Bei s pie I 2 : 2-[2- (2, 6- Dichlorphenyl) -acetylimino ]-hexahydropyrimidin
Th.).

The invention relates to a process for the preparation of novel heterocyclic compounds of the general formula RH # CH-CO-N = C # (I), where R1 is hydrogen, chlorine or methyl and R2 is chlorine or methyl, and n is 1 or 2, which is representative of phenacetylguanidines , and their acid addition salts.
According to the invention, compounds of the general formula (I) are obtained by adding compounds of the general formula
EMI1.1
where R is as defined above and R is a reactive group which can be split off with a hydrogen of a diamine, namely a free, esterified or etherified hydroxyl or mercapto group, for example a lower alkoxy or alkyl mercapto group or an aralkylthio group optionally activated by substituents such as the nitro group , with diamines of the general formula

<Desc / Clms Page number 2>
EMI2.1
orwherein R and Ri have the above meaning, with compounds of the general formulas
EMI2.2
wherein Alk in each case denotes a lower alkyl group and Aralk denotes an optionally substituted aralkyl group,
z. B. represents the p-nitrobenzyl group, implemented.
EMI2.3
those of a lower alkoxy or alkyl mercapto group or an optionally substituted aralkylthio group can be prepared in an analogous manner.
The compounds of general formula (I) prepared according to the invention and their acid addition salts are distinguished by extraordinarily favorable pharmacodynamic properties, but in particular by an antihypertensive effect. They can therefore be used for the treatment of hypertension in general and in particular for the treatment of essential and renal hypertension.
The antihypertensive effect of the products obtainable according to the invention is expressed in animal experiments with oral or intravenous administration in a lowering of the blood pressure in experimentally hypertensive rats (cf. F. Gross, P. Loustalot and F. Sulser, Archiv für exper. Pathalogie und Pharmakologie 229, [ 1956] pp. 381 to 388).
The 2- [2- (2,6-dichlorophenyl) -acetylimino] - - imidazolidine, 2- [2- (2,6-dichlorophenyl) -acetylimino] -hexahydropyrimidine and their acid addition salts have proven to be particularly effective.
In order to achieve beneficial therapeutic success, it is advisable to administer the compounds of the general formula (I) in a daily amount of 5 to 20 mg. A favorable single dose should contain 5 to 10 mg of compounds of the general formula (I).
The compounds of general formula (I) prepared according to the invention and their acid addition salts can be used as medicaments alone or in appropriate medicament forms for oral administration, for example in the form of tablets or dragees, or for parenteral administration, for example in the form of injection solutions.
A preferably tablet composition consists of 10 mg 2- [2- (2, 6-dichlorophenyl) acetylimino] - imidazolidine, 108 mg lactose, 2 mg paraffin oil, 1 mg gelatin, 13 mg corn starch and 6 mg talc. This

Tablets should be scored with a score.
In the following examples, which explain the implementation of the process but are not intended to restrict the invention in any way, the temperature data are not corrected.
Example 1: 2- [2- (2, 6-dichlorophenyl) acetylimino] imidazolidine 2.8 g of S-methyl-N- (2, 6-dichlorophenylacetyl) isothiourea are added to 0.6 g of ethylenediamine, one slight warming occurs and methyl mercaptan and NHg escape. The solid obtained after cooling the reaction mixture is separated off, stirred with 20 ml of ethylenediamine, filtered off, washed with water and taken up in 100 ml of dilute hydrochloric acid. The hydrochloric acid suspension is filtered to remove the starting material and the filtrate is made alkaline with dilute sodium hydroxide solution.
The deposited base is separated off, washed and treated with isopropanol, giving 2- [2- (2,6-dichlorophenyl) acetylimino] imidazolidine in the form of a white powder with a melting point of 234 to 2370 ° C. (yield 60d. Th .).
The raw material used in this example is obtained as follows:
A solution of 5.6 g of 2,6-dichlorophenylacetic acid chloride in 10 ml of acetone is slowly added to a solution of 4.7 g of S-methylisothiourea in 120 ml of acetone. The precipitated S-methylisothiouronium chloride is filtered off and the filtrate is evaporated. The residue is crystallized twice from diisopropyl ether, S-methyl-N- (2,6-dichlorophenylacetyl) isothiourea in the form of white rhombuses with a melting point.
125 to 12 T'c received.
At pie I 2: 2- [2- (2,6-dichlorophenyl) -acetylimino] -hexahydropyrimidine
Using the method described in Example 1 and replacing ethylenediamine by 1,3-propylenediamine, 2- [2- (2,6-dichlorophenyl) acetylimino] hexahydropyrimidine is obtained, which after washing with diethyl ether from methanol / diethyl ether Contains 0.15 mol of hydrochloric acid, is crystallized. The 2- [2- (2,6-dichlorophenyl) acetylimino] hexahydropyrimidine hydrochloride obtained in this way melts at 285 to 287 ° C. (yield 5510 of theory).
