D−ペプチドの発生:方法および組成物
序論
背景
薬理学的活性を有する小さい分子、例えば、種々のアクセプター分子のアゴニ
ストまたはアンタゴニスト、例えば、細胞表面のレセプター、酵素または抗体を
生産しかつ同定することが必要であることの認識が増大しつつある。薬剤として
有用である小さい分子についての探索は、このような分子を収集物を発生させ、
生理的活性をもつ分子について収集物をスクリーニングし、そしてスクリーニン
グ工程において陽性の結果を提供する分子の構造を同定することを必要とする。
組合せライブラリーのアプローチを使用して、小さい分子の収集物を発生させる
ことができる。しかしながら、先行技術はD−抗体およびD−ペプチドを使用し
て潜在的薬理学的活性について小さい分子のこのような組合せライブラリーをス
クリーニングする意義、および鏡像の転換を使用して有用なD−ペプチドおよび
D−抗体をつくる能力を認識しなかった。
必須の20アミノ酸のうちの19は、「キラリティー」、すなわち、ハンデッドネ
スを有する。唯一のアキラルの必須アミノ酸はグリシンである。キラル化合物を
記載するために、接頭語DおよびLを使用してそのキラル中心の回りの分子の立
体配置を言及する。アミノ酸のキラル中心はアルファ炭素であり、そしてアミノ
酸がD立体配置またはL立体配置を有するかどうかはエミル・フィッシャーが確
立した立体異性体のコンペンションに依存する。キラルアミノ酸は立体異性体と
して存在することができ、これらは互いに鏡像である

天然に存在するキラルアミノ酸のすべてはL立体配置で存在し、一般にL−ア
ミノ酸と呼ばれる。L−立体配置の各キラルアミノ酸の立体異性体はD−アミノ
酸と呼ばれる。D−アミノ酸はグリセルアルデヒドの2つの立体異性体、L−グ
リセルアルデヒドおよびD−グリセルアルデヒドのうちのD−立体異性体に相当
する立体配置を有するものである。D−グリセルアルデヒドと同一の立体化学的
立体配置を有するすべてのD−アミノ酸、D−ポリペプチドまたはD−ペプチド

を有するものはL−と表示される。
ポリペプチドのリボソームの翻訳における酵素反応はL−アミノ酸について立
体特異的であるので、すべてのD−アミノ酸から成るペプチドは一般に天然に存
在しない。したがって、特定の酵素的手段によるか、またはリボソーム上の生合
成によるよりむしろ他の翻訳後の修飾により導入された制限された数のD−アミ
ノ酸を含有する、ある種の抗体および細菌の細胞壁のタンパク質を除外して、す
べての天然に存在するタンパク質およびポリペプチドはL−アミノ酸から成る。
D−アミノ酸は、また、ラセマーゼによる翻訳後の修飾の結果として、そして
長いin vivo寿命をもつタンパク質の自発的ラセミ化の結果として、人間におい
て天然に存在する。HelfmanおよびBada,PNAS,72:2891-2894(1975)を参照のこ
と。ラセミ化は天然に存在するプロセスであり、天然に存在するL−アミノ酸を
L−アミノ酸およびD−アミノ酸の双方のラセミ混合物に経時的に変換する。
いくつかの承認されたペプチド様薬剤の構造物の制限された部分
はわずかのD−アミノ酸を含む。このような産物は広く知られている抗生物質の
バリノマイシン、グラミシジンA、グラミシジンS、およびまたバソプレシン類
似体のデソムプレシン(RPR)、ルプロン(Abbott)、シナレル(Syntex)、サン
ドスタチン(Sandoz)、SK&-110679(Smithkline Beecham)、およびデカペプチ
ル(Ipsen-Beuafor/Akzo)を包含する。このような構造物は小さく、そしてD−
アミノ酸のみから構成されていない。
現在、D−ペプチドは、この分野においてよく知られている技術を使用して、
化学的合成により作られる。例えば、D−ペプチドは、適当な保護基の使用を含
む固相合成法において、D−アミノ酸の段階的付加を使用して合成することがで
きる。L−ペプチドについて普通に使用されている固相ペプチド合成技術は、下
記の文献に記載されている:Meinhofer,Hormonal Proteins and Peptides,vol

D−ペプチドおよび混合L−およびD−アミノ酸を含有するペプチドはこの分

野において知られているリガンドの類似体は小さい有機分子、L−ペプチド、お
よび修飾されたL−ペプチドである。しかしながら、レセプターまたはリガンド
または基質に特異的に結合するD−抗体は文献に記載されてきていず、そしてこ
のようなD−抗体およびリガンドおよ
びレセプターの類似体であるD−ペプチドならびにそれらの同定および生産の方
法が要求されている。
発明の要約
したがって、本発明の目的は、ホルモンおよび神経ペプチドを包含するが、こ
れらに限定されない、生物学的に活性なペプチドおよびタンパク質の類似体に関
する方法および組成物を提供することであり、ここで類似体はD−アミノ酸から
もっぱらまたは本質的に構成されており、そして生物学的に機能的である。
本発明の1つの面は、天然のまたは人工的L−ペプチドまたはL−タンパク質
の標的、例えば、身体における生物学的レセプターと相互作用するD−アミノ酸
から完全に構成されたD−ペプチドおよびD−タンパク質を発生させることに関
する。
D−アミノ酸から構成された抗体様実在物(D−抗体、またはリガンドまたは
レセプターの類似体であるD−ペプチド)および操作された誘導体の形態を含ん
でなる新規な組成物、および必要な生物学的および製剤学的機能を達成するよう
な方法において、それらの新規な抗体様実在物を生産する方法が提供される。こ
の一般的方法は、D−ペプチド(すなわち、D−抗体またはそれらのフラグメン
トを包含する、D−アミノ酸から完全にまたは大部分構成されたポリペプチドま
たはタンパク質)の分子認識表面(例えば、ファン・デル・ワールスおよび静電
的表面)がL−アミノ酸から構成された天然の生物学的リガンドの分子認識表面

都合には消化管中のタンパク質分解に対して耐性であり、血清および組織のプロ
テアーゼに対して耐性であり、そして比較的免疫学的
に不活性である。本発明のD−抗体またはそれらの抗原結合ループを包含する、
それらのフラグメント、またはそれらの再設計された成分は、消化管中において
、また身体を通してタンパク質分解に対して耐性である。本発明のD−抗体およ
び他のD−ポリペプチドを含んでなる組成物が考えられる。特性がもはや抗体様
でない比較的小さい構造物が天然の生物学的リガンドの類似体として本質的に機
能するように、抗原結合ループの結合特異性を保存するL−抗体のD−ペプチド
類似体は可能である。この分野において知られており、かつ特定の抗原に対して
抗体を発生させる標準的モノクローナル抗体法の別法であるFAbフラグメントの
ファージ発生を包含する、免疫学的アプローチを洗練する別のスクリーニング法
が提供される。
通常のL−抗体の使用を越えた追加の利点は、抗体を著しく修飾することがで
きることであり、例えば、D−類似体中のFAbフラグメントのみの保持、または
基本的抗原の認識の保持を含み、そして結合ループはもとの抗原におけるのと同
一の向きにスカホールド(ここでは、D−アミノ酸の)上に配置される。設計は
抗体認識領域を直接修飾する必要がなく、生ずるD−抗体またはD−ポリペプチ
ドは形態が抗体とかなり異なる。しかしながら、D−ポリペプチドは通常タンパ
ク質分解に対して耐性であり、かつ対応するL−ポリペプチドよりも免疫原性が
非常に低い。
好ましい態様において、エピトープ、ドメインまたは全タンパク質のL−アミ
ノ酸配列が以前に実験的またはコンピューター手段により同定されている場合、
このようなエピトープ、ドメインまたは全タンパク質のL−アミノ酸配列に相当
するD−アミノ酸配列に対してモノクローナル抗体を発生させる。タンパク質リ
ガンド上の結合部位は、前述のエピトープ、ドメイン、またはタンパク質からな
る構造物として典型的な重要性をもつ1例であろう。エピトープ、ドメイン、ま
たはタンパク質に対して発生したモノクローナル抗体の配列を次いで決定し、そ
して全抗体または抗原結合部位またはその一部分を含む全抗体の一部分をモノク
ローナル抗体における配列または配列の一部分に相当するD−アミノ酸配列とし
て合成する。D−アミノ酸から完全に構成された生ずる抗体またはそのサブフラ
グメントは、前述のエピトープ、ドメイン、または全タンパク質のもとの天然の
生物学的L−形態と一般に相互作用する。ヒト化抗体と異なり、減少した大きさ
のペプチド鎖を含む、本発明のD−ポリペプチドの高度の修飾は、ヒト化を維持
するために典型的に要求される設計なしに、可能であり、そして生ずるD−ポリ
ペプチドは、ホルモンおよび神経ペプチドおよび天然の抗体のようなタンパク質
の薬理学的に機能的な類似体よりも、in vivoにおいてより長く生き残ることが
できる。
他の面において、本発明は前述の方法を含んでなり、この方法において、モノ
クローナル抗体に相当するD−ペプチドの合成は、モノクローナル抗体の結合部
位および前記結合部位のL−アミノ酸配列の決定、およびL−アミノ酸配列また
はモノクローナル抗体の結合部位の配列の合成を含む、すなわち、D−ペプチド
がモノクローナル抗体の結合部位のL−アミノ酸の代わりにD−アミノ酸を有す
る以外、D−ペプチドは結合部位のL−アミノ酸配列と同一のアミノ酸配列を有
する。
なお他の面において、本発明は、L−アミノ酸から成るL−抗体のL−アミノ
酸配列に相当するD−アミノ酸配列から構成されたポリペプチドまたはペプチド
を含んでなる合成されたD−抗体を提供する。本発明のD−抗体は、D−アミノ
酸またはアミノ酸類似体の対応する鏡像異性体から完全にまたは本質的に構成さ
れている。ま
た、本発明のD−抗体は1またはそれ以上のアキラルグリシンアミノ酸残基を含
有することができる。
本発明のD−抗体は、レセプター、酵素上の基質結合部位、レセプター抗体が
結合するとき、リガンドの結合を妨害するレセプターのエピトープ、レセプター
のリガンド結合部位、酵素上のコファクター結合部位およびタンパク質上の糖結
合部位を含むことができる。他の態様において、D−抗体は、レセプターのリガ
ンド、酵素結合部位の基質、レセプターのペプチドホルモン、レセプターの非ペ
プチドホルモン、レセプターの神経伝達物質、酵素上のコファクター結合部位の
コファクター、およびタンパク質上の糖結合部位の糖を含むことができる。
なお他の面として、スクリーニングおよび分子活性の方法、および生物学的操
作法または化学的合成法、例えば、質量または組合せのスクリーニング法を包含
するかどうかにかかわらず、この方法において認識表面を産生することに等しい
方法は、また、本発明の範囲内に入る。後述するように、モノクローナル抗体の
生産およびFAbフラグメントのファージ発生の既知の方法を使用して、D−ペプ
チドの抗原に応答して発生するL−抗体を生産することができる。
詳細な説明
一般に、リガンドまたはレセプターに結合するD−ペプチドを発生させる方法
は、リガンドまたはレセプターのD−バージョンをつくり、L−ペプチドから作
られたライブラリーを前記リガンドまたはレセプターのD−バージョンでプロー
ビングまたはスクリーニングし、「hits」、すなわち、前記リガンドまたはレセ
プターのD−バージョンに結合するL−ペプチドを検出し、次いでL−ペプチド
のD−バージョンを合成することを包含する。L−ペプチドのD−
バージョンは、リガンドまたはレセプターのL−バージョンに結合することがで
きる。このようなL−ペプチドのD−バージョンは、本明細書においてD−ペプ
チドまたはD−抗体としばしば呼ばれる。これらの工程は順次に実施するか、ま
たは次の工程に進行する前に反復する(例えば、スクリーニング工程)ことがで
きる。さらに、工程のあるもののみの完結は活性分子、例えば、治療薬の設計を

通常、第1工程はタンパク質の標的(または非ペプチドのリガンド)、例えば

とする酵素を選択することである。典型的には、このようなリガンドのD−ペプ
チドはアンタゴニストとして通常の機能を阻害するか、またはある場合において
アゴニストとして通常の機能の活性化を阻害するであろう。標的のタンパク質ま
たはタンパク質の成分(例えば、リガンド結合部位)は、それらをD−アミノ酸

通常、第2工程は、D−レセプターまたはD−ポリペプチドでプロービングす
ることによって、L−ペプチドのライブラリーを組合せスクリーニングを実施す
ることである。時には、標的が既知である場合、第1工程を使用しないことが望
ましいであろう。組合せスクリーニングにより、ペプチドの多様な組をつくるこ
とができ、次いでこれらのペプチドをD−ペプチドに変換することができる。ま
た、抗体の免疫学的生産または抗体の組換え生産を使用して、対応するD−ペプ
チドに変換することができるL−ペプチドの多様な組をつくることができる。合
成ペプチドのライブラリーを前もって組合せ的に製造し、次いでD−レセプター
に対してスクリーニングすることができる。合成ペプチドのライブラリーの1つ
の利点は、天然に存在しないアミノ酸、例えば、天然に存在するアミノ酸と同一
の電荷をもつが、電荷がペプチドの主鎖から位置する距離が異なるアミノ酸を使
用するとき、それらがより大きい化学的多様性を提供することである。距離の変
更は陰性または陽性の電荷の1,2または3原子のエクステンダーを使用して達
成することができ、好ましくは炭素原子を使用する。この分野において知られて
いるように、組換えペプチドのライブラリーを同様によく使用することができる
。
好ましい組合せライブラリーの1つのタイプは、ファージ表示D−ペプチドの
アプローチである。典型的には、まずこの分野において知られている方法を使用
して通常のL−ペプチドをもつバクテリ
オファージ表示を調製する。また、L−ペプチドの生成にプラスミドを用いる他
の組換えライブラリーを使用することができる。ここで、タンパク質を発現する
DNAのヌクレオチドセグメントのランダム突然変異または部分的ランダム突然変
異または組合せ選択により、多数の異なるペプチドまたはタンパク質の組合せ提
示を間接的に実行する。好ましくは、組換えライブラリー、例えば、ファージ表
示ライブラリーにおいて、DNAの短い区画は、L−ペプチドの一部分またはすべ
てとなる突然変異した領域をコードする。好ましくは、突然変異した領域はラン
ダムに突然変異せず、そして3〜10または5〜8アミノ酸の突然変異した領域に
ついてすべての可能なアミノ酸配列を含有しない。その代わり、突然変異は選択
的であり、通常、前もって決定したアミノ酸の位置において、保存的突然変異、
例えば、異なる極性のアミノ酸について交換する極性アミノ酸、異なる陰性に帯
電したアミノ酸について陰性に帯電したアミノ酸および異なる疎水性アミノ酸に
ついて疎水性アミノ酸を導入する。このような突然変異した領域は合成ペプチド
のライブラリーとともに使用され、好ましくは、D−抗体のファージ表示ととも
に使用される。
バクテリオファージのライブラリーは安価であり、操作が容易である。バクテ
リオファージ表示または「ファージ表示」は広く使用される。N−末端において
ランダムアミノ酸を表示するコートタンパク質のキメラの特質を記載するために
、それは本来「融合ファージ」技術として開発された(S.F.ParmleyおよびG.P.Sm
ith,Gene,73:305-318,1988、引用することによって本明細書の一部とされる

目的のための技術として開発された、1990年において多数の研究所において出現
し、そして典型的にはfdまたはM13フ
ィラメント状バクテリオファージの遺伝子IIIの中にDNAの合成片をクローニング
することによって実施された(J.K.ScottおよびG.P.Smith,Science 249:386-39
0,1990;J.L.Devlin,L.C.Panganiban,P.E.Devlin,Science 249:404-406,1
990;S.E.Cwiria,E.A.Peters,R.W.Barrett,W.J.Dower,Proc.Natl.Acad.Sci

セプターに結合するペプチドの配列を発生した遺伝子を同定する。
組合せスクリーニングの他の利点は、スクリーニングのそれ以上のサイクルに
よりスクリーニングをますます洗練させることである。好ましくは、スクリーニ
ングの追加のサイクルは、より強い結合、すなわち、より低い見掛けのKdについ
て選択するためにより高いストリンジェンシイの条件を提供する。より高いスト
リンジェンシイの条件は、温度の増加、イオン強度の低下または増加、カオトロ
フィック剤およびその他の増加、またはそれらの組み合わせにより達成すること
ができる。例えば、アフィニティー精製されたファージの集団で細胞を感染させ
、これにより、アフィニティーによる次の選択において、もとの選択の100万ま
でのコピーを使用し、バクテリオファージを回収し、増幅できるようにすること
ができる。このような典型的な場合において、この方法はほぼ1mlの体積の管に
おいて少なくとも108の異なるペプチドに対するアクセスを可能とする。
通常、第3工程において、L−ペプチドを検出し、次いで検出または同定の手
段を使用してL−ペプチドが表示する配列に関して配列決定または同定する。検
出または同定の手段の多数は組合せライブラリーについてこの分野において知ら
れている。組合せペプチドのライブラリーの使用において、それぞれ、結合する
ペプチドまたは濃縮されたペプチドを、例えば、エドマン分解法によりアミノ酸
を配列決定するか、または他の手段により同定することができる。例えば、組合
せがコントロールされた方法において発生したグリッド上の位置によるか、また
は放射性標識化標的を包含する化学的標識化標識、ユニーク結合核酸標識、質量
分析により測定された最終の質量(PCT/US95/03355明細書を参照のこと、これは

サブミクスチュアーの反復再合成およびスクリーニング(Zuckermann et al.,J
.of Medicinal Chem.,37(17):2678-2685(1994)を参照のこと)(引用すること
によって本明細書の一部とされる)によるか、またはこれらの任意の組合わせま
たは他の既知の技術により、「hits」を同定することができる。バクテリオファ
ージ表示の使用において、通常、問題のペプチドをコードするDNA配列の増幅お
よび検査により、配列を推定する。
通常、第4工程において、L−ペプチドまたはL−タンパク質の配列を記録し
、次いでL−アミノ酸よりむしろD−アミノ酸を使用して、同一のアミノ酸配列
を使用してD−ペプチドまたはD−タンパク質を作る。これらのD−ペプチドま

本発明の重要な面は、ファージの中にクローニングして、標的に対する結合部
位(認識ループ)を精製するばかりでなく、かつまたフレーム、特に抗原結合部
位を有するFabフラグメントを精製することによって、L−抗体またはその部分
、例えば、Fabフラグメントを表示することである。特に、独占的ではないが、
ファージ上に表示されたペプチドおよびタンパク質のすべては、もとのD−抗体
に対して発生したモノクローナル抗体の抗原結合ループを少なくとも含有し、そ
してこれはもとのL−型の抗原、例えば、身体における天然のレセプターと相互
作用する認識を有する。前述したように、本発明は、これらのループが精製の目
的でファージ表示により修飾できることを包含し、また、抗原結合部位の配列の
精製がファージにおいて発生するように、ある程度の設計を実施できることを包
含する。ファージ表示により考えられる修飾は、フレームの広範な修飾、認識ル
ープを担持するための非抗体フレームによる置換(他のタンパク質の折りたたみ
)、および可能ならばシスチンの交差結合の挿入を含むフレームの除去を包含す
る大きさの高度の減少を包含する。ファージ表示による抗体成分の修飾および選
択は、D−抗体を除外して、この分野において知られており、そして「半合成抗
体のライブラリー」の使用として知られている(C.Barbas et al.,Proc.Natl.A
cad.Sci.USA,89:4457,1992)。これらの修飾および最終産物について有益な
効果をもってこれらの修飾を実施する能力は、D−ペプチドまたはD−タンパク
質に必要な認識表面を割り当てる、本明細書において記載する本発明に関して、
特別な、前例
のない意義を有することに注意することが重要である。前述したように、D−抗
体またはそれらのフラグメントはタンパク質分解に対して結果であり、免疫原性
が低い、ヒト化の有効な形態であろう。したがって、抗体は、著しく修飾された
ときでさえ、D−型においてヒト化を保持する。これはファージ表示により発生
し、D−アミノ酸から作られた任意の形態を包含する。この天然のヒト化は、半
合成抗体のライブラリーのファージ表示の通常の適用における著しい修飾と異な
る。それゆえ、本発明によれば、活性が保持されるように選択して、含まれかつ
スクリーニングされた突然変異で、Fabフレームの大きさを漸進的に減少させる
ことができる。単一のジャンプにおける大きさの減少はフレームとの重要な相互
作用をかなり喪失させ、結合ループを変形させることがある。FabフラグメントM
cPC603からの抗原認識ループH2のコンピューターシミュレーション研究(V.P.C
ollura,P.J.GreanyおよびB.Robson,Protein Engineering,7:2211-223,199
2)において、抗原の認識ループとFabフラグメントの残部との間に重要な相互作
用が存在することが認められた。また、抗原認識ループはそれら自体は、重鎖上
において3つでありかつ軽鎖上において3つであり、結合のために提示させかつ
精製することができ、支持する分子のスカホールドを使用しないでD−ペプチド
として、個々に合成するか、または、例えば、オリゴグリシンのスペーサーを使

1つの態様において、D−アミノ酸から完全にまたは本質的に構成された抗体
は、L−アミノ酸から作られた対応するL−抗体がD−アミノ酸から作られたペ

D−抗体の結合を望むキラルまたはアキラルの、天然のまたは天然に存在しな

子についての情報ならびに、存在する場合、このような分子のアミノ酸配列に関
する情報を使用して達成される。L−ポリペプチドまたは天然に存在するタンパ
ク質のリガンド結合部位の選択は、既知のリガンド結合部位、レセプターまたは
リガンドのアミノ酸配列に関して文献を参照することによって達成される。この
ような分子がまだ配列決定されていないペプチドであ
る場合、当業者はよく知られているペプチド配列決定法を使用してペプチドを配
列決定することができる。
ペプチドのリガンド、レセプターまたは他の分子のL−アミノ酸配列に相当す
るD−ポリペプチドの抗原の合成は化学的合成を使用して達成される。好ましく
はL−ペプチドの合成について開発された既知の固相の段階的メリフィールド型
ペプチド合成技術を使用するが、この場合において段階的合成においてD−アミ
ノ酸または保護されたD−アミノ酸を使用して、D−ポリペプチドの抗原を合成
する。D−ポリペプチドの抗原を合成する他の方法、例えば、段階的合成により
製造されたモノマーの共有結合が考えられる。本発明のD−ポリペプチドの抗原
および他のD−ポリペプチドまたはD−抗体を合成する手段はこの分野において
知られており、そして本発明はなお開発すべきD−ポリペプチドの合成法を使用

一般に、D−ペプチドの固相合成は下記の工程からなる:D−アミノ酸の未保

抗体のファージ表示を使用する場合、抗体または表示された抗体の部分を、そ
れらの結合認識ループまたは支持する抗体の折りたたみにおいてファージ表示に
より精製することができる;このような抗原は「半合成」と呼ばれる(C.F.Barb
as et al.,Proc.Natl.Acad.Sci.USA 89-4457,1992)。通常、抗体またはFab
フラグメントのフレームに対する修飾は、最も典型的にはヒト化が喪失されるの
で、臨床的使用において利点を与えない。しかしながら、前述したように、本発
明の主要な意義は、D−抗体のプロテアーゼ耐性特性および免疫原性の減少が、
フレームに対する修飾が一般に制限でないような程度の組込みのヒト化を意味す

本発明の抗体および1またはそれ以上の所望の分析物の精製のためのアフィニ
ティー選択を使用して、無数の分析物を検出することができる。主として、分析
