Verfahren zur Herstellung von neuen Acylderivaten
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von neuen Acylderivaten des 5, 6- Dihydro-llH-dibenzo[c, f][1, 2]diazepins und des 5, 6, 1I, 12- Tetrahydro-dibenzo[c, g] [l, 2]diazocins mit wertvollen pharmakologischen Eigenschaften, sowie ihrer Salze mit anorganischen und organischen Basen.
Diese neuen Typen von kondensierten Heterocyclen sollten auf Grund der bestehenden NomenklaturRegeln (IUPAC-Nomenc1ature of Organic Chemistry-1957, Butterworths Scient. Publ. London 1958, B, 51 ff) als 13-substituierte l2, 14-Dioxo-5, 6, 13, 14-tetrahydro-12H-pyrazolo[1, 2-a]dibenzo[c, g][1, 2]- diazocine

Die Kondensation von Malonsäurediestern der allgemeinen Formel II mit 5, 6-Dihydro-llH-dibenzo[c, f][1, 2]diazepin bzw. mit 5, 6, 11, 12-Tetrahydro-dibenzo [c, g] [1, 2] diazocin erfolgt vorzugsweise in organischen Lösungsmitteln wie Benzol, Toluol, Xylol, Butanol oder Dibutyläther bei erhöhten Temperaturen, z. B. zwischen 80-160 C, wobei der freigesetzte Alkohol gegebenenfalls fortlaufend abdestilliert wird.
Die Kondensation wird mit Vorteil in einer Stickstoffatmosphäre durchgeführt. Als alkalische Kondensationsmittel eignen sich dabei allgemein solche, die ein bewegliches Wasserstoffatom durch ein Metallatom zu ersetzen fähig sind, z. B. Alkalimetalle oder deren Alkoholate, Amide, Hydride und metallorganische Verbindungen, wie Natrium, Kalium, Lithium, Natriumalkoholat, Kaliumalkoholat, Natriumamid, Lithiumamid, Natriumhydrid, Lithiumhydrid, Phenyllithium und Methyllithium.
Als säurebindende Mittel für die Umsetzung von Malonsäurederivaten der allgemeinen Formel III mit 5, 6-Dihydro-11 H-dibenzo[c, f][1, 2]diazepin bzw. mit 5,6, 11, 12-Tetrahydro-dibenzo[c, g]diazocin eignen sich insbesondere tertiäre organische Basen wie Pyridin, Dimethylanilin, Triäthylamin oder Tributylamin in An- oder Abwesenheit zusätzlicher organischer Lösungsmittel wie Chloroform, Diäthyläther oder Diisopropyläther. Der Ringschluss erfolgt in diesem Falle vorzugsweise zwischen Raumtemperatur und zirka 1000 C.
Anstatt die Kondensation in Gegenwart eines säurebindenden Mittels durchzuführen, kann man solche Mittel, welche die N-ständigen Wasserstoffatome des 5, 6-Dihydro-llH-dibenzo- [c, f] [1, 2] diazepins bzw. des 5, 5, 11, 12-Tetrahydro-dibenzo [cg] [1, 2] diazocins durch Metallatome zu ersetzen geeignet sind, wie z. B. Methyllithium oder Äthylmagnesiumbromid, auch unmittelbar vor der Kondensation z. B. zur 5, 6-Dilithiumverbindung oder 5, 6-Bis-magnesiumbromidverbindung umsetzen und auf letztere Malonsäurederivate der allgemeinen Formel III einwirken lassen.
Der Ringschluss zwischen einer Malonsäure der allgemeinen Formel V und 5, 6-Dihydro-11H-dibenzo- [c, f][1, 2]diazepin, bzw. 5,6, 11, 12-Tetrahydro-dibenzo[c, g][1, 2]diazocin vollzieht sich unter der Einwirkung von z. B. N. N'-Dicyclohexyl-carbodiimid in einem inerten organischen Lösungsmittel, vorzugsweise in einem solchen mit gutem Lösungsvermögen für die Ausgangsstoffe und das Reaktionsprodukt, aber schlechtem für N, N'-Dicyclohexylharnstoff, wie z. B. Dioxan, bereits bei Raumtemperatur.
Als Mittel für die Oxydation von Verbindungen mit durch S unterbrochenem Rest R'der allgemeinen Formel I zu entsprechenden Verbindungen mit durch SO, unterbrochenem Rest R eignet sich insbesondere

z. B. durch Kochen der Komponenten in Eisessig erfolgen und die Reduktion der erhaltenen Verbindungen der allgemeinen Formel VII z. B. mittels Wasserstoff in Gegenwart von Palladiumkohle bei zirka 20-60 C unter Atmosphärendruck in Methanol oder Äthanol.
Zur Kondensation mit 5, 6-Ma10nyl-5, 6-dihydro-11H-dibenzo[c, f][1, 2]diazepin bzw. mit 5, 6-Malonyl- 5, 6, 11, 12-tetrahydro-dibenzo[c, g][1, 2]diazocin geeignete reaktionsfähige Ester von Hydroxyverbindungen sind insbesondere die Halogenide. Am besten eignen sich Jodide, doch liefern auch Bromide in Gegenwart von weniger als der stöchiometrischen Menge Kaliumjodid annähernd gleiche Ergebnisse. Als säurebindendes Mittel dient beispielsweise Natriumhydroxyd in wässerigem oder wässerig-organischem Medium, z. B. in wasserhältigem Dioxan bei Temperaturen bis 100 C.
Ferner kann man beispielsweise als säurebindendes Mittel trockenes Kaliumcarbonat in Dimethylformamid als Reaktionsmedium verwenden und gegebenenfalls die Malonylverbindung zuerst mit dem säurebindenden Mittel in ein Salz überführen und letzteres mit einem reaktionsfähigen Ester einer Hydroxyverbindung der allgemeinen Formel VIII umsetzen.
Zahlreiche Ausgangsstoffe der allgemeinen Formeln II, III, IV und V sind bereits bekannt und weitere in analoger Weise herstellbar. Das 5, 6-Dihydro-llH-dibenzo [c, f] [1, 2] diazepin wird durch Reduktion des 11H-Dibenzo [cf] [1, 2] diazepins z. B. mittels Zinkstaub und Natronlauge erhalten. Der auch als 2, 2'Azo-diphenylmethan bezeichnete Ausgangsstoff ist seinerseits z. B. durch Reduktion des 0, 0'- Dinitro- diphenylmethans mit Lithiumaluminiumhydrid in Tetrahydrofuran zugänglich (vgl. Tetrahedron Letters No. 18, 5 (1959).
Das 5, 6, 11, 12-Tetrahydro-dibenzo[c, g][1, 2]diazocin wird nach H. Duval, Bull. Soc. Chim. [4], 7, 727 (1910) durch geeignete Reduktion des 2, 2'-Dinitro-dibenzyls erhalten.
Die neuen Verbindungen der allgemeinen Formel I bilden mit Alkalihydroxyden ziemlich leicht bis leicht lösliche einbasiche Salze. Lösungen derselben können direkt durch Auflösen der neuen Verbindungen in der berechneten Menge Alkalilauge, d. h. in Lösungen von Lithium-, Natrium- oder Kaliumhydroxyd hergestellt werden.
Die erfindungsgemäss hergestellten neuen Verbindungen bzw. deren Alkalisalze oder Salze mit organischen Basen, wie z. B. Dimethylaminoäthanol, Diäthylaminoäthanol oder Piperazin können durch Kombination mit geeigneten pharmazeutischen Trägerstoffen in Arzneimittel übergeführt werden. Die oben genannten Lösungen der Alkalisalze der neuen Verbindungen haben eine neutrale bis schwach alkalische Reaktion und eignen sich deshalb gut zur Injektion. Die Arzneimittel können auch Kapseln, Pulver, Tabletten u. a. zur peroralen Applikation geeignete Formen darstellen und können z.

der Wirkstoffe, d. h. der 5, 6-Malonyl-5, 6-dihydro-IIH-dibenzo [cf] [1, 2] diazepine, 5, 6-Malonyl-5, 6, 11, 12- tetrahydro-dibenzo[c, g][1, 2]diazocine und analoger Verbindungen mit substituiertem Malonrest, mit pharmazeutischen Trägerstoffen wie Stärke, Milchzucker, Stearinsäure, Talk, Magnesiumstearat usw. hergestellt werden. Sie können aber auch mit halbfesten Stoffen von geeignetem Schmelzbereich, z. B.
Kakaobutter, zu Suppositorien verarbeitet werden.
Die nachfolgenden Beispiele erläutern die Herstellung der neuen Verbindungen näher. Teile bedeuten darin Gewichtsteile, diese verhalten sich zu Vol.-Teilen wie g zu cm3. Die Temperaturen sind in Centigraden angegeben.
Beispiel 1 : 7 Teile 2, 2'-Azodiphenylmethan (HH-Dibenzo [c, f] [1, 2] diazepin) werden in 200 Vol.- Teilen Benzol gelöst und mit 36 Vol.-Teilen 4-n. Natronlauge unterschichtet. Die Luft wird mit Stickstoff verdrängt und das Reaktionsgemisch auf 700 erwärmt. Unter Rühren werden 7, 2 Teile Zinkstaub allmählich eingetragen. Nach rund 2 h verschwindet die orange Farbe. Das Gemisch wird in Stickstoffatmosphäre in einen Scheidetrichter filtriert und die Natronlauge abgetrennt. Die benzolische Lösung wird mit wasserfreiem Natriumsulfat getrocknet und stets noch in einer Stickstoffatmosphäre in einen Reaktionskolben filtriert.
Nach Abdestillieren von 50 Vol.-Teilen des Benzols werden 7, 8 Teile n-Butylmalonsäure-diäthylester und anschliessend eine Lösung von 0, 83 Teilen Natrium in 100 Vol.-Teilen abs. Äthanol zugegeben. In Gegenwart von 100 Vol.-Teilen Xylol wird unter Rühren und einer Badtemperatur von 150 C das Benzol und der vorhandene und entstehende Alkohol während 4 h abdestilliert. Die zurückgebliebene xylolische Lösung wird bei Raumtemperatur zuerst mit verdünnter Salzsäure, dann mit verdünnter Natriumhydrogencarbonatlösung und schliesslich mit Natriumcarbonatlösung ausgeschüttelt.
Die Natriumcarbonatlösung wird kongosauer gestellt, wobei Kristalle des 5, 6-n-Butylmalonyl-5, 6-dihydro- 11H-dibenzo[c, f][1, 2]-diazepins ausfallen, welche aus verdünntem Äthanol umkristallisiert bei 1740 schemel- zen.

eine Lösung von 1, 97 Teilen n-Butylmalonsäure-dichlorid in 10 Vol.-Teilen Chloroform zugetropft. Das Reaktionsgemisch wird 6 h bei 20-25'stehen gelassen und anschliessend in 20 Vol.-Teile Eiswasser gegossen. Die Chloroformphase wird zweimal mit verdünnter Salzsäure, dann zweimal mit verdünnter Natriumhydrogencarbonatlösung und zum Schluss dreimal mit verdünnter Natriumcarbonatlösung ausgeschüttelt. Die Natriumcarbonatauszüge werden vereinigt und mit Salzsäure kongosauer gestellt, wobei Kristalle ausfallen.
Diese werden abfiltriert und aus Äthanol umkristallisiert. Das so erhaltene 5, 6-n-

Beispiel 3 : 6, 68 Teile 5, 6, 11, 12-Tetrahydro-dibenzo [cg] [1, 2] diazocin werden in 33, 4 Vol.-Teilen Chloroform gelöst und dann 2, 83 Vol.-Teile Pyridin zugesetzt. Bei 0-5'wird unter Rühren eine Lösung von 6, 57 Teilen n-Butyl-malonsäure-äthylesterchlorid in 33, 4 Vol.-Teilen Chloroform zugetropft und das Reaktionsgemisch anschliessend 12 h bei Raumtemperatur stehen gelassen. Hierauf wird es dreimal mit verdünnter Natriumcarbonatlösung extrahiert, die Chloroformlösung über Natriumsulfat getrocknet und bei 11 Torr eingedampft. Der Rückstand wird in 250 Vol.-Teilen Benzol gelöst.
Die Lösung wird bei Raumtemperatur mit 0, 764 Teilen Natriumhydrid (als homogenisierte Suspension in Mineralöl) versetzt, wodurch sich das Gemisch auf zirka 60 0 erwärmt. Nach 12stündigem Stehenlassen bei Raumtemperatur wird das Gemisch mit verdünnter Natriumcarbonatlösung ausgeschüttelt, dieser Auszug mit Salzsäure kongosauer gestellt, und die ausgefällten Kristalle werden nach gründlichem Waschen mit Wasser aus Äthanol umkristallisiert, wobei man die im Beispiel 2 angegebene n-Butyl-Verbindung erhält.
Der Ringschluss kann z. B. auch wie folgt durchgeführt werden : Der Rückstand der Chloroformlösung wird mit 31, 8 Vol.-Teilen 1-n. NaOH und anschliessend mit soviel Äthanol (zirka 400 Vol.-Teile) versetzt, bis eine klare Lösung entsteht. Die Lösung wird 12 h bei Raumtemperatur stehen gelassen und dann zur Entfernung des Äthanols bei 11 Torr eingeengt. Der Rückstand wird mit Benzol extrahiert, die alkalischwässerige Phase abgetrennt, mit Salzsäure kongosauer gestellt und das ausgeschiedene Reaktionsprodukt wie oben aufgearbeitet.
Beispiel4 : 1, 60 Teile n-Butylmalonsäure und 2, 08 Teile 5, 6, l1, 12-Tetrahydro-dibenzo [c, g] [1, 2] diazocin werden in 50 Vol.-Teilen abs. Dioxan gelöst. Bei 20-25'werden auf einmal 4, 5 Teile Dicyclohexylcarbodiimid zugegeben, worauf das Reaktionsgemisch 12 h stehen gelassen wird. Der ausgeschiedene Dicyclohexyl harnstoff wird abgesaugt und das Filtrat im Vakuum eingedampft. Der Rückstand wird, wie in Beispiel 2 angegeben, aufgearbeitet, wobei man die dort angegebene n-Butylverbindung erhält.
In analoger Weise kann auch das 5,-6-Malonyl-5, 6, 11, 12-tetrahydro-dibenzo [c, g] [1, 2] diazocin vom Smp. 2030 hergestellt werden.
Beispiel 5 : 2, 56 Teile (P-Phenylsulfinyl)-äthyl-malonsäure in 50 Vol.-Teilen Dioxan werden mit 2, 08 Teilen 5, 6, 11, 12-Tetrahydro-dibenzo [cg] [1, 2] diazocin versetzt. Nach Zugabe von 4, 5 Teilen Di- cyclohexylcarbodiimid wird das Gemisch 12 h stehen gelassen. Nach Absaugen des ausgeschiedenen Dicyclohexylharnstoffes wird das Filtrat unter vermindertem Druck eingeengt und der Rückstand analog

Beispiel 2 aufgearbeitet. Man erhält so das 5,6-(ss-Phenylsulfinyl-äthyl)-malonyl-5,6,11,12-tetrahydro- dibenzo [c, g] [l, 2]-diazocin.

Eisessig werden mit 0, 68 VoL-Teilen Anisaldehyd versetzt und während l h auf 95-100'erhitzt. Nach dem Abkühlen wird die orange-rote Lösung bei Raumtemperatur in Gegenwart von 5%igem Palladium-
Kohle-Katalysator unter gewöhnlichem Druck hydriert. Nach Aufnahme der berechneten Menge Wasser- stoff schlägt die Farbe der Lösung nach hellgelb um. Der Katalysator wird abfiltriert und das Filtrat im Wasserstrahlvakuum möglichst weitgehend eingeengt. Der Rückstand wird aus Äthanol kristallisiert und ergibt das 5,6-(p-Anisyl-methyl)-malonyl-5,6,11,12-tetrahydro-dibenzo[c,g][1,2]diazocin.
Beispiel 7: 26,6 Teile 5,6-Malonyl-dihydro-11H-dibenzo[c,f][1,2]diazepin werden in 52, 2 ml 2-n.
NaOH gelöst, worauf 100 ml Dioxan und 11 ml n-Butyljodid zugegeben werden. Die entstandenen 2 Pha- sen werden mit einem Vibromischer bei einer Temperatur von 80-90 gut vermischt. Das Reaktionsge- misch wird dann bei 10 Torr stark eingeengt, der Rückstand zwischen 0, 5-n. Sodalösung und Äther ver- teilt, die wässerige Phase mit verdünnter HCI kongosauer gestellt und das ausgeschiedene Öl in Essigester aufgenommen. Der getrocknete Auszug wird eingeengt und der Rückstand in warmem 80% igen Äthanol gelöst. Die beim langsamen Erkalten ausgeschiedenen Kristalle werden mit 50 ml 80%igem Äthanol bei
Raumtemperatur während 20 min verrührt und abgesaugt.
Das Filtrat wird mit der oben erhaltenen Mutter- lauge vereinigt, unter reduziertem Druck eingeengt und der Rückstand in 100 m1 Äther gelöst. Diese Lösung wird durch eine zirka 3 cm dicke AIOg-Schicht (Aktivität I nach Brockmann) filtriert, das Filtrat eingedampft und der Rückstand aus 80%igem Äthanol umkristallisiert. Man erhält so das 5, 6-n-Butylmalonyl-5, 6- dihydro-11H-dibenzo[c,f][1,2]diazepin vom Smp. 174 .
