る。DETAILED DESCRIPTION OF THE INVENTION The present invention relates to a method for producing steroid aldehydes. Specifically, intermediates for the production of progesterone (progesterone), adrenal corticosteroids (hydrocortisone, cortisone, prednisolone, prednisone, etc.), namely 22.23-bisnorcholar 4-en-3-one-22-al and 22. 23-bisnorcholar 1,4-diene-3-
The present invention relates to a method for producing ON-22-R.

It has been found that 22,23-bisnorcholar 1,4-dien-3-one can be produced in larger quantities and at lower cost by microorganisms belonging to the genus Mycobacterium than sterols such as cholesterol or sitosterol. (For example, Japanese Patent Application Laid-Open No. 54-67094,
-67095 publication, West German patent publication No. 2571560) On the other hand, 22・23-bisnorcholar 4-en-3-
It is a well-known fact that on-22-al is an important intermediate in the production of progesterone (progesterone), and since brogesterone is an intermediate in the production of hydrocortisone, it is also an intermediate in the production of adrenocortical hormone. (See, e.g., LF Fieser and M. Fieser, “Steroids,” p. 675 (1959) USA) Therefore, 22-hydroxy-22,23-bisnorcholar-4-ene-3- The preparation of 22,23-bisnorcholar 4-en-3-one-22-al from 1 is of great industrial importance.

3363 pages (1968), British National and Journal of Organic Chemistry (J.Org.
Chem.), vol. 26, p. 4814 (1961) and the same American journal, vol. 35, p. 4000 (1970), it was reported that the corresponding aldehyde could be obtained from aliphatic alcohol in a yield of over 90%. ing. but,
60 in the case of 22-hydroxy-22,23-bisnorcholar 4-en-3-one, 22-hydroxy-22,23-bisnorcholar 1,4-dien-3-one
The corresponding aldehyde is formed only in ~70% yield. Furthermore, due to the toxicity of chromium compounds, disposal costs are high, and this method cannot be said to be advantageous as an industrial production method.

- a solvent that dissolves hydroxysteroids;
Particularly preferred are halogenated hydrocarbons such as dichloromethanechloroform and cyclic ethers such as tetrahydrofuran and dioxane. Of course,
Both may be used together. In other solvents, such as aromatic hydrocarbons such as toluene and benzene, only about 0.3% of 22-hydroxysteroids are dissolved, making it difficult to obtain a sufficient conversion rate of 22-hydroxysteroids.

い。 When a halogenated hydrocarbon or cyclic ether is used as the solvent for dissolving 22-hydroxysteroid, the reaction solvent is a mixed solvent of an aromatic hydrocarbon solvent and a halogenated hydrocarbon or cyclic ether. It was found that the composition of this mixed solvent had a significant effect on the reaction results. When the proportion of aromatic hydrocarbons is high, the solubility of 22-hydroxysteroid decreases, so there is a risk that 22-hydroxysteroid will precipitate. On the other hand, when the proportion of highly polar solvents such as halogenated hydrocarbons and cyclic ethers, which have a large dissolving power for 22-hydroxysteroids, is high, the A large amount of methyl ethers is produced as a by-product, significantly reducing the yield of the desired steroid aldehyde. In order to obtain steroid aldehyde in good yield, the composition of aromatic hydrocarbons, halogenated hydrocarbons and cyclic ether should be 1:1 to 4:1, especially
The ratio is preferably 1.5:1 to 2.5:1 (volume ratio).
