod- - alkylnitrilen, vorzugsweise in einem Losungsmittel, in Gegenwart einer Base, Umsetzen des Reaktionsproduktes mit Br, Behandeln mit H SO und Anlagerung einer Verbindung der Formel
W-S-R4, (IV) worin
W H oder ein Äquivalent eines Alkali-oder Erdalkalimetallatoms bedeutet, und
R4 die oben angegebene Bedeutung hat.
Amide der Formel (II) (Ri' = CONR5R6) erhält man z. B. nach an sich bekannter Methode aus 2-Oxocy- clopentylcarbonsäureäthylester durch Umsetzen mit #-Jod-alkancarbonsäureamiden, vorzugsweise in einem Lösungsmittel, in Gegenwart einer Base, Umsetzen des Reaktionsproduktes mit Br, Behandeln mit H2S04 und Anlagerung einer Verbindung der Formel (IV).
Verbindungen der Formel (II), in denen der Rest RI 1 eine funktionelle abgewandelte Carboxylgruppe ist,

können nach in der Literatur beschriebenen Methoden zu den Verbindungen der allgemeinen Formel (I) hy- drolysiert werden. Die Hydrolyse wird in saurem oder gegebenenfalls alkalischem Medium bei Temperaturen zwischen-20 und 300 C, vorzugsweise bei der Siedetemperatur des gewählten Lösungsmittels, durchgeführt.
Als saure Katalysatoren eignen sich beispielsweise anorganische Säuren, wie Salz-, Schwefel-, Phos- phor-oder Bromwasserstoffsäure ; als basische Katalysatoren verwendet man zweckmässig Metallhydroxyde oder basische Salze, wie Natrium-, Kalium- oder Calciumhydroxyd, Natrium- oder Kaliumcarbonat.
Als Lösungsmittel wählt man vorzugsweise Wasser ; niedere Alkohole wie Äthanol, Methanol ; Äther wie Dioxan, Tetrahydrofuran ; Amide wie Dimethylformamid ; Nitrile wie Acetonitril ; Sulfone wie Tetramethylen- sulfon ; oder deren Gemische, besonders die Wasser enthaltenden Gemische.
Man kann aber die Säureabkömmlinge z. B. auch in Äther oder Benzol unter Zusatz von starken Basen wie Kaliumcarbonat oder ohne Losungsmittel durch Verschmelzen mit Alkalien wie KOH und/oder NaOH oder Erdalkalien zu Verbindungen der Formel (I) verseifen.
Insbesondere werden die Verbindungen der Formel (I) durch Solvolyse von Nitrilen der Formel (II) (RI = CN) erhalten. Diese Solvolyse kann als Hydrolyse in an sich bekannter Weise in wässerigen Medien durchgeführt werden und führt dann zu den Säuren der Formel (I), oder analog in der Literatur beschriebenen Methoden unter Ausschluss von Wasser und in Gegenwart eines Alkohols ausgeführt werden und ergibt dann Ester der Formel (I). DieReaktion wird in Gegenwart eines sauren oder basischen Katalysators durchgeführt.
Die Hydrolyse der Nitrile (IIj R5 = CN) zu den Säuren (Ij R1 = H) erfolgt vorzugsweise in Gegenwart eines sauren Katalysators, insbesondere Schwefelsäure oder Salzsäure. Die Reaktion erläuft zwischen-10 und 1000C. Vorzugsweise lässt man zunächst das Reaktionsgemisch bis zu 6 Tagen bei Raumtemperatur stehen und kocht abschliessend in der Regel bis zu 6 h.
Verwendet man einen basischen Katalysator, so kommt vorzugsweise ein Alkalimetall- oder Erdalkalimetallhydroxyd wie NaOH, KOH oder Ba (OH) 2 oder auch ein basisches Salz wie KCO in Frage. Man arbeitet in HO oder H O/Alkohol-Gemischen bei erhöhter Temperatur, vorzugsweise beimSiedepunktdesReaktionsgemisches, gegebenenfalls auch im Autoklaven bei erhöhtem Druck.
DieSolvolyse vonAmiden der Formel (II) (R'= CONR ) erfolgt unter den bei den Nitrilen angegebenen Reaktionsbedingungen.
Auch Ester der Formel (I) können so verseift oder gegebenenfalls umgeestert werden.

Man arbeitet nach den in der Literatur beschriebenen Umesterungsmethoden, insbesondere in Gegenwart basischer oder saurer Katalysatoren, z. B. Natriumäthylat oder Schwefelsäure, bei Temperaturen zwischen etwa 00C und Siedetemperatur. Vorzugsweise arbeitet man so, dass nach Einstellung des Gleichgewichtes ein Reaktionspartner dem Gleichgewicht durch Destillation entzogen wird.
Die Verbindungen der Formel (I) haben, wie bereits erläutert, im allgemeinen mehrere Asymmetriezentren, stets aber wenigstens zwei. Sie werden daher meist als Gemische verschiedener stereoisomere Formen erhalten, d. h. als Racemate oder in der Regel als Gemische von Racematen. Da verschiedene Racemate zueinander diastereomer sind, können sie auf Grund ihrer unterschiedlichen physikalischen Eigenschaften aus ihren Gemischen isoliert und rein erhalten werden, beispielsweise durch Umkristallisieren aus geeigneten Lösungsmitteln (wobei insbesondere an Stelle der Verbindungen selbst, gut kristallisierende Derivate eingesetzt werden können), durch destillative Trennung, insbesondere aber mit Hilfe chromatographscher Methoden, wobei sowohl adsorptionschromatographische oder verteilungschromatographische Methoden als auch Mischform in Frage kommen.
Die Racemate können nach einer Vielzahl bekannter Methoden, wie sie in der Literatur angegeben sind, in ihre optischen Antipoden getrennt, werden. Die Methode der chemischen Trennung ist zu bevorzugen. Danach werden aus dem racemischen Gemisch durch Umsetzung mit einem optisch aktiven Hilfsmittel Diastereomeren gebildet. So kann man gegebenenfalls eine optisch aktive Base mit der Carboxylgruppe einer Verbindung der Formel (I) (R1 = H) umsetzen.
Zum Beispiel kann man diastereomere Salze der Verbindungen der Formel (I) mit optisch aktiven Ami-

nen, wie Chinin, Chinchonidin, Brucin, Cinchonin, Hydroxyhydrindamin, Morphin, 1-Phenyläthylamln, 1-Na- phthyläthylamin, Phenyloxynaphthylmethylamin, Chinidin, Strychnin, basischen Aminosäuren, wie Lysin,
Arginin oder den Aminosäureestern bilden.
In ähnlicher Weise lassen sich Ester-Diastereomeren durch Veresterung von Verbindungen der For- i mel (I) (R =H) mit optisch aktiven Alkoholen, wie Borneol, Menthol, Octanol-2, herstellen. Der UnterschIed in der Löslichkeit der anfallenden diastereomeren Salze bzw. Ester erlaubt die selektive Kristallisation der einen Form und die Regeneration der jeweiligen optisch aktiven Verbindungen aus dem Gemisch.
Die Hydroxysäuren und Hydroxyester der Formel (I) (R2 = [H, OH], R3 =H, R - [H, OH], R -OH,
R2 = 0, R3 - OH) können ferner durch Veresterung mit einer optisch aktiven Säure wie (+)-und (-)-Weinsäu-

phensäure oder (+) - und (-) -Milchsäure in geeignete diastereomere Ester umgewandelt werden, die sich auf
Grund ihrer unterschiedlichen Löslichkeiten trennen lassen. Die optisch aktiven Verbindungen der Formel (I) werden dann jeweils durch Verseifen des reinen Diastereomeren gewonnen.
Man kann aber auch zunächst mit Phthalsäure- oder Bernsteinsäureanhydrid die sauren Phthalsäure- bzw. Bernsteinsäureester darstellen und die so gewonnenen zweibasigen Säuren bzw. deren Monoester mit einer der oben angegebenen, optisch aktiven Basen in die diastereomeren Salze überführen und aus diesen die reinen Enantiomeren gewinnen.
Aus den Ketosäuren und den Ketoestern der Formel (I) (R2 = 0) können durch Umsetzung mit optisch ak- tiven Keton-Reagentlen, wie z. B. Menthylhydrazin oder Menthylsemicarbazid, die entsprechenden diastereo- meren Hydrazone bzw. Semicarbazone dargestellt werden, aus denen man ebenfalls die reinen Entaniomeren gewinnen kann.
Besonders vorteilhaft ist die Trennung der Racemate bzw. Racemat-Gemische mit Hilfe von chromato- graphischen Methoden. Man kann entweder optisch aktive Trägermaterialien, wie z. B. Weinsäure, Stärke,
Rohrzucker, Cellulose oder acetylierteCellulose und optisch inaktive und/oder optisch aktive Laufmittel zur
Trennung in die reinen Enantiomeren verwenden oder ein optisch inaktives Trägermaterial, wie z. B. Kiesel- gel oder Aluminiumoxyd in Kombination mit einem optisch aktiven Laufmittel.
Die optischen Antipoden können auch biochemisch unter Anwendung selektiver, enzymatischer Reaktio- nen getrennt werden. So kann man die racemischen Säuren der Formel (I) (R1 = H) einer Oxydase oder ge- gebenenfalls Decarboxylase aussetzen, die durch Oxydation oder Decarboxylierung einer Formzerstört. wäh- rend die andere Form unverändert bleibt.
Möglich ist ferner die Verwendung einer Hydrolase bei einem funktionellen Säurederivat des racemi- schen Gemisches zur bevorzugten Bildung einer optisch aktiven Form. So kann man Ester oder Amide der
Säuren der Formel (I) (rut = H) der Einwirkung einer Hydrolase aussetzen, welche das eine Enantiomere selektiv verseift und das andere unverändert lässt.
Weiterhin ist es natürlich möglich, optisch aktive Verbindungen nach den beschriebenen Methoden zu er- halten, indem man Ausgangsstoffe verwendet, die bereits optisch aktiv sind.
Wird nun eine Verbindung der Formel (I) oder eine Vorstufe mit bestimmten sterischen Verhältnissen an einem oder mehreren C-Atomen in eine Verbindung der Formel (I) bzw. eine andere Verbindung der For- mel (I) unter Ausbildung eines neuen Asymmetriezentrums umgewandelt, so lässt sich durch geeignete Reak- tionsführung erreichen, dass dieses C-Atom überwiegend eine bestimmte, vorzugsweise die gewünschte Kon- figuration aufweist.
Verbindungen der Formel (I) (rut = H) können durch Umsetzung mit einer Base in eines ihrer physiolo- gisch unbedenklichen Metall- bzw. Ammoniumsalze übergeführt werden. Als Salze kommen insbesondere die
Natrium-, Kalium-, Magnesium-, Calcium- und Ammoniumsalze in Betracht, ferner substituierte Ammoniumsalze, wie z. B. die Dimethyl- und Diäthylammonium-, Monoäthanol-, Diäthanol- und Triäthanolammonium-, Cyclohexylammonium-, Dicyclohexylammonium- und Dibenzyläthylendiammonium-Salze.
Umgekehrt können Verbindungen der Formel (I) aus ihren Metall- und Ammoniumsalzen durch Behand- lung mit Säuren, vor allem Mineralsäuren wie Salz-oder Schwefelsäure, in Freiheit gesetzt werden.
Die neuen Verbindungen können im Gemisch mit festen, flüssigen und/oder halbflüssigen Arzneimittelträgern als Arzneimittel in der Human- oder Veterinärmedizin verwendet werden. Als Trägersubstanzen kommen solche organischen oder anorganischen Stoffe in Frage, die für die parenterale, enterale oder topikale Applikation geeignet sind und die mit den neuen Verbindungen nicht in Reaktion treten, wie beispielsweise Wasser, pflanzliche Öle, Benzylalkohole, Polyäthylenglykole, Gelatine, Lactose, Stärke, Magnesium- stearat, Talk, Vaseline, Cholesterin.
Zur parenteralen Applikation dienen insbesondere Lösungen, vorzugsweise ölige oder wässerige Lösungen, sowie Suspension, Emulsionen oder Implantate. Für die enterale Applikation eignen sich Tabletten, Dragees, Sirupe, Säfte oder Suppositorien, für die topikale Anwendung Salben, Cremes oder Puder. Die angegebenen Zubereitungen können gegebenenfalls sterilisiert oder mit Hilfsstoffen, wie Gleit-, Konservierungs-,

Stabilisierungs- oder Netzmitteln, Emulgatoren, Salzen zur Beeinflussung des osmotischen Druckes, Puffer- substanzen, Farb-, Geschmacks-und/oder Aromastoffen und/oder andern Wirkstoffen, beispielsweise Vi- taminen, versetzt werden.
Die Substanzen werden vorzugsweise in einer Dosierung von 0, 1 bis 2000 mg pro Dosierungseinheit veri abreicht.
Die im folgenden angegebenen IR-Spektren wurden mit Perkin Eimer 6 als Film aufgenommen, und die
NMR-Spektren mit Varian HA 100 oder A 60 in CDCl3 gegen Tetramethylsilan als inneren Standard gemes- sen.
