Verfahren zur Herstellung von Cycloserin
EMI1.1
EMI1.2

Process for the preparation of cycloserine
EMI1.1
EMI1.2
Heretofore, cycloserine has been successfully produced by fermentation using a cycloserine-producing microorganism in an aqueous fermentation medium composed of a nitrogen source, a carbohydrate source and calcium carbonate. Several sources of nitrogen, such as alpha alpha meal, amino acid cake, milk solids, whey nutrient, dried wheat sludge, and casein, have been used previously. However, the best yields of cycloserine have been obtained when using soybean products such as soybean oil, flour and dry soybean meal-like products. However, soybean materials do not have a defined standardized composition. Their use therefore makes it difficult to obtain the antibiotic in predetermined yields.
Fermentation batches of cycloserine, in which soybean products are used as a nitrogen source, also contain many impurities that can only be removed by expensive and time-consuming cleaning measures. For example, the fermentation wort has a dark color due to pigments produced by cycloserine-producing microorganisms when soybean products are used as a nitrogen source, and it contains a large amount of residue of unassimilable soybean material.
Impurities such as these are currently removed from cycloserine by a long process, the adjustment of the pH of the fermentation medium to about 3 - 6, filtration of the pH-regulated fermentation medium, absorption of the cycloserine in the medium thus filtered on an ion exchange resin,

len, or the cycloserine can be crystallized from the aqueous solution by adding a water-miscible solvent to the solution in which cycloserine is insoluble.
The following examples serve to illustrate the process according to the invention for the preparation of cycloserine.

0.48% sterile urea was then added to the sterilized medium. The fermentor was then inoculated with 800 gallon (3028 L) inoculum prepared as described above and the medium was incubated at 300 ° C for 86 hours while aerating at a rate of 800 cubic feet (22650 l) per minute . At the end of the 86-hour fermentation period, the medium showed a cyclose content of 2600 g / ml.
Example 2: The procedure of Example 1 was followed with the modification that 7% starch and 0.62% urea were used to give titers of 3000 g / ml cycloserine at the end of a 72 hour fermentation period.
Example 3: The procedure of Example 1 was followed with the modification that 0.48% ammonia was used to give results similar to those of Example 1. The procedure of Example 1 was followed with the modification that 0.48% ammonium carbonate was used to give results similar to Example 1.

iel 4: AgNO added. The mixture was then thoroughly stirred for 30 minutes and then filtered. The filtrate was then adjusted to pH 6.8 with dilute aqueous sodium hydroxide solution in order to precipitate the silver salt of cycloserine. The precipitated silver cycloserine was then dried to give 1011 g of silver cycloserine containing 442 g of cycloserine. This means a yield of 74.50/0 of the cycloserine in the fermentation medium.
Deionized water was then added to the 1,011 grams of silver cycloserine and the resulting mixture was slurried. Hydrochloric acid was then added to this slurry to precipitate solid silver chloride and thereby liberate cycloserine in aqueous solution. The solid silver chloride was then removed by filtration. Hydrogen sulfide was bubbled through the aqueous cycloserine solution to precipitate residual amounts of silver, and the precipitated silver sulfide was removed by filtration. The resulting filtrate was then poured into cold isopropyl alcohol to obtain 369 g of white crystalline cycloserine. This means a 620 / liter yield of cycloserine.
