縮合ピリミジンライブラリーの組合せ的製造法
本願は、1996年12月18日に出願された米国仮特許出願第60/034
,295号の利益を主張するものである。
発明の分野
本発明は、縮合ピリミジン化合物のライブラリー、そのようなライブラリーの
作製方法、および多様な縮合ピリミジン化合物の容易に利用できる供給源を保存
および提供するための装置に関する。本組合せ的ライブラリーを格納する装置は
、薬物開発のために化合物を同定するための分析系の有用な構成要素である。
発明の背景
薬品工業において研究開発費は大きな資本支出の原因である。化合物の合成は
、研究開発における費用のかかる時間をくう段階である。歴史的には、研究に従
事する化学者は、生物学的スクリーニングのための高純度の化合物を個々に合成
および分析して医薬品の先導物を開発してきた。そのような方法は、市場に新し
い薬物を提供することに成功はしたけれども、個々の合成と完全な化合物の特徴
付けという制約が、新規の薬学的に活性な化合物の発見を少なからず遅らせた。
より迅速で費用のかからない薬物発見の方法論に対する必要性が、今日の薬品工
業の競争においてますます重要になってきている。
現在の薬物発見は、組合せ的化学反応を用いて、一般に「ライブラリー」と呼
ばれる大量の(102〜106)の化合物を作製する。組合せ的化学反応の重要な
目的は、スクリーニングして医薬品探索のリード化合物を生じ得る大量の新規化
合物を作製することである。
理論的には、所定のライブラリーについて生成し得る化合物の総数は、そのラ
イブラリーの分子骨格における可変の位置で置換基を形成するのに利用できる試
薬の数によってのみ決まる。本組合せ的方法は、化合物の生成およびそれらの生
物学的スクリーニングの両方において自動化に適しており、それによって薬物発
見の機会と効率が大きく増大する。
組合せ的化学は、化合物のライブラリーを混合物として生成させ、ポジティブ
なスクリーニング結果が得られた後まで個々の化合物の完全な同定をあとに延ば
すような形式で行うことができる。しかし、組合せ化学反応の好ましい形態は、
「並列合成」であり、これは個々の反応生成物が、同時に合成されるが分離され
た容器に入れられているものである。好ましい態様では、単一の反応生成物が各
々の分離された容器内で調製される。例えば、個々のライブラリー化合物を、マ
イクロタイタープレートの分離されたウェル内で、調製し、保存し、そして分析
することができ、各ウェルは並列の1つの構成員を含んでいる。標準化されたマ
イクロタイタープレートまたはそれと等価な装置の使用は、ライブラリー合成の
間およびライブラリーのサンプリング/分析の間の両方で、プログラムされたロ
ボット機械によりそのような装置に容易にアクセスできるので有利である。
組合せ的化学反応は、固体支持体、通常はポリマー上で典型的に行う。ライブ
ラリー骨格は、化学的に開裂可能な2価の連結基を介して、固体支持体に遊離可
能なように束縛する。反応を行って、骨格を固体支持体に束縛されている間に修
飾する。最終の工程で、生成物を開裂して固体支持体から遊離させる。しかし、
組合せ的ライブラリーは溶液相中でも調製することができる。溶液相での反応の
完結を確実にするために、典型的に1つの試薬をかなり過剰に加える。このよう
に、反応の完結によって、少なくとも1つの可溶な未反応の反応物とともに可溶
な生成物が生じる。したがって、最終の工程として、過剰の試薬と反応する機能
的な基を有するポリマースカベンジャーを使用して過剰の試薬を除く。
組合せ的化学反応は、薬物開発の2つの別個な段階で使用することができる。
発見段階では、多様なライブラリーを作製して、リード化合物を見つける。第2
の至適化段階では、強力なリード化合物をより厳密に修飾して、最適な分子の立
体配置を見つける。
本発明の方法は、新規のリード化合物の同定に有用な縮合ピリミジンの新規の
多様なライブラリーの調製に基づく。ライブラリーを作製し、保存し、二次元的
配列の貯留器(reservoir)を含む装置として使用する。各貯留器は、横に隣り
合った貯留器内の化合物とは1つの置換基のみが異なっている、そのライブラリ
ーの1つのあらかじめ決定された化合物を含んでいる。発明の要約
本発明は、一般式(IまたはII):

の縮合ピリミジン骨格を有する構造的に関連する化合物の組合せの配列を提供す
る。
本発明はさらに、一般に以下に記載する反応式1または反応式2にしたがう、
縮合ピリミジンライブラリーを作成する方法を提供する。
本発明の1っの態様は、薬学的リード縮合ピリミジン化合物の同定のための分
析キットを提供し、該キットは分析物質および二次元的配列の画定された貯留器
を提供するウェルプレート装置またはそれと等価な装置を含む。ウェルプレート
装置は多様な組合せ的ライブラリーを提供し、各ウェル(貯留器)は縮合ピリミ
ジンライブラリーの唯一の構成員を含む。ウェルプレート装置を使用して、ライ
ブラリーを製造するための複数の反応領域を提供し、ライブラリーの構成員であ
る化合物の容易に利用可能な供給源を保存および提供する。
発明の詳細な記載
本発明にしたがって使用される「分析キット」なる語は、2つの連携した要素
、即ち(1)ウェルプレート装置および(2)生物学的分析物質の集合体を意味
す
る。
「生物学的分析物質」は、選択された疾患の状態に関係するスクリーンにおけ
るいずれかのライブラリー化合物の効果の生物学的評価を行うに必要な物質であ
る。
「目的ライブラリー(directed library)」は、リード化合物の活性の至適化
を目的として組合せ的化学反応過程によって作製された化合物の集まりであり、
各ライブラリー化合物は共通の縮合ピリミジン骨格を有し、ライブラリーが、全
体として考えてリード化合物に対する密接に関連したホモローグまたはアナログ
の集まりである(「多様性ライブラリー」との比較で)。
「多様性ライブラリー(diverse library)」は、縮合ピリミジン組合せ的ラ
イブラリー骨格上の置換基が、構成原子、分子量、および構造において変化に富
み、ライブラリーが、全体として考えて密接に関連したホモローグまたはアナロ
グの集まりではない(「目的ライブラリー」との比較で)ことを意味する。
「リード化合物」は、分析キットによってその化合物が選択された疾患の状態
に関して有意に活性であることが分かった、選択された組合せライブラリー内の
ある化合物を意味する。
「ライブラリー」は、組合せ的化学反応の過程により作製された化合物の集ま
りであり、その化合物は1またはそれ以上の可変の置換基を有する共通の骨格を
有している。本発明の骨格は、縮合ピリミジン環系である。
「ライブラリー化合物」は、組合せ的ライブラリーの個々の反応生成物(通常
単一の化合物)である。
「並列合成」は、ライブラリーの組合せ的化学合成を行う方法を意味し、その
方法では、個々の組合せ的ライブラリー反応生成物を別々に製造して保存し、前
もっておよび後で意図的に混合しない。
「反応領域」は、本発明の組合せ的化学反応ライブラリー化合物製造過程が行
われ、そしてそこで個々のライブラリー化合物が合成される、個々の容器の位置
を意味する。適当な反応領域は、ウェルプレート装置の個々のウェルである。
本発明にしたがって使用される「骨格」なる用語は、組合せ的ライブラリーの
構成員である化合物の不変領域(本発明の縮合ピリミジンコア)を意味する。
「同時合成」は、組合せ的方法の1つの製造サイクル内での化合物のライブラ
リーの作製を意味する。(同じ瞬間にすべてのライブラリー化合物を作製するの
ではない)。
「置換基」は、組合せ的合成過程によって骨格に結合したかまたは導入された
化学的残基である。異なる機能性基がライブラリー全体を通して分子の多様性の
原因であり、多様性ライブラリーの場合には生物学的活性の多様性を骨格に与え
るよう、そして目的ライブラリーの場合には特定の生物学的活性の至適化を与え
るよう選択する。
「試薬」は、反応物を意味し、組合せ的合成に使用して置換基をライブラリー
の骨格に導入するためのいずれかの化合物である。
「ウェルプレート装置」は、決まった寸法の、画定された位置の多数のライブ
ラリー化合物を保持することができる構造物を意味する。
「非妨害性置換基」は、本発明の反応過程を立体的に妨害せず、安定な縮合ピ
リミジンライブラリー化合物を生じる置換基である。適当な非妨害性置換基には
以下のものが含まれるがこれらに限られない:ハロ、C1−C10アルキル、C2−
C10アルケニル、C2−C10アルキニル、C1−C10アルコキシ、C7−C12アル
アルキル、C7−C12アルカリール、C3−C10シクロアルキル、C3−C10シク
ロアルケニル、フェニル、置換されたフェニル、トルオイル、キシレニル、ビフ
ェニル、C2−C12アルコキシアルキル、C1−C6アルキルスルフィニル、C1−
C10アルキルスルホニル、−(CH2)m−O−(C1−C10アルキル)、アリール
、置換されたアリール、置換されたアルコキシ、フルオロアルキル、アリールオ

、ここでmは1〜8。好ましい非妨害性残基は、C1−C10アルキル、C2−C10
アルケニル、C1−C10アルコキシ、C7−C12アルアルキル、C7−C12アルカ

(CH2)m−O−(C1−C10アルキル)、アリール、および置換されたアリール
である。
「アリール」は、それぞれ炭素原子5または6の1またはそれ以上の芳香族環
を意味する。複数のアリール環は、ナフチルのように縮合してよいし、またはビ
フェニルのように縮合していなくてもよい。
「置換されたアリール」は、1またはそれ以上の非妨害性置換基を置換基とし
て有するアリールである。
「アルキル」は、典型的には炭素原子1〜20の直鎖または分岐鎖または環状
の炭化水素を意味する。
「置換されたアルキル」は、1またはそれ以上の非妨害性置換基を有するアル
キルである。
「ハロ」は、クロロ、フルオロ、ヨードまたはブロモを意味する。
「ヘテロ環」または「ヘテロ環の」は、原子数5、6または7の、不飽和また
は芳香族の特徴を有するかまたは有さない、少なくとも1つの炭素でない環原子
を有する、1またはそれ以上の環を意味する。好ましいヘテロ原子には、硫黄、
酸素、および窒素が含まれる。ベンゾフランのキノリノンのように複数の環が縮
合してもよい。
「置換されたヘテロ環」は、非妨害性置換基から形成された1またはそれ以上
の側鎖を有するヘテロ環である。
縮合ピリミジンの多様性ライブラリーは、本発明にしたがって提供される。本
発明の装置として組み込まれた縮合ピリミジンライブラリーは、薬物/農薬候補
のスクリーニング分析による新規の生物学的に活性な化合物の同定において使用
するための、構造活性関連性試験を明らかにすることにおいて使用するための、
および/または臨床的な研究において使用するための、構造的に関連した広い多
様性を有する化合物の容易に利用できる供給源として役立つ。
本発明の製造方法の記載において用いる略語は以下の通りである:
DMFはジメチルホルムアミドである。
THFはテトラヒドロフランである。
式(I)または式(II)で示される縮合ピリミジン化合物を、以下の反応式
に示した組合せ的化学反応並列合成技術を用いて製造する:
反応式I:縮合ピリミジンライブラリーの製造のための一般式(ホルムアミジン
経由)反応式2:縮合ピリミジンライブラリーの製造のための一般式(ホルムアミダー
ト経由) この方法は、示したシアノアミン化合物から芳香族または脂肪族のホルムアミ
ジンまたはホルムアミダート中間体を製造する工程と、各中間体を一連の第一級
アミン(RNH2)と反応させて縮合ピリミジン化合物のライブラリーを製造す
る工程を含む。ジメチルホルムアミドおよびトリメチルオルトホルメートは、そ
れぞれの中間体を形成するための好ましい反応物であるが、他のホルムアミジン
およびホルムアミダートを形成する反応物、例えばジ(C1−C4アルキル)ホル
ムアミドアセタールおよびトリ(C1−C4アルキル)オルトホルメートを使用す
ることができる。各ホルムアミジン/ホルムアミダート中間体を製造し、分離反
応チャンバー内でさらに第一級アミンと反応させ(即ち、並列合成)、反応生成
物を反応ウェル番号によって同定する。
式:で示される出発物質であるシアノアミン化合物は、商業的に入手可能であるかま
たは商業的に入手可能な出発物質から製造する。上記式において、縮合環Aは芳
香族環または非芳香族環であり、いずれもヘテロ環であってよい。上記のおよび
上記の式(I)および(II)を含む本発明を定義する他の式において、環構造
を形成する2価の基Aの例には、以下の基が含まれるがこれに限られない:

本縮合ピリミジンライブラリーの製造において本発明にしたがって使用される
第一級アミンは、式RNH2(式中、Rはアルキルまたはピリミジン形成反応を
妨害しない置換基を有する置換されたアルキル基である)で示される化合物であ
る。好ましくは、第一級アミン反応物は、分子量約30〜約600である。本発
明の定義内のR基の例には、以下の基が含まれるが以下の基に限定することを意
図しない: [式中、Lは、その基が結合する窒素原子に結合する点である]。このように、本
発明の製造方法に有用な第一級アミンの例は、Lが−NH2である式で示される
化合物である。

縮合ピリミジンライブラリーを製造するために本発明の製造方法において使用す
るのに適当な、さらなる第1級アミンは以下の式で示されるものである:
式中、CBzはベンジルオキシカルボニルである。
式II示される縮合ピリミジンライブラリー化合物の製造は、反応式1にした
がって、ホルムアミジン中間体が最初に形成し、次に第1級アミンと反応させて
広範な縮合ピリミジン化合物の配列を製造する、2つの工程を含む。別法として
、それら化合物をホルムアミダート中間体と式Iのライブラリー化合物の熱転位
を経由する反応式2にしたがって製造することができる。本発明の製造方法の1
つの態様では、式Iで示される縮合ピリミジンのライブラリーを製造し、各生成
化合物の一部を式IIで示される対応する化合物に変換して別のライブラリーを
製造する。第1級アミンとホルムアミジン/ホルムアミダート中間体との間の反
応の完結と、式Iで示される化合物の式IIで示される化合物への変換は、薄層
クロマトグラフィー(TLC)などを含む数多くの慣用の技術によって測定する
ことができる。
本発明の製造方法は、液体の反応媒体が入っており、入口および出口手段を有
するいずれかの容器中で行うことができる。1つの態様では、本発明の製造方法
を並列合成に適合した容器中で行う。
本発明の製造方法は複数の反応領域、最も典型的には二次元的配列の画定され
た貯留器を提供する装置において行い、本発明の縮合ピリミジンライブラリーの
1つの構成員がそれぞれの貯留器において製造される。このように本発明の多様

5−クロロ−2−(ジメチルアミノ)メチレンアミノベンゾニトリル
無水テトラヒドロフラン(10mL)中の2−アミノ−5−クロロベンゾニトリ
ル(3.82g,25mmol)およびジメチルホルムアミドジメチルアセタール(
4.7mL)を2時間還流した後、減圧濃縮した。残留物を塩化メチレンとヘキサ

4:1のテトラヒドロフラン/DMF(50mL)中の4−アミノ−3−シアノ
−1,2,5,6−テトラヒドロピリジン(4.31g,35mmol)およびトリエチ
ルアミン(7mL,50mmol)の氷冷(5℃)した懸濁液を4−フルオロベンゾイ
ルクロリド(6.66g,42mmol)をポット温度が20℃以下に保たれるよう

ドロピリジン(8.44g,98%)を灰白色の固体として得た。無水テトラヒ
ドロフラン(20mL)中のこの固体(7.36g,30mmol)の懸濁液をDMF
ジメチルアセタール(7.0mL,49mmol)で処理した後、還流温度に4時間加
熱した(固体が溶解した)。混合物を減圧濃縮し、酢酸エチル/ヘキサンから再

実施例1

96ウェルの1mLディープのウェルガラスウェルタイタープレートに、各ホル
ムアミジン溶液50μMolを、すべての液体がはね上がりが最小限なるようにウ
ェルの底に加わるよう気を付けながらウェルに行ごとに入れた。溶媒を除いた後
、アセトニトリル(200μL)を各ウェルに加え、プレートを96ウェルヒー
トブロック(USA Scientific)中で、栓をせずに60℃にて適度に振盪しながら
約15分間静かに加熱し、大部分の物質を溶解させた。適当なアミン溶液(各ウ
ェル50μL)をウェルに列ごとに加え、次いでさらにアセトニトリル(各ウェ
ル300μL)を加え、プレートをプレートキャップ全体の下部のストリップキ
