Description
適当な組成物として膣管に挿入された場合、多
様の化学薬品が、哺乳類の精子細胞の固有運動性
を減少すること(すなわち、精子不動化作用)又
は細胞毒性(すなわち、殺精子作用)のいずれか
により、哺乳類の精子細胞を阻害し、しかも前記
哺乳類の精子細胞の受精能力を減少することは以
前から知られている。一般に、かかる目的に対し
最も広く知られている薬品は、アルキルフエノキ
シポリエトキシエタノール類、第四級アンモニウ
ム化合物および有機水銀化合物である。最近、殺
精子作用を有する一連の1,2―ベンゾイソチア
ゾール誘導体が米国特許第4093730号に開示され
ている。
しかるに、ただ一種の該殺精子剤、ポリ(エチ
レングリコール)p―ノニルフエニルエーテル
{INN名:ノンオキシノール(nonoxynol)}が、
米国内において一般に使用されている。しかし、
この物質は失敗率が高く、一般に基準以下と見な
されている。
更に、膣内投与用のかかる精子不動化化合物お
よび殺精子化合物は、雄性の哺乳動物に全身的に
投与する場合、精子細胞に対する影響はたとえあ
るにしても小さい。
本発明は、通常使用されている殺精子剤よりも
より強力とみられ、しかも膣内に投与する場合の
みならず精子製造の雄性に投与する場合更に有効
な作用を有する一群の殺精子剤に関する。
本発明は、殺精子剤化合物および妊娠調節剤化
合物の調製に対し、1―置換イミダゾール誘導体
の一群から選択れた化合物の使用に関するもので
あり、該一群の誘導体は抗真菌性、抗菌性および
抗原虫作用を有するとして先に報告されている。
例えば、米国特許第4123542号参照。今や、驚く
べきことに、この類の化合物が、哺乳動物の精子
の運動性を減少することにより、又はそれらの通
常の細胞機能を無能力化することにより、又はそ
れらを殺すこと(殺精子作用)のいずれかによ
り、哺乳動物の精子を阻害し、そしてそれらが受
精する能力を阻止することを見出した。該化合物
は被投与体に対しては一般に非毒性である。
本発明は、一面において、精子不動化剤又は殺
精子剤として、その中に含有されている1―置換
イミダゾール類の使用に関する。又他の面におい
ては、本発明は膣内投与用に適した1―置換イミ
ダゾール類又はそれらの塩類を含有する、避妊に
有用な組成物の調製および製剤に関する。
更に、第三の面から、本発明は経口投与、特に
雄性の哺乳動物に対し経口投与に適した1―置換
イミダゾール類又はそれらの塩類を含有する、避
妊に有用な組成物の製造および製剤に関する。
更に、第四の面から、本発明は非経口的投与、
特に雄性の哺乳動物に対し非経口的投与に適した
1―置換イミダゾール類又はそれらの塩類を含有
する、避妊に有用な組成物の調製および製剤に関
する。
第五の面から、本発明は、本発明中説明される
如き化合物および/又は組成物を用いる避妊法に
関する。
上記の全ての面において、1―置換イミダゾー
ルは遊離塩基又は医薬として許容され得る酸付加
塩である。医薬として許容され得る塩は、遊離塩
基の製薬上の性質を維持する主題の塩基の塩であ
り、更に該塩は生物学的方法ではなく別な方法で
例えば以下の如き酸と共に好ましく形成される。
すなわち、塩酸、臭酸、硫酸、硝酸もしくは燐酸
の如き無機酸;又は酢酸、プロピオン酸、グリコ
ール酸、乳酸、ピルビン酸、修酸、マロン酸、コ
ハク酸、リンゴ酸、マレイン酸、フマル酸、酒石
酸、クエン酸、安息香酸、ケイ皮酸、マンデル
酸、メタンスルホン酸、エタンスルホン酸、p―
トルエンスルホン酸、サリチル酸等の如き有機酸
である。
抗真菌性、抗菌性および/又は抗原虫作用を示
すものとして先に説明した公知の多種類の1―イ
ミダゾール化合物は、雄性および雌性の哺乳動物
の双方において著るしい精子不動化作用および殺
精子作用を示すことが見出された。本発明の一部
分を形成する1―置換イミダゾール類は、次の一
般式によつて表わされる化合物(並びに医薬とし
て許容されうるそれらの酸付加塩)および/又は
後記の特許明細書に開示される化合物である:
すなわち、下記式()
で表わされる化合物、又は医薬的許容され得る該
化合物の酸付加塩。上記式()において、R1
およびR2はそれぞれ独立に1ないし12の炭素原
子を有するアルキル基、3ないし7の炭素原子を
有するシクロアルキル基、未置換もしくは置換フ
エニル(該置換フエニルは、ハロゲン原子および
1ないし4の炭素原子を有する低級アルキル基、
1ないし4の炭素原子を有する低級アルコキシ
基、およびトリフルオロメチル基から選ばれた1
ないし3個の基によつて置換されたフエニルであ
る)、又は、未置換もしくは置換フエニル(該置
換フエニルは、ハロゲン原子および1ないし4の
炭素原子を有する低級アルキル基、1ないし4の
炭素原子を有する低級アルコキシ基、およびトリ
フルオロメチル基から選ばれた1ないし3個の基
によつて置換されたフエニルである)で置換され
た1ないし4の炭素原子を有するアルキル基であ
り、R3は水素原子又は1ないし4の炭素原子を
有する低級アルキル基であり、Xは酸素原子又は
硫黄原子であり、そしてnは1から4までの整数
である。
本発明の好ましい態様は、上記式{式中、R1
およびR2は各々独立に1ないし12の炭素原子を
有するアルキル、3ないしの炭素原子を有するシ
クロアルキル、又は未置換もしくは置換フエニル
基(該置換フエニルは、1ないし4の炭素原子を
有する1個又は2個の低級アルキルにより又は1
個又は2個のハロゲン置換基によつて置換されう
る)、又はフエニル低級アルキル(C1〜4)(該フエ
ニルは、1ないし4の炭素原子を有する1個又は
2個の低級アルキルにより、又は1個又は2個の
ハロ置換基によつて置換されていてもよい)であ
り、R3は水素原子であり、nは1又は2である}
で表わされる一群の化合物、および該化合物の医
薬として許容され得る塩である。
更に好ましい態様は、置換基の次の組合せの内
で一つを示す化合物、およびそれらの組合せ((a)
〜(d))において医薬として許容され得る該化合物
の酸付加塩からなる:
(a) R1はアルキル又はシクロアルキルであり、
R2はアルキル、置換もしくは未置換フエニル、
又は置換もしくは未置換ベンジルであり、R3
は水素原子であり、nは1又は2である;
(b) R1は置換もしくは未置換フエニルであり、
R2は置換もしくは未置換のベンジルであり、
R3は水素原子であり、nは1である;
(c) R1は置換又は未置換フニネチルであり、R2
は置換又は未置換フエニルであり、R3は水素
原子であり、nは1である;および
(d) R1は置換又は未置換ベンジルであり、R2は
置換又は未置換フエニル又はベンジルであり、
R3は水素原子であり、nは2である。
このセツトにおける特に好ましい化合物の一群
は、R1が3ないし7の炭素原子、更に好ましく
は4ないし6の炭素原子、最も好しくは5の炭素
原子を有するアルキルであり、R2が置換又は未
置換のフエニル、好しくはハロゲン原子もしくは
低級アルキルで置換されたフエニルであり、R3
が水素原子であり、nが2であるような一群およ
び医薬として許容され得るそれらの酸付加塩であ
る。
第二の特に好ましい化合物群は、R1が4ない
し8の炭素原子を有するアルキルであり、R2が
置換又は未置換ベンジルであり、R3が水素原子
であり、nが1であるような一群および医薬とし
て許容され得るそれらの酸付加塩である。
本発明の特に好ましい態様は、
1―〔3―(n―オクチルチオ―n―オクチ
ル〕イミダゾール修酸塩、
1―〔3―(4―クロロフエニルチオ)―n―
オクチル〕イミダゾール修酸塩、
1―〔2―(4―クロロベンジルオキシ)―2

上記化合物の調製は、米国特許第3717655号、
第3839574号、第3658813号、第4055652号、第
4078071号、第4045568号、第4059705号および第
4123542号に述べられている。このクラスの内で
代表的化合物には、サルコナゾール
(sulconazole)、すなわち1―〔2―(4―クロ
ロベンジルチオ)―2―(2,4―ジクロロフエ
ニル)エチル〕イミダゾール、ミコナゾール
(miconazole)、すなわち1―〔2―(2,4―
ジクロロベンジルオキシ)―2―(2,4―ジク
ロロフエニル)エチル〕イミダゾール、エコナゾ
ール(econazole)、すなわち1―〔2―(4―
クロロベンジルオキシ)―2―(2,4―ジクロ
ロフエニル)―エチル〕イミダゾール、オルコナ
ゾール(orconazole)、すなわち1―〔2―
(2,6―ジクロロベンジルオキシ))―2―(4
―クロロフエニル)エチル〕イミダゾール、およ
び1―〔2―アリルオキシ―2―(2,4―ジク
ロロフエニル)―エチル〕イミダゾールが含まれ
る。
避妊薬組成物の調製および製剤において殺精子
剤としての上記化合物の使用に関し、当業者に周
知の賦形剤および技術が用いられる。製剤におけ
る適当な医薬担体およびそれらの使用は、E,
W.Martin著の「Remington′S Pharmaceutical
Science」(15版(1975年);マツク出版社・ニユ
ーヨーク)に説明されている。かかる組成物は、
全ての場合において、被投与者(体)に好ましい
投与をする為、適当な剤型を調製するため適切な
量の担体と共に有効量の活性化合物を含む。
膣内投与用に適する上記引用化合物を含有する
組成物の製剤および調製に関する本発明におい
て、多様の形態、例えば、膣坐薬、膣錠剤、膣ク
リーム、膣噴霧―泡沫剤、膣可溶ワツフル、膣ス
ポンジ等、並びに植込剤の如き有効成分の緩慢な
解放剤が当業者において典型的なものである。
各々のこれらの組成物は、かかる製剤に対し通
常用いられる医薬として許容され得る非毒性の担
体又は賦形剤中に有効量を含有する。固体の製剤
に対する典型的賦形剤は、例えば、澱粉、グルコ
ース、ラクトース、マンニツト、ステアリン酸マ
グネシウム、タルク、繊維素、炭酸マグネシウ
ム、炭酸水素ナトリウム、くえん酸等である。坐
薬の如き半固体製剤に対して、ポリアルキレング
リコール類、変性植物油又は例えば植物油、鉱物
油又はポリアルキレングリコールの配合物を含有
する軟ゼラチンカプセルの如き賦形剤が使用され

用できる。医薬組成物は、0.1ないし10.0重量%、

し、更に所望により他の活性成分を含有する。
1回の投与量は、平均的成人に対する1回の膣
内投与に対し1mgないし100mg、又は体重1Kg当

与量となる前記組成物の量からなる。
経口投与に適した上掲の化合物を含有する組成
物の調製および製剤に関する本発明において、最
終の製剤には、錠剤、丸剤、カプセル剤、粉末
剤、有効成分持続的解放剤、溶液、懸濁液、エリ
キシル剤等が含まれる。担体は石油、動物性油、
植物性油、合成油(ピーナツツ油、大豆油、鉱物
油、ゴマ油等)を含む種々の油から選ばれる。
水、塩水、水性ブドウ糖およびグリコールは液体
担体、特に注射液として好ましい。適当な医薬賦
形剤としては、澱粉、繊維素、タルク、ブドウ
糖、乳糖、蔗糖、ゼラチン、麦芽、米、小麦、白
亜、シリカゲル、ステアリン酸マグネシウム、ス

毎日雄性哺乳動物に化合物を投与することが賢明
である。
非経口的投与に適した上掲の化合物を含有する
組成物の調製および製剤に関する本発明におい
て、組成物は注射に即応した溶液又は懸濁液、溶
液もしくは注入直前懸濁液もしくは乳濁液に適し
た乾燥製品である。非経口的投与のごく最近の方
法は、当業者に一般に知られている植込タイプの
製剤、特に生体可溶性もしくは生体放解性を有す
る植込タイプの製剤の如き緩徐な有効成分持続的
解放の皮下埋込剤を介する方法である。この方法
において、長期間、例えば1ヵ月間活性成分を供
給するのにわずか1回の投与が必要とされるだけ
である。この方法に関し例えば米国特許第
3279996号および第4096239号参照。
固体製剤に対する典型的な賦形剤には、例えば
ステアリン酸マグネシウム、澱粉、ラクトース、
ゼラチン等が含まれ、液体製剤に対しては、例え
ばポリアルキレングリコール、水、植物源の油お
よびイソプロパノールおよび水素化ナフタレンの
如き低沸点溶剤があげられる。
組成物内の活性成分のパーセントは、液体製剤
において約0.1ないし10%の範囲、および注射直
前にあつて溶液又は懸濁液に適した固体組成物に
おいて約50%ないし95%の範囲にわたつている。
内移植に適した製剤は、活性成分0.1〜10%の範
囲内にある。活性成分の好ましい含量は、投与形
式に依存する。何故なら、非経口投与には、筋肉
内、皮下内および静脈内ルートが含まれるからで
ある。
本発明において説明された組成物および/又は
化合物を用いる避妊方法に関する本発明におい
て、雌性もしくは雄性のいずれか又は両性が該組
成物および/又は化合物の受容体である。
膣内投与に適した適量の組成物が、好ましくは
性交前12時間以内に一回投与又は(好ましくは)
くりかえして投与のいずれかで、雌性に適用され
る。例えば、かかる適用は、組成物がクリームも
しくはゼリーの場合注射器により、組成物が泡沫
状であればエアゾール装置により、又はワツフル
もしくは錠剤であれば手によりもしくは適当な適
用器によつて行なわれる。
雄性の適用に対しては、組成物は、性交前1週
間以内、好ましくは24時間前に経口投与され、更
に活性成分の適当量を組み合わせた1日単位の適
量形態において毎日投与されるのが好ましい。本
発明方法に関しての受精を防止するために十分な
活性成分好ましい量が以下に述べられる。他の方
法においては、皮下植込剤が使用され、化合物は
数週間又は数ヵ月にわたつて適用者に連続的に供
給される。
使用すべき方法は、勿論、投与される適用者、
特に使用される化合物、選ばれる組成物のタイプ
および処方する医者の判断に依存する。以下の実
施例は、本発明の方法および組成物を説明する。

以下に膣内投与用の製剤について説明する。
