Description
Descrizione dell'invenzione industriale dal titolo: Description of the industrial invention entitled:
"Composizione farmaceutica costituita da un sale di paroxetina ed una sostanza poliidrossilata” "Pharmaceutical composition consisting of a paroxetine salt and a polyhydroxylated substance"
TECNICA ANTERIORE FRONT TECHNIQUE
Paroxetine is a pharmacologically active substance characterized by inhibitory activity of the neuronal reuptake of serotonin (5-HT). Thanks to this activity, paroxetine is used as a therapeutic agent for various pathological forms and in particular for depression and Parkinson's disease.
Paroxetine is commonly used as paroxetine hydrochloride in crystalline hemihydrate form with a melting point of 143.5 ° C. This product is described in US patent 4721723.
Crystalline forms of anhydrous paroxetine hydrochloride are instead described in GB patent 2,297,550.
It is known that crystalline products are more slowly absorbed into the body than the corresponding amorphous products due to the different dissolution rate.
Paroxetine HCI with amorphous characteristics can be produced by lyophilization from an aqueous solution of paroxetine HCI.
However, the product obtained by lyophilization of the solution of paroxetine HCI alone has a rubbery appearance which makes it difficult to use for the preparation of solid pharmaceutical forms; it is also extremely hygroscopic as after storage for 24 h at 25 ° C / 60% relative humidity it collapses leaving a rubbery residue. In storage conditions at a higher temperature in the presence of humidity it loses the characteristics of an amorphous product, recrystallising in the hemihydrate form.
Also known are compositions comprising amorphous paroxetine hydrochloride and hydroxylated compounds (WO 99/16440), however these compositions are obtained by a process that uses ethanol as a solvent and contain significant amounts of ethanol.
The presence of ethanol constitutes a disadvantage for the pharmaceutical composition and furthermore the use of ethanol constitutes a disadvantage in the preparation process of the product.

This analysis was performed to confirm that the paroxetine-HCI in the compositions of the invention has different characteristics with respect to the forms of paroxetine described in the patent GB 2,297,550.
Gli spettri ottenuti mediante questa analisi sono riportati nelle figure da 5 a 9 in cui: Patent GB 2.297550 also describes a crystalline form of anhydrous paroxetine HCI with a melting temperature of 164 ° C, a temperature close to the thermal event (165 ° C) of the composition of the invention. The spectra obtained by this analysis are shown in figures 5 to 9 in which:
Fig. 5: Spettro FTIR della composizione DELL’ESEMPIO 1 (KBr); Fig. 5: FTIR spectrum of the composition OF EXAMPLE 1 (KBr);
Fig. 6: Spettro FTIR del mannitolo liofilizzato (KBr); Fig. 6: FTIR spectrum of lyophilized mannitol (KBr);
Fig. 7: Spettro FTIR come differenza della composizione dell’esempio 1 rispetto al mannitolo liofilizzato (rispettivamente figure 5 e 6) (KBr); Fig. 7: FTIR spectrum as the difference in the composition of example 1 with respect to lyophilized mannitol (respectively figures 5 and 6) (KBr);

Fig. 9: FTIR spectra, difference in the composition of example 1 and lyophilized mannitol (Nujol).
By eliminating by difference the bands relating to mannitol, the peaks of paroxetine in the composition of Example 1 were obtained.
The table below shows the frequency values of the most significant peaks in the interval between 800 and 1300 cm'1 in comparison with the same values reported in the GB patent 2.297.550 for the crystalline forms A, B, C, D of paroxetine Anhydrous HCI, for which the melting point in ° C is reported in the last line.
FTIR Paroxetina HCI: picchi principali FTIR Paroxetine HCI: main peaks
Based on these data, it is evident that the IR spectrum of paroxetine contained in the product of example 1 does not correspond to any of the products of said prior art. In particular, there is no correspondence with form C, which is the only form hypothetically present on the basis of the DSC data shown in the previous example.
11). About 50 mg of product made as described in example 1 are compressed in a press for the preparation of samples used for infrared analysis at a pressure of 10T for 5 minutes. The thermograms of the product are shown immediately after compression (fig. 10) and after 3 days of storage at room temperature (fig. 11).
The thermograms indicate that after compression the product substantially retains its characteristics with the formation of a negligible quantity of crystalline anhydrous paroxetine HCI (Pf.118).
