Verfahren zur Herstellung von neuen Sulfonamiden
Die Erfindung betrifft ein Verfahren zur Herstellung von neuen 3-Sulfanilamido-4. 5-di (niederalkyl) - - isoxazolen, insbesondere des neuen 3-Sulfanilamido-4, 5-dimethyl-isoxazols, sowie von Salzen solcher Sulfonamide, insbesondere von Salzen mit starken Basen.
Die erfindungsgemäss erhältlichenneuen 3-Sulfanilamido-4, 5-di (niederalkyl)-isoyazole entsprechen der allgemeinen Formel I :

worin R und Rl zwei gleiche oder verschiedenartige, niedere Alkylgruppen, wie Methyl, Äthyl, Propyl, insbesondere aber je eine Methylgruppe bedeuten.
Das erfindungsgemässe Verfahren ist dadurch gekennzeichnet, dass man ein 3-Amino-4, 5-di (nieder- alkyl)-isoxazol mit einem Benzolsulfohalogenid umsetzt, das in para-Stellung einen in die Aminogruppe überfiihrbaren Substituenten trägt und, nach erfolgter Umsetzung, den p-Substituenten in die Aminogruppe umwandelt, sowie, erwünschtenfalls, das erhaltene 3-Sulfanilamido-4, 5-di (niederalkyl) -isoxazol in ein Salz überführt.
Die Umsetzung der Amino-isoxazol-Komponente mit der'Benzolsulfohalogenid-Komponente und die Überführung des p-Substituenten in die Aminogruppe nach erfolgter Kupplung, wie schliesslich auch die Salzbildung können in an sich bekannter Weise erfolgen. So kann man z. B. 3-Amino-4, 5-dimethyl-isox- azol in Gegenwart von Pyridin mit p-Acetaminobenzolsulfochlorid bei Raumtemperatur oder leicht erhöhter Temperatur umsetzen und das erhaltene 3- (N4, -Acetylsulfanilamido) -4, 5-dimethyl-isoxazol mit einer wässerigen Lösung eines Alkalihydroxyds, z. B. Natriumhydroxyd, unter Erwärmen zum 3-Sulfanilamido-4,5-dimethyl-isoxazol verseifen.
Als p-Substituenten der Benzolsulfohalogenidkomponente kommen ausser dem oben genannten Acetaminorest weitere Acylaminoreste in Betracht, ferner ; Carbalkoxyamino, wie Carbäthoxyamino, Nitro ; Arylazo, wie Phenylazo. Diese p-Substituenten lassen sich durch Hydrolyse oder Reduktion leicht in die Aminogruppe überführen.
Die erfindungsgemäss erhältlichen, neuen Sulfonamide der Formel I können mit stärkeren Basen, wie Alkalihydroxyden, z. B. Kalium- oder Natriumhydroxyd, Alkalicarbonaten, z. B. Natriumcarbonat, oder Aminen, wie Äthanolamin, Diäthanolamin, in Salze übergeführt werden.
Die Sulfonamide der Formel 1 sowie ihre Salze mit starken Basen sind gegen eine Vielzahl von grampositiven und gramnegativen Bakterien hoch wirksam. Sie können dementsprechend zur Behandlung z. B. von E. Coli-, Pneumokokken-, Streptokokken- oder Staphylokokken-Infektionen verwendet werden. Von Vorteil ist dabei die überraschend hohe Wasserlöslichkeit ihrer Alkalimetallsalze, wie z. B. des Natriumsalzes von 3-Sulfanilamido-4, 5-dimethyl-isoxazol..

finden. welche sie oder ihre Salze in Mischung mit einem für die enterale oder parenterale Applikation geeigneten pharmazeutischen, organischen oder anorganischen inerten Trägermaterial, wie z. B. Wasser, Gelatine, Milchzucker, Stärke, Magnesiumstearat, Talk, pflanzliche Öle, Gummi, Polyalkylenglykole, Vaseline usw. enthalten. Die pharmazeutischen Präparate können in fester Form z. B. als Ta- bletten, Dragees, Suppositorien, Kapseln, oder in flüssiger Form, z. B. als Lösungen, Suspensionen oder Emulsionen, vorliegen.
Gegebenenfalls sind sie sterilisiert und bzw. oder enthalten Hilfsstoffe, wie Konservierungs-, Stabilisierungs-, Netz- oder Emulgiermittel, Salze zur Veränderung des osmotischen Drukkes oder Pfuffer. Sie können auch noch andere therapeutisch wertvolle Stoffe enthalten.
Beispiel : Zu einer Lösung von 8, 4 g 3-Amino-4, 5-dimethyl-isoxazol in 42 ml wasserfreiem Pyridin fügt man unter Schütteln 16, 8 g p-Acetamino-benzoesulfochlorid. Die Temperatur steigt dabei auf 50 C. Nach 1 h fügt man 225 ml Wasser zu und überlässt das Gemisch 2 h der Kristallisation. Die ausgeschiedenen Kristalle werden filtriert, mit Wasser gut gewaschen und im Vakuum über Kaliumhydroxyd getrocknet. Man erhält so 18, 4 g 3- (N, -Acet lsulfanilamido) -4, 5-dimethyl-isoxazol vom Schmelzpunkt 227-230 C. Nach Umkristallisation aus Äthanol beträgt der Schmelzpunkt 228-2300C.
Eine Mischung von 17, 4 g der erhaltenen N.-Acetylverbindung und 174 ml 10% figer Natronlauge wird unter Rühren 1 h auf dem Wasserbad erhitzt. Man giesst die erhaltene Lösung in 348 ml siedendes Wasser, fügt 42 ml Essigsäure zu und überlässt die Mischung der Kristallisation. Nach Filtration, Waschen mit Wasser und Trocknen im Vakuum über Kaliumhydroxyd erhält man 14,3 g 3-Sulfanilamido-4, 5-dimethyl-isoxazol vom Schmelzpunkt 193 -195, 5OC. Das Natriumsalz wird durch Behandlung mit wässeriger Natriumhydroxydlösung erhalten.

Toluolschicht wird zweimal mit je 225 ml Wasser extrahiert und die schwere Phase in den vereinigten wässerigen Extrakten gelöst.
Die wässerige Lösung wird mit 54 g Hydroxylamin-hydrochlorid versetzt. Dann fügt man im Verlaufe von 45 min 112 g 50% igue Natronlauge bei einer Temperatur unterhalb von 150C zu. Die resultierende Lösung, die dann praktisch neutral ist, wird 1 h im Eisbad weitergerührt, das Eisbad dann entfernt, das Rühren 1/2 h fortgesetzt, wobei die Lösung Raumtemperatur annimmt. Die Mischung wird dann auf 85 C erwärmt und 15 min bei dieser Temperatur gehalten. Dabei wird die Lösung trüb und es scheidet ein Öl aus, das nach Kühlung unter Rühren zu einer kristallinen Masse, dem 3-Methylamino-4, 5-dimethyl- - isoxazol, erstarrt (50 g) ; Schmelzpunkt 71-73 C.
Man stellt Mangandioxyd her, indem man im Verlaufe von 20 min eine Lösung von 180 g MnSO. H O in 400 ml Wasser zu einer Lösungvon 105g Kaliumpermanganat und 5 ml konz. Schwefelsäure in 2 l Wasser unter Rühren zugibt. Das Mangandioxyd wird filtriert, mit Wasser gewaschen, im Vakuumofen bei 700C 10 h und dann einen Tag im Vakuumexsiccator über Kaliumhydroxyd getrocknet. Ausbeute 177 g.
Das so hergestellte Mangandioxyd wird unter Rühren zu einer Lösung von 3 g 3-Methylamino -4, 5- - dimethyl-isoxazol in 300 ml Chloroform zugefügt. Die Temperatur steigt auf 4loC. Das Rühren wird 2 1/2 h fortgesetzt, wobei die Temperatur auf Raumtemperatur abfällt. Das Gemisch wird filtriert, das unlösliche Material mit frischem Chloroform gewaschen, und die Waschflüssigkeit mit dem Filtrat vereinigt. Das Gemisch wird unter reduziertem Druck zur Trockene eingedampft, wobei man einen Rück- stand von l, 15 g mit dem Schmelzpunkt 96 - 1000e erhält. Nach Umkristallisation aus Äthanol schmilzt das so erhaltene 3-Formamido-4, 5-dimethyl-isoxazol bei 112-113 C.
Eine Mischung von 0, 5 g 3-Formamido-4, 5-dimethyl-isoxazol, 1, 5 ml Wasser und 1, 5 ml 30igem Wasserstoffsuperoxyd wird auf 600C erwärmt und unter gelegentlichem Rühren 1 h bei dieser Temperatur gehalten. Nach Abkühlung erhält man 0, 43 g 3-Amino-4, 5-dimethyl-isoxazol vom Schmelzpunkt 112 bis 115 C. Nach Umkristallisation steigt der Schmelzpunkt auf 115-117 C.
