Verfahren zur Herstellung von neuen Benzimidazolderivaten
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von neuen Benzimidazolderivaten der allgemeinen Formel I :

Ar einen gegebenenfalls durch Halogenatome, die Nitrogruppe, den Trifluormethylrest, niedere Alkylreste und bzw. oder niedere Alkoxyreste substituierten Phenylrest oder einen Nyphtylrest, und
X und Y unabhängig voneinander Wasserstoff, ein Chloratom oder den Methylrest bedeuten, sowie von deren Salzen mit anorganischen oder organischen Säuren, pharmakologisch wertvolle Eigenschaften aufweisen. Die erfindungsgemäss herstellbaren Verbindungen üben erregende Wirkungen auf die glatte Muskulatur aus, insbesondere besitzen sie uteruskontrahierende Wirksamkeit.

weiteren Umsetzung, z. B. durch Behandlung mit Natriumamid, Lithiumhydrid, Kaliumamid, Natrium oder Kalium.
Ausgangsstoffe der allgemeinen Formel II sind bereits bekannt, und weitere in analoger Weise her-

erfindungsgemäss herstellbaren Verbindungen der allgemeinen Formel I Salze, wovon ein Teil in Wasser leicht löslich ist.
Das nachfolgende Beispiel soll die erfindungsgemässe Herstellung der neuen Verbindungen näher erläutern. Teile bedeuten darin Gew.-Teile, diese verhalten sich zu Vol.-Teilen wie g zu cm3. Die Temperaturen sind in Celsiusgraden angegeben.
Beispiel : a) 32 Teile o-Phenylendiamin werden in 95 Vol.-Teilen Äthanol gelöst und bei 00 langsam 36 Teile p-Toluylaldehyd zugetropft. Anschliessend versetzt man die Mischung mit 90 Teilen Nitrobenzol und erwärmt zum Sieden unter azeotropem Abdestillieren des Äthanols und des gebildeten Wassers. Wenn das Reaktionsgemisch den Siedepunkt des Nitrobenzols erreicht hat, wird es noch 15 min bei dieser Tem- peratur gehalten. Dann kühlt man es ab und versetzt es mit der äquivalenten Menge abs. äthanolischer Salzsäure. Die ausgeschiedenen Kristalle des 2- (p-Tolyl)-benzimidazol-hydrochlorid werden abgesaugt und mit wenig abs. Äthanol gewaschen. Das Hydrochlorid wird in Wasser suspendiert, die Suspension mit konz. Natriumcarbonatlösung versetzt und die ausgeschiedene Base abgesaugt.
Man wäscht sie mit Wasser und kristallisiert sie aus Äthanol um. Smp. 276-278 o. b) 6 Teile 2- (p- Tolyl) -benzimidazol werden in 200 V 01. - Teilen Toluol gelöst und mit einer Suspension von 1, 5 Teilen Natriumamid in Toluol versetzt. Das Reaktionsgemisch wird 18 h unter Rückfluss gekocht, dann auf 500 abgekühlt, die benzolische Lösung der Base aus 7 Teilen 2-Chlormethyl-2-imidazolinhydrochlorid zugegeben und das Ganze 2 h auf 50-60 C und anschliessend noch 2 h zum Kochen unter Rückfluss erhitzt. Nach dem Abkühlen wird das Reaktionsgemisch mit Wasser versetzt und der Benzolphase die basischen Anteile durch dreimaliges Ausschütteln mit 2-n. Essigsäure entzogen.
Die sauren Auszüge werden alkalisch gestellt und die ausgefallenen Kristalle aus Äthylacetat umkristallisiert. Das so erhaltene 1-[2'- Imidazolinyl- (2') -methyl-2- (p-tolyl) -benzimidazol schmilzt bei 198-199 .
In analoger Weise wird unter Verwendung von 32 Teilen Benzaldehyd in a) das 1-[2'-Imidazolinyl- (2')-methyl-2-phenyl-benzimidazol vom Smp. 175 , und unter Verwendung von 42, 4 Teilen p-Chlorbenzaldehyd in a) das 1-[2'- Imidazolinyl- (2') -methyl-2- (p-chlor-phenyl) -benzimidazol erhalten.
