Description
項1. (A)ナットウキナーゼ、(B)野菜粉砕物、並びに(C)エイコサペンタエン酸及び/又はドコサヘキサエン酸を含有することを特徴とする、経口組成物。
項2. 前記(C)成分として、エイコサペンタエン酸及びドコサヘキサエン酸を含有する、項1に記載の経口組成物。
項3. 前記(C)成分として、エイコサペンタエン酸及びドコサヘキサエン酸を含有する魚油を使用する、項1又は2に記載の経口組成物。
項4. 前記(B)成分が、野菜の乾燥粉末である、求項1〜3のいずれかに記載の経口組成物。
項5. 前記(B)成分を乾燥重量換算で1.7〜33.3重量%含有する、項1〜4のいずれかに記載の経口組成物。
項6. 前記(A)成分10000FU当たり、前記(B)成分を0.08g以上含有する、項1〜5のいずれかに記載の経口組成物。
項7. 前記(A)成分10000FU当たり、前記(C)成分を総量で0.063〜4.43g含有する、項1〜6のいずれかに記載の経口組成物。
項8. エイコサペンタエン酸100重量部当たり、ドコサヘキサエン酸を4.2〜2667重量部含有する、項2〜7のいずれかに記載の経口組成物。
項9. 血栓溶解剤として使用される、項1〜8のいずれかに記載の経口組成物。
項10. ソフトカプセル剤である、項1〜9のいずれかに記載の経口組成物。
項11. (A)ナットウキナーゼ、(B)野菜粉砕物、並びに(C)エイコサペンタエン酸及び/又はドコサヘキサエン酸を含有することを特徴とする、血栓溶解用の食品添加剤。
本発明の経口組成物は、ナットウキナーゼ(以下、(A)成分と表記することもある)、野菜粉砕物(以下、(B)成分と表記することもある)、並びにエイコサペンタエン酸及び/又はドコサヘキサエン酸(以下、(C)成分と表記することもある)を含有することを特徴とする。以下、本発明の経口組成物について詳述する。
本発明の経口組成物にはナットウキナーゼが含まれる。ナットウキナーゼは、納豆菌が産生するキナーゼであり、フィブリン分解作用を有する酵素である。
本発明の経口組成物には、更に野菜粉砕物が含まれる。本発明の経口組成物で使用される野菜粉砕物の種類については、特に制限されず、葉類、根菜類、芋類、豆類等のいずれの野菜であってもよい。具体的には、野菜粉砕物に含まれる野菜の種類として、人参、キャベツ、ニンニク、大麦若葉、ブロッコリー、生姜、玉葱、ウコン、ムラサキイモ、ケール、赤キャベツ、セロリ、南瓜、トマト、モロヘイヤ、ほうれん草、パセリ,ボタンボウフウ、明日葉、イチョウ葉、梅、オリーブ、杜仲、シソ、桑葉等が挙げられる。これらの野菜は、1種単独で使用してもよく、また2種以上を組み合わせて使用してもよい。
本発明の経口組成物には、前記(A)及び(B)成分に加えて、エイコサペンタエン酸及び/又はドコサヘキサエン酸を含む。このように、(A)〜(C)成分を一体不可分の関係で含有させることによって、ナットウキナーゼの活性低下を抑制することが可能になる。
本発明の経口組成物は、前記(A)〜(C)成分の他に、必要に応じて、他の栄養成分や薬理成分を含有していてもよい。このような栄養成分や薬理成分としては、飲食品や医薬品に使用可能なものであれば特に制限されないが、例えば、ビタミンB1、ビタミンB2、ビタミンB6、ビタミンB12、ビタミンC、ビタミンA、ビタミンD、ビタミンE、ビタミンK、ナイアシン、パントテン酸、葉酸、ビオチン、リコペン等のビタミン類;塩酸ベタイン、塩化カルニチン、塩化ベタネコール等の健胃剤;カルシウム、イオウ、マグネシウム、亜鉛、セレン、鉄等のミネラル類;大豆タンパク、卵白粉末、乳清タンパク等のタンパク質;グリシン、アラニン、アルギニン、アスパラギン酸、シスチン、フェニルアラニン、タウリン、トリプトファン等のアミノ酸;リノール酸、γ−リノレン酸、α−リノレン酸等の脂肪酸類;カラメル色素、クチナシ色素、アントシアニン色素、アナトー色素、パプリカ色素、紅花色素、紅麹色素、フラボノイド色素、コチニール色素、アマランス、エリスロシン、アルラレッドAC、ニューコクシン、フロキシン、ローズベンガル、アシッドレッド、タートラジン、サンセットイエローFCF、ファストグリーンFCF、ブリリアントブルーFCF、インジゴカルミン等の色素;各種フルーツのフレーバーやエッセンス等の香料;クエン酸及びその塩、リンゴ酸及びその塩、酒石酸及びその塩、酢酸及びその塩、乳酸及びその塩、食塩、グルタミン酸及びその塩、みりん、食酢、天然果汁、植物抽出エキス、果実・海産物等の裁断物又は粉末化物等の調味剤;アガリクス、シイタケエキス、レイシ、ヤマブシタケ等のキノコ類又はそのエキス;食物繊維、ローヤルゼリー、プロポリス、ハチミツ、コンドロイチン硫酸、グルコサミン、セラミド、ヒアルロン酸等のその他機能性素材等が挙げられる。これらの添加成分は、1種単独で使用してもよく、また2種以上を組み合わせて使用してもよい。また、これらの添加成分の含有量については、使用する添加成分の種類や経口組成物の用途等に応じて適宜設定される。
本発明の経口組成物の剤型については、経口摂取又は経口投与が可能であることを限度として特に制限されず、固体状、半固体状、又は液体状のいずれであってもよく、該経口組成物の種類や用途に応じて適宜設定すればよい。
本発明の血栓溶解用の食品添加剤は、(A)ナットウキナーゼ、(B)野菜粉砕物、並びに(C)エイコサペンタエン酸及び/又はドコサヘキサエン酸を含有することを特徴とする。
本発明の抑制方法は、ナットウキナーゼの活性低下を抑制する方法であって、(A)ナットウキナーゼに、(B)野菜粉砕物、並びに(C)エイコサペンタエン酸及び/又はドコサヘキサエン酸を共存させることを特徴とする。
ナットウキナーゼ粉末:「納豆菌培養エキス末BNK4000」(株式会社日本生物化学研究所製);1g当たりのナットウキナーゼの活性は43300FU
玉葱乾燥粉末1:「さらさらレッドパウダー(タマネギ乾燥粉末)」(こだま食品株式会社製);玉葱の乾燥粉末
玉葱乾燥粉末2:「玉葱パウダーS」(こだま食品株式会社製);玉葱の乾燥粉末
野菜ミックス乾燥粉末:「野菜ミックスパウダー」(こだま食品株式会社製);人参、キャベツ、ニンニク、大麦若葉、ブロッコリー、生姜、玉葱、ウコン、ムラサキイモ、ケール、赤キャベツ、セロリ、南瓜、トマト、モロヘイヤ、ほうれん草、及びパセリの乾燥粉末の混合物
サツマイモ乾燥粉末:「アヤムラサキパウダー」(こだま食品株式会社製);アヤムラサキの乾燥粉末
魚油1:「精製魚油 EPA−28」(日本水産株式会社製);イワシから得られた魚油、エイコサペンタエン酸26.74重量%及びドコサヘキサエン酸11.8重量%含有
魚油2:「DDオイル DHA−46」(日本水産株式会社製);マグロの頭部から得られた魚油、エイコサペンタエン酸5.12重量%及びドコサヘキサエン酸41.56重量%含有
魚油3:「TUNA OIL 25DHA」(Polaris nutritional lipids製);マグロから得られた魚油、エイコサペンタエン酸5.12重量%及びドコサヘキサエン酸25.51重量%含有
魚油4:「OMEGAVIE DHA70TG」(Polaris nutritional lipids製);マグロから得られた魚油を精製し得られた魚油。エイコサペンタエン酸5.35重量%及びドコサヘキサエン酸65.52重量%含有
魚油5:「OMEGAVIE EPA70TG」(Polaris nutritional lipids製);イワシ、サバから得られた魚油を精製し得られた魚油。エイコサペンタエン酸62.49重量%及びドコサヘキサエン酸4.98重量%含有
表1及び2に示す組成の経口組成物を調製し、各経口組成物1g当たりのナットウキナーゼの活性値(FU/g)を以下の方法で測定した。測定したナットウキナーゼの活性値から、以下の算出式に従って、ナットウキナーゼの活性維持率(%)、及びナットウキナーゼの活性向上率(%)を算出した。
1.試薬
(1)ホウ砂緩衝液
0.05モル/Lのホウ砂及び0.15モル/Lの塩化ナトリウムを含有する緩衝液(pH8.5)
(2)トリクロロ酢酸溶液
0.2モル/Lのトリクロロ酢酸を含有する水溶液
(3)フィブリノーゲン溶液
前記ホウ砂緩衝液に、牛血漿由来フィブリノーゲン(シグマ社製)を0.7重量%となるように溶解した溶液
(4)トロンビン溶液
前記ホウ砂緩衝液に、牛血漿由来トロンビン(シグマ社製)を20U/mlとなるように溶解した溶液
(5)希釈緩衝液
2ミリモル/Lの硫酸カルシウム、10モル/Lの塩化ナトリウム、2ミリモル/Lの酢酸ナトリウム、0.005重量%のTriton X−100を含む水溶液(pH6.0)
希釈緩衝液約8mLに各経口組成物1g添加してスパテールでよく撹拌した後に、4000×g、20分間遠心分離して上清を回収し、希釈緩衝液で10mLに調整して試料溶液とした。なお、各試料溶液は測定時の吸光度差が0.04〜0.08(活性値が活性値は0.67〜1.33FU/mL)の範囲内となるように調整した。
試験管にホウ砂緩衝液1.4mL及びフィブリノーゲン溶液0.4mLを量り取り、37℃の恒温水槽で5分間加温した後、トロンビン溶液0.1mLを加えて撹拌した。この液を37℃で10分間静置した後、試料溶液0.1mLを加え撹拌し、37℃で静置した。試料溶液を添加してから一定時間毎に撹拌し、60分後にトリクロロ酢酸溶液2mLを加えて撹拌し、更に20分間37℃で加温しながら静置した。この液(反応液)をマイクロテストチューブに入れ、15000×g、5分間遠心した。次いで、パスツールピペットを用いて慎重に遠心上清1mLを回収し、275nmにおける吸光度(Aγ)を測定した。
下記組成の内容液及び剤皮を用いて、ソフトカプセル剤(1剤当たり、内容液300mg、剤皮150mg含有)を調製した(製剤例1〜14)。得られたソフトカプセル剤を前記試験例1と同様の方法で試験したところ、いずれも、ナットウキナーゼの活性低下を十分に抑制できていた。
下記組成の錠剤(1錠当たり700mg、製剤例15〜17)及び顆粒剤(製剤例18〜20)を製造した。得られた錠剤及び顆粒剤を前記試験例1と同様の方法で試験したところ、いずれも、ナットウキナーゼの活性低下を十分に抑制できていた。
下記組成のドリンク剤(製剤例21)及びゼリー剤(製剤例22)を製造した。得られたドリンク剤及びゼリー剤を前記試験例1と同様の方法で試験したところ、いずれも、ナットウキナーゼの活性低下を十分に抑制できていた。
Claims (9)
- (A)ナットウキナーゼ、(B)野菜粉砕物、並びに(C)エイコサペンタエン酸及びドコサヘキサエン酸を含有し、
前記(A)成分が納豆菌の培養物の精製品又は粗精製品であり、
前記(B)成分を乾燥重量換算で1.7〜33.3重量%含有することを特徴とする、経口組成物。 - 前記(C)成分として、エイコサペンタエン酸及びドコサヘキサエン酸を含有する魚油を使用する、請求項1に記載の経口組成物。
- 前記(B)成分が、野菜の乾燥粉末である、請求項1又は2に記載の経口組成物。
- 前記(A)成分10000FU当たり、前記(B)成分を0.08g以上含有する、請求項1〜3のいずれかに記載の経口組成物。
- 前記(A)成分10000FU当たり、前記(C)成分を総量で0.063〜4.43g含有する、請求項1〜4のいずれかに記載の経口組成物。
- エイコサペンタエン酸100重量部当たり、ドコサヘキサエン酸を4.2〜2667重量部含有する、請求項1〜5のいずれかに記載の経口組成物。
- 血栓溶解剤として使用される、請求項1〜6のいずれかに記載の経口組成物。
- ソフトカプセル剤である、請求項1〜7のいずれかに記載の経口組成物。
- (A)ナットウキナーゼ、(B)野菜粉砕物、並びに(C)エイコサペンタエン酸及びドコサヘキサエン酸を含有し、
