Abstract
Compuestos derivados de 7-(4-fluorofenil)imidazo[1,2-b][1,2,4]triazina, sustituidos en la posición meta del anillo fenilo con un anillo fenilo o piridinilo opcionalmente sustituido con flúor o ciano, son ligandos selectivos para receptores GABAA, en particular tienen buena afinidad para la subunidad alfa2 y/o alfa3 de los mismos, y son en consecuencia de beneficio en el tratamiento y/o prevención de condiciones adversas del sistema nervioso central, incluyendo ansiedad y convulsiones. Se describen también composiciones farmacéuticas y el uso de los compuestos en la preparación de medicamentos. Dichos compuestos son de fórmula (1), en donde: Z representa un anillo fenilo o piridinilo, opcionalmente sustituido con uno o dos sustituyentes seleccionados entre flúor y ciano; y R1 representa hidroxipropilo, fluoropropilo o trifluorometilo.Compounds derived from 7- (4-fluorophenyl) imidazo [1,2-b] [1,2,4] triazine, substituted at the meta position of the phenyl ring with a phenyl or pyridinyl ring optionally substituted with fluorine or cyano, are ligands selective for GABAA receptors, in particular they have good affinity for the alpha2 and / or alpha3 subunit thereof, and are consequently of benefit in the treatment and / or prevention of adverse central nervous system conditions, including anxiety and seizures. Pharmaceutical compositions and the use of the compounds in the preparation of medicaments are also described. Said compounds are of formula (1), wherein: Z represents a phenyl or pyridinyl ring, optionally substituted with one or two substituents selected from fluorine and cyano; and R1 represents hydroxypropyl, fluoropropyl or trifluoromethyl.
