Abstract

Método para producir un precursor análogo de insulina humana y cepa de levadura metilotrófica que lo produce, más específicamente el método comprende preparar un inóculo de una cepa de levaduras metilotrófica recombinantes establemente trasformada por al menos una copia de un primera construcción de ADN y al menos una segunda construcción de ADN, donde dichas construcciones de ADN son capaces de dirigir la expresión y liberación al medio de un precursor análogo de insulina humana, y donde dichas construcciones de ADN comprenden la SEQ ID N° 5 o la SEQ ID N° 12, o variantes de las mismas que codifican un precursor análogo de insulina humana; sembrar el inóculo de la etapa en un fermentador haciendo crecer la cepa en un proceso en lote que comprende una primer etapa en presencia de una fuentes de carbono hasta que dicha fuente de carbono ha sido totalmente consumida y una segunda etapa de lote alimentado que comprende la adición de la dicha fuente de carbono y metanol; y asilar un precursor análogo de insulina del medio de cultivo de dicha cepa.
