Abstract

Procedimiento de seleccion implementada por ligandos en exceso que comprende un método estocástico basado en la probabilidad de existencia de un alto numeros de ligandos posibles contra un mismo blanco. El objetivo de este protocolo es el de proveer bibliotecas con concentraciones de ligandos compatibles a las concentraciones de equilibrio en solucion. La presente provee ligandos consistentes en oligonucleotidos y cada uno de ellos posee una estructura definida por su secuencia, capaz de reconocer el blanco utilizado para la seleccion. La ventaja de este método sobre los otros es la capacidad de obtener una gran cantidad de ligandos específicos en un solo paso y en unas pocas horas. Cuando se utilizan proteínas como blancos y bibliotecas combinatorias de oligonucleotidos, los ligandos obtenidos son de una afinidad del rango nanomolar y son sensibles a la conformacion de la misma. Además, este método permite reconocer blancos diferenciales de mezclas complejas, sin necesidad de sustancias puras.
