Заявляемое изобретение относится к области медицины, в частности к способам хранения тромбоцитов путем криоконсервирования, т.е. замораживания в жидком азоте, и может применяться в отделениях реанимации, гематологии, в онкологических клиниках и т.д. The claimed invention relates to medicine, in particular to methods for storing platelets by cryopreservation, i.e. freezing in liquid nitrogen, and can be used in intensive care units, hematology, oncology clinics, etc.
Известны способы криоконсервирования богатой тромбоцитами плазмы, заключающиеся в том, что плазму помещают в пластиковый контейнер, располагают этот контейнер между двумя металлическими пластинами и замораживают в жидком азоте или его парах. Перед замораживанием в плазму вводят криоконсервант, который предупреждает разрушение клеток и утрату их функциональной активности в процессах замораживания, хранения и размораживания. В качестве криоконсерванта обычно используют диметил сульфоксид (ДМСО). Однако после размораживания лишь 40-60% тромбоцитов сохраняют функциональную активность. Это является причиной низкой клинической эффективности плазмы, что естественно не отвечает запросам практической медицины. Кроме того, присутствие криоконсерванта вызывает тяжелые осложнения у больных. Контейнер располагают между двумя металлическими пластинами и заливают жидким азотом. При этом температура охлаждения составляет -120oC, а скорость охлаждения 8-10oC/мин. Время замораживания составляет от 45 мин до 1 часа. Плазму хранят при -120oC до трех лет. Плазму размораживают, используя водяную баню при температуре 37oC. Технический результат выражается в том, что предлагаемые режимы ведения процессов замораживания и размораживания плазмы позволяют сохранить 74-98%, т. е. в среднем 92% функционально активных тромбоцитов. Другим техническим результатом является устранение токсичности плазмы, т.к. предлагаемые режимы способа позволяют сохранить вышеуказанное количество полноценных тромбоцитов без использования токсичных криоконсервантов. Поставленная задача достигается тем, что в известном способе криоконсервирования богатой тромбоцитами плазмы, заключающемся в том, что плазму помещают в контейнер, располагают его между двумя металлическими пластинами, после чего последовательно подвергают процессам замораживания, хранения и размораживания, новым является то, что процесс замораживания плазмы ведут при скорости 2-3oC/сек.The specified technical result is provided by the totality of the claimed features. Заявляемые режимы замораживания и размораживания были получены опытным путем. При этом скорость замораживания примерно равнялась скорости размораживания. После этих процедур проводили исследования сохранения количества тромбоцитов в плазме. Было выявлено, что наибольшее число клеток сохраняется при скорости замораживания 2-3oC/сек и при скорости размораживания 1,5-2oC/сек (см. таблицу 1).The inventive modes of freezing and thawing were obtained experimentally. Для изучения влияния найденных опытным путем режимов замораживания и размораживания на функциональную активность тромбоцитов были проведены следующие исследования. Брали две пробы крови от одного донора. Путем центрифугирования крови получали богатую тромбоцитами плазму. Плазму 1-ой пробы помещали на хранение в водяной термостат при температуре 37oC (контрольная проба). Плазму 2-ой пробы (исследуемая проба) замораживали и размораживали по предлагаемому способу. После размораживания плазму вновь смешивали с эритроцитами (то же самое делали с контрольной пробой) и проводили сравнительные исследования 1-ой и 2-ой проб на функциональную активность тромбоцитов. Для этого пластикатный контейнер "Blood Bag CPD 450", содержащий донорскую богатую тромбоцитами плазму (число тромбоцитов в ней 432 • 109/л) поместили между двумя металлическими пластинами. Таким образом обеспечивали ровность слоя плазмы при замораживании. После этого контейнер помещали в емкость и заливали жидким азотом. Температура жидкого азота составляла -196oC, скорость замораживания ≈2,2oC/сек. Контейнер с плазмой находился в среде жидкого азота в течение 20 суток. На 20-е сутки произвели размораживание плазмы. Для этого контейнер поместили в водяной термостат, в котором температура была на уровне 90-100oC. При этой температуре плазма находилась в течение 62 сек до начала ее таяния. Скорость размораживания составляла ≈2oC/сек. Затем контейнер погружали в другой водяной термостат, температура которого была 37oC, и выдерживали в нем плазму до полного размораживания. Количество тромбоцитов в плазме после размораживания составило 417 • 109/л, т.е. 97% от исходного. Функциональная активность тромбоцитов, т.е. их прокоагулянтная функция, активировалась по сравнению с исходной (после предварительной инкубации с каолином время свертывания плазмы укоротилось на 19% исходного).Example.
