Abstract
Se describe una desaturasa de ácido graso capaz de catalizar la conversion de ácido oleico a ácido linoleico, ácido linoleico a ácido gama-linolenico oácido alfa-linolénico a ácido estearidonico, la cual ha sido codificada por una secuencia denucleo tidos seleccionada del grupo que consiste en los NO ID SEC:1, 3, 19, 21, 23, 25, 27, 28, 29, 30, 31, 32 y 33; y la cual comprende una secuencia de polipéptidos seleccionada del grupo que consiste en los NO. ID. SEC:2, 4, 5, 6, 7, 8, 9, 10,11, 20, 22 , 24, 26,34, 35, 36, 37, 38, 39, 4O y una secuencia de polipéptidos que comprende los residuos de aminoácidos 50-53, 39-43,172-176, 204-213 y 390-402 del NO. ID. SEC: 2; una célula huésped empleada para expresar la desaturasa de ácidograso preferent emente una célula Saccharomycesde levadura; y el uso de una célula huésped que expresa la desaturasa de ácido graso en un método destinado a la produccion de ácido gama-linolenico en uncultivo de células huesped, que comprende:desarrollar un cultivo huésped que tenga una multiplicidad de células huésped recombinantes, donde las células de lalevadura o un ancestro de las mencionadas células de la levadura se transformaron con un vector que comprende ADN fungal que codificaun polipéptido el cual convierte el ácido linolenico a ácido gama-linoleico, donde el citado ADN está operativamente asociado con una secuencia de control de expresion funcional enlas mencionadas células huésped, en condiciones por las cuales seexpresa dicho ADN, donde el ácido gama-linolenico se produce a partir de ácido linoleico enel mencionado cultivo huésped. SEC: 2; a host cell used to express the fatty acid desaturase preferentially a Saccharomyces yeast cell; and the use of a host cell expressing fatty acid desaturase in a method intended for the production of gamma-linolenic acid in a host cell culture, comprising: developing a host culture having a multiplicity of recombinant host cells, where yeast cells or an ancestor of the aforementioned yeast cells were transformed with a vector comprising fungal DNA encoding a polypeptide which converts linolenic acid to gamma-linoleic acid, where said DNA is operatively associated with a control sequence of functional expression in said host cells, under conditions whereby said DNA is expressed, where gamma-linolenic acid is produced from linoleic acid in said host culture. The use of a host cell that expresses stearidonic acid desaturase in a mouse-grown culture is also described; the use of a host cell expressing fatty acid desaturase in a method intended for the production of linoleic acid in a host cell culture; an isolated nucleic acid which encodes the acid fatty acid desaturase, the use of the host cell in a method intended to obtain the altered biosynthesis of the long chain polyunsaturated fatty acid; an oil or a fraction thereof has been produced according to this last method;
