Abstract
La presente se refiere a procesos para epimerizar alcoholes de compuestos que tienen un sustituyente hidroxilo en un carbono alílico asimétrico, tales como compuestos utiles para la síntesis de análogos de la vitamina D en donde el sustituyente hidroxilo epimérico está en la posicion 24. La presente se refiere además a métodos para producir productos intermedios utiles para la síntesis de calcipotriol por medio de dichos procesos de epimerizacion. Reivindicacion 1: Un método para epimerizar un epímero o mezcla epimérica de un compuesto que comprende una cadena lateral de estructura general A y/o B, en la pocsicion del átomo de carbono al que se unen el grupo hidroxi y R3; poniendo en contacto dicho epímero o mezcla epimérica con un ácido en presencia de agua; en donde R3 es alquilo, alquenilo, alquinilo o cicloalquilo, opcionalmente sustituido con uno o más sustituyentes seleccionados del grupo que consiste en alquilo, alcoxicarbonilo, halogeno, hidroxi, metoxi y etoxi; en donde el carbono marcado con un asterisco está unido con un enlace simple al átomo de carbono C-17 de un fragmento de un análogo de la vitamina D; o en donde el carbono marcado con un asterisco está unido con un enlace simple a un fragmento de un precursor para la síntesis de un análogo de la vitamina D en su posicion análoga C-17.This refers to processes for epimerizing alcohols of compounds having a hydroxyl substituent on an asymmetric allyl carbon, such as compounds useful for the synthesis of analogs of vitamin D wherein the epimeric hydroxyl substituent is at position 24. Claim 1: A method for epimerizing an epimer or epimeric mixture of a compound comprising a side chain of general structure A and / or B, in the position of the carbon atom to which the hydroxy group and R3 are attached; contacting said epimer or epimeric mixture with an acid in the presence of water; wherein R3 is alkyl, alkenyl, alkynyl or cycloalkyl, optionally substituted with one or more substituents selected from the group consisting of alkyl, alkoxycarbonyl, halogen, hydroxy, methoxy and ethoxy; wherein the carbon marked with an asterisk is attached with a single bond to the C-17 carbon atom of a fragment of a vitamin D analog; or wherein the carbon marked with an asterisk is attached with a single bond to a fragment of a precursor for the synthesis of a vitamin D analog in its analogous position C-17.
