Description
The present invention relates to a balloon catheter, a method for manufacturing the same, and a composition for balloon coating of a balloon catheter.
A catheter is a device that refers collectively to medical means that are inserted directly into the body. In the medical field, a variety of catheters for inserting the body are used, such as vasoconstriction, coronary artery expansion, urethral insertion, airway insertion, and laparoscopic surgical catheter.
Among them, a balloon catheter for coronary artery dilation has an inflation balloon formed near the end of the guide tube inserted into the blood vessel, for example, and a fluid is introduced through the fluid infusion tube communicated with the balloon to inflate the balloon do.
On the other hand, the balloon of such a balloon catheter can be coated with a drug such as an anticancer drug on its surface. Such a medicament is configured so that when the balloon is inflated in the blood vessel and abuts against the inner wall of the blood vessel, it penetrates the inner wall of the blood vessel and can directly act on the blood vessel and its adjacent tissues.
However, according to the balloon catheter configured as described above, the amount of the anticancer drug that actually penetrates the inner wall of the blood vessel during the inflation time of the balloon is very low, resulting in poor therapeutic efficiency and, consequently, the result is poor. In addition, the drug coated on the balloon may be dissolved by the blood flow in the blood vessel while the balloon catheter is inserted and guided along the blood vessel, so that a sufficient amount of the drug can not reach the inner wall of the blood vessel at the target point.
It is an object of the present invention to provide a balloon catheter capable of improving the degree of absorption of an anticancer drug against the inner wall of a blood vessel while reducing the solubility of an anticancer drug against blood flow, To provide a composition.

Anticancer drugs may include paclitaxel. Parklitaxel is an anticancer chemotherapeutic agent extracted from the bark of tree bark. It has been approved by the US Food and Drug Administration (FDA) in 1993 for use as an anti-cancer drug. Since paclitaxel is difficult to synthesize and exists only in a small amount in the tree of interest, a large amount of baccatin III component, which is commonly found in various plants belonging to the order Trees, is obtained by separation and then subjected to a semi-synthesis process Is being produced. The paclitaxel acts on the M phase, the last stage of cell division, and binds to a substance called tublin, which inhibits the cell cycle. As shown, this pucklit cell is brought into abutment with the inner wall of the blood vessel by the expansion of the
Absorption enhancers are a factor for promoting the absorption of paclitaxel to the inner wall of blood vessels. Absorption accelerators may be composed of vitamin E-based materials. Specifically, the absorption enhancer may include vitamin E d-a-tocopheryl polyethylene glycol 1000 succinate (hereinafter, referred to as 'vitamin E TPGS'). Such vitamin E TPGS may have a structure represented by the following formula (1).
[Chemical Formula 1]
The dissolution inhibitor is a factor for preventing the dissolution of paclitaxel in blood. Specifically, the dissolution inhibitor can prevent the phenomenon that the paclitaxel is dissolved by the blood and the like in the blood vessel while the

As shown in the figure, first, paclitaxel, vitamin E TPGS, and shellac are mixed in a solvent (S1), and the mixture is stirred through a stirrer to produce a liquid medicine for balloon coating (S3). Here, the content of paclitaxel may be in the range of 1 to 5 parts by weight based on 100 parts by weight of the chemical solution for balloon coating.
The drug solution for balloon coating is applied to the surface of the
The content of shellac and vitamin E TPGS can be adjusted within a set range, which will be described later with reference to FIG.

Content ratio
Type 2-1: 100 parts by weight of paclitaxel, 70 parts by weight of vitamin E TPGS, 30 parts by weight of shellac
Type 2-2: 100 parts by weight of paclitaxel, 50 parts by weight of vitamin E TPGS, 50 parts by weight of shellac
Type 2-3: 100 parts by weight of paclitaxel, 30 parts by weight of vitamin E TPGS, 70 parts by weight of shellac
Crystalline structure: crystalline morphology
Intravascular balloon inflation time: 30 seconds each
Result: Average value after 3 repetitions
As a result, it was confirmed that the absorption of paclitaxel to the inner wall of the blood vessel did not show a large change when the content ratio between vitamin E TPGS and shellac was controlled within a predetermined range. Specifically, if the content ratio of vitamin E TPGS is adjusted between 30 parts by weight and 70 parts by weight with respect to 100 parts by weight of paclitaxel, and the content of shellac is adjusted between 30 parts by weight and 70 parts by weight, Respectively.
Together with this, it was confirmed that Type 2-1, Type 2-2, and Type 2-3 in the present experiment all exhibited a considerably high level of blood vessel uptake. This suggests that the absorbance of paclitaxel is significantly increased due to the complex action of vitamin E TPGS and crystalline structure, even though shellac acts to lower the absorption of paclitaxel (see Experiment 2).
Such a balloon catheter, a method for producing the same, and a composition for balloon coating of the balloon catheter are not limited to the construction and operation of the embodiments described above. The embodiments may be configured so that all or some of the embodiments may be selectively combined so that various modifications may be made.
