【発明の属する技術分野】
本発明は電気化学センサを用いアンペロメトリー(Amperometry)法により血液などに含まれるグルコース、アルコール等を分析、測定する電気化学センサ測定装置及びその測定方法に関する。

【従来の技術】
従来から、バイオセンサ等の電気化学センサを簡易血糖装置で電流測定し、この電流値と検量線を用いてグルコース濃度を表示させる様に成した血液等の定量分析方法とその装置が特許文献1に開示されている。

図10は上述の特許文献1に示された簡易血糖装置の系統図を示すもので、図11は動作説明用のフローチャートである。

図10に於いて、1は簡易血糖装置を示すものであり、2は較正センサ11等の電極パット13等が挿入可能なコネクタであり、該コネクタ2は電流/電圧変換器3及び電極パット13等の電極挿入検知スイッチ4に接続されている。

電流/電圧変換器3の出力はアナログ/デジタル変換器6を介してデジタル化されたデータをコンピュータ等の情報処理装置7(以下CPUと記す)に供給すると共に電極挿入検知スイッチ4の出力もCPU7に供給されている。

5は反応電圧設定回路であり、CPU7に送られた電極挿入スイッチ4の電極挿入・装着データに基づいて電流/電圧変換器3を介してコネクタ2に電圧を印加し、センサの電極パット13に電圧を供給する。

8は液晶表示装置(LCD)等の表示器、9はCPU7に広く利用される電気的消去書込み可能読み出し専用メモリ(EEPROM)等の不揮発性メモリ、10はRAM等の揮発性メモリである。

上述の簡易血糖装置1による血糖測定時には少なくとも較正センサ11及び測定センサ12を必要とし、これらセンサを簡易血糖装置1に装着させる。これらセンサには夫々固有の抵抗値を持たせ、装着時に簡易血糖装置1が夫々のセンサの抵抗値を検出することで動作状態を設定し、抵抗値に基づき電極が装着されていることを検知して、装着されたセンサに応じた機能を実行する様に成されている。

上述の簡易血糖装置1の血糖測定方法を以下図10と共に図11で説明する。

図11に於いて、第1ステップS1 では較正センサ11の電極パット13を簡易血糖装置1のコネクタ2に装着する。

次の第2ステップS2 では簡易血糖装置(以下装置と記す)1内の内部処理に入り、電極挿入検知スイッチ4により、各センサの抵抗値の違いから電極パット13の装着を検知する。この検知信号はCPU7に送信される。CPU7はいづれのセンサが装着されたかをセンサに付加した夫々の抵抗値から認識する。

第3ステップS3 に於いて、CPU7は反応電圧設定回路5に信号を送り、反応電圧設定回路5はコネクタ2に嵌着された較正センサ11の電極パット13に所定電圧を供給する。

第4ステップS4 では較正センサ13の夫々の抵抗値に基づいた電流が発生する。この電流を電流/電圧変換器3に供給し、電圧に変換し、A/D変換器6からCPU7にデジタルデータが供給される。

然し、上記した様な簡易血糖装置の如き電気化学センサ測定装置では、測定センサ12の特性にバラツキがあり、これを補正するためには測定センサ12の他に較正センサ11を使用するためユーザは測定センサでの血糖値測定以外に較正センサ11による較正を行なう煩わしさを生ずる。

本発明は上述の課題を解消するために成されたもので測定センサのみで較正及び測定、分析可能で、較正値は電気化学センサに電圧印加した際の電流値変化を測定することで簡単に測定可能な電気化学センサ測定装置及びその測定方法を提供する。

【課題を解決するための手段】
第1の本発明の電気化学センサ測定装置は、少なくとも2個の電極23及び24を有する電気化学センサ素子21と、この電気化学センサ素子21を保存水溶液中に浸漬し、2個の電極22と24間に一定電圧を供給する電位制御手段26と、電気化学センサ素子21への電圧供給時に発生する過度応答電流を検出する電流検出手段25と、この電流検出手段25の過度応答出力の経過時間を計数し、経過時間と較正値の関係を参照して、電気化学センサ素子の較正値を決定する様に成したものである。

【発明の実施の形態】
以下、本発明の1形態例を図1乃至図6によって詳記する。

図1の電気化学センサ測定装置20を説明する前に図4乃至図6によって本発明に用いる電気化学センサについて説明する。

一般に電気化学センサを用いて溶液中の特定の物質を測定することは化学工業、臨床検査等で利用されている。例えば、臨床検査等での検体中の基質測定として尿中のグルコースの濃度測定等は広く行なわれている。

この様な基質の測定に利用する電気化学センサは一般には図4(A)(B)に示す様に構成されている。図4(A)は電気化学センサの平面図、図4(B)は図4(A)のA−A′断面矢視図である。図4(A)(B)で絶縁性基板36上には作用極22、対極23、参照極24の3電極が構成されている。絶縁性基板36はシリコン上に酸化シリコン膜を形成した基板、ガラス、石英、ポリイミド、ポリカーボネート、ガラスエポキシ基板等が用いられ、例えば、その大きさは6×9mmの長方形状に形成される。

絶縁性基板36上に形成する作用極22及び対極23はチタン層38及び白金層41で構成する。これらチタン層38及び白金層41は例えば、スパッタ法で膜厚0.02〜0.2μm及び0.1〜1.0μmに形成し、その大きさを例えば夫々作用極22で1.2mm×2.0mm、対極22で0.4mm×2.0mmの長方形状に選択形成されている。

上述の作用極22及び対極23の電極材料は白金層41の他にカーボン、金、イリジウム等のいづれか1つを選択することが出来る。又、電極の形成法はスパッタ法に限らず蒸着法、スクリーン印刷、鍍金等で形成可能である。

次に、参照極24は絶縁性基板36上に対極23と対向した位置に銀層37を形成し、次に銀層37の表面を処理して塩化銀層39を形成する。塩化銀層39は塩化鉄(III)若しくは塩化クロム(III)水溶液で処理するか、塩化物水溶液中で電解する方法で製作される。参照極24の下地として銀層37の下に絶縁性基板36との密着性を高めるために、例えばスパッタ法等でチタン層を0.1〜1.0μm厚に形成する様に成してもよい。参照極24の大きさは例えば0.4mm×2.0mmの長方形状と成されている。

作用極22、対極23、参照極24に連通するリード(0.2mm×2.8mm)34及びコンタクト電極(1.6mm×0.8mm)35c,35b,35aも絶縁性基板36上に形成されている。この構成の3電極式では作用極22と参照極24間の電位差を一定に保ち、作用極22と対極23間の電流を測定する様に成される。

上述の構成では、3電極式の構成を説明したが、作用極22と参照極24とから構成される2電極式もあり、この場合は作用極22と参照極24の電位を一定に保ち、その時の作用極22と参照極24に流れる電流を測定する様に成されている。

次に絶縁性基板36上に形成した作用極22、対極23、参照極24上に選択透過膜40が形成される。この選択透過膜は、過酸化水素を選択的に透過し、妨害物質を除去する機能を有し、例えばアセチルセルローズ膜或はイオン交換樹脂等から形成される。

選択透過膜40上には膜中に酵素を担持する機能を有する酵素膜42が形成される。この酵素膜42は、例えば酵素とアルブミンの様な蛋白質をグルタルアルデヒドの様な架橋剤で固定化することで形成する。

上述の選択透過膜40、酵素膜42、制限透過膜43からなる有機薄膜の各層の厚みは0.1〜数μm程度に選択されている。

制限透過膜43は、汚染物質の吸着を防ぐと共に、グルコース等の透過を制限し測定範囲を拡大する機能がある。選択透過膜40は例えば過酸化水素のみ透過し、妨害物質の透過を防ぐ機能がある。
制限透過膜43及び選択透過膜40には微小な孔が形成されており、この孔の大きさによって、物質の透過を制限している。

上述の電気化学センサ21をスパッタ法を用いて製造するプロセスの概要は以下に記す通りとなる。
(1) 絶縁性基板(60×70mm)36上にチタン層38及び白金層41をスパッタ法により成膜し、さらに銀層37をスパッタ法により成膜する。
(2) フォトリソグラフィにより作用極22、対極23、参照極24、リード34、コンタクト電極35a,35b,35cを形成する。
(3) 塩化鉄(III)水溶液で処理して銀層表面に塩化銀層39を形成し、参照極24を形成する。
(4) 選択透過膜40、酵素膜42、制限透過膜43を形成する。
(5) 絶縁性基板36を個々の電気化学センサ21(6×9mm)に分割する。

作用極22と参照極24間に一定電圧を印加すると保存液に依存する電流が作用極22と対極23間に流れて、定常状態になると殆ど流れなくなる。この現象については電気化学及び工業物理化学Vol68,No1.p63〜68に詳しく記載されている。作用極22と参照極24の間に電圧を印加した場合、今までこれら電極と静電気的につりあって分布していた電極近傍の陽イオンや陰イオンが、電極電位の急激な変化にともなって、新しい平衡状態を達成するため移動を開始する。作用極22の電位がこれまでよりも正になるように電位を印加すると、陽イオンは作用極22から遠ざかり、逆に陰イオンは近づき、平衡分布に落ち着いたところで新しい電気二重層が形成される。電荷を持った粒子が動くので、この動きは外部回路を流れる電流として観測される。この現象は、電気二重層という一種のコンデンサに溶液という抵抗体を通して電流を流し、電荷を充電(あるいは放電)する過程と似ている。

この様な電流値を第4ステップST4 では電流検出回路25で検出する様にする。図1では電流検出回路25を作用極22側に接続させたが、一定電圧印加後に流れる電流は作用極22と対極23間に流れるので電流検出回路25は対極23側に接続させる様にしてもよい。

電流検出回路25で検出された電流はROM32に保存したファームウェアに従って、CPU30へA/D変換器29を介して取り込み、第5ステップST5 の様にRAM33に格納する。

このRAM33に格納された過渡応答時間と較正値との関係を図2に示す。この図から解る様に電気化学センサ21は過渡応答時間が1分未満の場合は、曲線56の様に配線部53の断線、電極22〜24の不活性あるいは制限透過膜43の欠陥などの原因で不良である。一方、過渡応答時間が10分より大きい曲線57の場合は、制限透過膜43及び選択透過膜40が緻密であり不良と成り、過渡応答時間が2分、4分、5分の曲線58,59,60では較正値が40nA、20nA、10nAと成されている。作用極22、対極23及び参照極24から構成される測定電極上に機能性有機薄膜を形成した場合、イオンの流れは有機薄膜中の拡散によって制限される。したがって、この電流が流れる時間(過渡応答時間)は、有機薄膜の膜質に依存する。

第8ステップST8 では電流値が定常状態に達するまでの過渡応答時間を決定して、第9ステップST9 では、この決定した過渡応答時間をRAM33に格納する。

(1) 作用極22等の測定電極と酵素膜42等の機能性有機薄膜から成る電気化学センサ21は機能性有機薄膜の膜質のバラツキが大きいため、個々の電気化学センサ21での出力のバラツキが大きくなるが基準液となる保存液に浸漬することで例えばグルコース濃度と出力値との関係を知るための較正値を、CPUが電気化学センサの装着を認識し、電圧印加後に発生する電流が定常状態になるまでの過渡応答時間をカウントすることで機能性有機薄膜の膜質(厚みや透孔状態)に依存するファクタをカウント時間から簡単に決定することが可能と成って電気化学センサの較正値を自動的に決定することが出来る。
(2) (1)と同様に電気化学センサの良否判定も簡単に行なうことが可能と成る。
