Abstract
The present invention discloses an extraction method of seabuckthorn leaves extract, including: step one, drying seabuckthom leaves, crushing it, and sieving it to obtain seabuckthom leaves powder, step two, performing subcritical extraction of the seabuckthom leaves powder, and extracting it for 3 times to obtain an extracting solution, step three, separating the extracting solution to obtain seabuckthom leaves extract. The extraction method of the present invention has simple process, rapid extraction speed, mild extraction conditions, and high extraction yield, and solves the problems of long extraction time, high extraction temperature, low quality of extract and high cost in prior extraction technologies.
Description
EXTRACTION METHOD OF SEABUCKTHORN LEAVES EXTRACT
The present invention relates to the field of the extraction of extract. More particularly, the present invention relates to an extraction method of seabuckthorn leaves extract.
Seabuckthorn leaves are extremely enriched with nutrients and physiologically active substances, mainly including proteins, polysaccharides, organic acids, alkaloids, flavonoids, amino acids, carotenoids, chlorophyll, trace elements, etc., which can regulate blood lipids, inhibit bacteria, scavenge free radicals, remove sputum, relieve cough and relieve asthma, etc., and have very significant medicinal value. Extract refers to perfumery products obtained by extracting perfume plant tissues (such as flowers, leaves, branches, stems, barks, roots, fruits, etc.) that do not contain exudates with organic solvents, which is enriched with nutrients and active substances, and 1s an ideal base material for the separation of active ingredients.
Currently, the extraction methods of extract include traditional extraction and supercritical extraction. However, the traditional extraction has complicated process, long extraction time, high extraction temperature, low extraction efficiency, and low quality of extract, and the supercritical extraction has high production cost and cannot meet the requirements of markets.
An object of the present invention is to solve at least above problems, and to provide, at least, the advantages that will be described later.
An another object of the present invention is to provide an extraction method of seabuckthorn leaves extract, which adopts subcritical extraction with 1,1,1,2-tetrafluoroethane, solves the problems of long extraction time, high extraction temperature, low quality of extract and high cost in prior extraction technologies, and provides high-quality samples for the subsequent separation and purification of active 1 components.
In order to achieve the above object, the present invention provides an extraction method of seabuckthorn leaves extract, including the following steps of: step one, drying seabuckthorn leaves, crushing it, and sieving it to obtain seabuckthorn leaves powder; step two, performing subcritical extraction of the seabuckthorn leaves powder, and extracting it for 3 times to obtain an extracting solution; step three, separating the extracting solution to obtain seabuckthorn leaves extract.
Preferably, in the step two, wherein an extraction pressure of the subcritical extraction is 0.7 MPa, and an extraction temperature is 50-55°C.
Preferably, the step two specifically includes: placing the seabuckthorn leaves powder packed with an extraction bag in an extraction kettle, closing a feeding valve, opening an air exhaust valve, closing the air exhaust valve after exhausting air of the extraction kettle, opening a liquid inlet valve, adding an extraction solvent into a solvent tank, heating it to reach a subcritical state, introducing it into the extraction kettle, adjusting an extraction pressure of the extraction kettle to a preset pressure, and performing extraction.
Preferably, the extraction solvent is 1,1,1,2-tetrafluoroethane, which is delivered by a high-pressure plunger pump until the seabuckthorn leaves powder is immersed.
Preferably, the extraction kettle is subjected to exhausting air until a pressure in the extraction kettle is reduced to -0.5 MPa.
Preferably, in the step two, the subcritical extraction is completed for 3 times within 60-90 min.
Preferably, in the step one, drying seabuckthorn leaves includes drying the seabuckthorn leaves at room temperature until the moisture content is below 5%.
Preferably, in the step one, crushed seabuckthorn leaves are subjected to sieving with a 20-40 mesh sieve to obtain the seabuckthorn leaves powder.
Preferably, the step three specifically includes: introducing the extracting solution into a separation kettle, increasing temperature under reduced pressure for vaporizing and separating the solvent, and mixing separated products of three subcritical extractions to obtain the seabuckthom leaves extract. 2 wherein the vaporized solvent can be pressurized, condensed, liquefied and re-entered into the solvent tank for recycling.

The extraction method of seabuckthorn leaves extract of the present invention has simple process, rapid extraction speed, mild extraction conditions, and high extraction yield more than 2.5%. The extraction solvent can be recycled, and the extraction solvent has high recycling rate in a production process, which can effectively protect the ecological environment and reduce production costs.
Other advantages, objects and features of the present invention will be partially reflected by the following description, and will be partially understood by those skilled in the art through researching and practicing the present invention.

Embodiment 1
An extraction method of seabuckthorn leaves extract, including the following steps of: drying seabuckthorn leaves at room temperature until a moisture content is below 5%, crushing it with a crusher, sieving it with a 20 mesh sieve to obtain seabuckthorn leaves powder, weighing 500 g, packing it into an extraction bag, placing it in an extraction kettle with a capacity of 3 L, closing a feeding valve, opening an air exhaust valve, exhausting air in the extraction kettle through a vacuum pump until a pressure in the extraction kettle is 3 reduced to -0.5 MPa, closing the air exhaust valve, opening a liquid inlet valve, delivering
R134a (1,1,1,2-tetrafluoroethane) into a solvent tank through a high-pressure plunger pump, heating it with a plate heat exchanger to 50°C until R134a reaches a subcritical state, then injecting it into the extraction kettle through the high-pressure plunger pump until the seabuckthorn leaves powder is immersed by observing a liquid level of the solvent through a sight glass, adjusting an extraction pressure of the extraction kettle to 0.7 MPa, and performing extraction for 3 times within 60 min to obtain an extracting solution. After the extraction, the extracting solution is transferred to a separation kettle, and is subjected to increasing temperature under reduced pressure for vaporizing and separating the solvent, wherein the solvent after being pressurized, condensed, liquefied can be re-entered into the solvent tank for recycling. Separated products of the subcritical extraction for 3 times are mixed to obtain the seabuckthom leaves extract with a yield of 2.5%.
Embodiment 2
An extraction method of seabuckthorn leaves extract are the same as that in the
Embodiment 1, and the difference is: performing extraction for 3 times within 90 min, and obtained seabuckthorn leaves extract has a yield of 3.1%. Other processes and parameters are exactly the same as that in the Embodiment 1.
Embodiment 3
An extraction method of seabuckthorn leaves extract are the same as that in the
Embodiment 2, and the difference is: sieving it with a 40 mesh, and obtained seabuckthorn leaves extract has a yield of 3.5%. Other processes and parameters are exactly the same as that in the Embodiment 2.
Embodiment 4
An extraction method of seabuckthorn leaves extract are the same as that in the
Embodiment 3, and the difference is: heating it with a plate heat exchanger to 55°C, and obtained seabuckthorn leaves extract has a yield of 3.9%. Other processes and parameters are exactly the same as that in the Embodiment 3.
Comparative example 1
An extraction method of seabuckthorn leaves extract are the same as that in the
Embodiment 1, and the difference is: sieving it with a 10 mesh, heating it with a plate heat 4 exchanger to 40°C, and performing extraction for 3 times within 30 min. Obtained seabuckthom leaves extract has a yield of 1.8%. Other processes and parameters are exactly the same as that in the Embodiment 1.
Comparative example 2
An extraction method of seabuckthorn leaves extract are the same as that in the
Embodiment 1, and the difference is: sieving it with a 100 mesh, heating it with a plate heat exchanger to 60°C, and performing extraction for 3 times within 100 min. Obtained seabuckthorn leaves extract has a yield of 2.2%. Other processes and parameters are exactly the same as that in the Embodiment 1.
Although the embodiments of the present invention have been disclosed above, they are not limited to the applications previously mentioned in the specification and embodiments, and can be applied in various fields suitable for the present invention. For ordinary skilled person in the field, other various changed model, formula and parameter may be easily achieved without creative work according to instruction of the present invention, changed, modified and replaced embodiments without departing the general concept defined by the claims and their equivalent are still included in the present invention.
