Verfahren zur Herstellung von neuen Pyridylmethylaminen Die Erfindung betrifft ein Verfahren zur Herstellung von neuen Pyridylmethylaminen der Formel :

und deren Säureadditionssalzen.
Das erfindungsgemässe Verfahren besteht darin, dass man ein Pyridylmethyl-benzylamin mit einem Benzoyl-halogenid behandelt und das gebildete N-Benzyl-N- (pyridylmethyl)-benzamid reduziert und dass man gewünschtenfalls das gebildete Pyridylmethyl-dibenzylamin in ein Säureadditionssalz überführt.
Die Reduktion der N-Benzyl-N- (pyridylmethyl)-benzamide wird vorteilhafterweise mit Lithiumaluminiumhydrid durchgeführt. Als Benzoylhalogenide verwendet man im erfindungsgemässen Verfahren mit Vorteil die entsprechenden Chloride. Die Säureadditionssalze können durch Umsetzung der Pyridylmethyldibenzylamine mit etwa der 1-2fachen molaren Menge einer anorganischen oder organischen Säure hergestellt werden. Solche Säureadditionssalze sind z. B. Mineralsäuresalze, wie Hydrochloride, Hydrobromide, Hydrojodide, Phosphate, Sulfate, Nitrate, usw. ; Arylsulfonsäuresalze, wie Benzolsulfonate, Toluolsulfonate, usw. und andere Salze organischer Säuren, wie Citrate, Tartrate, Ascorbate, Salicylate, Acetate, Benzoate, usw.
Die Endprodukte des erfindungsgemässen Verfahrens eignen sich zur Behandlung von Magen-Darmgeschwüren. Sie können oral oder parenteral in Form der Base oder eines medizinisch verwendbaren Säureadditionssalzes verabreicht werden.
Die folgenden Beispiele erläutern das erfindungsgemässe Verfahren. Alle Temperaturen sind in Celsius-Graden angegeben.
Beispiel 1 : 100 g Pyridinaldehyd- (2) und 100 g Benzylamin werden in 1000 ml Äthanol gelöst und mit Wasserstoffgas bei 68 atm unter Verwendung von 15 g 10% Palladium enthaltender Kohle als Katalysator hydriert. Nach Vervollständigung der Hydrierung wird der Katalysator durch Filtration und der überschüssige Alkohol im Vakuum entfernt. Das erhaltene Öl wird im Vakuum destilliert, wobei bei 150-1550/3-4 mm das [Pyridyl- (2)-methyl]-benzylamin übergeht.
Man lässt 136 g [Pyridyl- (2)-methyl]-benzylamin und 110 g Benzoylchlorid in 750 ml Pyridin auf einem Wasserbad während 4 Stunden reagieren. Man entfernt das Pyridin im Vakuum und behandelt den Rückstand mit Wasser und sättigt mit Natriumbicarbonat. Das dabei gebildete N-Benzyl-N-[pyridyl- (2)-methyl]-benzamid wird mit Äther extrahiert. Der Ätherextrakt wird eingedampft, wobei ein fester Rückstand erhalten wird. Dieser Rückstand von N-Benzyl-N- [pyridyl- (2)-methyl]-benzamid wird aus Äthylacetat umkristallisiert und schmilzt bei 95-96 .
87 g N-Benzyl-N- [pyridyl- (2)-methyl]-benzamid in Form eines feinen Pulvers werden in eine Lösung von 37, 5 g Lithiumaluminiumhydrid in 2, 3 kg trockenem Äther eingetragen. Man rührt die Lösung während 14 Stunden, zersetzt hierauf das überschüssige Lithiumaluminiumhydrid mit 250 ml Äthylacetat und rührt die Mischung weiter während 2 Stunden. Hierauf setzt man 150 ml Wasser zu und rührt während einer weiteren Stunde. Die Mischung wird hierauf filtriert und der unlösliche Filterkuchen mit 2 1 heissem Benzol extrahiert. Die Äther- und Benzollösungen werden vereinigt und im Vakuum eingedampft.
Hierauf destilliert man das erhaltene Öl bei 220-225 /3-5 mm. Der Vorlauf (unter 220 ) und die bei 225-2300 übergehende Fraktion wird verworfen. Das erhaltene N- [Pyridyl- (2)-methyl]-dibenzylamin wird aus Äthylacetat umkristallisiert und schmilzt bei 47-48 .
Beispiel 2 : 212 g Benzaldehyd und 216 g Pyridyl- (3)-methylamin in 800 ml Äthanol werden mit Wasserstoff unter einem Druck von 34 atm bei 60 in Anwesentheit von 20 g 10% Palladium enthaltender Kohle als Katalysator reduziert. Der Katalysator wird durch Filtration entfernt und das Äthanol im Vakuum abdestilliert. Der erhaltene N- [Pyridyl- (3)-methyl]-benzylamin destilliert bei 178-180 /2 mm.

100 g N-[Pyridyl- (3) -methyl]-benzyla1nin werden in 500 ml Pyridin gelöst und 105 g Benzoylchlorid im Verlaufe einer halben Stunden zugesetzt. Hierauf erhitzt man die Lösung während 4 Stunden am Rückfluss, kühlt und versetzt mit 100 ml Wasser. Die Lösung wird hierauf zur Trockne eingedampft, der Rückstand in 1, 51 Eiswasser dispergiert, mit 200 ml konzentriertem Ammoniumhydroxyd alkalisch gestellt und 5mal mit je 250 ml Äther extrahiert. Die Ätherextrakte werden vereinigt, mit 250 ml Eiswasser gewaschen und hierauf auf 200 ml eingeengt. Aus dem Äther kristallisiert N-Benzyl-N- [pyridyl- (3)methyls-benzamid mit dem Schmelzpunkt von 75-76 .
Man löst 30 g Lithiumaluminiumhydrid in 1250 g trockenem Äther und kühlt die Lösung im Eisbad.
Zu dieser Lösung setzt man in kleinen Anteilen 70 g N-Benzyl-N- [pyridyl- (3)-methyl]-benzamid im Verlaufe einer halben Stunde zu. Nach Rühren während 2 Stunden lässt man die Mischung über Nacht stehen.
Hierauf zersetzt man das überschüssige Lithiumaluminiumhydrid durch Zusatz von 200 ml Äthylacetat. Der Reduktionskomplex wird durch Zusatz von 100 ml Eiswasser zerstört. Man trennt den Äther vom unlöslichen Material und extrahiert den Filterkuchen einmal mit 200 ml heissem Benzol. Die Benzol,' Äthylacetat/Äther-Lösung wird durch Destillation im Vakuum eingeengt und das Öl hierauf in einer Vigreaux-Kolonne destilliert. Die bei 200-212012 mm übergehende Fraktion wird gesammelt und in 45 ml Äthylacetat gelöst. Die Äthylacetatlösung wird in einem Kohlensäure-Aceton-Bad gekühlt, wobei Kristallisation eintritt.
Nach rascher Filtration und Waschen mit eiskalter Skellysolve B (Siedebereich 60-700) erhält man reines kristallisiertes N-[Pyridyl- (3) -methyl]-dibenzylamin vom Schmelzpunkt 60-61 o.

The process according to the invention consists in treating a pyridylmethylbenzylamine with a benzoyl halide and reducing the N-benzyl-N- (pyridylmethyl) benzamide formed and, if desired, converting the pyridylmethyl-dibenzylamine formed into an acid addition salt.
The reduction of the N-benzyl-N- (pyridylmethyl) -benzamides is advantageously carried out with lithium aluminum hydride. The corresponding chlorides are advantageously used as benzoyl halides in the process according to the invention. The acid addition salts can be prepared by reacting the pyridylmethyldibenzylamines with about 1-2 times the molar amount of an inorganic or organic acid. Such acid addition salts are e.g. B. mineral acid salts such as hydrochlorides, hydrobromides, hydroiodides, phosphates, sulfates, nitrates, etc .; Arylsulfonic acid salts such as benzenesulfonates, toluenesulfonates, etc. and other salts of organic acids such as citrates, tartrates, ascorbates, salicylates, acetates, benzoates, etc.

Example 1: 100 g of pyridine aldehyde (2) and 100 g of benzylamine are dissolved in 1000 ml of ethanol and hydrogenated with hydrogen gas at 68 atm using 15 g of 10% palladium-containing carbon as a catalyst. After the hydrogenation is complete, the catalyst is removed by filtration and the excess alcohol is removed in vacuo. The oil obtained is distilled in vacuo, the [pyridyl (2) methyl] benzylamine passing over at 150-1550 / 3-4 mm.
136 g of [pyridyl (2) methyl] benzylamine and 110 g of benzoyl chloride in 750 ml of pyridine are allowed to react on a water bath for 4 hours. The pyridine is removed in vacuo and the residue is treated with water and saturated with sodium bicarbonate. The N-benzyl-N- [pyridyl- (2) -methyl] -benzamide thus formed is extracted with ether. The ether extract is evaporated to give a solid residue. This residue of N-benzyl-N- [pyridyl- (2) -methyl] -benzamide is recrystallized from ethyl acetate and melts at 95-96.
87 g of N-benzyl-N- [pyridyl- (2) -methyl] -benzamide in the form of a fine powder are added to a solution of 37.5 g of lithium aluminum hydride in 2.3 kg of dry ether. The solution is stirred for 14 hours, then the excess lithium aluminum hydride is decomposed with 250 ml of ethyl acetate and the mixture is stirred for a further 2 hours. 150 ml of water are then added and the mixture is stirred for a further hour. The mixture is then filtered and the insoluble filter cake is extracted with 2 liters of hot benzene. The ether and benzene solutions are combined and evaporated in vacuo.
The oil obtained is then distilled at 220-225 / 3-5 mm. The forerun (below 220) and the fraction passing over at 225-2300 are discarded. The N- [pyridyl- (2) -methyl] -dibenzylamine obtained is recrystallized from ethyl acetate and melts at 47-48.
Example 2 212 g of benzaldehyde and 216 g of pyridyl (3) methylamine in 800 ml of ethanol are reduced with hydrogen under a pressure of 34 atm at 60 in the presence of 20 g of 10% palladium-containing carbon as a catalyst. The catalyst is removed by filtration and the ethanol is distilled off in vacuo. The N- [pyridyl- (3) -methyl] -benzylamine obtained distills at 178-180 / 2 mm.

100 g of N- [pyridyl- (3) -methyl] -benzyla1nin are dissolved in 500 ml of pyridine and 105 g of benzoyl chloride are added over the course of half an hour. 70 g of N-benzyl-N- [pyridyl- (3) -methyl] -benzamide are added in small portions to this solution over the course of half an hour. After stirring for 2 hours, the mixture is left to stand overnight.
The excess lithium aluminum hydride is then decomposed by adding 200 ml of ethyl acetate. The reduction complex is destroyed by adding 100 ml of ice water. The ether is separated from the insoluble material and the filter cake is extracted once with 200 ml of hot benzene. The benzene, ethyl acetate / ether solution is concentrated by distillation in vacuo and the oil is then distilled in a Vigreaux column. The fraction passing over at 200-212012 mm is collected and dissolved in 45 ml of ethyl acetate. The ethyl acetate solution is cooled in a carbonic acid-acetone bath, whereupon crystallization occurs.
After rapid filtration and washing with ice-cold Skellysolve B (boiling range 60-700), pure crystallized N- [pyridyl- (3) -methyl] -dibenzylamine with a melting point of 60-61 o.
