Abstract
Se describe un proceso de cultivo masivo de células de mamífero recombinantes, productoras de EPO. El proceso productivo sigue diferentesetapas de expansion, partiendo de células viables conservadas congeladas (Master y Workingban ks). Posteriormente, se pasa a una etapa productiva en laque se utilizan medios de cultivo esencialmente formulados para evitar el agregado de suplementos proteicos. El sistema consigue obtener una elevadaproductividad de EPO que seencuentra c on alta pureza en el medio de cultivo cosechado. La suplementacion con insulina es uno de los aspectos clave delmétodo. El sobrenadante de cultivo es finalmente concentrado para obtener mayor concentracion de EPO en una formaapropiada para ser utilizada tal cualse obtiene o purificada ulteriormente para los usos que así lo requieran.A massive culture process of recombinant mammalian cells, producing EPO, is described. The production process follows different stages of expansion, starting from viable cells preserved frozen (Master and Workingbanks). Subsequently, it goes to a productive stage in which culture media essentially formulated to avoid the addition of protein supplements are used. The system achieves a high EPO productivity that is found with high purity in the harvested culture medium. Insulin supplementation is one of the key aspects of the method. The culture supernatant is finally concentrated to obtain a higher concentration of EPO in a suitable form to be used as it is obtained or further purified for uses that require it.
