【発明の詳細な説明】
カルシウムチャネル拮抗薬としてのアミン誘導体類
本発明は、アミン誘導体、さらに詳しくは、アルキル部分にヘテロ原子を有す
るアリールオキシ−、アリールチオ−およびアロイル−アルキルアミノ化合物、
該アミン誘導体の製造方法、該アミン誘導体を含有する医薬組成物、および治療
における、特に、虚血性発作の治療における該アミン誘導体の使用に関する。
発作は、先進国における一般的な死亡原因の第3位であることが報告されてい
る。虚血性発作についての現行治療法は、制限されており、出血を悪化させる危
険などの多くの欠点を有している。したがって、虚血性発作の新規でかつ改善さ
れた治療が必要とされている。
英国特許第924961号には、抗線虫薬(anti-nematodal agent)製造用中
間体として、式R.W.CH2.CH2.NXY(式中、Rは、所望により、とりわけ
、ハロゲン、アルキルまたはアルコキシにより置換されていてもよいフェニルで
あり、XおよびYは、とりわけ、アルキルであるか、または、NXYは、ピロリ
ジノ、ピペリジノまたはモルホリノ基を表し、Wは、炭素原子16個および非隣
接酸素原子1〜3個を含有する直鎖状の飽和鎖である)で示される化合物が開示
されている。
本発明者らは、ある種のアミン誘導体がカルシウムチャネル拮抗薬としての活
性を示すことを見いだした。
したがって、本発明は、第1の態様において、治療薬として使用するための式
(I):

R1およびR2は、各々独立して、水素、C1-8アルキル、C3-8シクロアルキル
、C3-8シクロアルキルC1-4アルキル、アリールC1-4アルキル、C2-6ヒドロキ
シアルキルまたはR3R4NC2-6アルキルを表すか(ここで、R3およびR4は、独
立して、HまたはC1-4アルキルを表す)、または
NR1R2は、環原子4〜9個を含有する飽和複素環式環を表し、ここで、該環
原子のうち1つは、所望により、O、SまたはNR5から選択されるさらなるヘ
テロ原子であってもよく(ここで、R5は、H、C1-4アルキルまたはアリールC1 -4
アルキルである)、該飽和複素環式環は、所望により、C1-6アルキルおよびC1-6
アルコキシから選択される1または2個の置換基によって置換されていても
よく;
Xは、O、SまたはNR6を表し(ここで、R6は、水素、C1-4アルキルまた
はアリールC1-4アルキルである)、
Yは、O、S、C=Oまたは結合を表し、
nは、2〜5であり、
mは、1〜5であり(ただし、mが1である場合、Yは、結合を表す)、
Arは、所望により、ハロ、C1-8アルキル、C1-8アルコキシ、C1-2アルキレ
ンジオキシ、例えば、メチレンジオキシ、トリフルオロメチル、トリフルオロメ
チルオキシから選択される1〜3個の置換基によって、または、基Ph(Alk1)p
A(Alk2)q−(ここで、Phは、所望により置換されていてもよいフェニルであ
り、Aは、結合、O、S、−C=OまたはCH=CHであり、Alk1およびAlk2
は、独立して、直鎖状であっても分枝鎖状であってもよいC1-4アルキルを表し
、pおよびqは、独立して、0または1であり、ただし、(Alk1)A(Alk2)の長
さは、原子5個を超えない)によって置換されていてもよいフェニルを表すか、
または、Arは、所望により置換されていてもよい三環式ヘテロアリール基:

で示される化合物またはその医薬的に許容される塩を提供するものである。
さらなる態様では、本発明は、カルシウム拮抗薬を必要とする症状の治療薬の
製造における前記定義の式(I)で示される化合物またはその医薬的に許容される
塩の使用を提供するものである。
式(I)で示される化合物およびそれらの医薬的に許容される塩は、好ましくは
、哺乳動物の脳細胞におけるカルシウムの蓄積に関連する症状の治療薬の製造に
おいて使用される。
式(I)で示される化合物において、nおよびmは、好ましくは、独立して、n
+mの合計が4〜8となるような2〜4である。Yが結合である場合、n+mは
、好ましくは、7であり、Yが結合以外である場合、n+mは、好ましくは、6
である。
Yは、好ましくは、酸素を表す。
Xは、好ましくは、酸素またはNR6を表し、R6は、好ましくは、水素を表す
。
Arが基Ph(Alk1)pA(Alk2)q−によって置換されている場合、Aは、好まし
くは、酸素、C=Oまたは結合である。Alk1およびAlk2は、好ましくは、独立
して、CH2、または、分枝鎖状の場合にはC(H)(CH3)もしくはC(CH3)2を
表す。Aが酸素である場合、qは、好ましくは、0であり、pは、好ましくは、
0または1である。Aが結合である場合、p+qの合計は、好ましくは、1また
は2である。AがCH=CHまたはC=Oを表す場合、pおよびqは、好ましく
は、共に0である。
三環式ヘテロアリールの例としては、ジベンゾフラニル、ジベンゾチエニル、
カルバゾール、N−メチルカルバゾール、アクリジンおよびジベンゾキセピンが
挙げられる。該三環の部分は、適切な環原子を介して式(I)の残部に結合するこ
とができる。
Phおよび三環式ヘテロアリール基についての適切な置換基としては、例えば
、ハロゲン、トリフルオロメチル、トリフルオロメトキシ、C1-4アルキルおよ
びC1-4アルコキシから選択される1〜3個の置換基が挙げられる。
好ましくは、Arは、フェノキシ、ベンジル、ベンジルオキシ、ベンゾイル、
ハロベンゾイルまたはハロによって一置換されているフェニル;ハロによって二
置換されているフェニルであるか;または、Arは、2−ジベンゾフラニルであ
る。最も好ましくは、Arは、ベンジル、ベンゾイル、フルオロベンゾイルまた
はベンジルオキシによって置換されているフェニルである。該置換基は、好まし
くは、フェニル環の3位または4位にある。
単独または別の基の一部として式(I)で示される化合物にあるアルキル基は、
直鎖状であっても分枝鎖状であってもよい。かくして、C1-6アルキル基は、例
えば、メチル、エチル、n−プロピル、n−ブチル、n−ペンチル、n−ヘキシ
ル、またはイソプロピル、t−ブチルまたはsec−ペンチルなどのそれらの分枝
鎖状異性体である。
医薬用については、式(I)で示される化合物の塩は、医薬的に許容されるべき
であると思われる。医薬的に許容される塩の例としては、塩酸塩、臭化水素酸塩
、硫酸塩、リン酸塩、酢酸塩、フマル酸塩、マレイン酸塩、クエン酸塩、乳酸塩
、酒石酸塩、シュウ酸塩、メタンスルホン酸塩などの無機および有機の酸付加塩
または類似の医薬的に許容される無機または有機酸付加塩が挙げられる。他の医
薬的に許容されない塩は、例えば、最終生成物の単離において使用され、本発明
の範囲内に含まれる。
式(I)で示される化合物は、1個以上の不斉中心を含有すると思われる。かか
る化合物は、光学異性体(鏡像異性体)として存在するであろう。両方の純粋な
鏡像異性体、ラセミ混合物(各鏡像異性体50%)および2つの不均等混合物は
、本発明の範囲内に含まれる。さらに、可能な全てのジアステレオマー形(純粋
な鏡像異性体およびその混合物)は、本発明の範囲内である。
式(I)で示されるある種の化合物は、新規であると思われる。かくして、さら
なる態様では、本発明は、式(IA):

びAraは、R1、R2、X、YおよびArの定義と同じである;

で示される新規化合物およびその塩を提供するものである。
新規化合物であると思われる本発明の特定の化合物としては、
N−[3−(4−ベンジルオキシフェノキシ)プロピル]−N−[3−(1−ピペリ
ジノ)プロピル]アミン、
1−{3−[3−(4−ベンジルオキシフェノキシ)プロピルオキシ]プロピル}ピ
ペリジン、
N−[3−(3,4−ジクロロフェノキシ)プロピル]−N−[3−(1−ピペリジ
ノ)プロピル]アミン、
N−[3−(4−ベンジルフェノキシ)プロピル]−N−[3−(1−ピペリジノ)
プロピル]アミン、
N−{3−[4−(4−フルオロベンゾイル)フェノキシ]プロピル}−N−[3−(
1−ピペリジノ)プロピル]アミン、および
それらの医薬的に許容される塩が挙げられる。
本発明化合物は、当該技術分野で知られている製造方法と類似の製造方法によ
って製造することができる。したがって、本発明は、さらなる態様において、

が−CH2−を表す化合物に還元し、
次いで、所望により、塩を形成することからなることを特徴とする、式(I)で
示される新規化合物の製造方法を提供するものである。
工程(a)において、式(II)で示される化合物および式(III)で示される化合
物の間の反応は、標準的な条件下で行うことができる。例えば、離脱基L1は、
ハロゲン原子またはスルホニルオキシ基、例えば、メタンスルホニルオキシもし
くはp−トルエンスルホニルオキシであってもよい。この場合、該反応は、ジメ
チルホルムアミドまたはメチルエチルケトンなどの溶媒の不在または存在下、所
望により水素化ナトリウムまたは炭酸カリウムなどの塩基の存在下、0〜200
℃の範囲の温度で行われる。別法として、L1がヒドロキシであり、yが0であ
る場合、該反応は、ジエチルアゾジカルボキシラートおよびトリフェニルホスフ
ィンの存在下で行われる。かかる反応は、ミツノブ(Mitsunobu)反応として知
られている[シンセシス(Synthesis)1981、1に開示されている]。この反
応は、所望により、テトラヒドロフランなどの溶媒の存在下で行われてもよい。
工程(b)に従った式(IV)で示される化合物と式(V)で示される化合物との反
応は、慣用手段で、例えば、溶媒として過剰のアミンを用いて、あるいは、別法
として、エタノールまたはジメチルホルムアミドなどの有機溶媒を用いて行われ
る。離脱基L2は、例えば、臭化物もしくは塩化物などのハロゲン化物、アセト
キシもしくはクロロアセトキシなどのアシルオキシ基またはメタンスルホニルオ
キシもしくはp−トルエンスルホニルオキシなどのスルホニルオキシ基である。
該反応は、炭酸カリウム、水素化ナトリウムまたはカリウムt−ブトキシドなど
の塩基の存在下で行われるか、または、過剰のアミンが用いられる。
工程(c)に従った式(VI)、(VII)、(VIII)または(IX)で示されるアミドの還
元は、水素化アルミニウムリチウムなどの適切な還元剤を用いて行われる。この
工程では、基Yおよび/またはAの同時還元が望まれない限り、YまたはAは、
一般的にC=Oではないと思われる。
工程(d)では、アルデヒド(X)の還元アミノ化は、当該技術分野でよく知ら
れている方法に従って、式(V)で示される化合物の存在下、シアノホウ水素化ナ
トリウムなどの還元剤を用いて行われた。XがNHを表す場合、窒素原子は、好
ましくは、以下に説明するように保護されるべきであると思われる。
工程(e)では、化合物(XI)および(XII)は、前記工程(a)と同様の方法で
行われる。
工程(f)に従ったピリジニウム誘導体(XIII)の還元は、好適にはエタノー
ルのようなアルコールなどの溶媒中、パラジウム−炭、白金または酸化白金(ア
ダムス触媒)などの貴金属触媒を用いて水素添加などによって行われる。
工程(g)では、式(XIV)および(XV)で示される化合物間での反応は、基L2
の性質およびyの数値に依存する条件下で起こり得る。例えば、yが1である
場合、L2は、ハロゲンまたはトシラートもしくはメシラートなどのスルホン酸
残基であり、該反応は、ジメチルホルムアミドなどの溶媒中、標準的な条件下、
所望により水素化ナトリウムなどの塩基の存在下で行われる。yが0である場合
、L2は、化合物(XV)がフルオロ置換アリール化合物であるようにフルオロで
あり、該反応は、好ましくは、水素化ナトリウムなどの強塩基の存在下、ジメチ
ルスルホキシドまたはジメチルホルムアミドなどの極性有機溶媒中で行われる。
工程(h)に従った転換反応は、標準的な方法を用いて行われる。かくして、
例えば、Aが−CH=CH−を表す化合物(I)からAが−CH2CH2−を表す化
合物(I)への転換は、接触水素添加によって行われ、Yおよび/またはAがC=
Oである化合物の還元は、水素化アルミニウムリチウムなどの好適な還元剤を用
いて行われる。
式(II)で示される化合物は、標準的なアルキル化条件下、式(XVI):
[式中、L1、L2、X、m、nおよびxは、前記定義と同じである]
で示される化合物を前記定義の式(V)で示される化合物と反応させることによっ
て製造することができる。該反応は、好適には、工程(b)について前記した条
件と同様の条件下で行われる。XがNHである場合、窒素基は、例えば、以下に
説明するように保護されるべきであると思われる。
式(XVI)で示される化合物において、離脱基L1およびL2は、好ましくは、式
(V)で示される化合物がL2と選択的に反応するように選択される。例えば、式(
XVI)で示される化合物では、L1は、好適には、ヒドロキシであり、L2は、好
適には、ハロである。
式(III)で示される化合物は、市販品であるか、または、当該技術分野でよく
知られている標準的な方法を用いて、例えば、工程(a)について記載した方法
と同様の方法を用いて製造してもよい。
式(IV)で示される化合物は、前記定義の式(III)で示される化合物を前記定義
の式(XVI)で示される化合物と反応させることによって製造することができる。
この反応では、L1およびL2は、共に同一であってもよく、例えば、ハロである
が、この場合、nおよびmも同一の数値であるべきであると思われる。該反応は
、好適には、炭酸カリウムなどの弱塩基の存在下で行われる。別法として、該反
応は、水酸化カリウムなどの強塩基を用いて相転移条件下で行われる。別法とし
て、YがC=Oを表す場合、式(IV)で示される化合物は、化合物HArの、式
L2(CH2)nX(CH2)mC(O)Hal(式中、Halは、ブロモまたはクロロなどの
ハロゲン原子を表す)のアシル化剤によるフリーデル−クラフトアシル化によっ
て製造することができる。XがNHを表す場合、好ましくは、窒素保護を用いる
べきであると思われる。得られた式(IV)で示される化合物におけるカルボニル基
は、所望により、次いで、還元されてもよい。
式(V)および(XVI)で示される化合物は、市販品であるか、または、標準的な
方法によって製造してもよい。
式(VI)で示される化合物は、式(II)または(V)に対応する適切なアミドを用い
て、前記の一般的な工程(a)および(b)に従って製造される。
式(VII)で示される化合物は、式(V)で示される化合物の適切な酸塩化物また
はエステルとのアシル化によって製造される。ここで、該酸塩化物またはエステ
ルは、適切な市販のブロモアルキルエステルまたは酸との反応によって、次いで
、酸塩化物への転換によって式(III)で示される化合物から製造される。別法と
して、化合物(VII)は、工程(a)と同様の方法によって製造される。
式(VIII)および(IX)で示されるアミドは、例えば、各々、対応するカルボン酸
、または、酸ハロゲン化物、エステルもしくは無水物などのその活性化誘導体の
、式ArY(CH2)m(R6)NHまたはR1R2N(CH2)n(R6)NHで示されるアミ
ンとの反応によって製造される。
式(X)で示されるアルデヒドは、例えば、トルエンなどの不活性溶媒の存在下
、水素化アルミニウムジイソブチルなどの還元剤を用いて、対応するニトリルの
還元によって製造される。好都合には、アルデヒドの還元アミノ化は、in situ
で行われる。すなわち、式(I)で示される化合物は、中間体アルデヒドを単離せ
ずに一浴反応でニトリルから得られる。該ニトリルは、L2がハロである式(IV)
で示される化合物をシアン化カリウムと反応させることによって製造される。化
合物(X)は、また、エステルの還元またはアルコールの酸化などの他の標準的な
方法によって製造される。
式(XI)で示される化合物は、前記工程(a)〜(d)のいずれかと同様の方法
によって製造される。別法として、化合物(XI)は、Arがベンジルオキシフェニ
ル基を表す式(I)で示される対応する化合物の接触水素添加によって得られる。
したがって、これは、式(I)で示される化合物の式(I)で示される別の化合物へ
のさらなる転換方法を提供する。
式(XII)で示される化合物は、市販品であるか、または、文献公知である。
式(XIII)で示される化合物は、アミンとして所望により置換されていてもよい
ピリジンを用いて前記工程(b)と同様の方法で製造される。
式(XIV)で示される化合物は、式(II)について前記したと同様に製造される。
前記工程(a)〜(h)の一定の間および中間体の製造の間、次段階で当該技
術分野でよく知られている方法によって除去されるN−保護基などで分子中に存
在する非置換窒素原子を保護するのが必要であると思われる。好適な保護基とし
ては、ベンジル、ジフェニルメチルまたはトリフェニルメチルなどのアラルキル
基およびアセチル、トリフルオロアセチル、ベンゾイル、メトキシカルボニル、
エトキシカルボニル、t−ブトキシカルボニルまたはベンジルオキシカルボニル
などのアシル基が挙げられる。ベンジルなどのアラルキル基は、水素化分解によ
って切断され、ベンゾイルなどのアシル基は、加水分解によって切断される。
式(I)で示される化合物が鏡像異性体の混合物として得られる場合、これらは
、分割剤の存在下での結晶化、または、例えば、キラルHPLCカラムを用いる
クロマトグラフィーなどの慣用の方法によって分離される。
本発明は、本明細書に記載の新規中間体、特に、式(II)、(IV)、(VI)、(VII)
、(VIII)、(IX)および(XII)の中間体をも包含する。
本発明化合物は、例えばニューロンにおいて、高カルシウム流入ブロック活性
を示すことが判明した。当該化合物自体は、哺乳動物、特に、ヒトの脳細胞にお
けるカルシウムの蓄積に関連する症状および疾患を治療する際に治療において有
用であると思われる。例えば、当該化合物は、無酸素症、例えば発作を含む虚血
、片頭痛、内臓痛、癲癇、外傷性頭部もしくは脊髄損傷、AIDS関連痴呆、ア
ルツハイマー病および年齢関連記憶障害(age-related dementia)などの神経変
性疾患(neurodegenerative disease)、気分障害、ならびにエタノール嗜癖禁断
症状などの薬物嗜癖禁断症状の治療において有用であると思われる。
本発明は、治療を必要とする患者に、前記定義の式(I)で示される化合物また
はその医薬的に許容される塩の有効量を投与することからなる哺乳動物の脳細胞
中のカルシウム蓄積に関連する(例えば、該カルシウム蓄積によって生じるか、
または悪化する)症状または疾患の治療方法を提供するものでもある。
かくして、例えば、本発明は、治療を必要とする患者に、式(I)で示される化
合物またはその医薬的に許容される塩の有効量を投与することからなる、無酸素
症、例えば発作を含む虚血、片頭痛、内臓痛、癲癇、外傷性頭部もしくは脊髄損
傷、AIDS関連痴呆、アルツハイマー病および年齢関連記憶障害などの神経変
性疾患、気分障害ならびにエタノール嗜癖禁断症状などの薬物嗜癖禁断症状の治
療方法を提供するものである。
医薬における使用について、本発明化合物は、通常、標準的な医薬組成物で投
与される。したがって、本発明は、別の態様では、前記定義の式(I)で示される
化合物またはその医薬的に許容される塩および医薬的に許容される担体または賦
形剤からなる医薬組成物を提供するものである。
本発明化合物は、例えば、経口、非経口、口腔内、直腸内または経皮投与によ
る好都合な方法によって投与されてもよく、それに応じて、該医薬組成物は、適
合させられる。非経口投与が一般的に好ましい。
経口投与の場合に活性である式(I)で示される化合物およびそれらの医薬的に
許容される塩は、液剤または固形剤、例えば、シロップ剤、懸濁液剤もしくは乳
剤、錠剤、カプセル剤およびロゼンジ剤として製剤化され得る。
液体製剤は、一般に、懸濁化剤、保存剤、フレーバーリング剤または着色剤を
含む、適切な液体担体、例えば、エタノール、グリセリン、非水性溶媒、例えば
、ポリエチレングリコール、油、または水中の当該化合物またはその医薬的に許
容される塩の懸濁液または溶液からなる。
錠剤の形態の組成物は、固体製剤を調製するために慣用に使用される適切な医
薬担体を用いて調製することができる。かかる担体の例としては、ステアリン酸
マグネシウム、デンプン、ラクトース、シュークロースおよびセルロースが挙げ
られる。
カプセル剤の形態の組成物は、慣用の被包方法を用いて調製することができる
。例えば、標準的な担体を用いて、活性成分を含有するペレットを調製し、次い
で、
ゼラチン硬カプセル中に充填する;別法としては、適切な医薬担体、例えば、水
性ガム、セルロース、シリケート、または油を用いて、分散物または懸濁液を調
製することができ、次いで、該分散物または懸濁液をゼラチン軟カプセル中に充
填する。
本発明化合物は、ボーラス注射または連続輸液によって、非経口投与されても
よい。典型的な非経口用組成物は、無菌水性担体または非経口的に許容される油
、例えば、ポリエチレングリコール、ポリビニルピロリドン、レシチン、落花生
油またはゴマ油中の当該化合物またはその医薬的に許容される塩の溶液または懸
濁液からなる。別法としては、該溶液を凍結乾燥させ、次いで、投与直前に好適
な溶媒で再構成することもできる。
液体および固体組成物は、共に、シクロデキストリンまたは可溶化剤、例えば
、クレモフォアなどの医薬技術の分野において知られている別の賦形剤を含有し
てもよい。
好ましくは、当該組成物は、錠剤、カプセル剤またはアンプル剤などの単位投
与形態である。

溶液
ピペット(内部溶液)は、CsOHでpH7.2に緩衝化された、CsCl 130
mM;HEPES 10mM;EGTA 10mM;MgCl2 4mM;ATP 2mMを
含有した。細胞を通常タイロード溶液中に浸漬した後、浸漬溶液がCa2+電流を
断路するものに変化した場合に全細胞の樹立を記録した。Ca2+チャネル電流を
記録するための外部溶液は、TEA−OHでpH7.3に緩衝化された、BaCl2
10mM;TEA−Cl 130mM;グルコース10mM;HEPES 10mM;

チャネル電流を、電荷担体としてBa2+ 10mMを用いて記録した。電流は、保
持電位−80mVから試験電位0または+10mVまで15秒間ごとに誘発された
。この試験電位は、電流電圧関係のピークであり、ここでのブロックを評価する
ことは、保持電位をドリフトすることによるエラーを減少させた。いくつかの細
胞は、Ca2+電流を測定する場合に一般的に見られるように、電流のゆっくりと

3−(4−ベンジルオキシフェノキシ)プロパン−1−オール
4−ベンジルオキシフェノール(2.0g)、80%水素化ナトリウム(0.30g)
、3−ブロモプロパン−1−オール(1.39g)およびジメチルホルムアミド(
30ml)の混合物を、窒素下、60℃で4時間撹拌した。該混合物を水で処理し
