Claims (1)
- 【特許請求の範囲】 1.液体試料中の細胞の少なくとも1つの選別集団又は亜集団を濃縮するため の方法で、前記試料が血液又は骨髄の一部分である方法であって、以下の: 示差的比重沈降を提供するのに十分な密度を有する複数の粒子を提供し、前記 粒子が前記試料から除去される少なくとも2つの予備選別集団の細胞と特異的に 結合するそこに結合された反応体を有し、前記集団が赤血球系統の細胞及び骨髄 性系統の細胞であり; 濃縮される前記選別集団又は亜集団を実質的に物理的に損傷することなく除去 される前記予備選別集団に前記粒子を結合させるために少なくとも1mlの容量を 有する前記試料の一部分を前記粒子と混合し、前記試料部分の実質的部分中に前 記粒子を繰り返し沈降させることにより前記粒子と前記試料部分を混合し; 前記試料部分中で前記結合集団を用いて前記粒子を示差的比重沈降させ;そし て 濃縮される前記選別集団又は亜集団を含めた前記試料部分の結果的に生じた上 清の少なくとも一部分を除去される前記粒子及び前記結合集団から分離する 工程からなる方法。 2.前記混合が前記試料及び前記粒子を転倒方式で回転させることにより実行 される請求項1記載の方法。 3.前記粒子がニッケルで構成される請求項1記載の方法。 4.前記粒子が約3〜35μの直径を有する請求項1記載の方法。 5.前記粒子が約5〜10μの直径を有する請求項1記載の方法。 6.前記粒子が約2g/cm3又はそれ以上の密度を有する請求項1記載の方法。 7.前記粒子が約9g/cm3の密度を有する請求項6記載の方法。 8.前記粒子が前記集団の密度の約2〜3倍の密度を有する請求項1記載の方 法。 9.前記反応体が抗体、薬剤、ホルモン、成長因子、レクチン又は酵素のうち の少なくとも1つである請求項1記載の方法。 10.前記反応体が抗体である請求項1記載の方法。 11.前記抗体が前記赤血球系統細胞に特異的な一次抗体及び前記骨髄系統細 胞に特異的な二次抗体である請求項10記載の方法。 12.前記一次抗体がKC16抗体であり、前記二次抗体がCD15型抗体で ある請求項11記載の方法。 13.前記一次及び二次抗体が別々にそして順次前記試料部分と混合され、そ してそれから除去される請求項11記載の方法。 14.前記混合が約4〜30分間実行される請求項1記載の方法。 15.前記混合が約4分間実行される請求項14記載の方法。 16. 前記比重沈降が約4〜30分間実行される請求項1記載の方法。 17.前記比重沈降が約4分間実行される請求項16記載の方法。 18.前記試料部分の実質的部分中で前記粒子を繰り返し沈降させることによ り前記粒子と前記試料部分を混合させることを含む請求項1記載の方法。 19.前記試料が未処理血液又は骨髄試料であることを含む請求項1記載の方 法。 20.前記分離工程が前記粒子並びに前記結合集団又は亜集団を伴わずに前記 試料部分の結果的に生じた上清の少なくとも一部分を除去することを含む請求項 1記載の方法。 21.前記上清を前記予備選別集団と結合する反応体を有する複 数の二次粒子と混合することにより前記上清の少なくとも一部分からの前記予備 選別集団の少なくとも1つの除去を反復し、前記粒子を上清中で沈降させ、そし てその結果生じる二次上清を前記上清部分から分離することを含めた請求項20 記載の方法。 22.濃縮される前記選別集団又は亜集団を結果的に生じた前記二次上清から ポジティブ選別することを含めた請求項21記載の方法。 23.濃縮される前記選別集団又は亜集団を結果的に生じた前記上清からポジ ティブ選別することを含めた請求項20記載の方法。 24.前記2つの一次予備選別集団とは異なる少なくとももう一つの予備選別 集団を結果的に生じた前記上清から除去することを含めた請求項20記載の方法 。
