Verfahren zur Herstellung neuer N-Polymethylderivate von Antibiotica
Die Erfindung betrifft ein Verfahren zur Herstellung neuer N-Polymethylderivate von Neomycinen und Kanamycinen sowie von pharmazeutisch verwendbaren Additionssalzen und quarternären Ammo- niumverbindungen dieser neuen N-Polymethylderivate.
In der Beschreibung wird für Neomycin A der Ausdruck Neamin verwendet.
Das erfindungsgemässe Verfahren besteht darin, dass man derartige Antibiotica durch Umsetzung mit Formaldehyd in Gegenwart von Ameisensäure einer reduktiven Alkylierung unterwirft und gegebenenfalls die erhaltenen Produkte in quarternäreAmmoniumverbindungen oder mit physiologisch verträg- lichen Säuren in Additionssalze überführt.
Die reduktive Polymethylierung kann entweder alle oder zumindest eine der in den Ausgangsantibiotica vorhandenen Aminogruppen angreifen. In ähnlicher Weise kann die reduktive Polymethylierung jede primäre Aminogruppe in eine sekundäre oder tertiäreAminogruppe überführen. DieseModifikationen stellen verschiedene Ausführungsformen der Erfindung dar.
Die erfindungsgemäss erhältlichen N-polymethylierten Verbindungen lassen sich leicht in die entsprechenden quarternären Ammoniumverbindungen unter Anwendung an sich bekannter Methoden überführen. Zu diesem Zweck wird die Reaktion vorzugsweise bei Zimmertemperatur in einem inerten Lösungsmittel, wie beispielsweise Acetonitril, durchgeführt. Die Herstellung der quarternären Ammoniumverbindungen gehört zum Bereich der Erfindung.
Zu Beispielen pharmazeutisch verwendbarer quarternärer Ammoniumverbindungen gehören Chlor-, Brom oder Jodmethylate oder -äthylate und Chlor- und Brombenzylate oder-allylate u. dgl.
Zu Beispielen pharmazeutisch verwendbarer Additionssalze gehören die Hydrochloride, Sulfate, Phosphate, Maleinate, Fumarate, Succinate, Tartrate, Oxalate, Citrate, Methansulfonate, Äthansulfonate u. dgl.
Es wurde überraschenderweise gefunden, dass die erfindungsgemäss darstellbaren neuen chemischen VerbindungenfreivonderToxizität der Ausgangsmaterialien sind und dass sie entweder von der antibiotischen Wirksamkeit der Ausgangsmaterialien vollständig frei sind oder eine stark herabgesetzte antibiotische Wirksamkeit im Vergleich zu den Ausgangsmaterialien zeigen. Ausserdem besitzen alle erfindungsgemäss erhältlichen neuen Verbindungen eine stark senkende Wirkung auf den Cholesterinspiegel im Serum.
Die erfindungsgemäss herstellbaren Produkte wurden Menschen und Tieren oral in Dosierungseinheitsform mehrere Wochen lang verabreicht. Während der Versuche wurde eine beträchtliche Herabsetzung der Cholesterinspiegel ohne irgendeine Spur von Toxizität erzielt.
Die minimale tägliche Dosis beträgt etwa 1 g (als Gewicht der Base) für den Menschen, doch können auch viel höhere Dosen verabreicht werden. Bevorzugte Dosen liegen zwischen 2 und 20 g (als Gewicht der Base) je Tag.

Die nach dem erfindungsgemässen Verfahren erhältlichen Verbindungen können oral in jeder für diese Verabreichung bekannten pharmazeutischen Form gegeben werden. Zu Beispielen pharmazeuti- scher Formen gehören Pulver, Kapseln, Tabletten, Sirupe, Präparatformen mit gesteuerter Freisetzung u. dgl., wobei die Kapselform in der Praxis bevorzugt wird.
Die folgenden Beispiele erläutern die Erfindung, ohne sie zu beschränken.
Beispiel l ; 7, l g Neamin werden langsam zu einem Gemisch von 1 g Ameisensäure (98%ig) und 2, 5 ml Wasser zugegeben. Dann wird ein Gemisch von 15 g 35%iger Formaldehydlösung und 19 g Ameisensäure (98% zig) zugegeben, und das Gemisch wird 3 h unter Rückfluss erhitzt. Anschliessend werden 2 ml Salzsäure zugesetzt und das Medium wird unter vermindertem Druck auf die Hälfte seines Volumens eingedampft. Weitere 4 ml Salzsäure in 5 ml Wasser werden zugegeben und das Medium wird unter vermindertem Druck bis zur Bildung eines Gels eingeengt. Dieser Rückstand wird in einer Lösung von 6 ml Salzsäure in 25 ml Wasser aufgenommen und die erhaltene Lösung zur Trockne eingedampft.

Wird das N-octamethylneamin durch aufsteigende Papierchromatographie an Schleicher & Sehüll- Papier Nr. 2043 (mit Säure gewaschenes Papier) in dem System n-Propanol/Essigsäure/Pyridin/Wasser (9 :1:1:10) und anschliessendem Nachweis mit Hinhydrin geprüft, so wird ein blauvioletter Fleck mit Rf=0,60 (¯ 0,03) erhalten. Unter den gleichen Bedingungen gibt das als Ausgangsproduktverwendete Neamin einen violettbraunen Fleck mit Rf = 0, 42 ( 0, 03).
Das -Tetraaetyl-N-octamethylneamin wird wie folgt erhalten :

und dann auf Raumtemperatur abgekühlt. Nun werden 200 ems Wasser zugesetzt, die Lösung auf eineSäule aus 400 ems Dowex 2 X 8 Harz (OH--Form) gegossen, die Elution gesammelt und das Harzbett mit 3 l Wasser gewaschen.
Die gesammelten Fraktionen-Elution und Waschflüssigkeit - werden bei vermindertem Druck zur Trockne eingedampft.
Der Rückstand wird in 35 cn absolutem Äthanol aufgenommen und l, 3 l trockener Äther langsam zugesetzt. Nach etwa 5 h Stehen wird der Niederschlag abfiltriert und das Filtrat bei Unterdruck eingedampft. Der Rückstand wird mit Petroläther (Siedebereich 40 bis 60 C) verrieben, der anschliessend durch Filtration wieder abgetrennt wird.
Nach dem Trocknen wird das N-Dodeca-methylneomycin B erhalten, das sich bei 140 bis 1450C zersetzt, [α]D=+61 (c=1 in n/10 H2SO ).
Aus dem Niederschlag, der nach 5stündigem Stehen erhalten wurde, kann durch Lösen in 15 cm3 absolutem Methanol und Fällung mit trockenem Äther eine zweite Fraktion N-Dodeca-methylneomy- cin B gewonnen werden.

Die wässerige Lösung wird dann bei vermindertem Druck zur Trockne eingedampft und der Rückstand in 30 cms absolutem Äthanol gelöst. Bei Ätherzusatz entsteht ein Niederschlag, der abfiltriert und ge-

trocknet wird. Auf diese Weise werden 1, 5 gN-Dodeca-methylneomycin B-polybenzochlorid erhalten, das sich bei 168 Dis 172 C zersetzt, [ a] D = + 360 (c = 1 in Wasser).
Bei Prüfung des N-Dodeca-methylneomycin B-polybenzochloridsdurch aufsteigende Papierchromatographie an Schleicher & Schüll-Papier Nr. 2043 (mit Säure gewaschenes Papier) wird in dem System n-Propanol/Essigsäure/Pyridin/Wasser (9 :1:1:10) und anschliessendem Nachweis mit DragendorffReagenz ein orangegelber Fleck mit Rf = 0, 87 (0, 02) erhalten.
B e i s p i e l 4: 9,8 g des gemäss Beispiel 2 erhaltenen N-Dodeca-methylneomycins B werden in einer Mischung aus 200 cms absolutem Äthanol und 600 cm ? Acetonitril gelöst und der Lösung bei Raumtemperatur 85 g Methyljodid zugesetzt.
DieMischung bleibt 3 Tage bei Raumtemperatur stehen und wird dann beiunterdruck auf ein Viertel ihres Anfangsvolumens konzentriert. Der während der Konzentrierung entstehende Niederschlag wird abfiltriert, mit Methylenchlorid gewaschen und getrocknet. Man erhält 13,2 g N-dodeca-methylneomycin B -polymethojodid.
Dieses Produkt wird in 200 cm3 Wasser gelöst und die Lösung auf eine Säule (Durchmesser 1, 5 cm) aufgegeben, die 50 cms Dowex 1 x 2 (stark basischer Polystyrolanionenaustauscher mit quarternären Ammoniumgruppen mit Vernetzungsgrad 2 in Cl'-Form) enthält und das Harzbett mit 200 cm ? Wasser gewaschen. Die gesammelten Elutionen werden zu einem schweren Sirup eingedickt.
Dieser Sirup wird in 100 cm absolutem Äthanol gelöst, das dann verdampft wird. Der Rückstand wird mit trockenem Äther bedeckt und verrieben, wobei eine Suspension entsteht. Nach dem Abfiltrieren des Niederschlages und Trocknen werden 8, 9 g N-Dodeca-methylneomycin B-polymethochlorid er-

Beispiel 7 : Nach der im Beispiel 4 beschriebenen Arbeitsweise erhält man bei Verwendung von 9, 8 g N -Dodeca-methylneomycin an Stelle der dort angegebenen 9, 8 g N -Dodeca-methylneomycin B das N-Dodeca-methylneomycin-polymethochlorid.
Beispiel 8 : In 19,2 ml Ameisensäure (98 bis 100%ig) werden 15,5 g Kanamycinbase. welche durch Leiten über ein Ionenaustauscher-harz "DOWEX 2 x 8" erhalten wurde, gelöst. Dieser Lösung werden 8, 42 g Paraformaldehyd, das 2,2 mMol je m Äquivalent primärer Aminogruppen entspricht, hinzugefügt. Das Gemisch wird während 2 1/2 h in einem Ölbad von 100 bis 105 C kräftig gerührt.
Nach dem Erkalten wird die Lösung mit 100 ml Wasser verdünnt und über eine Kolonne von 500 ml Harz "DOWEX 2 x 8" in OH- -Form (100 bis 200 Maschen) geleitet.
Die so erhaltene N-polymethylierte Kanamycinbase wird mit Wasser verdünnt, das Eluat unter vermindertem Druck zur Trockne verdampft und der Rückstand in absolutem Äthanol aufgenommen, das seinerseits dann verdampft wird. Der Rückstand wird neuerlich in wenig absolutem Äthanol aufgenommen, aus welchem die Base durch Abkühlen auskristallisiert. Konzentration durch Verdampfen liefert eine zweite Fraktion des Produktes.

The invention relates to a process for the production of new N-polymethyl derivatives of neomycins and kanamycins and of pharmaceutically usable addition salts and quaternary ammonium compounds of these new N-polymethyl derivatives.
In the description, the term neamine is used for neomycin A.
The process according to the invention consists in subjecting such antibiotics to reductive alkylation by reaction with formaldehyde in the presence of formic acid and optionally converting the products obtained into quaternary ammonium compounds or into addition salts with physiologically acceptable acids.
