本発明は、イミド誘導体の製造方法に関する。

【従来の技術】
例えば下記式(4)
で示される2−[[2−[[4−(1,2−ベンズイソチアゾール−3−イル)−1−ピペラジニル]メチル]シクロヘキシル]メチル]ヘキサヒドロ−4,7−メタノ−1H−イソインドール−1,3(2H)−ジオンに代表される一般式(3)
(式中、Aは、置換されていてもよい炭素数2〜4のアルキレン基または置換されていてもよい炭素数2〜4のアルケニレン基を表わし、Dは、カルボニル基またはスルホニル基を表わす。Yは、置換されていてもよい炭素数1〜2のアルキレン基を表わし、Zは、置換されていてもよいメチレン基または置換されていてもよいイミノ基を表わす。)
で示されるイミド誘導体は、例えば精神分裂病、老年性精神疾患、そううつ病、神経症等の治療薬の中間体として有用である(例えば特開平5−17440号公報等)。

で示される第四級アンモニウム塩とを、炭酸カリウムおよびジベンゾ−18−クラウン−6−エーテルの存在下に反応させる方法が知られている(特開平5−17440号公報)が、高価で回収が困難なクラウンエーテルを用いる方法であるため、さらに工業的に有利な製造方法の開発が望まれていた。

【発明が解決しようとする課題】
このような状況のもと、本発明者らは、上記一般式(3)で示されるイミド誘導体をさらに工業的に有利に製造する方法について鋭意検討したところ、上記一般式(1)で示されるイミド化合物と一般式(2)で示される第四級アンモニウム塩との反応を、固体無機塩基および水の存在下に実施することにより、収率よく、目的とする上記一般式(3)で示されるイミド誘導体が得られることを見いだし、本発明に至った。

【課題を解決するための手段】
すなわち本発明は、芳香族炭化水素系溶媒中、一般式(1)
(式中、Aは、置換されていてもよい炭素数2〜4のアルキレン基または置換されていてもよい炭素数2〜4のアルケニレン基を表わす。Dは、カルボニル基またはスルホニル基を表わす。)
で示されるイミド化合物と一般式(2)
(式中、Yは、置換されていてもよい炭素数1〜2のアルキレン基を表わし、Zは、置換されていてもよいメチレン基または置換されていてもよいイミノ基を表わし、X-は、対アニオンを表わす。)
で示される第四級アンモニウム塩とを、固体無機塩基および水の存在下に反応させることを特徴とする一般式(3)

【発明の実施の形態】
まず、本発明の原料化合物について、説明する。
一般式(1)
で示されるイミド化合物(以下、イミド化合物(1)と略記する。)の式中、Aは、置換されていてもよい炭素数2〜4のアルキレン基または置換されていてもよい炭素数2〜4のアルケニレン基を表わし、Dは、カルボニル基またはスルホニル基を表わす。

炭素数2〜4のアルキレン基としては、例えばエチレン基、トリメチレン基、テトラメチレン基等が挙げられ、炭素数2〜4のアルケニレン基としては、例えばエテニレン基、プロペニレン基、1−ブテニレン基、2−ブテニレン基、ブタジエニレン基等が挙げられる。

かかる炭素数2〜4のアルキレン基および炭素数2〜4のアルケニレン基は、置換基を有していてもよく、置換基としては、例えば低級アルキル基、低級アルコキシ基、低級アルケニル基、低級アルコキシ基で置換された低級アルキル基、低級アルコキシ基で置換された低級アルケニル基等が挙げられる。かかる置換基は、一緒になって酸素原子を有していてもよいアルキレン鎖または酸素原子を有していてもよいアルケニレン鎖を形成してもよい。

続いて、本発明について説明する。
本発明は、芳香族炭化水素系溶媒中、イミド化合物(1)と第四級アンモニウム塩(2)とを、固体無機塩基および水の存在下に反応させるものである。芳香族炭化水素系溶媒としては、例えばトルエン、キシレン、メシチレン、クロロベンゼン、ジクロロベンゼン等の単独または混合溶媒が挙げられる。かかる芳香族炭化水素系溶媒の使用量は、イミド化合物(1)に対して、通常5重量倍以上、好ましくは10重量倍以上であり、その上限は特にないが、あまり多すぎると容積効率が悪くなるため、実用的には、イミド化合物(1)に対して、30重量倍以下である。

以下、実施例により本発明をさらに詳細に説明するが、本発明はこれら実施例に限定されない。なお、分析は高速液体クロマトグラフィ(以下、LCと略記する。)を用いた。

4’−(1,2−ベンズイソチアゾール−3−イル)−(3aR,7aR)−オクタヒドロ−スピロ[2H−イソインドール−2,1’−ピペラジニウム]メタンスルホン酸塩17.6gを含むトルエン溶液に、ヘキサヒドロ−(3aS,4R,7S,7aR)−4,7−メタノ−1H−イソインドール−1,3(2H)−ジオン10.4g、炭酸カリウム7.0gおよび水0.76gを加え、2時間還流させた。有機層を水洗し、活性炭処理した後、トルエンを一部濃縮除去し、濃縮後の溶液45.5gに、メタノール113gを加え、種晶を添加した後、さらにメタノール76gを加え、冷却した。析出した結晶を濾取し、洗浄、乾燥し、2−[[(1R,2R)−2−[[4−(1,2−ベンゾイソチアゾール−3−イル)−1−ピペラジニル]メチル]シクロヘキシル]メチル]ヘキサヒドロ−(3aS,4R,7S,7aR)−4,7−メタノ−1H−イソインドール−1,3(2H)−ジオン17.1gを得た。収率:83%(4’−(1,2−ベンズイソチアゾール−3−イル)−(3aR,7aR)−オクタヒドロスピロ[2H−イソインドール−2,1’−ピペラジニウム]メタンスルホン酸塩基準)、純度:99.1%(LC修正面積百分率値)。
