Verfahren zur Herstellung von neuen Chinazolin-Derivaten Die Erfindung betrifft ein Verfahren zur Herstellung von neuen Chinazolin-Derivaten der Formel

Der in dieser Beschreibung verwendete Ausdruck"Alkyl" (allein oder in Kombinationen wie"Al- koxy") bezieht sich auf sowohl geradkettige als auch verzweigte, gesättigte Kohlenwasserstoffe mit 1 bis 7 Kohlenstoffatomen wie Methyl, Äthyl, Isopropyl u. dgl. Der Ausdruck Halogen, Halogenid usw. bezieht sich auf alle vier Halogene, d. h. Brom, Chlor, Fluor und Jod. Der Ausdruck Halogenalkyl umfasst Alkylgruppen, bei welchen ein oder mehrere Wasserstoffatome durch Halogenatome ersetzt sind.
Falls mehr als ein Halogenatom vorhanden ist, können es die gleichen oder verschiedene Halogenatome sein. Falls die Gruppen RI'Ra oder R4 Halogen oder eine halogenhaltige Gruppe bedeuten, ist Brom oder Chlor bevorzugt. Falls die Gruppe R2 ein Halogenatom ist, ist Chlor oder Fluor bevorzugt. Falls die Gruppe Rg oder Ri ein Halogenalkylrest ist, so sind ex-Monohalogen-a1kyle und o, ex-Dihalogen-alkyle wie a-Monochlormethyl, o, a-Dichlormethyl u. dgl. bevorzugt. Der Ausdruck Aminoalkyl umfasst unsubstituierte und auch mono- und disubstituierte Aminoalkylgruppen, vorzugsweise Monoalkyl- oder Dialkyl-amino-alkylgruppen.
Der Ausdruck Azacycloalkyl-alkyl umfasst gesättigte, stickstofhaltige, he- terocyclische Ringe, vorzugsweise 5-oder6-gliedrigeRinge, wiePiperidino od. dgl., welche zumKohlen- stoffatom in 2-Stellung des Chinazolin-Ringes über ein Alkylgruppe gebunden sind. Der Alkylenrest für Rs und R4 zusammengenommen umfasst geradkettige oder verzweigte Kohlenwasserstoffreste, welche zusammen mit dem Kohlenstoffatom in 2-Stellung der Formel I einen Cycloalkylrest bilden, z. B.
Cyclohexyl. Ähnlich bedeutet der Rest Aza-alkylen für R und R zusammengenommen eine Gruppe, welche

zusammen mit dem Kohlenstoffatom in 2-Stellung der Formel I einen gesättigten, stickstoffhaltigen, heterocylischenRing bildet, wie Piperidin, z. B. ein 4-Piperidino-Radikal wie die Gruppe 1-Methyl- 4-piperidino.

Eine spezielle Ausführungsform des erfindungsgemässen Verfahrens besteht darin, dass man ein
Oxim der allgemeinen Formel IV wählt, worin Rl Wasserstoff, Halogen, Nitro, Trifluormethyl, Cyan oder Alkylthio, R Wasserstoff oder Halogen und Ra Wasserstoff oder Alkyl bedeuten.
Eine andere spezielle Ausführungsform des erfindungsgemässen Verfahrens besteht darin, dass man eine Carbonyl-Verbindung der Formel V wählt, worin Ra und R. je Wasserstoff, Alkyl, Halogenalkyl,
Aza-cycloalkyl-alkyl oder zusammengenommen Alkylen, Aza-alkylen oder N-Alkyl-aza-alkylen be- deuten.
Bevorzugte Ketone der Formel V sind z. B. Aceton, Chloraceton, Dichloraceton, 1-Methyl-4 -pi- peridon, 1, 3-Dimethyl-4-piperidon, Piperidinoaceton und Cyclohexanon. Speziell bevorzugte Ketone für die später erwähnteumwandlung in ss-Oxime sind Alkylketone wie Aceton und Methyläthyl-ketonund Cycloalkanon wie Cyclohexanon. Bevorzugte Aldehyde der Formel V sind Acetaldehyd, Chloracetaldehyd, Dichloracetaldehyd.
Das erfindungsgemässe Verfahren, wobei ein Oxim der Formel IV mit einem Keton oder Aldehyd der Formel V unter Bildung eines 1, 2-Dihydro-chinazolin-3-oxyds der Formel I kondensiert wird, kann unter Verwendung des Ketons oder Aldehyds der Formel V als Reaktionsmedium durchgeführt werden. Das Verfahren kann jedoch auch in einem inerten organischen Lösungsmittel durchgeführt werden, vorzugsweise in einem solchen, in welchem das Oxim im wesentlichen löslich ist, wie z. B. einem Alkanol wie Methanol, Äthanol od. dgl., Äther, Dioxan. Tetrahydrofuran, w, w- Dimethoxy- diätbyl -Ilther, einem Kohlenwasserstoff, wie Benzol, Toluolod. dgl. Die Reaktion kann bei Raumtemperatur, erhöhten Temperaturen oder unterhalb Raumtemperatur durchgeführt werden.
Wenn man ein a- Oxim der Formel IV einsetzt, ist es zweckmässig, dem Reaktionsmedium ein Schwermetallsalz zuzusetzen. Der anionische Teil des Schwermetallsalzes ist nicht wesentlich und kann entweder von organischen oder anorganisehen Säuren, vorzugsweise den letzteren, abgeleitet sein. Es hat sich als besonders zweckmässig herausgestellt, ein Cuprisalz, z. B. Kupfersulfat, zu verwenden. Die Menge des vorhandenen Kupfersulfates ist nicht wesentlich, es wurde jedoch festgestellt, dass Cuprisalze, wie Kupfersulfat, einen günstigen Einfluss auf die Reaktion eines ex-oximes der Formel IV mit einem Keton oder Aldehyd der Formel V aus- üben.
Die Reaktion einer Verbindung der Formel V mit einem ss-Oxim der Formel IV wird zweckmä- ssigerweise in der vorstehend für das a-Oxim angegebenen Weise in Gegenwart eines Schwermetallsalzes durchgeführt oder in Gegenwart eines basischen oder sauren Katalysators. Die Menge des Katalysators ist nicht wesentlich. Der Katalysator kann ausserdem entweder anorganisch oder organisch sein ; ein geeigneter basischer Katalysator ist z. B. Pyridin od. dgl. und geeignete saure Katalysatoren sind Mire- ralsäuren, z. B. Halogenwasserstoffsäuren, wie Chlorwasserstoffsäure, Bromwasserstoffsäure, Essigsäure od. dgl.
Die erfindungsgemäss erhältlichen 1, 2-Dihydrochinazolin-3-oxyde der Formel I können durch saure Hydrolyse in h-oxime der Formel IV umgewandelt werden. Diese saure Hydrolyse wird zweckmässiger- weise bei Raumtemperatur, unterhalb Raumtemperatur oder bei erhöhten Temperaturen, in Gegenwart von Wasser durchgeführt (die Temperatur soll jedoch nicht so hoch sein, dass das 0-Oxim zerstört wird). Die Hydrolyse kann erwünschtenfalls in einem organischen Lösungsmittel, wie z. B. einem niederen Alkanol, z. B. Methanol, Äthanol od. dgl., Dioxan, Tetrahydrofuran, Dimethylsulfoxydod. dgl. oder in organischen Lösungsmitteln, welche nicht durch saure Hydrolyse angegriffen werden, durchgeführt werden.
Auf der Spaltbarkeit von Verbindungen der Formel I durch Säure beruht die Möglichkeit eines Austausches der Carbonylkomponente. Falls man nämlich die Spaltung in Gegenwart eines Ketons oder Aldehyds der Formel V durchführt, so kann man Verbindungen der Formel I erhalten, wobei die durch die Symbole Ra und R4 dargestellten Reste zusammen mit dem Kohlenstoffatom in 2-Stellung in Formel 1

durch einen andern Rest ersetzt werden. Man kann z. B. eine Verbindung der Formel I, worin R3 und R4 je Methyl bedeuten, in Gegenwart von Chloraceton sauer hydrolysieren, wobei man eine Verbindung der Formel I erhält, worin einer der Reste R 3 und R4 Methyl und der andere Chlormethyl ist.
1, 2-Dihydrochinazolin-3-oxyde der Formel I, worin R3 Wasserstoff ist, können N1-alkyliert werden. Die Bildung derartiger 1-Alkyl-Derivate wird zweckmässigerweise durch ein Alkylierungsmittel üblicher Art bewirkt. Zum Beispiel kann eine Verbindung der Formel I durch Reaktion mit einem Alkylhalogenid (vorzugsweise einem Bromid oder Jodid) in Gegenwart einer starken Base, wie Kalium- oder Natriumalkoxyd oder-hydrid, alkyliert werden. Dialkylsulfate können auch zur Alkylierung herangezogen werden.
Die durch Kondensation eines Oxims der Formel IV mit einem Keton oder Aldehyd der Formel V erhaltene Verbindung der Formel I kann mit einem Phosphortrihalogenid, wie Phosphortrichlorid, oder mit Wasserstoff in Gegenwart eines Katalysators, wie Raney-Nickel, behandelt werden, wobei das entsprechende 3-Desoxyd der allgemeinen Formel

Verbindungen der Formeln I, lI, III und VI sind wertvolle chemische Zwischenprodukte. Verbindun- gen der Formeln I - III können durch Säurespaltung in 2-Aminobenzophenon-ss-oxime oder durch Säurespaltung in Gegenwart eines Aldehyds oder Ketons der Formel V in andere Verbindungen der Formeln I bis III umgewandelt werden, von denen einige pharmazeutisch wertvoll sind. Verbindungen der Formel III und ihre pharmazeutisch verwendbaren Säureadditionssalze sind hypotensiv wirksam. Verbindungen der Formel VI und der Formel VII und ihre Säureadditionssalze sind antikonvulsiv und anorektisch wirksam.
2-Aminobenzophenon-ss-oxime ihrerseits sind wertvolle Zwischenprodukte zur Herstellung von pharmazeutisch wirksamen 5-Phenyl-l, 3-dihydro-2H-1, 4-benzodiazepin-2-on-4-oxyden.
Verbindungen der Formel L worin Ra und R s Wasserstoff und R4 eine α,α-Dihalogenalkyl-Gruppe bedeuten, sind von besonderem Interesse, da sie auf vielfältige Weise in wertvolle, bekannte Verbin-

Durch Behandlung mit Ammoniak oder einem primären Amin können sie in entsprechende 2-Amino-oder 2-substituierte-Amino-5-phenyl-3H-1, 4-benzodiazepin-4-oxyde umgewandelt werden. Eine andere bevorzugte Gruppe von Verbindungen der Formel I sind solche, worin R, und R3 beide Wasserstoff und R4 Alkyl, α-Monohalogenalkyl oder α, α-Dihalogenalkyl bedeuten. Oxydation dieser letzteren Verbindungen mittels Mercurioxyd führt zu einer Mischung von 2-R-3-R-5- (R -Phenyl)-7-R- 1, 2-dihydro-5H-l, 4-benzodiazepin-4-oxyden und 2-R16-3-R17-5-(R2-Phenyl0-7-R1-2,3-dihydro-1H- 1, 4-benzodiazepin-4-oxyden. Diese zwei Gruppen von Verbindungen können durch Phosphortrichlorid desoxydiert werden.
Eine andere bevorzugte Gruppe von Verbindungen der Formel I sind solche, worin R, Wasserstoff, R Alkyl und R Halogenmethyl sind. Derartige Verbindungen und ihre 3-Desoxy-Derivate können mit einer starken Base, wie Kalium-t-butoxyd oder Natriumhydrid, behandelt werden, wobei man 3-A1kylen-1,2-dihydro-3H-1,4-benzodiazepin-4-oxyde bzw. ihre 4-Desoxy - Derivate erhält. Derartige Verbindungen können ihrerseits, z. B. durch katalytische Hydrierung mit Raney-Nickel, zu 3-Alkyl- 1, 2-dihydro-3H-l, 4-benzodiazepinen reduziert werden. Falls man Chinazolinderivate der zu Beginn dieses Absatzes definierten Art mit wässerigem Alkali behandelt, erhält man 2-Alkyl-3H-l, 4- benzodiazepin-4-oxyde, welche ihrerseits weiteren Umwandlungen zugänglich sind.
Die pharmazeutisch aktiven Verbindungen des erfindungsgemässen Verfahrens können als Heilmittel, z. B. in Form pharmazeutischer Präparate, welche sie oder ihre pharmazeutisch verwertbaren Salze in Mischung mit einem für die enterale, perkutane oder parenterale Applikation geeigneten pharmazeutischen organischen oder anorganischen inerten Trägermaterial, wie z. B. Wasser, Gelatine, Gummiara- bicum, Milchzucker, Stärke, Magnesiumstearat, Talk, pflanzliche Öle, Polyalkylenglykole, Vaseline, usw. enthalten. Die pharmazeutischen Präparate können in fester Form, z. B. als Tabletten, Dragees, Suppositorien, Kapseln ; in halbfester Form, z. B. als Salben ; oder in flüssiger Form, z. B. als Lösungen, Suspensionen oder Emulsionen, vorliegen.
Gegebenenfalls sind sie sterilisiert und bzw. oder enthalten Hilft stoffe, wie Konservierungs-, Stabilisierungs-, Netz- oder Emulgiermittel, Salze zur Veränderung des osmotischen Druckes oder Puffer. Sie können auch noch andere therapeutisch wertvolle Stoffe enthal-

Die folgenden Beispiele veranschaulichen das erfindungsgemässe Verfahren. Alle Temperaturen sind in c) C angegeben. Wenn von einem Oxim"unbestimmter Konfiguration"gesprochen wird, so wird ein

und 0,5 g fein gemahlenes Kupfersulfatpentahydrat wird 2 h zum Rückfluss erhitzt. Es beginnt bald ein gelbes Reaktionsprodukt auszukristallisieren. Die Reaktionsmischung wird auf Raumtemperatur gekühlt und das Reaktionsprodukt abfiltriert. Man suspendiert dann in 75 ml Wasser, filtriert und wäscht mit Wasser zur Entfernung des Kupfersulfats. Man erhält 6-Chlor-l, 2-dihydro-2, 2-dimethyl-4-phenylchinazolin-3-oxyd in Form von gelben Prismen vom Schmelzpunkt 200 - 2200.
A) 200 g 6-Chlor-l, 2-dihydro-2,2-dimethyl-4-phenyl-chinazolin-3-oxyd und 200 ml Chlor-propanon- (2) werden 2 l Äthanol, 0,5 1 Benzol und 2 ml konz. Salzsäure zugesetzt. Man destilliert sodann Lösungsmittel aus der Reaktionsmischung übereineVigreaux-Kolonne. Im Verlaufe von 80min gehen 1, 5 l über. Die Reaktionsmischung wird gekühlt. mit 2 n Ammoniak neutralisiert und das Reaktionsprodukt mit 11 Eiswasser ausgefällt. Der Niederschlag wird abfiltriert mit Äthanol und anschliessend mit

3-oxyd, 100 ml Äthanol, 50 ml Benzol und 10 ml (etwa 100 mMole) rohem destillierten Dichloracetaldehyd erhitzt man 45 min, wobei man 100 ml Lösungsmittel abdestilliert. Der vorhandene Dichloracetaldehyd ist genügend sauer, um die Reaktion zu katalysieren.
Die Reaktionsmischung wird gekühlt und das gebildete 6-Chlor-2-dichlormethyl-1, 2-dihydro-4-phenyl-chinazolin-3-oxyd abfiltriert. Das Produkt schmilzt bei 195 - 1980 (Zers. ).
C) Eine Mischung von 14, 3 g (50 mMole) 6-Ghlor-1, 2-dihydro-2, 2-dimethyl-4-phenyl-chinazolin- 3-oxyd, 150 ml Äthanol und 6,78 g (enthaltend 60 mMole monomeren Dichloracetaldehyd) Dichloracetaldehydpolymer wird 30 min erhitzt. wobei 70 ml Lösungsmittel abdestilliert werden. Die erhaltene Reaktionsmischung wird gekühlt und das erhaltene gelbe kristallisierte 6-Chlor-2-dichlor-methyl- 1,2-dihydro-4-phenyl-chinazolin-3-oxyd abfiltriert und mit Äther und Petroläther gewaschen. Das Produkt schmilzt bei 205 - 2070.

Stäbchen vom Schmelzpunkt 221 - 2220.
A) Aus einer Mischung von 200 g 6-Chlor-l, 2-dihydro-2,2-dimethyl-4-phenyl-chinazolin-3-oxyd,
200 g 1, 3-Dichlor-propanon, 2 1 Äthanol, 400 ml Benzol und 2 ml konz. Salzsäure destilliert man Lö- sungsmittel ab. Während 1 h gehen 1, 21 Lösungsmittel über. Die Reaktionsmischtung wird in Eis ge- kühlt und mit 40 ml 2 n Ammoniak neutralisiert. Das erhaltene feste Produkt wird abfiltriert und mit
200 ml Äther/Äthanol (l : l) gewaschen. Man erhält 6-Chlor -2, 2 - bis (chlor-methyl) -l, 2 -dihydro-
4-phenyl-chinazolin-3-oxyd vom Schmelzpunkt 169 -1710.
B) 57,4 g 6-Chlor-l, 2-dihydro-2,2-dimethyl-4-phenyl-chinazolin-3-oxyd werden in 2 l Tetra- hydrofuran suspendiert. Die Reaktionsmischtung wird auf 70 abgekühlt und sodann mit 25 g Kalium-t.- butoxyd versetzt. Die Reaktionsmischung wird 3 min gerührt und sodann mit 50 ml Methyljodid ver- setzt. Die Reaktionsmischung wird weitere 15 min gerührt, wobei die Farbe von rot auf gelb verblasst.
Man filtriert sodann über Kieselgur, dampft das Filtrat im Vakuum ein und behandelt den Rückstand mit
Hexan, wobei man ein dickes gelbes Produkt erhält, das aus Äthanol/Wasser umkristallisiert rohes
6-Chlor-l, 2-dihydro-l, 2, 2-trimethyl-4-phenyl-chinazolin-3-oxyd liefert.
C) 10g6-Chlor-l, 2-dihydro-l, 2, 2-trimethyl-4-phenyl-chinazolin-3-oxyd, 150 ml Äthanol,
10 ml Chloraceton und 0,3 ml konz. Salzsäure werden zusammen erhitzt, wobei man langsam während 1 1/2 h Lösungsmittel abdestilliert. Insgesamt werden 100 ml Destillat erhalten. Der Rückstand wird mit 250 ml Äther verdünnt und dreimal mit je 250 ml Wasser gewaschen. Die Ätherlösung wird über Natriumsulfat getrocknet und im Vakuum zu einem gelbbraunen harzartigen Material eingeengt, das aus Äther umkristallisiert 6-Chlor-2-chlormethyl- 1, 2-dihydro -1, 2 -dimethyl-4-phenyl-chinazolin- 3-oxyd liefert. Nach zweimaligem Umkristallisieren aus Cyclohexan erhält man gelbe Prismen vom
Schmelzpunkt 118 - 1200.
Beispiel 8 : Man vermischt 300 g 2-Amino-5-chlor-benzophenon-oxim unbestimmter Konfiguration mit 3 l Aceton, welches 15 g pulverisiertes Kupfersulfatpentahydrat und 3 ml Eisessig enthalt. und erhitzt unter Rühren 7 - 8 h zum Rückfluss. Nach 2 - 3 h beginnen aus der braunen Lösung gelbe Kristalle von 6-Chlor-l, 2-dihydro-2,2-dimethyl-4-phenyl-chinazolin-3-oxyd auszufallen. Man lässt die Reaktionsmischung über Nacht auf Raumtemperatur abkühlen, filtriert den Niederschlag ab und wäscht zweimal mit je etwa 200 ml Wasser, um das Kupfersulfat zu entfernen. Das Reaktionsprodukt bildet gelbe Prismen vom Schmelzpunkt 230 - 2340.
A) 5 g 6-Chlor-l, 2-dihydro-2,2-dimethyl-4-phenyl-chinazolin-3-oxyd werden in 100 mlChloroform und 2 ml Phosphortrichlorid in 20 ml Chloroform gelöst und die Mischung 1/2 h zum Rückfluss erhitzt. Die erhaltene rote Lösung wird in 100 ml 1, 2 n Natronlauge gegossen. Man trennt die Phasen und wäscht die Chloroformschicht mit 50 ml einer 10% eigen Natriumbicarbonatlösung und mit 50 ml gesättigter Kochsalzlösung und trocknet über Natriumsulfat. Die Lösung wird sodann durch 50 g Aluminiumoxyd filtriert und das Aluminiumoxyd mit 100 ml Methylenchlorid gewaschen.
Das Eluat wird im Vakuum eingeengt und der Rückstand aus Hexan kristallisiert, wobei man 6-Chlor-l, 2-dihydro-2, 2-di- methyl-chinazolin als gelbes Produkt erhält, das nach wiederholter Kristallisation aus Äthanol/Wasser und Hexan/Äther gelbe Nadeln vom Schmelzpunkt 142 -144, 50 bildet.
3, 3 g der erhaltenen Base werden in 30 ml Äthanol gelöst und sodann mit 2 ml 10 n methanolischer Salzsäure und 200 ml Äther versetzt. Es scheidet sich 6-Chlor-l, 2-dihydro-2, 2-dimethyl-4-phenyl- chinazolin-hydrochlorid in Form von orange gefärbten Nadeln vom Schmelzpunkt 120 - 1250 (Zers.) ab.
B) 5,0 g 6-Chlor-1, 2-dihydro-2,2-dimethyl-chinazolin werden in 400 ml Methanol gelöst Die Lösung wird in Eis gekühlt und mit 2,5 g Natriumborhydrid versetzt. Die Reaktionsmischung wird 6 h in einem Eisbad gerührt. Hierauf neutralisiert man mit Essigsäure und engt im Vakuum zur Trockne ein.
Der Rückstand wird in 200 ml Methylenchlorid gelöst und die Lösung mit 100 ml Wasser gewaschen und über Natriumsulfat getrocknet. Nach Einengen im Vakuum erhält man ein schwach gefärbtes Öl. Ein Anteil von 2,73 g dieses Öls wird in 10 ml Methanol gelöst und mit 1 ml 10 n methanolischer Salzsäure

Während 17 h werden 2, 8 I Wasserstoff aufgenommen. Der Katalysator wird sodann entfernt und das Filtrat im Vakuum zu einem gelben Harz eingeengt. Dieses wird mit Hexan/Äther behandelt und das unlösliche Material abfiltriert. Das Filtrat wird vom Lösungsmittel befreit und der Rückstand aus Hexan kristallisiert, wobei man 6'-Chlor- 1-methyl-4'-phenyl-spiro[piperidin-4,2'-(1'H)-chinazolin]vom Schmelzpunkt 120 - 1270 erhält.
Nach Reinigung durch Filtration einer Ätherlösung durch Aluminiumoxyd und Umkristallisieren aus Methylenchlorid/Hexan erhält man gelbe Prismen vom Schmelzpunkt 127 - 1290.
B) Eine Lösung von 3,27 g (10 mMole) 6'-Chlor-1-methyl-4-'-phenyl-spipro[piperidin-4,2'-(1'H)chinazolin] in 50 ml Äther wird mit überschüssiger methanolischer Salzsäure behandelt. Das ausgeschie-

Beispiel 12 : Man stellt Piperidinoaceton her, indem man 8 ml (etwa 75 mMole) Chloraceton tropfenweise zu einer Lösung von 20 ml (0, 202 Mole) Piperidin in 50 ml wasserfreien Äther zutropft. Die Reaktionsmischung wird 1/2 h zum Rückfluss erhitzt, das Piperidin-hydrochlorid abfiltriert und mit Wasser gewaschen und das Filtrat im Vakuum eingeengt. Der erhaltene ölige Rückstand wird ohne weitere Reinigung weiter verarbeitet. Man versetzt mit 17 g 2-Amino-5-chlor-benzophenon-ss-oxim in Äthanol und anschliessend mit 10 ml konz. Salzsäure, um den PH-Wert der Lösung auf 1 - 3 zu bringen.
Schliesslich versetzt man mit 100 ml Benzol. Man destilliert 150 ml des Lösungsmittels ab und lässt die Reaktionsmischung über Nacht stehen. Der gelbe Niederschlag wird abgetrennt und zwischen Methylenchlorid und 10%iger Natriumcarbonatlösung verteilt. Der Methylenchloidextrakt wird getrocknet und im Vakuum eingeengt. Die Reaktionsmischung wird 5 min auf einem Dampfbad erwärmt. Sodann kühlt man, wobei sich 61-Chlor-41-phenyl-spiro [cyclohexan-1, 21- (1'H)-chinazolin]-3-oxyd ausseheidetdasabfiltriertwird und nach zweimaligem Umkristallisieren aus Äthanol gelbe Prismen vom Schmelzpunkt 215-221 bil- det.
Beispiel 15 : EineMischungvonlOg5-Chlor-2-methyl-amino-benzophenon-oxim (Mischung von Stereoisomeren), 0, 1 g Kupfersulfatpentahydrat, 100 ml Aceton und 100 ml Chloroform wird über
Nacht zum Rückfluss erhitzt, wobei man das Destillat durch einen Soxhlet-Extraktor leitet, welcher wasserfreies Natriumsulfat enthält. 50 g Raney-Nickel frei als Katalysator hydriert. Während 20 min werden 2,4 1 Wasserstoff aufgenom- men. Man neutralisiert die Reaktionsmischung mit konz. Ammoniak, filtriert den Katalysator nach Zu- satz von Benzol ab und engt das Filtrat im Vakuum zu einem roten Öl ein. Dieses wird in250 ml Äther gelöst und mit 250 ml einer 5%igen Natriumbicarbonatlösung gewaschen. Die Ätherlösung wird über Na- triumsulfat getrocknet und das Lösungsmittel entfernt. Man erhält ein gelbes Öl, welches aus Äther/He- xan kristallisiert und durch Filtration mit 100 ml Methylenchlorid durch 40 g Aluminiumoxyd gereinigt wird.
Man erhält 6-Chlor-l, 2-dihydro-l, 2 ; 2-trimethyl-4-phenyl-chinazolin vom Schmelzpunkt 80 bis
89 . Nach Umkristallisieren aus Hexan erhält man gelbe Prismen vom Schmelzpunkt 86, 5 - 88, 50.

The amount of copper sulfate present is not essential, but it has been found that cupric salts, such as copper sulfate, exert a beneficial influence on the reaction of an ex-oxime of the formula IV with a ketone or aldehyde of the formula V.

The 1,2-dihydroquinazoline-3-oxides of the formula I obtainable according to the invention can be converted into h-oximes of the formula IV by acid hydrolysis. Example 20: 3.7 g (15.6 mmol) of 2-amino-5-nitro-benzophenone-ss-oxime are added to a filtered solution of 10 g of polymeric dichloroacetaldehyde in 50 ml of ethanol. The reaction mixture is heated to boiling and then allowed to cool to room temperature. The yellow precipitate of 2-dichloromethyl-1,2-dihydro-6-nitro-4-phenyl-quinazoline-3-oxide is filtered off. Recrystallization from chloroform / hexane gives yellow needles of 1,2-dihydro-2,2,6-trimethyl-4-phenyl-quinazoline-3-oxide with a melting point of 205-2120 (decomp.).
A) A mixture of 25 g (94 mmol) of 1,2-dihydro-2, 2, 6-trimethyl-4-phenyl-quinazoline-3-oxide, 25 ml of technical grade chloroacetone, 0.25 ml of conc. Hydrochloric acid, 50 ml of benzene and 500 ml of ethanol are heated up and about 300 ml of solvent are evaporated off. The residue is cooled in an ice bath and the reaction product is separated off. Additional reaction product is obtained after concentrating the mother liquors. Recrystallization from methylene chloride / hexane gives yellow prisms of 2-chloromethyl-1,2-dihydro-2,6-dimethyl-4-phenyl-quinazoline-3-oxide with a melting point of 172-1740.
