Die Erfindung betrifft ein Verfahren zur Herstellung von 3-Hydroxy-#1,3,5(10)-19-nor-steroiden oder 3-Äthern davon der allgemeinen Formel

Nor-steroidverbindungen.- steroid mit einer Persäure zur Bildung der entsprechenden 5, 10-Epoxyverbindung ; diese wird zuerst durch Umsetzung mit Alkalien in ein 10ss-Hydroxy-#4-3-keto-19-nor-steroid und dann durch Behandeln mit Säuren ins 3-Hydroxy-1, 3, 5 (10)-östratrien umgewandelt (J. Org. Chem. Bd. 23 [1958], S. 1744). Nach den bekannten Methoden wird also die Aromatisierung des A-Ringes durch eine zweistufige Reaktion aus 3-Keto-5, 10-oxido-steroiden über eine Zwischen-lOss-hydroxy--3-ketoverbindung durchgeführt.
Es wurde nun gefunden, dass die Aromatisierung des A-Ringes des Steroidmoleküls einstufig durchgeführt werden kann, indem man 5, 10-Oxido-steroidverbindungen, die in 3-Stellung eine gegebenenfalls keta1ysierteKetogruppe enthalten, mit starken Säuren in einem inerten Lösungsmittel behandelt.
Ferner wurde gefunden,, dass man, wenn man die Behandlung der genannten Epoxyde mit Säuren in alkoholischem Medium oder in Gegenwart von Alkoholen durchführt, unmittelbar Äther von 3-Hydroxy- - östratrienen erhält, deren Ätherreste dem Kohlenwasserstoffrest des bei der Reaktion verwendeten Alkohols entsprechen

Anschliessend wurde überraschendgefunden, dass nicht nur die obengenannten 5, 10-Oxidosteroide, aber eine beliebige ss, y-disubstituierte 3-Keto-19-nor-steroidverbindung durch Behandeln mit Säuren in ein Phenolsteroid, wenn man in einem inerten Lösungsmittel arbeitet, oder in einen Äther davon, wenn man in Gegenwart des geeigneten Alkohols arbeitet, umgewandelt werden kann.

Im allgemeinen können starke organische, wie Ameisensäure oder Säuren mit pK < 1, 5, z. B. Salzsäure, Bromwasserstoffsäure, Schwefelsäure, Perchlorsäure, Oxalsäure, Dichloressigsäure, Trichloressigsäure und Sulfonsäuren, wie Methan-, Benzol- oder p-Toluolsulfonsäure in einem nicht-alkoholischen Lösungsmittel, wie z. B. ein Kohlenwasserstoff, Aceton, Eisessig, Äther, Dioxan, Tetrahydrofuran oder Chloroform, gegebenenfalls in Gegenwart eines aliphatischen, cycloaliphatischen oder araliphatischen Alkohols angewendet werden. Die Temperatur, wobei die saure Behandlung durchgeführt wird, kann zwischen Raumtemperatur und dem Siedepunkt des Lösungsmittels schwanken ; eine Temperatur von 50 bis 1000 C ist für die Aromatisierung optimal.
Am Ende der Reaktion scheiden sich die Phenolsteroide aus ; insbesondere, wenn man in nicht-alkoholischen Lösungsmitteln arbeitet, erhält man 3-Hydroxy- - 1, 3, 5 (10) -östratriene, während man, wenn man in Gegenwart von Alkoholen oder in einem alkoholsehen Lösungsmittel arbeitet, die mit dem verwendeten Alkohol verätherten 3-Hydroxy-l, 3, 5 (10) -östra- triene erhält.
Die erhaltenen Endprodukte können aus dem Reaktionsgemisch üblicherweise, z. B. durch einfache Verdünnung mit Wasser und Abfiltrierung des entstehenden Niederschlages oder durch Eindampfen des Lösungsmittels und nachfolgendes Umkristallisieren, abgetrennt werden.
Die als Ausgangsstoffe verwendeten Verbindungen der Formel II, von welchen einige neue Produkte darstellen, können nach bekannten Methoden leicht erhalten werden.
Die 5, 10- und 5, 6-Oxidoverbindungen und Mischungen davon können hergestellt werden, indem man die entsprechenden )-und/oder/ ( )-Verbindungen, die ihrerseits durch direkte Synthese aus den Derivaten der Normalreihe über die lOss-Cyan-19-nor-steroide leicht hergestellt werden können, mit einer Persäure, wie Perameisensäure, Peressigsäure, Perbenzoesäure, Monoperphthalsäure oder Wasser- stoffperoxyd, in einem organischen Lösungsmittel bei 0 - 300 C umsetzt. Geeignete Lösungsmittel sind die gegebenenfalls halogenhaltigen Kohlenwasserstoffe, wie Benzol, Toluol, Hexan, Petroläther, Chloroform oder Äther, Tetrahydrofuran oder Dioxan. Die am Ende der Reaktion erhaltenen Epoxyde können üblicherweise, z.
B. durch einfaches Abfiltrieren oder durch Abstumpfen des Reaktionsgemisches, Ausziehen, Eindampfen und Umkristallisieren, isoliert werden. Verwendet man als Ausgangsmaterial eine Mischung von (")-und A )-Verbindungen, so können die am Ende der Epoxydierung erhaltenen Epoxyverbindungen auseinander getrennt werden. Solche Trennung kann durch fraktionierte Kristallisierung oder

durch Chromatographie der Mischung der Epoxyde durchgeführt werden ; so kann eine befriedigende Trennung aller möglicher Isomere erzielt werden, die in Phenolsteroide einzeln umgewandelt werden können.
Jedoch ist es praktisch vorteiliger, der Aromatisierung dieselbe, am Ende der Persäure-Behandlung erhaltene Mischung von Epoxyden ohne weitere Reinigung zu unterwerfen.
Bevorzugte Epoxyde, die neue Produkte darstellen, sind durch die folgenden Formeln gekennzeichnet :

Durch Behandeln mit einer starken Säure aromatisieren die obigen Verbindungen mit praktisch quantitativer Ausbeute mit gleichzeitiger Hydrolyse der vorhandenen Ketale zur Bildung der entsprechenden Phenolsteroide, die in 17-oder 20-Stellung einen Ketosauerstoff tragen. Die Verbindungen III, IV und V und die Mischungen derselben durch kurzes Erhitzen mit einer Säure liefern quantitativ Östron, während die Verbindungen VI und VII liefern das 3-Hydroxy-170-acetyl-l, 3, 5 (10)-östratrien. Selbstverständlich durch Arbeiten in Gegenwart von Alkoholen erhält man die Äther der entsprechenden 3-Hy- droxy- östratriene.
Die verschiedenen Halogenhydrine können durch Spaltung des Epoxyringes von 3-Keto-5, 10- oder - 5, 6-oxidoverbindungen oder durch Behandlung der entsprechenden A -oder ( -ungesättigten Verbindungen mit den geeigneten Untersäuren, wie beispielsweise mit unterchloriger oder unterbromiger Säure, oder auch durch Behandlung der 3-Hydroxy-#5(10)- und 3-Hydroxy-#5(6)-19-nor-verbindungen

Die Dihalogenverbindungen werden durch Halogenierung der entsprechenden ungesättigten Verbindungen oder vorzugsweise durch Halogenierung von 3-Hydroxy-#5(6)-19-nor-steroiden und nachfolgende Oxydierung der 3-Ketogruppe hergestellt.
Die folgenden Beispiele erläutern das erfindungsgemässe Verfahren.
Beispiel 1 : 2 g einer Mischung von 3,17-Bis-äthylendioxy-19-nor-#5(10)-androsten und 3,17-Bis-äthylendioxy-19-nor-#5(6)-androsten, (die durch Behandeln von 3,17-Bis-äthylendioxy- -10ss-cyan-19-nor-#5(6)-androsten mit Natrium in Äthanol erhalten wurde), werden in 200 cm3 Äther gelöst und mit 12 cm3 einer 15% eigen Lösung von Monoperphthalsäure bei Raumtemperatur behandelt Nach Stehen über Nacht wird die Lösung zuerst mit einer 5% igen Natriumbicarbonadösung, dann mit Wasser neutral gewaschen, anschliessend getrocknet und im Vakuum eingedampft. Der aus einer Mischung von Epoxyden bestehende Rückstand wird auf"Florisil" (synthetisches Aluminium-Magnesium-sili- cat) chromatographiert.
Durch Eluieren mit Benzol-Äther = 3 : 2 erhält man 1, 35 g 3, 17-Bis-äthylen- dioxy-5ss, 10ss-oxido-19-nor-androstan, das, nach Umkristallisieren zuerst aus Methanol und dann aus Hexan, bei 116-117 C schmilzt ; [a] g = +100 (Chloroform, c = 0, 50/0). Besondere Infrarotabsorptionsmaxima: 1170, 1107, 1057, 1043, und 952 cm-1.

- 1700Cyclopentanol gelöst und mit 2 Tropfen konz. Salzsäure behandelt. Man kocht 1 h am siedenden Wasserbade und destilliert den überschüssigen Alkohol im Vakuum ab.
