Abstract
Un proceso para la obtencion de sertindol, el cual es una droga antipsicotica. Dicho proceso comprende la preparacion de N-(4-fluorfenil)-N-(2-carboxi-4-clorofenil)glicina haciendo reaccionar una sal de metal alcalino del ácido 2,5-dic lorobenzoico con una sal de metal alcalino de N-(4-fluorfenil)glicina en un medio alcalino acuoso en presencia de un catalizador de cobre; ciclando la N-4-(flourfenil)-N-(2-carboxi-4-clorofenil)glicina de modo deformar el correspondiente 3-acetoxi -indol; reduciendo el 3-acetoxi-indol y luego eliminando el H2O de modo de obtener 5-cloro-1-(4-fluorofenil)indol, elcual se hace reaccionar con 4-piperidona, en una mezcla de ácido acético y HCI concentrado; reduciendo el 5-cloro-1-(4- fluorfen il)-3-(1,2,3,6-tetrahidropiridin-4-il)indol y haciendo reaccionar este compuesto con 1-(2- cloroetil-2-imidazolidinona, a fin de obtener sertindol. En forma alternativa,el 5-cloro-1-(4-fluorofenil)-3-(1,2,3,6-tetrahidropiridin-4)indolpuede prime ro reaccionar con 1-(2-cloroetil)-2-imidazolidinona y luego se puede reducir elproducto a fin de obtener sertindol. Este proceso utiliza reactivos y solventes que son utiles y pueden utilizarse para la fabricacion a gran escala.Además,se obtienen bu enos rendimientos totales.A process for obtaining sertindole, which is an antipsychotic drug. Said process comprises the preparation of N- (4-fluorphenyl) -N- (2-carboxy-4-chlorophenyl) glycine by reacting an alkali metal salt of 2,5-dic lorobenzoic acid with an alkali metal salt of N- (4-fluorophenyl) glycine in an aqueous alkaline medium in the presence of a copper catalyst; cycling the N-4- (flourphenyl) -N- (2-carboxy-4-chlorophenyl) glycine so as to deform the corresponding 3-acetoxy-indole; reducing 3-acetoxy-indole and then removing H2O so as to obtain 5-chloro-1- (4-fluorophenyl) indole, which is reacted with 4-piperidone, in a mixture of acetic acid and concentrated HCI; reducing 5-chloro-1- (4- fluorphenyl) -3- (1,2,3,6-tetrahydropyridin-4-yl) indole and reacting this compound with 1- (2- chloroethyl-2-imidazolidinone, in order to obtain sertindole. Alternatively, 5-chloro-1- (4-fluorophenyl) -3- (1,2,3,6-tetrahydropyridin-4) indole can first react with 1- (2-chloroethyl) -2-imidazolidinone and then the product can be reduced to obtain sertindole.This process uses reagents and solvents that are useful and can be used for large-scale manufacturing.In addition, good total yields are obtained.
