Abstract

PURPOSE:To obtain a calcitonin pharmaceutical easily administrable of the noninjection type, by dispersing calcitonin with an amino acid derivative in a suppository base. CONSTITUTION:A calcitonin pharmaceutical for rectal administration prepared by dispersing calcitonin and an amino acid derivative, e.g. sodium N-(1-methyl-2- ethoxycarbonyl-vinyl)-DL-phenylglycinate, in a suppository base, e.g. cacao butter, uniformly. The calcitonin is a single chain polypeptide hormone consisting of 32 amino acids secreted from the thyroid gland of mammals, and acts on the bone, intestinal tract or kidney to reduce the calcium or inorganic phosphorus in blood. Therefore, the pharmaceutical is effective against hypercalcemia, particularly senile osteoporosis, however, has been only used as an injection. The use of the calcitonin with the amino acid derivative in the suppository permits the good absorption by the rectum and facilitates the administration of the calcitonin.
