Description
好ましい実施態様では、本発明のコンジュゲートは、ステロールがコレステロールであるものである。
別の好ましい実施態様では、コンジュゲートは、ガイディングリガンドがペプチドであるものである。別のより好ましい実施態様では、ガイディングリガンドは、RGD配列を含む。別のさらにより好ましい実施態様では、ガイディングリガンドは、配列番号1:RGDFKの配列を有するペプチドであり、ここで、RGD及びK残基はL−アミノ酸で、F残基はD−アミノ酸である。
好ましい実施態様では、上に記載のリポソームは単峰性の粒子径分布を有する。
別の好ましい実施態様では、上に記載のリポソームは、25から500ナノメートルまでの平均粒子径を有し、平均Z電位の絶対値は20から90mVまでである。
別の好ましい実施態様では、上に記載のリポソームは、治療剤を更に含有する。より好ましい実施態様では、治療剤は、α−ガラクトシダーゼ(又は略してGLA)である。
本発明の第四の態様は、医薬として使用するための上に記載のリポソームに関する。

ヒトを含む、治療を必要とする対象に、薬学的に許容される賦形剤又は担体と共に、上に記載のリポソームの治療有効量を投与することを含む、ファブリー病の治療及び/又は予防の方法もまた本発明の一部と考えられる。
本発明の第七の態様は、上に記載のコンジュゲートの調製方法であって、a)ステロールを塩基及び溶媒の存在下でハロゲン化スルホニルと反応させて、対応するスルホニルエステルを得る工程;b)工程a)で得られた化合物を、溶媒の存在下で2から10の繰返し数を持つポリエチレングリコールと反応させる工程;c)工程b)で得られた化合物を炭酸ジスクシンイミジルで活性化する工程;d)工程c)で得られた化合物を、塩基及び溶媒の存在下、RGD配列を含むペプチドと反応させる工程を含む、方法を提供することである。好ましくは、スルホニルエステルは、メシレート、ベシレート、又はトシレートである。
ここで使用される「リポソーム」及び「小胞体」との用語は交換可能である。当業者には、リポソームと呼ばれる小胞体は、その膜中に幾らか存在するリン脂質を有している一方、小胞体との用語はリポソームがリン脂質を欠いている場合もまた使用されることが通常理解される。何れにせよ、本発明の一部であるコンジュゲートは、リン脂質を含むか又は欠く小胞体に含ませることができる。
ここで使用される「治療的有効量」との用語は、投与されると、疾患又は状態の一又は複数の症状の発症を予防するか、又は何らかの形で軽減するのに十分である化合物の量を意味する。本発明に従って投与される化合物の特定の用量は、投与化合物、投与経路、治療されている疾患、及び類似の考慮事項を含む、それぞれの場合に関連する状況によって明らかに設定されるであろう。
「薬学的に許容される賦形剤」との用語は、薬学的に許容される物質、組成物又はビヒクル、例えば液体又は固体の充填剤、賦形剤、溶媒、又は封入材料を意味する。各成分は、薬学的組成物の他の成分と適合性があるという意味で薬学的に許容可能でなければならない。それは、あまりに多い毒性、刺激又はアレルギー反応、免疫原性又は他の問題もしくは合併症がなく妥当なリスク/ベネフィット比で、ヒト及び動物の組織又は器官と接触させられて使用されるのに適していなければならない。

コレステロールトシル(A)の調製:
フラスコ中にコレステロール(1当量、1.006g)を入れ、無水ピリジン(12mL)に溶解した。ついで、塩化トシル(2当量、1.004g)を添加し、溶液を室温で24時間撹拌したままにした。その溶液に5mLの水を添加し、粗生成物をDCM(3×6mL)で抽出した。Et2O(全溶液)を有機相に添加した後、MgSO4で乾燥させ、濾過し、溶媒を除去して乾固させた。粗生成物を石油エーテルから再結晶し、白色固形物(0.969g、70%)を得た。
フラスコ中に0.503gのコレステロールトシル(A)を加え、5mLの無水1,4−ジオキサンに溶解した。3.591gのテトラエチレングリコール(20当量)をこの溶液に加え、混合物をアルゴン雰囲気、還流下で4時間反応させた。得られた溶液を濃縮乾固した後、20mLのDCMに溶解し、2×20mLの飽和NaHCO3、3×20mLのH2O、1×20mLの飽和NaClで洗浄した。得られた水性相を20mLのDCMで再び抽出した。最後に、有機相を合わせた後、MgSO4で乾燥させ、濾過し、溶媒を蒸発させ乾固させた。粗生成物の精製は塩基性アルミナを用いたクロマトグラフィー(DCM/MeOH0−5%)によって実施した。黄色がかった油として0.260gの生成物を得た(50%)。
化合物B(1当量、0.2746g)を9mLのDCM:ACN:DIPEA(1:1:1)の混合物に溶解し、DSC(3当量、0.2746g)を加えた。混合物をアルゴン雰囲気下で撹拌したところ、16時間後に出発生成物Bの消失が認められた。溶媒を蒸発させて乾固させ、粗生成物を5mLのDCMに溶解させた。この有機相を5mLの水で洗浄した。有機相をMgSO4で乾燥させ、濾過し、最後に溶媒を蒸発させ乾固させた。粗生成物を、更に精製することなく次の反応に使用した。
D−ペプチドの合成を、僅かに変更してDai X.等 "An improved synthesis of a selective αvβ3-integrin antagonist cyclo (-RGDfK)" Tetrahedron Letters 2000, vol. 41, pp. 6295-6298に記載されたようにして実施した。簡単に説明すると、最終の環状ペプチドを、TFA/TIS/H2O(95:2.5:2.5)の混合物を用いて脱保護し、RP−HPLCによって精製し、D−ペプチド(180.9mg、15%)を白色固形物として得た。
化合物C(1.5当量、69.00mg)を5mLの無水DMFに溶解させ、DIPEA(2当量、32μL)とD−ペプチド(1当量、65.95mg)を添加した。反応物を、D−ペプチドが消失するまで(HPLC−MSにより管理)16時間攪拌したままにした。溶媒を除去し、粗生成物をTMBE(3×)から沈殿させた。化合物Eを白色固形物(55.5mg、74%)として得た。HPLC-MS: (対称300 C4, 5-100% B, A: ACN B: H2O, 30分, 1 mL/分, λ= 210 nm) rT: 17.31分, m/z = 1192,8 [M+H]+, 算定質量: 1192,53。HPLC-PDA: (C4, 5-100% B, A: ACN B: H2O, 30分, 1 mL/分, λ= 210 nm) rT: 20.1分(98% 210 nm)。アミノ酸の分析: 0.99, Gly: 1.18, Arg: 1.15, Lys: 0.92, Phe: 1.04
コレステロール−DCI(F)の調製:
コレステロール(1当量、200.2mg)を9mLのDCM/DIPEA/ACN(1:1:1)の混合物に溶解し、これにCDI(10当量、852.1mg)を加えた。一晩反応させたところ、所望の生成物の出現が観察された。溶媒を蒸発させて乾固させ、粗生成物を5mLのDCMに溶解させた。この有機相を水5mLで洗浄した。有機相をMgSO4で乾燥させ、濾過し、最後に溶媒を蒸発させて乾固させた。粗生成物を更に精製することなく次の反応に用いた。
化合物F(1当量、109.6mg)を取り上げ、DCMに溶解させ、この溶液に、1−アミノ−3,6,9−トリオキサウンデカニル−11−オール(4.5当量、152.8mg)を加え、ついでその溶液を、DIPEAを使用してpH8にした。その混合物を反応させ、出発生成物Fの消失が観察されるまで、HPLC−MSによってその進行をモニターした。粗生成物を洗浄し、水で有機相から抽出を行った。有機相をMgSO4で乾燥させ、濾過し、最後に溶媒を蒸発させて乾固した。最終の粗生成物をシリカカラム(均一濃度AcOEt)により精製して、80.2mgの化合物Gを黄色がかった油として得た(60%)。HPLC-MS: (XSelect C18, 5-100% B, A: ACN B: MeOH, 4.5分, 2 mL/分, λ= 210 nm) rT: 1.87分 m/z = 606.47 [M+H]+, 算定質量: 605.89。HPLC-PDA: (C4, 5-100% B, A: ACN B: H2O, 30分, 1 mL/分, λ= 210 nm) rT: 21.8分(95% 210 nm) 1H NMR: (400 MHz, CDCl3) δ: 0.680 (s, 3H); 0.86 (d, 1.8 Hz, 3H); 0.87 (d, 1.8 Hz, 3H); 0.91 (d, 6.5 Hz, 3H); 1.01 (s, 3H); 3.36 (m, 1H); 3.65 (m, 16H). 13C NMR: (100 MHz, CDCl3) δ: 156.58 (C29), 140.07 (C4), 122.53 (C3), 70.74 (C8), 70.56-70-25 (C33, C35, C36, C38, C39, C41), 61.79 (C42), 56.83 (C14), 56.27 (C15), 50.15 (C6), 42.45 (C13), 40.85 (C32), 39.88 (C22), 39.66 (C12), 38.75 (C7), 37.15 (C5), 36.71 (C10), 36.32 (C20), 35.93 (C18), 32.05 (C1), 32.02 (C2), 28.37 (9), 28.35 (C17), 28.15 (C16), 24.43 (C23), 23.97 (C21), 22.96 (C24), 22.70 (C25), 21.18 (C11), 19.48 (C19), 18.85 (C27), 12.00 (C26)。
化合物G(1当量、80.2mg)を取り上げ、9mLのDCM/DIPEA/ACN混合物(1:1:1)中のDSC(10当量、316.8mg)と16時間反応させた。化合物Gの消失が検出されたところで、溶媒を蒸発させて乾固させ、粗生成物を5mLのDCMに溶解し、水で洗浄した。有機相をMgSO4で乾燥させ、濾過し、溶媒を蒸発させ、更に精製することなく使用できる粗生成物を得た。
化合物H全部を5mLの無水DMFに溶解させ、DIPEA(2当量、60ml)とD−ペプチド(1当量、51.2mg)を加えた。反応物を、D−ペプチドが消失するまで(HPLC−MSにより管理)16時間攪拌したままにした。溶媒を除去し、粗生成物をTMBE(3×)から沈殿させ、最後に水で洗浄した。このようにして、17mgの生成物Iが白色固形物として得られた。
本発明のコンジュゲート(エーテル結合)又は従来技術のコンジュゲート(カルバメート結合)を運ぶ小胞体を、他の文献(Cano-Sarabia, M. et. al. "Preparation of Uniform Rich Cholesterol Unilamellar Nanovesicles Using CO2-Expanded Solvents" Langmuir 2008, vol. 24, pp. 2433-2437)に記載の手順に基づいて、以下に記載の通りに、調製した。
先ず、1.2mLのエタノール中の8mgのコレステロール、24mgのDPPC及び4mgのコンジュゲート(コレステロール−PEG−RGD)の溶液を、大気圧下、操作温度(TW=35℃)で、7.5mLの容積を有する高圧反応器に入れた。圧縮CO2を加え、モル比XCO2=0.8及び操作圧力PW=10MPaまで溶液の体積膨張を生じせしめた。全体の均質化と熱平衡を達成するため、系を、10MPaと35℃に約60分間放置した。最後に、膨張有機溶液を、24mLの水溶液の上で、操作圧から大気圧に減圧した。この最終工程では、10MPaのN2流を、減圧中に反応器内の操作圧を一定に維持するためにエタノール中のコレステロール溶液を押出すプランジャーとして使用した。ついで、小胞体を容器に移し、そこで、密封されて、使用するまで5±3℃で保存された。
平均粒子径、粒子径分布及びZ電位は25℃の温度でDLS(動的光散乱)によって決定された。
表1は、異なったバッチの小胞体DPPC:コレステロール:コレステロール−PEG−RGDの物理的外観、平均粒子径及びZ電位の結果を示す。
本発明のコンジュゲート運搬リポソームが、先行技術のコンジュゲート運搬リポソームと比較して、その内容物の細胞内送達の点で優れた性質を有しているかどうかを評価するため、細胞株中でのリポソームの内容物の内部移行の実験を実施した。送達される物質は、蛍光によって容易にモニターできる染料であった。
リポソーム標識化のために、500μLのプレーンリポソーム(DPPC:Colリポソーム)を25μLのDidエタノール溶液(1μM)と直接混合して、膜中50nmの染料の最終濃度にした。30分の混合後に、遊離のDiDを全試料からゲル濾過により分離した。このために、事前に充填されたカラム(PD SpinTrap G-25)をPBS緩衝液で3回平衡化した後、130μLの試料を添加した。分離は、800×gのスピン遠心分離によって行った。
上記の手順を、他のリポソーム型に対しても同様に繰り返した。
CDC/EU.HMEC−1(HMEC−1)細胞は疾病管理予防センター(CDC−NIDR)から提供された。HMEC−1は、正常なヒト微小血管内皮細胞の形態学的、表現型、及び機能的特徴を保持する不死化されたヒト微小血管内皮細胞株である。HMEC−1細胞を、5%CO2を含む37℃の加湿雰囲気下、50単位ml−1のペニシリン、50μg ml−1のストレプトマイシン、10mMのL−グルタミン及び10%のウシ胎児血清(FBS)を補充したMCDB131(Invitrogen)中に維持した。培地、血清及び抗生物質は全てInvitrogenから購入した。
HMEC−1細胞を、プレート当たり2×105細胞の密度でFluorodish培養プレート(World Precision Instruments, Sarasota, FL)に播種し、36−48時間増殖させた。50μlのDiD標識リポソーム(DPPC:CoL)又はDiD標識リポソーム−RGDコンジュゲート(DPPC:Col:Col−PEG−RGDエーテルリポソーム及びDPPC:Col:Col−PEG−RGDカルバメートリポソーム)(1.5mg/ml)を200μlのMCDB131培地と混合し、細胞中に添加し、5%のCO2を含む加湿雰囲気中で37℃で3時間インキュベートした。その後、細胞を無血清MCDB131で洗浄し、エンドソーム/リソソームコンパートメントを標識するためにリソトラッカーグリーンDND−26(50nM、分子プローブ、Eugene, Oregon)と共に5分間、37℃でインキュベートした。細胞を、60×1.42NA油浸対物レンズを使用して倒立ライカSP5レーザー走査型共焦点分光顕微鏡(Leica Microsystems Heidelberg GmbH, Mannheim, Germany)で調べた。2色の蛍光を同時に視覚化するために、我々はリソトラッカーグリーンにはアルゴンレーザーの514nm線を、DidにはHe−Neレーザーの630nm線を使用した。
DPPC:Col:Col−PEG−RGDエーテルリポソーム(上段)、DPPC:Col:Col−PEG−RGDカルバメートリポソーム(中段)及びDPPC:Col(下段)リポソームに対する共焦点画像を図3に示す。レッドチャネル(右側の列)に観察される斑点は、細胞によって内部移行された標識リポソームの存在を示す。DPPC:Col:Col−PEG−RGDエーテルリポソームでは、この内部移行が、他の2つに比べて高いことが明確に観察される。
HMEC−1細胞を、実験の36−48時間前にFluorodish培養プレート(World Precision Instruments, Sarasota, FL)上に2×105細胞ml−1の密度で播種した。細胞を37℃で3時間、FBSなしで10mMのL−グルタミンを補填したMCDB131中に再懸濁させたDiD標識リポソーム(DPPC:Col)又はDiD標識リポソーム−RGDコンジュゲート(DPPC:Col:Col−PEG−RGDエーテルリポソーム及びDPPC:Col:Col−PEG−RGDカルバメートリポソーム)(0.3mg/ml)と共にインキュベートした。その後、細胞をダルベッコのリン酸緩衝生理食塩水(DPBS)溶液で2回洗浄し、トリプシンを使用して剥離し、細胞培養培地に再懸濁させた後、蛍光活性化細胞選別分析に供した。データ収集と解析は、FACSスキャン(Beckton-Dickinson)及びBD FACSDivaソフトウェアを使用して実施した。10000生存細胞を各実験において評価した。
図4には、DID標識リポソームを内部移行した細胞に関連した蛍光強度を見ることができる。この内部移行は、DPPC:Col:Col−PEG−RGDカルバメート運搬リポソーム又はコンジュゲートを含まない単純なDPPC:Colリポソームを使用する場合よりもDPPC:Col:Col−PEG−RGDエーテル運搬リポソームを使用する場合に有意に高いことが示されている。
活性アッセイ:
GLA(αガラクトシダーゼ)欠損マウス(GlatmKul1)のマウス大動脈内皮細胞(MAEC)の初代培養を、以前に記載された手順(Shu L.等 “An in vitro model of Fabry disease” J. Am. Soc. Nephrol. 2005, vol. 16, pp. 2636-45)に従って単離した。単離細胞の内皮起源はCD105染色によって確認した。
活性アッセイのために、2〜5の継代の細胞を24ウェルプレートに播種し、37℃、5%CO2に維持した。播種から24時間後に8μMのNBD−Gb3(Matreya)を、特定の濃度の試験化合物(遊離酵素−α−ガラクトシダーゼ、酵素含有リポソーム又は空のリポソーム)と共に培養物に添加した。48時間のインキュベーション後、細胞をトリプシン処理し、Gb3−NBD蛍光シグナルをフローサイトメトリーメーター(FacsCalibur, Beckton Dickinson)により分析した。Gb3−NBDシグナルの割合を計算するために、コントロール細胞(処理なし)における蛍光シグナルを100%として設定し、値の残りをそれに応じて正規化した。α−ガラクトシダーゼ活性はそのGb3沈着物を減少させるので、Gb3損失割合(Gb3損失%=100−Gb3-NBDシグナル%)を使用して結果をプロットした。
また図6には、エーテル結合の透析濾過コンジュゲート運搬リポソームが、様々な濃度で、カルバメート結合のコンジュゲート運搬リポソームよりも高い活性を有していることが示されている。
組換えGLA(α−ガラクトシダーゼ)の生産
発現ベクターpOpinE−GLAはpOPINEプラスミド(Berrow NS, et al. “A versatile ligation-independent cloning method suitable for high-throughput expression screening applications” Nucleic Acids Res 2007, vol. 35, e45)中にクローニングしたヒトα−ガラクトシダーゼ(GLA)遺伝子の完全長型をコードする。懸濁液適応型HEK(ヒト胚性腎臓)細胞株FreeStyleTM293F(Gibco, Invitrogen社)を使用して、PEI媒介一過性遺伝子発現により組換えGLAを生成した。GLA生産及び精製の詳細は、先に記載されている(Corchero JL等, “Integrated approach to produce a recombinant, his-tagged human alpha-galactosidase a in mammalian cells”, Biotechnol Prog. 2011, vol. 27, pp. 1206-1217)。
リポソーム中への組換えGLAの導入を推定するために、1)初期の水中GLAのサンプル、2)脂質と混合したGLAのサンプル、3)精製したリポソームのサンプル及び4)遊離の非封入GLAを含む水のサンプルを、変性添加緩衝液と混合し、SDS−PAGEによって分析し、更にSanta Cruz Biotechnologyからのウサギポリクローナル抗GLA血清(a-gal A H-104: sc-25823)とヤギ抗ウサギIgG HRPコンジュゲート(Bio-Rad Laboratories社, カタログ# 170-6515)を二次抗体として用いてウエスタンブロットによって反応させた。上記の各サンプル中の組換えGLAの量を、我々の研究室において以前に産生され、精製され、定量された既知量(通常25〜125ngの範囲)の組換えGLAとの比較によって推定した。定量的に比較されるサンプルは、同じゲルで実験し、セットとして処理した。バンドの濃度測定分析はQuantity Oneソフトウェア(Bio-Rad Laboratories社)を用いて実施した。封入されたGLAの割合は、精製リポソーム画分中に見出されるGLAの量を添加されたGLAの全初期量と比較することによって得られた。
様々なサンプル中に見出されたGLAのα−ガラクトシダーゼ酵素活性を、Desnick等(Desnick RJ等 “Fabry's disease: enzymatic diagnosis of hemizygotes and heterozygotes. Alpha-galactosidase activities in plasma, serum, urine, and leukocytes”, J. Lab. Clin. Med. 1973, vol. 81, pp. 157-171)に記載のものにMayes等(Mayes JS.等 “Differential assay for lysosomal alpha-galactosidases in human tissues and its application to Fabry's disease” Clin. Chim. Acta 1981, vol. 112, pp. 247-251)の修正を加えた方法に従ってアッセイした。簡単に説明すると、酵素活性を、アッセイ緩衝液(0.01M酢酸、pH4.5)中2.46mMの濃度の4メチルウンベリフェリルαD−ガラクトシド(4MUG、Sigma Chemical)を基質として使用してアッセイした。典型的なアッセイ反応混合物は100μlの基質と25μlの酵素サンプルを含む。酵素反応は1時間37℃で攪拌下で起こり、1.25mLの0.2Mグリシン−NaOH緩衝液(pH10.4)で停止させた。放出された生成物(4−メチルウンベリフェロン又は4−MU)は、励起波長及び発光波長がそれぞれ365及び450nmの蛍光測定によって決定した。0.2Mグリシン−NaOH緩衝液(pH10.4)中に0から500ngの4−MU/mlの市販4−MU(Sigma Chemical)を含むサンプルを使用して、読み取り値を較正するための標準曲線を作成した。比酵素活性は、μmol4−MU/h/mgタンパク質として表される。
様々なサンプル中のGLAの安定性を、それらの比酵素活性を調べることによりアッセイした。それらの調製(「0日目」)後、各サンプルについてGLAタンパク質量と酵素活性を決定した(前述したようにして)。それらの値を用いて、初期の比酵素活性を決定した。サンプルを4℃で水中に維持し、様々な時点で、酵素活性をアッセイした。GLAの初期の量を使用して、比酵素活性を再計算し、基準として使用される「0日目」に決定されたものと比較した。
それぞれの封入実験では、次のサンプルを得て、分析した。
1.初期GLA(封入の前)。
2.全GLA、つまり、ポイント1に脂質を加え、初期GLAの画分を封入した結果物。全とは、封入GLAと非封入GLAの双方を意味する。溶液中に残存する遊離GLAからのGLA封入リポソームを分離するとポイント3及び4が得られる。
3.リポソーム封入GLA、つまり、封入されているGLAのみ。
4.遊離GLA、つまり封入されていないGLA。
これらの結果によれば、小胞体中へのGLA封入効率は、27+/−6.8%(平均+/−標準偏差)である。
分かるように、小胞体又はその成分とまだ結合されていない初期GLAと比較すると、「全」サンプル(小胞体を形成する脂質と混合されたGLA)の比酵素活性は明らかに増加する(4から18倍)。

- i)ステロール;

iii)標的細胞中に存在する一又は数種のレセプターに選択的に結合可能なガイディングリガンドであって、ii)の遠位端に共有結合されるガイディングリガンド
を含むコンジュゲートであって、ガイディングリガンドがペプチドである、コンジュゲート。 - ステロールがコレステロールである、請求項1に記載のコンジュゲート。
- ポリエチレングリコール鎖が2から10の繰返し数を有する、請求項1又は2に記載のコンジュゲート。
- ガイディングリガンドがRGD配列を含む、請求項1から3の何れか一項に記載のコンジュゲート。
- ガイディングリガンドが配列番号1のペプチドである、請求項4に記載のコンジュゲート。
- 請求項1から5の何れか一項に記載のコンジュゲートを含むリポソーム。
- 単峰性の粒子径分布を有する、請求項6に記載のリポソーム。

b)工程a)で得られた化合物を、溶媒の存在下で2から10の繰返し数を持つポリエチレングリコールと反応させる工程;
c)工程b)で得られた化合物を炭酸ジスクシンイミジルで活性化する工程;
d)工程c)で得られた化合物を、塩基及び溶媒の存在下、RGD配列を含むペプチドと反応させる工程
を含む、方法。 - 請求項6から11の何れか一項に記載のリポソームの調製方法であって、
a)界面活性剤を任意選択的に含んでいてもよい水溶液を調製する工程;
b)請求項1から5の何れか一項に記載のコンジュゲートと、任意選択的に、有機溶媒に溶解させたリン脂質を含有する溶液を調製する工程であって、該有機溶液が圧縮流体で膨張せしめられている工程;
c)任意選択的に、工程a)の溶液か、工程b)の溶液の何れかに、該溶液を膨張させる前に、治療剤を加える工程;及び
d)工程a)で得られた溶液に対して工程b)で得られた溶液を減圧する工程
を具備する方法。
