Abstract

Ambos plásmidos tienen como unicos elementosde control de la expresion alpromotor te mprano de virus SV40 y su senal de poliadenilacion. Este sistema permite efectuar una doble seleccion de lascélulas recombinantes, ya que confieren resistencia a geneticina y a metotrexate (MTX). Una vez efectuada laseleccion se efect uo la amplificaciongenética con MTX. Con un inmunoensayo específico para EPO se efectuo el testeo de la capacidad productiva de las células recombinantes elegidas.The gene coding for human erythropoietin (EPO) was obtained from human genomic DNA. This gene does not include genetic elements homologous to 5 of the first translated ATG codon. Eukaryotic cells were cloned into an expression vector. Hamster ovarian cells (CHO) were cotransfected with this vector and another expression plasmid for dehydrofolatoreductase (DHFR). Both plasmids have as their only elements to control the early expression of the SV40 virus and its polyadenylation signal. This system allows a double selection of the recombinant cells, since they confer resistance to geneticin and methotrexate (MTX). Once the selection was made, the genetic amplification was performed with MTX. With a specific immunoassay for EPO, the productive capacity of the selected recombinant cells was tested.
