Sobald sich eine Säuerung des Nährbodens einstellte, wurde mit Natronlauge auf den PH-Wert 7, 6 eingestellt.
Die normale Serie zeigte nach Beendigung des Wachstums einen Streptokinasewert von 48Christensen-
Einheiten Streptokinase pro crois, während die Polyoxyäthylensorbitanmonooleat enthaltende Kolbenreihe einen maximalen Streptokinasewert von 250 Christensen-Einheiten pro ems erreichte.
Beispiel 2: Es wurden 5 Kolben mit je 5 1 eines Mediums, bestehend aus 0, 5% Fleischextrakt,
1% Caseinpepton - pankreatisch verdaut - und 20/0 Traubenzucker steril gefüllt und auf einen PH-Wert von 7, 4 eingestellt. Eine weitere Reihe von 5 Kolben mit je 5 l enthielt das gleiche Medium mit einem
Zusatz von 0,05% Polyoxyäthylensorbitanmonooleat, ebenfalls eingestellt auf pH-Wert 7, 4. Nach Be- impfung mit dem Streptokokkenstamm H 46 A und laufender Neutralisation gebildeter Säure wurde die
Streptokinasebildung verfolgt.
Als Höchstwerte fanden sich bei dem einfachen Nährboden 10 Christensen-Einheiten pro cm in der das genannte oberflächenaktive Mittel enthaltenden Kulturflüssigkeit jedoch maximale Werte von 320
Christensen-Einheiten pro cm3.
Beispiel 3 : 1 Fermenter wurde mit 100 l eines Mediums, bestehend aus 0, 5 Fleischextrakt, 1% Caseinpepton-pankreatisch verdaut-und 2% Traubenzucker, und ein weiterer mit dem gleichen Nährmilieu, jedoch mit einem Zusatz von 0, 050/0 Polyoxyäthylensorbitanmonooleat, beimpft mit einer Kultur des Streptokokkenstammes H 46 A. Es wurde laufend gebildete Säure neutralisiert.
Die Ermittlung der maximalen Werte von gebildeter Streptokinase ergab, dass in der das genannte oberflächenaktive Mittel enthaltenden Kulturlösung 350 Christensen-Einheiten pro cm3 festzustellen waren, wogegen in dem Nährboden ohne den obigen Zusatz sich nur weniger als 40 Einheiten pro cm3 maximal finden liessen.
Beispiel 4: Es wurden 5 Kolben à 5 1 eines Nährmediums, das dem von Müller angegebenen (vgl.
World Hlth. Org. Techn. Rep. Ser. 61[1953], S.46) entsprach, mit 0, 05% Polyoxyäthylensorbitanmono- oleat versetzt ; 5 weitere Kolben mit dem gleichen Medium erhielten keinen solchen Zusatz.
Nach Beimpfung mit dem Streptokokkenstamm H 46 A und laufender Neutralisation gebildeter Säuren wurden in derdas oberflächenaktive Mittel enthaltenden Kulturflüssigkeit 530 Christensen-Einheiten pro cm3, im Nährmedíum ohne den obigen Zusatz 50 Christensen-Einheiten pro cm3 festgestellt.
Beispiel 5: 1 Fermenter wurde mit 250 1 einer Nährlösung - hergestellt nach Müller - beschickt, ein weiterer mit demselben Medium und Zusatz von 0, 05% Polyoxyäthylensorbitanmonooleat.
Die nach Ende der Bebrütungszeit und laufender Neutralisation erhaltenen Maximalwerte von Streptokinase betrugen mit Zusatz der oberflächenaktiven Substanz 800 Christensen-Einheiten Streptokinase pro cms, ohne den genannten Zusatz 80 Christensen-Einheiten pro cnA PATENTANSPRÜCHE :
1. Verfahren zur biologischen Gewinnung von Streptokinase, dadurch gekennzeichnet, dass man den verwendetenKulturlösungen nichtionogene, oberflächenaktive Substanzen in Konzentrationen von 0, 01 bis 0, 1% zusetzt und die Streptokinase in an sich bekannter Weise isoliert.

As soon as the nutrient medium became acidic, the pH was adjusted to 7.6 with sodium hydroxide solution.

Example 3: 1 fermenter was filled with 100 l of a medium consisting of 0.5 meat extract, 1% casein peptone - pancreatically digested - and 2% grape sugar, and another with the same nutrient medium, but with the addition of 0.050/0 polyoxyethylene sorbitan monooleate , inoculated with a culture of the streptococcal strain H 46 A. Acid formed continuously was neutralized.
The determination of the maximum values of streptokinase formed showed that 350 Christensen units per cm3 were found in the culture solution containing the surface-active agent mentioned, whereas in the nutrient medium without the above addition only less than 40 units per cm3 were found.
Example 4: 5 flasks of 5 l each of a nutrient medium similar to that given by Müller (cf.
World Hlth. Org. Techn. Rep. Ser. 61 [1953], p.46), mixed with 0.05% polyoxyethylene sorbitan monoleate; 5 further flasks with the same medium received no such addition.
After inoculation with the streptococcal strain H 46 A and ongoing neutralization of the acids formed, 530 Christensen units per cm3 were found in the culture liquid containing the surfactant, and 50 Christensen units per cm3 in the culture medium without the above addition.
Example 5: 1 fermenter was charged with 250 l of a nutrient solution - prepared according to Müller -, another with the same medium and the addition of 0.05% polyoxyethylene sorbitan monooleate.
The maximum streptokinase values obtained after the end of the incubation period and ongoing neutralization were 800 Christensen units of streptokinase per cms with the addition of the surfactant, and 80 Christensen units per cnA without the addition mentioned.
