Abstract

PURPOSE:To readily obtain a 5-hydroxyhydantoin derivative useful as a drug having excellent blood sugar lowering action and lipid reducing action, by oxidizing a readily obtainable hydantoin derivative nonsubstituted at the 5-position as a raw material. CONSTITUTION:A hydantoin derivative such as 1-methylhydantoin shown by formula I (R1 and R2 are H, alkyl or cycloalkyl) nonsubstituted at the 5-position is reacted with a metal acetate such as lead acetate (IV) in a solvent such as benzene to give a 5-acetoxyl derivative, which is then hydrolyzed with an acid such as sulfuric acid or treated with an alkyl hydroperoxide such as t-butyl hydroperoxide in the presence of a metallic salt such as Ca<+> or Co<2+> to convert the 5-position of the compound shown by formula I to hydroperoxy and then the hydroperoxy group is converted into hydroxyl group by catalytic reduction with a catalyst of Pd/C so that the compound shown by formula I is oxidized to give the aimed compound such as 5-hydroxy-1-methylhydantoin shown by formula II.
