Definitions
- the present application relates to the fields of pharmaceutical chemistry, biochemistry, and medicine.
- it relates to crystalline forms of the compound of Formula I and methods of making and using the same.
- the thyroid hormones play a critical role in growth, development, metabolism, and homeostasis. They are produced by the thyroid gland as thyroxine (T4) and 3,5,3'-triiodo-L-thyronine (T3). T4 is the major secreted form in humans and is enzymatically deiodinated by deiodinases to the more active form, T3, in peripheral tissues. THs exert their action by interacting with thyroid hormone receptors (TRs), which belong to the nuclear hormone receptor superfamily, and regulate the transcription of target genes.
- TRs thyroid hormone receptors
- TRs are expressed in most tissues and exist as two isoforms (TRa and TKb). Tissue distribution studies, mouse knockout studies, and evaluation of patients with resistance to thyroid hormone (RTH) syndrome have established that TRa is the predominant isoform in the heart and regulates most cardiac functions, while the TKb isoform predominates in the liver and the pituitary' and regulates cholesterol metabolism and thyroid stimulating hormone (TSH) production, respectively.
- RTH thyroid hormone
- TRJ3 agonists may be used for the treatment of adrenoleukodystrophy (ALD) and lipid disorders such as hypercholesterolemia and fatty liver diseases, for example non-alcoholic fatty liver disease (NAFLD), non-alcoholic steatohepatitis (NASH), and glycogen storage disease (GSD).
- ALD adrenoleukodystrophy
- NASH non-alcoholic fatty liver disease
- NASH non-alcoholic steatohepatitis
- GSD glycogen storage disease

The location of the peaks along the x- axis (° 2Q) in both the figures and the tables were determined using proprietary software and rounded to one or two significant figures after the decimal point based upon the above criteria. Peak position variabilities are given to within ⁇ 0.2° 2Q. For d-space listings, the wavelength used to calculate d-spaeings was 1.5405929 A, the Cu-Ka ⁇ wavelength.
