Abstract
Un método para la preparacion de citalopram que comprende los pasos de: a)hacer reaccionar un compuesto de formula IV donde R1 es H o alquilcarbonilo C1-6con un reactivo de Grignard de 4-halogeno-fluorfenilo, b)hacer reaccionar el compuestoresultant e de formula V donde R1 es, segun se define precedentemente,con un reactivo de Grignard de 3-halogeno-N,N-dimetilpropilamina; c)efectua el cierre de anillos del compuesto resultante de la formula VI donde R1 es segun sedefineprecedentemente, y d) co nvertir el compuesto resultante de formula VII donde R1 es, segun se define precedentemente, para el correspondiente 5-cianoderivado, es decir, citalopram, el cual es separado como base o sal farmacéuticamente aceptable delmismo; y una composicion f armacéutica antidepresiva quecomprende citalopram fabricado mediante el método.A method for the preparation of citalopram comprising the steps of: a) reacting a compound of formula IV where R 1 is H or C 1-6 alkylcarbonyl with a 4-halogen-fluorophenyl Grignard reagent, b) reacting the resulting compound of formula V where R1 is, as defined above, with a Grignard reagent of 3-halogen-N, N-dimethylpropylamine; c) effect the ring closure of the resulting compound of formula VI where R1 is as seemingly insignificant, and d) convert the resulting compound of formula VII where R1 is, as defined above, for the corresponding 5-cyanoderivative, that is, citalopram , which is separated as a base or pharmaceutically acceptable salt thereof; and an antidepressant pharmaceutical composition comprising citalopram manufactured by the method.
