Abstract
Método para producir un precursor análogo de insulina humana y cepa de levadura metilotrófica que lo produce, más específicamente el método comprende preparar un inóculo de una cepa de levaduras metilotrófica recombinantes establemente trasformada por al menos una copia de un primera construcción de ADN y al menos una segunda construcción de ADN, donde dichas construcciones de ADN son capaces de dirigir la expresión y liberación al medio de un precursor análogo de insulina humana, y donde dichas construcciones de ADN comprenden la SEQ ID N° 5 o la SEQ ID N° 12, o variantes de las mismas que codifican un precursor análogo de insulina humana; sembrar el inóculo de la etapa en un fermentador haciendo crecer la cepa en un proceso en lote que comprende una primer etapa en presencia de una fuentes de carbono hasta que dicha fuente de carbono ha sido totalmente consumida y una segunda etapa de lote alimentado que comprende la adición de la dicha fuente de carbono y metanol; y asilar un precursor análogo de insulina del medio de cultivo de dicha cepa. Reivindicación 2: El método de acuerdo con la reivindicación 1, caracterizado porque el precursor análogo de insulina es el polipéptido B(1-30)-Arg-Arg-A(1-20)-X1, en donde B(1-30) representa la cadena B de la insulina humana, A(1-20) corresponde a la cadena A de la insulina humana en donde se da el reemplazo en la posición 21 Asn por X1, en donde X1 es seleccionado del grupo de aminoácidos comprendido por Gly, Ala, Val, Thr, Ser y Phe.Method for producing a human insulin analog precursor and methylotrophic yeast strain that produces it, more specifically the method comprises preparing an inoculum of a recombinant methylotrophic yeast strain stably transformed by at least one copy of a first DNA construct and at least one second DNA construct, wherein said DNA constructs are capable of directing expression and release to the medium of a human insulin analog precursor, and wherein said DNA constructs comprise SEQ ID No. 5 or SEQ ID No. 12, or variants thereof encoding a human insulin analog precursor; sowing the inoculum of the stage in a fermenter by growing the strain in a batch process comprising a first stage in the presence of a carbon sources until said carbon source has been fully consumed and a second stage of fed batch comprising the addition of said source of carbon and methanol; and isolating an insulin analog precursor from the culture medium of said strain. Claim 2: The method according to claim 1, characterized in that the insulin analog precursor is polypeptide B (1-30) -Arg-Arg-A (1-20) -X1, wherein B (1-30) represents the B chain of human insulin, A (1-20) corresponds to the A chain of human insulin where replacement occurs at position 21 Asn by X1, where X1 is selected from the group of amino acids comprised by Gly , Ala, Val, Thr, Ser and Phe.
